Aristolochic acid (AA), extracted from Aristolochiaceae plants, plays an essential role in traditional herbal medicines and is used for different diseases. However, AA has been found to be nephrotoxic and is known to cause aristolochic acid nephropathy (AAN).AA-induced acute kidney injury (AKI) is a syndrome in AAN with a high morbidity that manifests mitochondrial damage as a key part of its pathological progression. Melatonin primarily serves as a mitochondria-targeted antioxidant. However, its mitochondrial protective role in AA-induced AKI is barely reported. In this study, mice were administrated 2.5 mg/kg AA to induce AKI. Melatonin reduced the increase in Upro and Scr and attenuated the necrosis and atrophy of renal proximal tubules in mice exposed to AA. Melatonin suppressed ROS generation, MDA levels and iNOS expression and increased SOD activities in vivo and in vitro. Intriguingly, the in vivo study revealed that melatonin decreased mitochondrial fragmentation in renal proximal tubular cells and increased ATP levels in kidney tissues in response to AA. In vitro, melatonin restored the mitochondrial membrane potential (MMP) in NRK-52E and HK-2 cells and led to an elevation in ATP levels. Confocal immunofluorescence data showed that puncta containing Mito-tracker and GFP-LC3A/B were reduced, thereby impeding the mitophagy of tubular epithelial cells. Furthermore, melatonin decreased LC3A/B-II expression and increased p62 expression. The apoptosis of tubular epithelial cells induced by AA was decreased. Therefore, our findings revealed that melatonin could prevent AA-induced AKI by attenuating mitochondrial damage, which may provide a potential therapeutic method for renal AA toxicity.
3.Identification model of tooth number abnormalities on pediatric panoramic radiographs based on deep learning

Objective:To identify tooth number abnormalities on pediatric panoramic radiographs based on deep learning.Methods:Eight hundred panoramic radiographs of children aged 4 to 11 years meeting the inclusion and exclusion criteria were selected and randomly assigned by writing programs in Python (version 3.9) to the training set (480 images), verification set (160 images) and internal test set (160 images), taken in Department of Pediatric Dentistry, Peking University School and Hospital of Stomatology between November 2012 to August 2020. And all panoramic radiographs of children aged 4 to 11 years taken in the First Outpatient Department of Peking University School and Hospital of Stomatology from June 2022 to December 2022 were collected as the external test set (907 images). All of the 1 707 images were obtained by operators to determine the outline and to label the tooth position of each deciduous tooth, permanent tooth, permanent tooth germ and additional tooth. The deep learning model with ResNet-50 as the backbone network was trained on the training set, validated on the verification set, tested on the internal test set and external test set. The images of test sets were divided into two categories according to whether there was abnormality of tooth number, to calculate sensitivity, specificity, positive predictive value and negative predictive value, and then divided into four types of extra teeth and missing permanent teeth both existed, extra teeth existed only, missing permanent teeth existed only, and normal teeth number, to calculate Kappa values. Results:The sensitivity, specificity, positive predictive value and negative predictive value were 98.0%, 98.3%, 99.0% and 96.7% in the internal test set, and 97.1%, 98.4%, 91.9% and 99.5% in the external test set respectively, according to whether there was abnormality of tooth number. While images were divided into four types, the Kappa value obtained in the internal test set was 0.886, and that in the external test set was 0.912. Conclusions:In this study, a deep learning-based model for identifying abnormal tooth number of children was developed, which could identify the position of additional teeth and output the position of missing permanent teeth on the basis of identifying normal deciduous and permanent teeth and permanent tooth germs on panoramic radiographs, so as to assist in diagnosing tooth number abnormalities.

Novel Coronavirus Pneumonia (NCP) is a class B infectious disease, which is prevented and controlled according to class A infectious diseases. Recently, children′s NCP cases have gradually increased, and children′s fever outpatient department has become the first strategic pass to stop the epidemic. Strengthening the management of the fever diagnosis process is very important for early detection of suspected children, early isolation, early treatment and prevention of cross-infection. This article proposes prevention and control strategies for fever diagnosis, optimizes processes, prevents cross-infection, health protection and disinfection of medical staff, based on the relevant diagnosis, treatment, prevention and control programs of the National Health and Health Commission and on the diagnosis and treatment experience of experts in various provinces and cities. The present guidance summarizes current strategies on pre-diagnosis; triage, diagnosis, treatment, and prevention of 2019-nCoV infection in common fever, suspected and confirmed children, which provide practical suggestions on strengthening the management processes of children′s fever in outpatient department during the novel coronavirus pneumonia epidemic period.
5.Feature exaction and classification of autism spectrum disorder children related electroencephalographic signals based on entropy.

