1.The Mechanism of Gongfa Static Training Regulating Mitophagy in Skeletal Muscle of Type 2 Diabetes Mellitus(T2DM)Mice via the PINK1/Parkin Pathway

OBJECTIVE To explore the mechanism by which Gongfa Static Training regulates mitophagy through the PTEN-in-duced kinase 1(PINK1)/Parkin pathway in skeletal muscle insulin resistance in type 2 diabetes mellitus(T2DM).METHODS T2DM mouse model was established using a high-fat diet combined with streptozotocin(STZ)intraperitoneal injection.Mice were ran-domly divided into a model group,a metformin group,an aerobic exercise group,and a Gongfa Static Training group.The intervention effects of Gongfa Static Training were evaluated by measuring fasting blood glucose,Homeostatic Model Assessment of Insulin Resist-ance(HOMA-IR),glycated hemoglobin(HbA1c),lipid metabolism indicators,mitochondrial function in the gastrocnemius muscle,and the expression of PINK1 and Parkin-related genes and proteins.RESULTS Gongfa Static Training significantly reduced fasting blood glucose,HbA1c,and insulin resistance index in T2DM mice,improved lipid metabolism,and enhanced insulin sensitivity.It improved the structure and function of mitochondria in the gastrocnemius muscle by upregulating the mRNA and protein expression of PINK1 and Parkin.CONCLUSION Gongfa Static Training improves mitochondrial function and insulin resistance in the skeletal muscle of T2DM mice by regulating the PINK1/Parkin pathway.

Based on the mechanism of modern immunology,Professor Ding Ying,a traditional Chinese medicine(TCM)master,uses the analogy of TCM to simplify the complex and calls the abnormal immune response that leads to the onset of immune thrombocy-topenia(ITP)"latent poison".She believes that the invasion of latent poison is the basis of ITP;blood damage and marrow injury are the pathological essence of ITP;and the transformation of"poison-deficiency-stasis"is the basic pathogenesis of ITP.She has formu-lated the treatment principle of"strengthening healthy qi,removing poison,and removing stasis",and attaches importance to the appli-cation of rattan drugs and charcoal drugs,which has achieved good results in clinical practice.
3.Study on Mechanism of Huiyang Shengji Decoction in Promoting Yin Syndrome Wound Healing in Diabetic Mice by Acceler-ating Dendritic Cell Efferocytosis

OBJECTIVE To explore the effect and mechanism of Huiyang Shengji Decoction on wound healing in mice with dia-betic yin syndrome.METHODS SPF C57BL/6 mice were selected,and 24 diabetic ulcer models were established by intraperitoneal injection of streptozotocin and skin defect method.The mice were randomly divided into model group,low-dose,medium-dose and high-dose Huiyang Shengji Decoction groups,with 6 mice in each group.Six mice were selected to establish a common wound model as the blank group.The blank group and model group were treated with distilled water by intragastric administration every day,and the Huiyang Shengji Decoction groups were treated with Huiyang Shengji Decoction by intragastric administration every day.The wounds were photographed every day,and the wound healing rate was recorded.HE staining was used to observe the growth of granulation tis-sue in the wounds.Western blot was used to detect the expression of Axl,Tyro3 and Mertk proteins related to wound efferocytosis.Im-munohistochemistry was used to detect the expression of CD11c in wound tissue.qPCR was used to detect the mRNA expression of TNF-α,IL-1β,IL-10 and TGF-β1 in the wounds.TUNEL staining was used to calculate the number of apoptotic cells(AC)in the wounds.Mouse primary bone marrow dendritic cells(BMDC)were resuspended in 1640 medium containing 10%blank serum or 5%,10%,and 20%Huiyang Shengji Decoction-containing serum.After culturing in a cell culture incubator at 37℃and 5%CO2 for 24 h,the wound endocytosis-related receptors were detected by Western blot.BMDC and AC were co-cultured in vitro,and the phag-ocytic rate of BMDC phagocytosis of AC was detected by flow cytometry.RESULTS The wound healing in each dose group of Huiyang Shengji Decoction was significantly faster than that in the model group(P<0.05).Compared with the model group,the protein expres-sion of Axl and Tyro3 in the wound tissue of mice in the high dose group of Huiyang Shengji Decoction was significantly increased(P<0.01,P<0.000 1),and there was no significant difference in the expression of Mertk protein in the wound tissue of mice in the low,medium and high dose groups(P>0.05).The Huiyang Shengji Decoction significantly inhibited the expression of IL-1β and TNF-α mRNA in the wound tissue(P<0.05),promoted the expression of TGF-β1 and IL-10 mRNA(P<0.05),and the number of AC in the wound after treatment with Huiyang Shengji Decoction was less than that in the model group(P<0.05).In the in vitro experiment,compared with the model group,the protein expression of Axl and Mertk in BMDC in the 20%Huiyang Shengji Decoction-containing serum group was significantly increased(P<0.01,P<0.001),and the protein expression of Tyro3 in the 5%,10%,and 20%Huiy-ang Shengji Decoction-containing serum groups was higher than that in the model group(P<0.01,P<0.001).The phagocytic rate of AC in the Huiyang Shengji Decoction group was higher than that in the model group(P<0.05).CONCLUSION Huiyang Shengji Decoction can enhance the efferocytosis function of dendritic cells(DC),reduce AC aggregation in the wound,promote the disappear-ance of wound inflammation and tissue repair,and accelerate the healing of skin wounds.
4.Investigation of the Mechanism of Cold Hyperalgesia in KOA Mice Relieved by Shangke Lengtongtie Based on HMGB1/CX-CL12/CXCR4 Signaling Axis

