1.Underlying target of bullatine A in treating rheumatoid arthritis based on LiP-SMap drug target proteomics

Aim To identify the underlying target of bullatine A(BA)against rheumatoid arthritis(RA)u-sing limited proteolysis-small molecule mapping(LiP-SMap)drug target proteomics and to provide a scientif-ic basis for clinical application of Aconiti brachypodi Radix in the treatment of RA.Methods LiP-SMap drug target proteomics was employed to perform bioin-formatics analysis for comparing and validating the dif-ferential protein expression after BA intervention.A collagen-induced arthritis(CIA)model was estab-lished in DBA/1 mice using bovine type Ⅱ collagen.The mice were then divided into the CIA model group,methotrexate-positive control group(MTX group),and BA groups(10 mg·kg-1 and 20 mg·kg-1)based on their clinical scores.After drug intervention,the thera-peutic efficacy against RA was assessed by joint index scores and foot thickness measurements.Histopatholog-ical changes in the arthritic joints of CIA mice were e-valuated using hematoxylin and eosin(HE)staining.Enzyme-linked immunosorbent assay(ELISA)was employed to detect inflammatory cytokines interleukin-17(IL-17)and total IgG and IgG3 anti-collagen-spe-cific antibodies levels from the serum of CIA mice.Flow cytometry was used to detect the expression levels of intracellular Th17 cells(IL-17+CD4+T cells)and Th1 cells(IFN-γ+CD4+T cells).Fluorescent quanti-tative PCR was performed to detect the expression of genes related to differential proteins.Results The proteomic analysis identified Serpinb1a as a protein with strong binding affinity to BA,and KEGG enrich-ment analysis indicated IL-17 signaling pathway was a crucial pathway of BA in against RA.BA treatment significantly reduced clinical scores and foot thickness,improved local arthritis symptoms in CIA mice,and al-leviated inflammatory cell infiltration into arthritic joints(P<0.05).Differential protein validation re-sults showed that BA had strong affinity with Serpinb1a(-5.92 kJ·mol-1)and downregulated the expres-sion of Serpinb1a mRNA.Furthermore,the administra-tion of BA markedly reduced serum IL-17 A levels from CIA mice,inhibited the expression of intracellular IL-17 A and IFN-γ cytokines in splenic CD4+T cells(P<0.05),and significantly downregulated the transcrip-tional expression of IL-17F(P<0.05).Conclusion BA exhibits therapeutic effects on collagen-induced arthritis,and its mechanism of action may involve the regulation of Serpinb1a and the IL-17 signaling path-way.
2.Recent Advances in Electrochemical Sensors for Detection of Disinfection By-Products in Drinking Water

Disinfection by-product(DBPs)are contaminants generated during drinking water treatment processes.Despite their low concentration level,these compounds exhibit high toxicity,posing threaten to both environmental safety and human health.Traditional DBPs analysis methods rely on chromatography/mass spectrometry techniques,which are limited by complex and time-consuming pretreatment processes,as well as expensive and non-portable instrumentation.Therefore,there is an urgent need to develop sensitive,fast,simple and low-cost in-situ detection technique and analysis instruments for DBPs.Electrochemical sensors,as a beneficial complement to the standard DBPs detection method,are expected to achieve on-site in-situ detection and remote real-time monitoring.This article provided a concise overview of regulatory indicators and standardized detection methods for DBPs,followed by an in-depth discussion of recent advancements in electrochemical detection of DBPs,focusing on two key aspects,recognition probes and analytical techniques.Finally,the current challenges and potential research directions in the field of electrochemical sensors for DBPs were summarized.
3.Mechanism of antidepressant action of kaji-ichigoside F1 based on metabolomics.

