1.Effect of miR-147a Targeting INHBA-TGF-β1/Smads Signaling Pathway on Proliferation,Migration and Invasion of Cervical Cancer HeLa Cells

Objective To investigate the role of miR-147a targeting inhibin subunit beta A(INHBA)in transforming growth factor-β1(TGF-β1)/Smads signaling pathway in the invasion and metastasis of cervical cancer HeLa cells.Methods The miR-147a mimics,miR-147a inhibitors and their negative controls(mimicsNC and inhibitor NC)were transfected into HeLa cells and divid-ed into five groups:control group(blank control),mimics NC group,miR-147a mimics group,inhibitor NC group,and miR-147a in-hibitor group.The proliferation activity of HeLa cells after miR-147a intervention was detected by the MTT method,the invasion and me-tastasis abilities of HeLa cells after the intervention of miR-147a were detected by Transwell and cell scratch assay,the mRNA expres-sion changes of miR-147a and INHBA in different groups were detected by real-time fluorescence quantitative polymerase chain reaction(RT-qPCR),the relationship between miR-147a and the target gene INHBA was detected by dual-luciferase reporter gene assay,the protein expressions of TGF-β1,TGFBR1,Smad3,Smad2 and INHBA in different groups were detected by Western blot,and the ex-pression changes of TGF-β1 were detected by enzyme-linked immunosorbent assay(ELISA).Results Compared with blank control and negative control,miR-147a mimics inhibited cell proliferation,decreased cell invasion and migration ability,and decreased cell healing ability.miR-147a inhibitor promoted cell proliferation,increased invasion and migration ability,and improved cell healing abili-ty.In the miR-147a mimics group,miR-147a was significantly increased,and the target gene INHBA was significantly decreased(P<0.05).In the miR-147a inhibitor group,miR-147a was significantly decreased,and the target gene INHBA was significantly in-creased(P<0.05).The fluorescence intensity ratio of miR-147a mimics+WT group was significantly lower than that of mimics NC+WT group(P<0.05).The expression levels of TGFBR1,TGF-β1,Smad3,Smad2 and INHBA in miR-147a mimics group were sig-nificantly decreased(P<0.05).The expression levels of TGFBR1,TGF-β1,Smad3,Smad2,and INHBA in miR-147a inhibitor group were significantly increased(P<0.05).Conclusion miR-147a targets the TGF-β1/Smads signaling pathway mediated by IN-HBA,which plays an important role in inhibiting the proliferation,invasion and migration of cervical cancer HeLa cells,and provides a potential molecular target and theoretical basis for the treatment of cervical cancer.
2.Effect of different intensity neuromuscular training on muscle strength and knee joint function of patients after anterior cruciate ligament reconstruction

Objective To compare the effect of different intensity of neuromuscular training(NMT)on muscle strength and knee joint function of patients after anterior cruciate ligament reconstruction(ACLR).Methods From January,2023 to January,2024,60 ACLR patients in Changhai Hospital were selected,and they received the same intensity of NMT from one to eight weeks after surgery.Eight weeks after surgery,they were randomly divided into low intensity group(n=30)and high intensity group(n=30),and then they received different inten-sities of NMT from nine to 16 weeks after surgery,each training session lasted one hour,with three sessions per week,totaly 48 sessions.The Lysholm score,knee flexor and extensor muscle strength and muscle endurance-were compared at eight weeks and 16 weeks after surgery.Results After group training,the Lysholm score significantly increased in both groups(|t|>13.739,P<0.001),and was higher in the high intensity group than in the low intensity group(t=-2.574,P<0.05);in the high intensity group,the relative peak torque and endurance of the extensor and flexor muscles improved at angular velocities of 60°/s,120°/s and 180 °/s(|t|>2.320,P<0.05);in the low intensity group,the flexor peak torque improved at all the three angular velocities(t>2.177,P<0.05),the extensor peak torque improved at angular velocities of 60°/s and 180°/s(|t|>1.715,P<0.05),and the extensor endurance improved at angular velocity of 60°/s(t=-2.293,P<0.05).However,there was no significant difference in the relative peak torque and endurance of the extensor and flexor muscles at all the three angular velocities(P>0.05).Conclusion Both high and low intensity NMT could improve the muscle strength,muscle endurance and knee joint func-tion.Maybe,high intensity is superior to low intensity.Further verification is still needed.

5.Microwave sensor for recognition of abnormal nodule tissue on body surface.

For the detection and identification of abnormal nodular tissues on the body surface, a microwave sensor structure loaded with a spiral resonator is proposed in this paper, a sensor simulation model is established using HFSS software, the structural parameters are optimized, and the actual sensor is fabricated. The S21 parameters of the tissue were obtained when nodules appeared by simulation, and the characteristic relationship between the difference of S21 parameters with position was analyzed and tested experimentally. The results showed that when nodules were present in normal tissues, the curve of S21 parameter difference with position change had obvious inverted bimodal characteristics, and the extreme value of S21 parameter difference appeared when the sensor was directly above the nodules, which was easy to identify the position of nodules. It provides an objective detection tool for the identification of abnormal nodular tissues on the body surface.

6.Development of a Portable Chronic Non-specific Low Back Pain Measurement System.