The early diagnosis of children with autism spectrum disorders (ASD) is essential. Electroencephalography (EEG) is one of most commonly used neuroimaging techniques as the most accessible and informative method. In this study, approximate entropy (ApEn), sample entropy (SaEn), permutation entropy (PeEn) and wavelet entropy (WaEn) were extracted from EEGs of ASD child and a control group, and Student's -test was used to analyze between-group differences. Support vector machine (SVM) algorithm was utilized to build classification models for each entropy measure derived from different regions. Permutation test was applied in search for optimize subset of features, with which the SVM model achieved best performance. The results showed that the complexity of EEGs in children with autism was lower than that of the normal control group. Among all four entropies, WaEn got a better classification performance than others. Classification results vary in different regions, and the frontal lobe showed the best performance. After feature selection, six features were filtered out and the accuracy rate was increased to 84.55%, which can be convincing for assisting early diagnosis of autism.

6.Detection and analysis of EBV DNA integration in NK/T cell lymphoma genome

To investigate the presence of integrated Epstein-Barr virus (EBV) DNA in the NK/T cell lymphoma (NKTCL) ge-nome and analyze the integration information in the genome of NKTCL cell lines. Methods: PCR and in situ hybridization were used to detect EBV infection in five EBV (+) NK/T samples and four EBV (-) NK/T samples provided by the biobanks of the First Affiliated Hospi-tal of Zhengzhou University. Whole-genome DNA of the samples was sequenced and subjected to bioinformatics analysis. Whole-ge-nome sequence alignment was used to identify the EBV integration sequence. BLAST analysis was used to compare EBV fasta files of the samples and EBV fasta library. CREST software was used to extract softclip reads, filter all paired reads, and enumerate their distri-bution on chromosomes. The integrated genomics viewer (IGV) was used to compare the distribution of reads in partial regions of chromosome. PCR was used to amplify the high-frequency integration region of the EBV DNA. The amplified fragments were sanger se-quenced. Results: EBV DNA and EBER expression were detected in five EBV (+) NK/T samples but not in the four EBV (-) NK/T samples. Sequencing depth, coverage depth, proportion of coverage, and proportion of alignment all met the requirements for subsequent re-search. Sequence alignment revealed that the captured sequences were viral sequences. Filtered reads were most numerous in EBV (+) NKTCL cell line SNK, YTS, and EBV (+) nasal NKTCL tissue. The reads were non-randomly enriched in chromosome 2. EBV DNA inte-gration in the 400 bp region of chr2:30234084-30234483 caused insertion or deletion in the chr2p23.1 site. Conclusions: EBV DNA is highly integrated in the chr2p23.1 site of EBV (+) NKTCL cells and may affect the expression of related genes.
7.Expression and clinical significance of PD-1/PD-Ls in EBV-positive T/NK lymphoprolif-erative disorders

Objective: To investigate the expression and clinical significance of programmed death-ligand 1 (PD-L1), programmed death-ligand 2 (PD-L2), and their receptor programmed cell death protein 1 (PD-1) in EBV-positive T/NK lymphoproliferative disease [Epstein-Barr virus-positive T/natural killer (NK)-cell lymphoproliferative disease, EBV(+)-T/NK-LPD]. Methods: The pathological paraffin-embedded tissues of 17 patients with EBV(+)-T/NK-LPD from the First Affiliated Hospital of Zhengzhou University from January 2013 to December 2017 were collected. These patients include 12 males and 5 females, aged 10-82 years old, the average age being 29 years, 4 people in gradeⅠ, 7 in gradeⅡ, 3 in gradeⅢ, and 3 people with hydroa vacciniforme-like lymphoproliferative disorders. Immunohistochemical SP method was used to detect the expression of PD-1, PD-L1, and PD-L2 in human EBV(+)-T/NK-LPD tissues. The relationship between PD-1, PD-L1, PD-L2 expression, and clinicopathological parameters, pathological grades and prognosis were analyzed by Fisher's exact probabilities and Spearman rank correlation. Result: After statistical analysis, the results showed that in 17 cases of tissue samples, there were 12 cases with positive PD-1 expression, 6 cases with positive PD-L1 expression and 5 cases with positive PD-L2 expression. There was no significant correlation between PD-1 and PD-L2 expression and prognosis (P>0.05). PD-L1 expression showed a positive correlation with prognosis (P<0.05). There was no significant correlation between the expression of PD-L1 and PD-L2 with age, sex, as well as LDH and Ki-67 levels (P>0.05). Moreover, there was no significant correlation of PD-1 and PD-L2 expression with pathological grade (r=0.141, r=-0.149, both P>0.05). However, there was a negative correlation between the PD-L1 expression and pathological grade (r=-0.563), and the correlation between the PD-L1 ex-pression and pathological grade was statistically significant (P<0.05). Conclusions: PD-1, PD-L1, and PD-L2 are abnormally expressed in the pathological tissues of EBV(+)-T/NK-LPD. Although there was no significant correlation between the expression of PD-1 and prognosis or pathological grade, it was significantly higher in EBV+T/NK-LPD. PD-1/PD-Ls associated signaling pathway is expected to be a potential new target for EBV(+)-T/NK-LPD immunotherapy.
8.Study on the application of extended care on coronary intervention therapy in patients with acute myocardial infarction

