1.Effects of polygonatum sibiricum polysaccharides on the growth performance,in-testinal morphology,antioxidant capacity,and intestinal function of chicks

Polygonatum sibiricum polysaccharide(PSP)is a polysaccharide with multiple pharma-cological activities that has been widely studied and used in the human body.However,there is cur-rently a lack of research investigating the potential advantages of PSP in poultry farming.This study investigated the effects of adding PSP to drinking water on the growth performance,antioxi-dant status,serum biochemical indicators,ileal tissue morphology,immune organs,and intestinal function of chicks.88 Hailan brown laying hens were randomly divided into 4 groups,with 22 hens in each group,namely the blank control group(CON),and fed with basic feed;The low-dose PSP group(250 mg/L),the medium dose PSP group(500 mg/L),and the high-dose PSP group(1 000 mg/L)were fed with corresponding doses of PSP through drinking water on the basis of basic feed,and the experimental period was 21 d.The initial and final body weight and immune or-gan relative quality of chicks,serum biochemical indicators,the activities of SOD,CAT,and GSH-Px,as well as the contents of T-AOC and MDA in the serum of chicks were measured;HE stai-ning method was used to observe the pathological changes of intestinal tissue slices in the ileum;Real time fluorescence quantitative PCR technology was used to detect the mRNA expression lev-els of cytokines ZO-1,Claudin-1,Occludin,Mucin-2,IL-1β,TNF-a,IFN-γ,IL-4,IL-8,and IL-10 in the ileum.The results showed that compared with the blank control group,the addition of medium dose PSP significantly increased the final relative quality(P<0.01),the final body weight and ADG of PSP500 group chicks significantly increased(P<0.01),and the F/G of PSP250 and PSP500 groups significantly decreased(P<0.05 or P<0.01).The villus height of the jejunum in the 200,500,and 1 000 mg/L PSP groups of chicks significantly increased(P<0.05).The SOD ac-tivity significantly increased(P<0.01),and the CAT activity in the PSP1000 group significantly increased(P<0.01).The PSP500 and PSP1000 groups significantly reduced the mRNA expression of cytokines IL-1β,IL-4,and IFN-γ in the ileum(P>0.05);PSP did not show significant changes in serum total protein(TP),albumin(ALB),glucose(GLU),cholesterol(T-CHO)content,and immune organ index(P<0.05).In summary,PSP can improve growth performance,enhance an-tioxidant capacity,improve ileal morphology and epithelial barrier function,and regulate mucosal immune status.Considering the overall economic benefits,the recommended level of PSP addition is 500 mg/L.

OBJECTIVES:
Peroxisome proliferator-activated receptor gamma coactivator 1α (PGC1α) controls mitochondrial biogenesis, but its role in cardiovascular diseases is unclear. The purpose of this study is to explore the effect of PGC1α on myocardial ischemia-reperfusion injury and the underlying mechanisms.
METHODS:
The transverse coronary artery of SD rat was ligated for 30 minutes followed by 2 hours of reperfusion. Triphenyltetrazolium chloride (TTC) staining was performed to measure the area of myocardial infarction. Immunohistochemistry and Western blotting were used to detect the PGC1α expression in myocardium. The rat cardiomyocyte H9C2 was subjected to hypoxia/reoxygenation (H/R) with the knockdown of PGC1α or hypoxia- inducible factor 1α (HIF-1α), or with treatment of metformin. Western blotting was used to detect the expression of PGC1α, HIF-1α, p21, BAX, and caspase-3. CCK-8 was performed to detect cell viability, and flow cytometry was used to detect apoptosis and mitochondrial superoxide (mitoSOX) release. RT-qPCR was used to detect the mRNA expression of PGC1α and HIF-1α. Besides, chromatin immunoprecipitation (ChIP)-qPCR and luciferase reporter gene assay were applied to detect the transcriptional regulation effect of HIF-1α on PGC1α.
RESULTS:

CONCLUSIONS
After I/R, HIF-1α up-regulates the expression of PGC1α, leading to an increase in ROS production and aggravation of injury. Metformin can inhibit the accumulation of HIF-1α during hypoxia and effectively protect myocardium from ischemia/reperfusion injury.

4.Afferent Arteriovenous Anastomosis For Finger Tip Replantation.