We developed a portable non-specific low back pain measurement system EasiLBP and evaluated its performance in collecting EMG signals:during the wearer's movement without the assistance of a doctor, the collection of EMG signals by portable devices met problems such as large noise interference, difficulty in accurately calibrating the start and end points of the action interval, and imbalanced samples for feature recognition, et al. To challenge these problems, we proposed a small group-based noise removal method, a dynamic dual-threshold automatic method for identifying the start and end points of the motion interval, and a sampling method to balance group samples, respectively. Portable device and a medical EMG acquisition equipment Thought Technology FlexComp Infiniti 10 were used to perform EMG measurements on 15 patients with non-specific low back pain and 15 normal people. Clinical experiments and statistical analysis show that the portable EMG acquisition system has significant differences in EMG signal characteristics between normal people and non-specific low back pain patients, and it has good measurement consistency and accuracy with the medical EMG acquisition equipment.

7.Quantitative study of intraoral scanners′ accuracy

Objective:To establish a standard method to evaluate the scanning accuracy of intraoral scanner (IOS) and to investigate six IOS′s scanning accuracy and the relationship between different scan span.Methods:Five simplified six abutments full arch model were fabricated by high accuracy (5 μm) milling machine with 7075 aluminum alloy. The machining accuracy, which was verified by a coordinate measuring machine with higher accuracy (0.7 μm), was considered as the reference accuracy. The model with the highest machining accuracy was considered as the test model in IOS′s scanning accuracy test, and computer-aided design (CAD) data of the model was used as the reference data. Six IOS scanned the test model 10 times with the same scanning path, obtained 60 test data. CAD data and test data were input into Geomagic Studio 2014. The preparation part above the margin of the abutments of the data was isolated and divided into 4 segments of interest: single crown, three-unit bridge, five-unit bridge, and full arch. The test data were then best-fit aligned to CAD data or each other followed by deviation analysis. Scanning trueness and precision were then calculated.Results:The mid-value of scanning trueness and precision of six IOS in single crown, three-unit bridge, five-unit bridge and full arch were 13.3-29.6 μm and 7.6-20.7 μm, 15.4-30.9 μm and 8.7-26.5 μm, 17.0-66.1 μm and 11.3-44.2 μm, 24.0-107.9 μm and 24.6-150.1 μm respectively.Conclusions:Long-span scanning can affect the accuracy of IOS to a varying extent.
8.Risk factors of perioperative myocardial infarction in the patients undergoing noncardiac surgery

Objective To investigate the risk factors and prognosis of perioperative myocardial infarction in the patients undergoing noncardiac surgery. Methods Clinical data of 562 patients who had accepted non-cardiac surgery was collected and retrospectively analyzed. The risk factors, treatments and outcomes of all these patients were recorded and analyzed. Results A total of 19 out of the 562 patients had perioperative myocardial infarction ( PMI) . The incidence was 3. 4% . The mean occurrence time was (43. 5 ± 12. 7)h after operation. Eleven PMI patients (11 ∕ 19) were non-ST-segment elevation myocardial infarction and eight patients (8 ∕ 19) were ST-segment elevation myocardial infarction. Thirteen PMI patients were left coronary artery occlusion and six patients were right coronary artery occlusion. Advanced age, history of myocardial infarction, unstable angina, change of ST-T segment on electrocardiography (ECG), multivessel diseases, diabetes,hypertension,and high risk non-cardiac surgery were the risk factors of PMI and positively correlated to PMI. Sixteen PMI (16 ∕ 19) patients accepted PCI treatment and three patients (3 ∕ 19) accepted drug conservative treatment. Two patients had unstable angina attack after treatment and one patient had arrhythmia. The heart function in two patients decreased by one or more than one class within the follow up of 1 year. No patient had recurrent acute myocardial infarction or deceased during follow-up. Conclusions Many factors could lead to PMI. Making preoperative assessment, recognizing patients of high risks and dealing with patients who had PMI in time was necessary.
9.Bridging antiplatelet strategies during temporary withdrawal of antiplatelet therapy for non-cardiac surgery after drug-eluting stent implantation

Objective To compare the rates of major adverse cardiovascular events(MACE)and bleeding events of three different antiplatelet strategies during temporary withdrawal of antiplatelet therapy for non-cardiac surgery within 1 year after drug-eluting stent (DES)implantation.Methods Retrospectively analyzed 42 patients who had accepted non-cardiac surgery and required temporary withdrawal of antiplatelet therapy within 1 year after drug-eluting stent implantation. The patients were divided into three groups according to the bridging antiplatelet strategies they received.All patients discontinued clopidogrel 5 to 7 days before the non-cardiac surgery. The tirofiban group was treated with intravenous tirofiban 0.4ug/kg·min in the first 30 min followed 0.1μg/(kg·min). The dosage was reduced by half for patients whose Creatinine clearance were less than 30 ml/min.The low molecular weight heparin group was treated with subcutaneous enoxaparin (Clexane 4000 AxaIU, once per day) .The asprin group was given only oral asprin(100 mg, once per day) . Tirofiban and low molecular weight heparin were continued until clopidogrel was resured. Perioperative cardiovascular events and serious bleeding were recorded. Results The rates of major adverse cardiac events in the tirofiban and the low molecular weight heparin group were lower than the aspirin group. Acute myocardial infarction caused by confirmed in-stent thrombosis was diagnosed in one patient in the aspirin group. One case of asymptomatic ST-T changes was found in the low molecular weight the aspirin group. 3 cases in the aspirin group presented ST-T changes on ECG and among them 1 case was STEMI due to LAD thrombosis requiring primary and 2 other cases were agina pectoris.There were no significant differences in bleeding events among the three groups.Conclusions Potential for the perioperative management with tirofiban or low molecular weight heparin is safe and feasible for patients who had recently undergone DES implantation and required noncardiac surgery with the interruption of antiplatelet therapies.