In this study, serum metabolomics techniques and molecular biology methods were used to investigate the intervention effect of kaji-ichigoside F1 on chronic unpredictable mild stress(CUMS) depression mouse model and its mechanism. The CUMS depression mouse model was constructed, and the mice were divided into blank group, model group, escitalopram(ESC, 10 mg·kg~(-1)) group, and low-dose, medium-dose, and high-dose kaji-ichigoside F1 groups(1, 2, and 4 mg·kg~(-1)). CUMS modeling was performed on all mice except the blank group, and the cycle was four weeks. At the end of modelling, ESC and kaji-ichigoside F1 were administered by gavage once a day for 28 days. After the end of the administration, behavioral testing(sucrose preference test, open field test, forced swimming test, and tail suspension test) was conducted to evaluate the improvement of depression symptoms of different doses of kaji-ichigoside F1 on CUMS depression mouse model. The morphology of neurons and the number of Nissl bodies in the hippocampus were observed by Nissl staining. Metabolomics technique was used to analyze the changes in serum differential metabolites in mice. Protein expression levels of P2X7 purinergic receptor(P2X7R), adenosine A1 receptor(A1R), and adenosine receptor A2A(A2AR) in mouse hippocampus were detected by Western blot. The results showed that compared with that in the blank group, the body weight of mice in the model group was significantly decreased, and the sucrose preference rate was significantly decreased. The immobility time was significantly increased in the forced swimming and tail suspension tests, and the total moving distance was significantly decreased in the open field test. The number of Nissl bodies was significantly decreased, and the depression-like behavior and the number of Nissl bodies in the hippocampus of mice were significantly improved after administration of kaji-ichigoside F1. In the metabonomics analysis, the purine metabolism of serum after kaji-ichigoside F1 administration was involved in the metabolic passage of depression, and Western blot analysis verified the expression of P2X7R, A1R, and A2AR proteins in purine metabolic pathways. The results show that kaji-ichigoside F1 significantly decreases the expression of P2X7R and A2AR proteins in the hippocampus of CUMS model mice and increases the expression level of A1R proteins. It is suggested that kaji-ichigoside F1 may play an antidepressant role by regulating the expression of P2X7R, A1R, and A2AR proteins in the purine metabolism pathway.

4.Targeted gene silencing in mouse testicular Sertoli and Leydig cells using adeno-associated virus vectors.

Researchers commonly use cyclization recombination enzyme/locus of X-over P1 (Cre/loxP) technology-based conditional gene knockouts of model mice to investigate the functional roles of genes of interest in Sertoli and Leydig cells within the testis. However, the shortcomings of these genetic tools include high costs, lengthy experimental periods, and limited accessibility for researchers. Therefore, exploring alternative gene silencing techniques is of great practical value. In this study, we employed adeno-associated virus (AAV) as a vector for gene silencing in Sertoli and Leydig cells. Our findings demonstrated that AAV serotypes 1, 8, and 9 exhibited high infection efficiency in both types of testis cells. Importantly, we discovered that all three AAV serotypes exhibited exquisite specificity in targeting Sertoli cells via tubular injection while demonstrating remarkable selectivity in targeting Leydig cells via interstitial injection. We achieved cell-specific knockouts of the steroidogenic acute regulatory ( Star ) and luteinizing hormone/human chorionic gonadotropin receptor (Lhcgr) genes in Leydig cells, but not in Sertoli cells, using AAV9-single guide RNA (sgRNA)-mediated gene editing in Rosa26-LSL-Cas9 mice. Knockdown of androgen receptor ( Ar ) gene expression in Sertoli cells of wild-type mice was achieved via tubular injection of AAV9-short hairpin RNA (shRNA)-mediated targeting. Our findings offer technical approaches for investigating gene function in Sertoli and Leydig cells through AAV9-mediated gene silencing.

5.Predictive value of uterine artery Doppler parameters combined with cerebral placental ratio for late-onset fetal growth restriction

Objective To investigate the application value of uterine artery Doppler parameters combined with cerebral placental ratio(CPR)in predicting late-onset fetal growth restriction(FGR).Methods A total of 82 pregnant women with singleton pregnancy of FGR who were admitted to Jiangyin Hospital of Traditional Chinese Medicine from May 2021 to May 2023 were enrolled.And 60 healthy pregnant women with singleton pregnancy who underwent physical examinations during the same period were enrolled as control.All participants underwent arterial ultrasound examination,and CPR was measured.The uterine artery Doppler parameters and CPR were compared between the FGR group and control group,and their correlation with the disease was analyzed.The predictive efficacy of combined detection was assessed through receiver operator characteristic(ROC)curve.Results The systolic peak velocity/end diastolic velocity(S/D),resistance index(RI),and pulsatility index(PI)of the uterine artery and umbilical artery(UA)in the FGR group were higher than those in the control group,whereas the S/D,RI,and PI of the middle cerabral artery(MCA)and CPR in the FGR group were lower than those in the control group(P<0.05).Spearman correlation analysis showed that the S/D,RI,and PI of the uterine artery and UA were positively correlated with late-onset FGR,and the S/D,RI,and PI of the MCA and CPR were negatively correlated with late-onset FGR(P<0.05).ROC curve analysis indicated that the AUC for predicting late-onset FGR by combining uterine artery Doppler parameters with CPR was 0.925,with a sensitivity of 79.3%and a specificity of 88.3%.Conclusion The uterine artery Doppler parameters combined with CPR can effectively predict late-onset FGR,providing significant clinical value in early identification of high-risk pregnant women and guiding clinical interventions.
6.Effects of esketamine combined with ultrasound-guided stellate ganglion block on stress response and postoperative immune function in elderly patients undergoing enterectomy