OBJECTIVE To explore the intervention mechanism of Shangke Lengtongtie on cold hyperalgesia in KOA mice based on the HMGB1/CXCL12/CXCR4 signaling axis.METHODS Monosodium iodoacetate(MIA)was used for the intra-articular injec-tion into the knee joint to establish mice model of knee osteoarthritis(KOA).Peripheral blood monocytes were extracted from mice,cultured,and then reinfused into the tail vein of the mice.Subsequently,in vivo animal imaging was used to observe the recruitment sites of these monocytes.The cold hyperalgesia threshold was measured at various time points in each group of mice.Hematoxylin and eosin(HE)staining was used to evaluate the level of synovial pathological changes.ELISA was employed to detect the expression of in-flammatory factors IL-1β,TNF-α,and pain mediators CGRP and Substance P in mouse serum.Western blot and qPCR methods were used to detect the protein and gene expression of cold hyperalgesia-related indicators such as TRPA1,TRPM8,HMGB1,CXCL12,CXCR4,Collagen Ⅰ,and Netrin-1 in synovial tissue,as well as DCC in dorsal root ganglia(DRG)tissue.RESULTS In vivo ima-ging showed that after the monocytes were reinfused into KOA mice,they were recruited to the knee joint area,with the HMGB1 group exhibiting a greater recruitment of circulating monocytes at the knee joint.Additionally,compared to the control group,the KOA group and HMGB1 group showed inflammatory pathological changes in the synovium,increased expression of serum inflammatory factors and pain mediators,reduced cold hyperalgesia threshold,and upregulated protein and gene expression of cold hyperalgesia-related indica-tors in synovial and DRG tissues.The changes were more significant in the HMGB1 group compared to the KOA group(P<0.05).Af-ter treatment with Shangke Lengtongtie or GL intervention,synovial inflammation was alleviated,serum inflammatory factors and pain mediators decreased,cold hyperalgesia threshold increased,and the upregulation of cold hyperalgesia-related indicator protein and gene expression levels was significantly reversed(P<0.05).CONCLUSION Shangke Lengtongtie exerts a beneficial effect on the mitigation of synovitis and cold hyperalgesia in KOA mice,a therapeutic mechanism that possibly mediated through the inhibition of the HMGB1/CXCL12/CXCR4 signaling axis.
5.Study on the Inhibitory Effect of Bruceine A on Wnt/β-catenin Signaling Pathway against Proliferation and Metastasis of Colorectal Cancer Cells