Objective To investigate the clinical effectiveness of extended care on coronary intervention therapy in patients with acute myocardial infarction. Methods 90 patients with acute myocardial infarction who had been through coronary intervention therapy successfully in our hospital from June 2014 to April 2015 were divided into study group and control group with 45 patients in each according random number table. Patients in control group were treated with routine nursing care intervention while patients in study group were treated with additional extended care. Clinical effectiveness of nursing care in two groups were observed. Results After the intervention of extended care, there were 2 cases with nonfatal myocardial infarction (4. 4%), 3 patients undergoing revascularization for a second time (6. 7%) and one death (2. 2%) in study group, which were significantly fewer than those in control group, the difference was statistically significant(P<0. 05). 30 cases were satisfied with extended care and 11 cases were somewhat satisfied in study group after intervention. The satisfactory rate in study group was 91. 1%, which was significantly higher than those in control group (66. 7%), the difference was statistically significant (P<0. 05). According to the results of follow-up visit, nursing compliance of diet, exercise, medication and review in patients of study group was better than that in control group, the differences were statistically significant(P<0. 05). Conclusion Extended care on coronary intervention therapy in patients with acute myocardial infarction could reduce the incidence of adverse reaction, improve the satisfactory rate of nursing care and improve patients ' quality of life. It was worth promotion.
9.Influence of standardized secondary prevention on prognostic outcome of patients with coronary heart disease

Objective:To explore influence of standardized secondary prevention on prognostic outcome of patients with coronary heart disease (CHD).Methods:A total of 128 CHD patients were randomly and equally divided into secondary prevention group and routine treatment group.Routine treatment group received routine CHD therapy during hospitalization,received routine discharge guidance,and they were followed up once/six months.Secondary prevention group were served by specific physicians and received secondary prevention intervention,and they were followed up once/month after discharge.CHD patients received questionnaire to know their knowledge about CHD risk factors and compliance taking secondary prevention drugs after discharge.Risk factor control and recurrence rate of CHD etc.were assessed.Results:Compared with routine treatment group,after one-year follow up,there were significant rise in awareness rate of risk factors (34.38% vs.78.18%);control of risk factors (blood pres-sure,blood lipids,smoking,diet and exercise etc.)and administration rate of secondary prevention drugs after dis-charge (18.75% vs.87.50%),and significant reductions in clinic events [heart failures (18.75% vs.4.69%),an-gina pectoris (28.13% vs.6.25%),rehospitalization (21.89% vs.3.13%)and myocardial infarction (15.63% vs. 4.69%)]in secondary prevention group,P <0.05 or <0.01. Conclusion:Standardized secondary prevention is ef-fective in patients with coronary heart disease,which is worth clinic extending.
10.Masticatory performance and assessment of life quality of children with ectodermal dysplasia after prosthetic rehabilitation

Objective To evaluate masticatory performance and life quality of children with ectodermal dysplasia(ED) after prosthetic rehabilitation.Methods Six children with ED received denture restoration and 18 healthy children were involved in this study.The surface electromyography(EMG) of masseter(MM) and anterior temporalis(TA) during clenching and chewing movement were recorded.The EMG amplitude,area,asymmetry index of total and activity index of MM/TA were compared at each stage.The masticatory efficiency was measured with spectrophotometer.The life quality was assessed using visual analogue scale questionnaire.Results The EMG amplitude of MM and TA during chewing in ED Group were 41.7% and 45.6% of the control group respectively,the area were 35.9% and 36.0% respectively.Significant difference in asymmetry index of total during clenching was observed between the two groups (P<0.05) but not during chewing(P>0.05).The differences of activity index of MM/TA during clenching and chewing between the two groups were not detected(P>0.05).The masticatory efficiency of ED group was 67.2% of the control group.The score of chewing function in children with ED after prosthetic rehabilitation was three times higher than before,and no difference was present between the two groups (P>0.05).Conclusions Early prosthetic rehabilitation can significantly improve the masticatory performance and life quality of children with ED.