Objective:To investigate effect of esketamine combined with ultrasound-guided stellate ganglion block(SGB)on stress response and postoperative immune function in elderly patients undergoing colorectal resection.Methods:A retrospective analy-sis was conducted in clinical data from 70 cases of elderly patients undergoing elective colorectal resection in Xuzhou Hospital of Tradi-tional Chinese Medicine,Nanjing University of Chinese Medicine from September 2020 to April 2022,who were randomly divided into observation group(n=35)and control group(n=35),control group was treated with ultrasound-guided SGB,observation group was treated with esketamine combined ultrasound-guided SGB.Heart rate(HR),mean arterial pressure(MAP),product of heart rate and systolic blood pressure(RPP)and stress response levels[cortisol(CORT),SOD,MDA,IL-6,TNF-α,IL-1β]were compared between two groups at 5 min before SGB(t0),immediately after tracheal intubation(t1),1 h after operation(t2)and end of operation(t3).CD3+T,CD4+T,CD8+T and CD4+T/CD8+T at t0,t3,24 h after operation(t4)and 72 h after operation(t5)were compared,respec-tively.Recovery time,first exhaust time,bowel sound recovery time and first oral feeding time and incidence of adverse reactions during anesthesia were compared between two groups.Results:HR,MAP,RPP,CORT,SOD,MDA,IL-6,TNF-α,IL-1β,CD3+T,CD4+T,CD8+T and CD4+T/CD8+T at different time points in two groups were compared by repeated measures analysis of variance,and there were significant differences between groups and at different time points(P<0.05).Compared with control group,observation group had shorter recovery time,first exhaust time,bowel sound recovery time,first oral feeding time,postoperative time out of bed(P<0.05),and less intraoperative pain medication consumption,incidence of adverse reactions during anesthesia was reduced[48.57%(17/35)vs 25.71%(9/35),P<0.05].Conclusion:Esketamine combined with ultrasound-guided SGB in elderly patients undergoing enterectomy can promote recovery of postoperative gastrointestinal function,reduce perioperative stress response,has little impact on immune function of patients,reduce perioperative adverse reactions.
7.Qualitative study on risk perception of additional injury during motor rehabilitation in stroke patients with hemiplegia

Objective:To explore the characteristics and influencing factors of risk perception related to additional injury during motor rehabilitation in stroke patients with hemiplegia, and to provide a foundation for the development of assessment tools and prevention strategies.Methods:A purposive sampling strategy combined with maximum variation sampling was employed to select 15 stroke patients with hemiplegia who were hospitalized in the Department of Neurology and Department of Rehabilitation at Northern Jiangsu People's Hospital between January and April 2024. Face-to-face, semi-structured in-depth interviews were conducted. Data were analyzed and thematically extracted using Colaizzi seven-step method.Results:Each interview lasted between 25 and 47 minutes, totaling 547 minutes of interview time and approximately 92 000 words of transcription. A total of 3 major themes were identified: characteristics of risk perception regarding additional injury during motor rehabilitation, multidimensional influencing factors of risk perception, and perceived severity of additional injury.Conclusions:Stroke patients with hemiplegia exhibit diverse and dynamic risk perception characteristics regarding additional injury during motor rehabilitation, which are influenced by multiple interrelated factors. A generally low and inaccurate level of risk perception was observed. Medical staff should pay greater attention to improving the accuracy of patients' risk perception, enhancing risk assessment mechanisms, and developing targeted prevention strategies to help patients avoid additional injuries and maximize the benefits of motor rehabilitation.