OBJECTIVE To investigate the inhibitory effects of Bruceine A(BA)on colon cancer and its underlying mechanisms.METHODS Human colon cancer HT-29 and HCT116 cells were treated with various concentrations of BA(0,1,2,5,10,20,40,80 μmol·L-1).Cell viability was assessed using the Cell Counting Kit-8(CCK-8).Flow cytometry,wound healing assays,and Transwell assays were employed to evaluate the effects of BA on cell apoptosis,cell cycle,invasion,and migration.Mo-lecular docking simulations were used to assess the binding of BA to GSK-3β protein,and Western blot analysis was used to examine protein expression related to the cell cycle,epithelial-mesenchymal transition,and the Wnt/β-catenin signaling pathway.An HT-29 cell subcutaneous xenograft mouse model was established.After tumor formation,mice were randomly divided into three groups(six mice per group):a blank group,a low-dose BA group(0.1 mg·kg-1),and a high-dose BA group(0.2 mg·kg-1).Mice were ad-ministered the drug for 19 d,then sacrificed,and tumor tissues were collected.Tumor volume changes over time were observed;Ki67 immunohistochemistry was used to assess cell proliferation in tumor tissues;Western blot analysis of Wnt/β-catenin signaling pathway protein expression was conducted.RESULTS Compared with the blank group,BA could significantly inhibit the proliferation of HT-29 and HCT116 cells,with IC50 values of 10.80 μmol·L-1 and 17.96 μmol·L-1,respectively.Flow cytometry results showed that BA significantly induced apoptosis of HT-29 cells(P<0.01,P<0.001),and arrested the cell cycle at the S phase,accompanied by de-creased expression of cycle-related proteins CDK2 and Cyclin A(P<0.05,P<0.01,P<0.001).BA inhibited cell migration and in-vasion ability(P<0.05,P<0.01,P<0.001),reduced the expression of EMT-related proteins Snail,Vimentin,and N-Cadherin(P<0.01,P<0.001),and upregulated the expression of E-Cadherin protein.In addition,BA inhibited the expression of β-catenin and p-GSK3β proteins.Wnt agonist LiCl could significantly antagonize the anti-colon cancer effect of BA;Wnt inhibitor XAV939 could enhance the anti-colon cancer effect of BA.In the in vivo experiment,compared with the blank group,the tumor volume of the low-dose and high-dose BA groups was significantly reduced(P<0.05,P<0.001).Immunohistochemistry results showed that compared with the blank group,the expression of Ki67 in tumor tissues of the low-dose and high-dose BA groups was significantly reduced(P<0.001).Western blot results further proved that BA inhibited the Wnt/β-catenin signaling pathway.CONCLUSION BA inhibits the viability,invasion,and migration of colon cancer HT-29 cells,induces apoptosis,and causes cell cycle arrest.Additionally,it significantly suppresses the growth of subcutaneous HT-29 cell xenografts in vivo,possibly related to the Wnt/β-catenin signaling pathway.
6.A New Phenolic Glycoside from the Stems and Leaves of Illicium Dunnianum

OBJECTIVE To study the constituents from the dried stems and leaves of Illicium dunnianum Tutcher.METHODS The compounds were isolated and purified by column chromatography of AB-8 macroporous resin,silica gel,HW-40C,ODS,Sephadex LH-20,and semi-preparative RP-HPLC.Their structures were elucidated by physicochemical properties,spectral analyses and ECD.RESULTS The 70%ethanol extract of Illicium dunnianum was subjected to AB-8 macroporous adsorption resin CC to yield 30%ethanol fraction.Five compounds were obtained and characterized as anisole glycol-7-O-β-L-funanarabifuranosyl-(1→6)-β-D-glucopyranoside(1),4-O-β-D-glucopyranosyloxy-benzaldehyde(2),benzyl-O-α-L-rhamnopyranosyl-(1→6)-β-D-glucopyranoside(3),β-D-glucopyranoside benzoate(4)and sachalinoside B(5),respectively.CONCLUSION Compound 1 is a new phenolic glycoside,2-5 are identified from Illicium dunnianum for the first time.
7.Exploring the Mechanism of Safflower in Alleviating Myocardial Injury during Low-Temperature Storage of Mouse Hearts Based on Network Pharmacology and Experimental Verification

OBJECTIVE To explore the protective mechanism of safflower on mouse hearts preserved at low temperatures,sum-marize and analyze the potential targets of safflower,and provide reference for the improvement of ex vivo myocardial injury by tradition-al Chinese medicine safflower.METHODS Databases such as TCMSP,TCMID,and Swiss Target Prediction were used to search for the chemical composition and target of safflower.Databases such as OMIM,TTD,and Genecards were employed to screen disease tar-gets for ischemic cardiomyopathy.A network diagram of"drug-ingredient-target-disease"was construct using Cytoscape,and the mechanism of the action of safflower was enriched and analyzed.A mouse ex vivo heart cold preservation model was constructed and the predicted targets of network pharmacology were validated using techniques such as ELISA detection,qPCR,Western blot,etc.RE-SULTS A total of 22 active ingredients and 421 drug targets were obtained from safflower;1 812 disease targets were identified in is-chemic cardiomyopathy,resulting in 117 drug-disease common targets.GO enrichment analysis yielded 1 906 entries,and KEGG pathway enrichment screening identified a total of 137 signaling pathways,including the Akt signaling pathway.Experimental verifica-tion showed that safflower might alleviate the damage to mouse hearts during low-temperature storage by regulating the PI3K-Akt path-way to regulate cell apoptosis.CONCLUSION This article uses network pharmacology to reveal the protective targets and pathways of safflower on the heart of mice subjected to low-temperature cold storage,and conducts relevant experimental verification.This pro-vides a basis for further exploring the mechanism of safflower in reducing myocardial injury in low-temperature cold stored hearts,and provides a theoretical basis for the clinical application and pharmacological research of safflower.
8.Clinical Study on Zhenshi Burn Ointment Promoting the Wound Healing of Postoperative Simple Anal Fistula with Damp-Heat Pouring Downward Syndrome