PURPOSE: The vessels of the finger tip are very small and fragile, have many branches, so the anastomosis of this vessel is very difficult. Guillotine type amputation of digits are rare, on the other hand, crush/avulsion injuries which require complex surgical solutions are more frequent. We often found that the arteries of distal amputee on crush/avulsion injuries are lost. In these cases, replantation is much more difficult even impossible. But when replantation is successes, the replantated finger tip provides adequate soft tissue, restoration of length, sensory and nail. MATERIALS AND METHODS: We report here our experience of finger tip replantation using afferent a-v anastomosis technique accumulated from March 2006 to March 2009. RESULT: The success rate was 96.4%. Complications occurred in eight patients. (5 digit atrophy, 1 osteomyelitis, 1 severe digit pain and 1 cold intolerance) CONCLUSIONS: We believe that the afferent a-v anastomosis replantation provides us wider range of indication of finger tip replantation on crush/avulsion injuries.

5.Voice Evaluation after Laser Resection of Early Glottic Cancer.

BACKGROUND AND OBJECTIVES: Voice quality may be an important issue for choosing treatment options of early glottic cancer. It has been reported that voice quality is worse after laser cordectomy compared to that of radiation therapy. But, with developed visualization tools, the precise localization of lesions became possible enabling surgeons to decide the appropriate surgical extent to preserve voice after laser cordectomy. The authors assessed the postoperative voice quality of patients who underwent laser cordectomy according to the classification by the European Laryngological Society. SUBJECTS AND METHOD: Thirty patients who were diagnosed with glottic cancer and treated with laser cordectomy between April, 1999 and January, 2006, were evaluated. All the patients were followed-up more than 6 months. Objective evaluations included maximal phonation time, fundamental frequency, jitter, shimmer, noise-to-harmonics ratio and subjective evaluation was performed by the GRBAS scale. RESULTS: The type II laser cordectomy group and some cases of the type III cordectomy showed no significant differences compared with the normal control group in both objective and subjective evaluations. CONCLUSION: This study shows that the group of laser cordectomy type II and some cases of type III had acceptable results compared to that of the normal control group. Thus, laser cordectomy could be considered as an alternative to radiation therapy in selected patients with early glottic cancer, even when preserving the quality of voice is an important issue.

6.The Seroepidemiologic Study for Human T-cell Lymphotropic Virus Type I(HTLV-I) Infection in Residents of Cheju-Do.

BACKGROUND: Human T-cell lymphotropic virus type I (HTLV-I) is a retrovirus that has been identified as a cause of adult T-cell leukemia/lymphoma and tropical spastic paraparesis. HTLV-I infection is highly endemic in the southwestern islands of Japan, Caribbean basin, South America, and Africa. In 1993, we showed that the seroprevalence of antibodies to HTLV-I was 0.13% among blood donors in Korea, but surprisingly, 0.80% in Cheju-Do adjacent to endemic areas of Japan. So this study was designed to reevaluate the seroprevalence of antibodies to HTLV-I among residents in Cheju-Do. METHODS: Total 2,372 residents in Cheju-Do were tested from December 1995 to March 1996. Anti-HTLV-I antibodies were detected by the microtiter particle agglutination test. RESULTS: Among total 2,372 residents, 19 were anti-HTLV-I positive. So the overall positive rate of anti-HTLV-I antibodies was 0.80%. The positive rate in females was higher than in males (0.82% vs 0.78%). The positive rate was 1.45% in the age group of 20-29 years, 1.41% in 40-49 years, 0.91% in 0-9 years, 0.70% in 30-39 years, and 0.54% in 50-59 years. The mean age of seropositive cases is 35.2 in males and 35.4 in females, with a mean of 35.3. Geographically, high positive rate was observed in Sogwipo-City (1.37%) and Namcheju-Gun (0.83%) compared to those of Pukcheju-Gun (0.64%) and Cheju-City (0.61%), which showed high seroprevalence in districts adjacent to endemic areas of Japan. Any specific risk factors or associated disorders of HTLV-I infection could not be found among the seropositive cases. CONCLUSION: The seroprevalence of antibodies to HTLV-I in Cheju-Do was noted to be very high by the microtiter particle agglutination test. So henceforth serosurvey by confirmative laboratory tests is needed, and if high seroprevalence is showed from it, screening of blood donors for HTLV-I in Cheju-Do should be considered to prevent transfusion-associated HTLV-I infection.
