ObjectiveIn the realm of forensic science, dust is a valuable type of trace evidence with immense potential for intricate investigations. With the development of DNA sequencing technologies, there is a heightened interest among researchers in unraveling the complex tapestry of microbial communities found within dust samples. Furthermore, striking disparities in the microbial community composition have been noted among dust samples from diverse geographical regions, heralding new possibilities for geographical inference based on microbial DNA analysis. The pivotal role of microbial community data from dust in geographical inference is significant, underscoring its critical importance within the field of forensic science. This study aims to delve deeply into the nuances of fungal community composition across the urban landscapes of Beijing, Fuzhou, Kunming, and Urumqi in China. It evaluates the accuracy of biogeographic inference facilitated by the internal transcribed spacer 2 (ITS2) fungal sequencing while concurrently laying a robust foundation for the operational integration of environmental DNA into geographical inference mechanisms. MethodsITS2 region of the fungal genomes was amplified using universal primers known as 5.8S-Fun/ITS4-Fun, and the resulting DNA fragments were sequenced on the Illumina MiSeq FGx platform. Non-metric multidimensional scaling analysis (NMDS) was employed to visually represent the differences between samples, while analysis of similarities (ANOSIM) and permutational multivariate analysis of variance (PERMANOVA) were utilized to statistically evaluate the dissimilarities in community composition across samples. Furthermore, using Linear Discriminant Analysis Effect Size (LEfSe) analysis to identify and filter out species that exhibit significant differences between various cities. In addition, we leveraged SourceTracker to predict the geographic origins of the dust samples. ResultsAmong the four cities of Beijing, Fuzhou, Kunming and Urumqi, Beijing has the highest species richness. The results of species annotation showed that there were significant differences in the species composition and relative abundance of fungal communities in the four cities. NMDS analysis revealed distinct clustering patterns of samples based on their biogeographic origins in multidimensional space. Samples from the same city exhibited clear clustering, while samples from different cities showed separation along the first axis. The results from ANOSIM and PERMANOVA confirmed the significant differences in fungal community composition between the four cities, with the most pronounced distinctions observed between Fuzhou and Urumqi. Notably, the biogeographic origins of all known dust samples were successfully predicted. ConclusionSignificant differences are observed in the fungal species composition and relative abundance among the cities of Beijing, Fuzhou, Kunming, and Urumqi. Employing fungal ITS2 sequencing on dust samples from these urban areas enables accurate inference of biogeographical locations. The high feasibility of utilizing fungal community data in dust for biogeographical inferences holds particular promise in the field of forensic science.

Objective:To investigate the composition of immune cells and fibroblastic reticular cells (FRCs) in the lymph nodes (LNs) follicles of human immunodeficiency virus (HIV)-infected individuals with varying immune statuses, and their association with HIV replication. Methods:Neck LNs samples were collected from 4 treatment-naive, newly diagnosed HIV-infected individuals with diverse immune statuses. RNA in situ sequencing was employed, with imaging achieved via rolling circle amplification and fluorescence labeling. By integrating cell segmentation and nuclear staining, single-cell data from up to one hundred thousand cells were generated per paraffin tissue section. Using lymphoid follicles as the unit of analysis, compositional changes in immune cells and FRCs were characterized, and their correlations with viral replication were evaluated. Results:The peripheral blood CD4 + T cell counts of samples LN_1, LN_2, LN_3, and LN_4 exhibited a sequential decrease. A total of 31, 15, 16, and 18 structurally intact follicles were identified in each sample, respectively. In the follicles of LN_1, the proportion of HIV-replicating cells positively correlated with cDCs abundance ( R2=0.2, P=0.011), and HIV RNA signals were spatially colocalized with cDCs and FRCs. In the follicles of LN_2, HIV RNA molecules showed preferential enrichment within FRCs. In sample LN_3, HIV RNA enrichment was observed in both cDCs and CD4 + T cells. In sample LN_4, the proportion of cells with HIV replication was positively correlated with the proportions of the following cells: cDCs ( R2=0.38, P=0.006 4), CD4 + T cells ( R2=0.28, P=0.025), and FRCs ( R2=0.26, P=0.029), and HIV RNA molecules were detected in cDCs, CD4 + T cells, and FRCs. LN_1 and LN_2 samples showed a trend toward negative correlation between HIV-replicating cell proportion and CD8 + T cells proportion. LN_4 sample demonstrated a significant positive correlation between HIV-replicating cell proportion and CD8 + T cells proportion ( R2=0.23, P=0.046). Conclusions:RNA in situ sequencing technology reveals unique distribution patterns of immune cells and viral replication in LNs follicles of HIV-infected individuals. The follicular immune microenvironment exhibits distinct characteristics associated with peripheral blood CD4 + T cell counts, providing novel insights into the spatial dynamics of HIV persistence and immune cell interactions during infection.