OBJECTIVE To observe the clinical efficacy of Zhenshi Burn Ointment on postoperative wound healing of simple anal fistula and explore its potential mechanism of action.METHODS A total of 60 patients with low-position simple anal fistula of damp-heat pouring downward type admitted to the Department of Anorectum,Nanjing Hospital of Chinese Medicine Affiliated to Nan-jing University of Chinese Medicine from May to December 2023 were selected as the research subjects and randomly divided into an experimental group and a control group,with 30 cases in each group.After surgery,the control group was given sterilized vaseline gauze for external application,and the experimental group was given Zhenshi Burn Ointment for external application.The course of treatment for both groups was 30 d.After treatment,the postoperative wound healing time,wound healing rate,pain score,wound granulation growth and secretion,and changes in TCM syndrome score were compared between the two groups.The pathological sec-tions of wound granulation tissue were compared 14 d after surgery.The transcription levels of COL1A1,type Ⅲ collagen(COL3A1),epidermal growth factor(EGF),and fibroblast growth factor(FGF2)in granulation tissue were detected by qPCR.The expression lev-els of proteins related to MEK/ERK signaling pathway were detected by Western blot.RESULTS Compared with the control group,the wound healing rates of the experimental group were significantly increased at 7,14,21,and 30 d after surgery(P<0.01),and the wound healing time was significantly shortened(P<0.01).At 7 d after surgery,the pain score of the experimental group was signifi-cantly lower than that of the control group(P<0.05).At 7 and 14 d after surgery,the granulation growth score of the wound in the ex-perimental group was significantly lower than that of the control group(P<0.01).At 14 d after surgery,the secretion score of the ex-perimental group was significantly lower than that of the control group(P<0.01).At 14 and 30 d after surgery,the total score of TCM syndrome in the experimental group was significantly better than that in the control group(P<0.05).At 14 d after surgery,the patho-logical improvement of granulation tissue in the wound in the experimental group was better than that in the control group,and the con-tent of collagen fiber and type I collagen(Collagen I)was significantly higher than that in the control group(P<0.01).The mRNA contents of COL1A1,COL3A1,EGF,and FGF2 were significantly higher than those in the control group(P<0.05,P<0.01);the levels of p-MEK1/2 and p-ERK1/2 proteins were higher than those in the control group(P<0.05).CONCLUSION Zhenshi Burn Ointment has a good effect on promoting postoperative anal fistula wound healing,which may be related to upregulating growth factor expression,promoting collagen regeneration,and activating MEK/ERK pathway.
9.From"Disease-Centered"to"People-Centered":Transformation and Development of Health Model

With the in-depth implementation of the national strategy of"Healthy China 2030",people's understanding and de-mand for health have been constantly evolving,promoting the transformation of the health model from"disease-centered"to"people-centered".This paper discusses the characteristics and limitations of the"disease-centered"health model,proposes the connotation of the"people-centered"health model and the method of maintaining health in traditional Chinese medicine,emphasizes overall health with prevention as the main focus and individual as the center,and specifically discusses the implementation measures of the"people-centered"health model:emphasize the transformation of ideas and concepts,give full play to the advantages of modern traditional Chi-nese medicine,and establish a system network of health models.It is also related to the transformation of the active health paradigm,in order to break through the limitations of the existing health model and achieve the goal of health for all.
10.Alienation and Enlightenment of the View of Life in TCM Health Preservation Theory

With the development of the times,the ancient view of life has been changing.Some excellent ideas,methods and tech-nologies with great ecological value have been or are being alienated under the impact of scientific and technological progress.The cog-nition of the value of life has gradually faded,and simple and easy technical methods have been distorted by Western values.In order to make the TCM health preservation knowledge system truly an important part of the Healthy China strategy,it is necessary to correct the cognition of the essence of life,clarify the definition of the connotation and extension of health preservation,set the philosophical premise of health preservation rules,examine the value and significance of the concept of life and death,and solve these basic theoreti-cal elements,so as to give full play to the guiding value of TCM life philosophy to human health.
