1.Establishment of a humanized tumor-immune microenvironment mouse model

Objective:This study aims to construct a human peripheral blood mononuclear cell(PBMC)-NPSG-PDX(patient-derived xenograft)model,by transferring PBMCs into NPSG(NOD-PrkdcscidIl2rgnull/Shjh)mice and transplanting human tumor tissues.This model mimics the human tumor microenvironment to investigate the interactions between tumors and the immune system.Methods:PBMCs from healthy donors were transferred to NPSG mice to generate hPBMC-NPSG model.Patient-derived xenografts(PDX)were established in nude(BALB/c-nu)mice.Third-generation PDX tumors were then transplanted into hPBMC-NPSG mice to establish the hPBMC-NPSG-PDX model.Mouse body weight was monitored,and flow cytometry was used to analyze immune reconstitution and T cell function.Tumor growth was evaluated,and immunohistochemistry was performed to analyze tumor morphology and immune cell infiltration.Results:Fourteen days after constructing the hPBMC-NPSG mouse model,the proportion of human hu-CD45+CD3+T cells reached 97%.The proportions of human CD8+T,CD4+T,CD56+natural killer(NK),and CD19+B cells were 64%,24%,4.6%,and 1.0%,respectively.Human CD4+and CD8+cells secreted various cytokines(IL-2,IFN-γ,and TNF-α)and expressed cytotoxic molecules(FasL,granzyme B,and perforin)28 days post reconstitution.Tumor growth in hPBMC-NPSG-PDX mice was rapid initially but then stabilized.Immunohistochemistry staining revealed typical tumor morphology,and tumor cell apoptosis was observed in areas with immune cell infiltration.Conclusion:This study successfully constructed a hPBMC-NPSG-PDX model that effectively simulates the human tumor microenvironment,providing an ideal platform for tumor immunology research.

3.Preliminary investigation on diagnostic value of polyethylene glycol precipitation and gel filtration chromatography in insulin autoimmune syndrome

A 65-year-old male presented with recurrent palpitation and fatigue over one year. Lab tests revealed him with hyperinsulinaemic hypoglycemia. Insulin autoimmune antibody was repeatedly negative. Imaging of the pancreas seemed to be normal. Insulin-insulin autoimmune antibody complexes were detected by polyethylene glycol precipitation and gel filtration chromatography, thus the diagnosis of insulin autoimmune syndrome was suggested. By adjusting diet and administration of acarbose, all the symptoms were evidently relieved.
4.Clinicopathologic features and differential diagnoses of non-involuting congenital hemangioma in children.

OBJECTIVETo investigate the clinicopathologic features and differential diagnoses of non-involuting congenital hemangioma (NICH) in children.
METHODSThe clinical, morphologic and immunophenotypic characteristics of 22 cases of NICH were retrospectively analyzed.
RESULTSThe mean patients' age at diagnosis was 4.2 years, with a male to female ratio of 1.75:1. The tumors were located in the head and face (5 cases), neck (3 cases), body (6 cases), upper limbs (5 cases), and lower limbs (3 cases). Histologically, the tumor was dominated by rather large lobules of small vessels that were mostly rounded, curved, small and thin-walled, and were lined by endothelial cells surrounded by one or more layers of pericytes. The center of the lobules was occupied by one or more thin or thick walled vessels, which were surrounded by fibrous and fatty tissue, which contained abnormal arterial and venous structures. At the edge of the lobules there were lymphatic vessels. Immunohsitochemical study showed that tumor cells in NICH were positive for CD34 (22/22), CD31 (22/22), SMA (22/22), vimentin (22/22) and Glut1 (0/22). D2-40 expression was located at the edge of the capillary lobules.
CONCLUSIONSNICH is a benign lesion. Clinically and pathologically, it needs to be differentiated from rapidly involuting congenital hemangioma, infantile hemangiomas, tufted angioma, vascular malformation, and others.

6.Multiple metallic foreign bodies accidentally detected in different body cavities: a case report.

We accidentally found an unusual case of a middle aged Tibetan woman who had eight metallic foreign bodies (eight needles) in her head, chest and abdomen. These needles were not related to any surgical intervention or trauma. The diagnosis "metallic foreign bodies" cannot usually be made in an acute setting. Some patients may present chronic symptoms, such as infection or pain. However, in some cases, the patients do not have any symptoms.

7.In vitro evaluation of antibacterial activity and cytocompatibility of antibacterial stainless steel containing copper.

To study the in vitro antibacterial activity and biocompatibility of 317L stainless steel containing 4.5% copper alloy (317L-Cu), we produced 317L-Cu stainless steel with epsilon-Cu phase. The cell proliferation of osteoblasts on material surface was detected in vitro. Escherichia coli was cultured with 317L-Cu for evaluating the antibacterial activity. We found that the 317L-Cu could effectively kill the Escherichia coli on material surface. The cell proliferation of osteoblasts on material surface was not different significantly, compared with titanium material. Our study clearly demonstrated that the 317L-Cu alloys had a significant antimicrobial activity and was biocompatible in vitro, so it would be suitable to be used as a new medical material with antibacterial activity.

8.Report on skin-sparing mastectomy with immediate breast reconstruction in 15 cases

BACKGROUND: Recently the prevalence rate of mass in mammary gland increases significantly than before. To use which kind of technology is not only related with the post-operation recurrence, but also related with the reconstruction of their confidence. Therefore, how to reduce the trauma physically and psychologically in patients to the maximal degree has become the chief factor to choose technology by surgeons.OBJECTIVE: To observe the effects of transverse rectus abdominis musculocutaneous (TRAM) flap, latissimus dorsi musculocutaneous flap with breast prothesis, local flap and simple breast prothesis after skin-sparing mastectomy with immediate breast reconstruction.DESIGN: Case analysis.SETTING: Department of Plastic Surgery, Shenzhen People's Hospital,Second Clinical Medical College of Jinan University.PARTICIPANTS: A total of 15 ductal carcinoma in situ (DCIS) and huge breast benign tumor patients were enrolled into Shenzhen People's Hospital between October 1997 and May 2004, aged from 20 to 46 years. They all got disease in single breast, and 8 of them had preoperative diagnosis of DCIS, T1N0M0 and 7 of them had huge breast benign tumor. Operation program was informed to the patients before operation so as to gain their agreement.METHODS: Skin-sparing mastectomy was performed with circular shape incision at areola and using TRAM flap, latissimus dorsi musculocutaneous flap with breast prothesis, local flap and simple breast prothesis to fill breast. Slight skin of musculo-cutaneous flap could compensate the nipple and areola when performing reconstruction. At the reoperation, nipple was made at the transferred flap.MAIN OUTCOME MEASURES: To observe the patients with or without recurrence or transfer after operation and local healing of wound.RESULTS: With the cooperation of general surgeons the operation was done in 15 cases and 14 cases of them were followed up for more than 1 year. The reconstructed breasts were with good symmetry with the opposite one, normal skin sensation, natural shape and inconspicuous scar.CONCLUSION: This is an excellent alternative method for mastectomy and breast reconstruction based on the avoidance of recurrence of breast cancer in severe operation.
