Objective To analyze differences of purified water, softened water, and concentrated softened water on the decontamination effects for surgical instruments,so as to provide a reliable reference for the selection of cleaning water for surgical instruments. Methods The physical and chemical indexes and components of concentrated softened water, softened water, and purified water were detected, and their cleaning effects for instrument were compared. The decontamination cleaning experiment was conducted in three hospitals with different types of cleaning water using mini cleaning machines, and the differences in the decontamination time were analyzed. The cleaning and decontamination experiments on contaminated instruments was performed using a spray cleaning and disinfection device, and the cleaning effects of instruments with different types of cleaning water were analyzed. Results Calcium content and hardness of the three types of cleaning water were all at a low level, which can avoid scale adhesion for cleaning surgical instruments. There were statistically significant differences in the pH value, conductivity, and contents of calcium, sodium and chloride among the three cleaning waters(P<0.05).The purified water was weakly acidic, and its conductivity, hardness, and contents of calcium, sodium and chloride were all at low levels. The softened water and concentrated softened water were weakly alkaline, with high levels of conductivity and sodium content and low level of chloride. There were statistically significant differences in the decontamination time of the purified water, softened water and concentrated softened water among hospitals(P<0.05).Under the same contamination condition of surgical instruments, there was a statistically significant difference in the qualified rate of instruments cleaning in the upper and lower cleaning baskets with different cleaning waters(P<0.05).Conclusion Concentrated softened water and softened water have high sodium content, and their decontamination and cleaning abilities are significantly stronger than those of the purified water.Purified water has poorer effects in instruments cleaning for its deionized property.Using concentrated softened water or softened water in the surgical instrument cleaning process can achieve effects of reducing consumption and increasing efficiency, which is conducive to improving cleaning efficiency and quality.

Journal of Regional Anatomy and Operative Surgery 2025;34(10):855-860
Objective To investigate the role and potential mechanism of protein arginine methyltransferase 1(PRMT1)in regulating the proliferation, apoptosis and migration of human oral squamous cell carcinoma SCC-25 cells. Methods Normal oral mucosal cells S-G and oral squamous cell carcinoma cells SCC-25,OECM-1,and HSC-3 were collected and cultured, and the transcription level of PRMT1 was detected.SCC-25 cells were divided into the untransfected group(SCC-25 group),empty vector control group(SCC-25+NC group),and transfection group(SCC-25+si-PRMT1 group).Protein expression levels were detected by Western blot;cell proliferation and migration abilities were assessed by MTT assay and cell scratch wound healing assay, respectively;and the apoptosis rates were determined by flow cytometry.Exogenous TGF-β stimulation experiment was conducted on the basis of constructed SCC-25+si-PRMT1 cells to verify the role of TGF-β/Smad pathway in the PRMT1 silencing effect. Results Compared with normal oral mucosal cells S-G,PRMT1 was highly expressed in oral squamous cell carcinoma cells SCC-25,OECM-1,and HSC-3(P<0.05),with the highest expression observed in SCC-25 cells(P<0.05).After silencing the PRMT1 gene, the expressions of pro-apoptotic proteins Bax and C-Caspase-3 in SCC-25 cells significantly increased, the apoptosis rate significantly increased, the expression of anti-apoptotic protein Bcl-2 significantly decreased, and the cell proliferation and migration abilities were inhibited.Simultaneous silencing of the PRMT1 gene can significantly downregulated the expression levels of TGF-β and its downstream effector p-Smad.Compared with the SCC-25+si-PRMT1 group, the cell proliferation and migration abilities in the SCC-25+si-PRMT1+TGF-β group were enhanced, and the apoptosis rate decreased, which suggesting that TGF-β could partially reverse the tumor suppressor effect caused by PRMT1 silencing. Conclusion PRMT1 regulates the proliferation, apoptosis and migration of SCC-25 cells through TGF-β/Smad signaling pathway.

Objective To elucidate the mechanisms of trauma-induced coagulopathy(TIC),clarify the specific pathogenic factors and pathophysiological processes, and discover the effective diagnostic indicators and therapeutic targets. Methods Transcriptomic data of traumatic hemorrhagic shock patients were obtained from the Gene Expression Omnibus(GEO)to identify differentially expressed genes(DEGs).Coagulation-related genes(CRGs)from the Kyoto Encyclopedia of Genes and Genomes(KEGG)were intersected with DEGs.Machine learning algorithms, including least absolute shrinkage and selection operator(LASSO)and random forest(RF),were applied to identify key genes. The CIBERSORT algorithm was used to analyze the correlation between key genes and immune cell infiltration.Through consensus clustering, subtype analysis was conducted on trauma patients to compare the infiltration of immune cells.A rat model of traumatic hemorrhagic shock was established to validate coagulation function and the expression of key genes. Results The dataset included samples from 17 healthy controls and 478 patients with traumatic hemorrhagic shock.A total of 6 315 DEGs were identified under the screening criterion of corrected P<0.05.Gene set enrichment analysis(GSEA)showed that the up-regulated DEGs were significantly enriched in the glucose metabolism pathway, while the down-regulated DEGs were enriched in the immune reaction-related pathways.Through cross-analysis of DEGs and CRGs,a total of 65 differentially expressed coagulation-related genes(DE-CRGs)were screened out.GO functional enrichment showed that these genes were mainly located in secreting granular membranes and platelet α-granules,and were involved in physiological processes such as blood coagulation, regulation of body fluid levels, and wound healing.KEGG pathway analysis revealed that these genes were significantly enriched in pathways such as platelet activation, complement and coagulation cascade reactions,Rap1 signaling pathway, and human cytomegalovirus infection.Six key DE-CRGs were identified through machine learning.Receiver operating characteristic(ROC)curve analysis indicated that these genes had good diagnostic efficacy.CIBERSORT analysis revealed a significant correlation between key genes and immune cell infiltration.Patients were classified into two subtypes based on the six key genes:subtype A was rich in CD8+T cells and activated NK cells, presented an immune-active state;subtype B was mainly composed of monocytes and resting NK cells, with insufficient activation of immune pathways.Animal experiments on rats showed that hemorrhagic shock can lead to coagulation dysfunction. The results of qRT-PCR further confirmed that the expression trend of key genes was consistent with the results of bioinformatics analysis. Conclusion In this study, through transcriptomics and machine learning methods, six key genes closely related to TIC were systematically screened out, namely GNA13,PIK3R3,ITGAM,MAPK14,PPP1CC and LYN,and their close connections with coagulation function and immune infiltration were revealed.Animal experiments have further verified the value of these genes as potential diagnostic and therapeutic targets.