Objective To investigate the effect and mechanism of cell migration-induced protein(KIAA1199)on the proliferation of osteosarcoma cells. Methods The KIAA1199 gene was knocked down or overexpressed in osteosarcoma cell lines(HOS and U2OS)with high KIAA1199 expression. The expression of KIAA1199 was confirmed by RT-PCR and Western blotting.CCK-8 and colony formation assays were used to determine the effect of KIAA1199 on cell proliferation.CCK-8 and cell viability assays were used to confirm that KIAA1199 promoted osteosarcoma cell proliferation by inhibiting ferroptosis. The BODIPY 581/591 probe was used to investigate the effect of KIAA1199 on cell lipid peroxidation. The effect of KIAA1199 on PTGS2 was investigated by RT-PCR,and the level of ferroptosis related proteins(GPX4,SLC7A11,DHODH,and FSP1)were determined by Western blotting. Results KIAA1199 knockdown inhibited osteosarcoma cell proliferation, which mainly worked by promoting ferroptosis in osteosarcoma cells.Ferrostatin-1(Fer-1,a ferroptosis inhibitor)could reverse the proliferation inhibition caused by KIAA1199 knockdown.Osteosarcoma cells with KIAA1199 knockdown were more sensitive to ferroptosis inducer Erastin, and their lipid peroxidation and PTGS2 mRNA increased significantly.KIAA1199 knockdown resulted in lower expression of GPX4 and SLC7A11 proteins and a lower GSH/GSSG ratio. On the other hand,KIAA1199 overexpression promoted osteosarcoma cell proliferation, made cells more tolerant to Erastin, significantly reduced lipid peroxidation and PTGS2 mRNA levels, and obviously increased the protein expression levels of GPX4 and SLC7A11.Conclusion KIAA1199 inhibits ferroptosis through SLC7A11/GSH/GPX4 pathway, thereby promoting osteosarcoma cell proliferation.

Background Accurate T-staging is critical for esophageal cancer therapy, but CT-based assessment has significant limitations.Large vision models(LVMs)hold promise, yet their zero-shot clinical diagnostic capability without fine-tuning remains unvalidated. Methods A retrospective analysis was conducted on the chest CT images from 98 esophageal cancer patients and 50 normal controls.Using radiologist-consensus as the gold standard, the zero-shot T-staging performance of 3 LVMs(GPT-5,Gemini,and MedGemma)was evaluated with prompts of varying complexity. Results GPT-5 exhibited the highest accuracy and stability.Significant biases were observed among models:Gemini tended to over-stage,while MedGemma showed a tendency to under-stage.All models faced challenges in identifying early-stage tumors, but structured prompts improved diagnostic performance for mid-to-late stage lesions. Conclusion LVMs have potential for zero-shot T-staging,but their performance highly depends on model choice and prompt design. The generic model GPT-5 show superior zero-shot generalization. However, current model performance is not yet clinically viable, especially for early diagnosis.Future work should focus on fine-tuning with high-quality clinical data and developing standardized prompt frameworks.
