1.Mogroside Ⅴ promotes osteogenic differentiation of bone marrow mesenchymal stem cells by modulating M1 polarization of macrophages under high glucose condition

BACKGROUND:The diabetic microenvironment can cause excessive M1 polarization of macrophages,and this hyperglycemic inflammatory state can inhibit osteogenic differentiation of bone marrow mesenchymal stem cells,thus affecting the healing of diabetic bone defects.Studies have indicated that mogroside V possesses anti-inflammatory,antioxidant,and hypoglycemic properties.However,its potential to modulate M1 polarization of macrophages and osteogenic differentiation of bone marrow mesenchymal stem cells under high glucose and inflammatory condition remains unclear. OBJECTIVE:To explore the effect of mogroside V on regulating M1 macrophage polarization and its effect on osteogenic differentiation of bone marrow mesenchymal stem cells under high glucose and inflammatory condition. METHODS:Murine diabetic models were established using C57BL/6 mice.Bone marrow-derived macrophages were isolated from tibia and fibula of normal and diabetic mice,and cultured in low-glucose and high-glucose media.Then M1 polarization of bone marrow-derived macrophages was induced using lipopolysaccharide and interferon-γ.Bone marrow-derived macrophages were treated with 160,320,and 640 μmol/L mogroside V.Flow cytometry was employed to determine the proportion of F4/80+CD86+cells.qRT-PCR was utilized to assess mRNA expression levels of inducible nitric oxide synthase,interleukin 1β,and interleukin 6.ELISA was employed to evaluate tumor necrosis factor-α secretion in bone marrow-derived macrophage supernatants.Bone marrow mesenchymal stem cells were isolated from tibia and fibula of C57BL/6 suckling mice,and induced osteogenic differentiation using low-or high-glucose osteogenic induction medium.Bone marrow mesenchymal stem cells were treated with M1 macrophage-conditioned mediums with or without 320 μmol/L mogroside V in osteogenic differentiation process.qRT-PCR was employed to assess the mRNA expression of alkaline phosphatase,Runt-related factor 2,osteocalcin,and osteopontin on day 14 after osteogenic induction.Alizarin red staining and quantitative analysis were conducted to evaluate calcium deposition on day 21 after osteogenic induction. RESULTS AND CONCLUSION:(1)Flow cytometry results showed that with the treatment of 320 and 640 μmol/L mogroside V,the proportion of F4/80+CD86+bone marrow-derived macrophages was significantly lower than that in the high-glucose control group(P<0.05).(2)qRT-PCR results showed that with the treatment of 160,320,and 640 μmol/L mogroside V,the mRNA expression levels of inducible nitric oxide synthase and interleukin 6 were significantly lower than that in the high-glucose control group(P<0.05).With the treatment of 320 and 640 μmol/L mogroside V,the mRNA expression level of interleukin 1β was significantly lower than that in the high-glucose control group(P<0.05).(3)ELISA results exhibited that with the treatment of 160,320,and 640 μmol/L mogroside V,the tumor necrosis factor-α secretion level was significantly lower than that in the high-glucose control group(P<0.05).(4)With the treatment of 320 μmol/L mogroside V,calcium salt deposition was increased in bone marrow mesenchymal stem cells under high glucose and inflammatory conditions(P<0.05),and the mRNA relative expression levels of alkaline phosphatase,Runt-related factor 2,and osteopontin were increased(P<0.05).These findings indicate that mogroside V can promote osteogenic differentiation of bone marrow mesenchymal stem cells by inhibiting the M1 polarization of bone marrow-derived macrophages under high glucose and inflammatory conditions and reducing the generation of inflammatory factors.

Objective To systematically examine research progress and evolutionary trajectories of public hospitals in China in digital intelligence era,providing foundational references for digital intelligence management studies.Methods Based on papers published in core journals indexed by China National Knowledge Infrastructure(CNKI)from January 2018 to Septem-ber 2024,including those from Peking University Core,CSCD,and CSSCI journals,we conducted a visualized analysis using CiteSpace 6.2.R6 software.The analysis covered publication trends,collaboration networks,research hotspots,and core topics in the field of public hospital management research in the digital intelligence era.Results A total of 385 papers were finally in-cluded,the publication exhibiting an upward trend in number.The authors and affiliations were relatively independent.Such top-ics as"high-quality development""informatization""big data",and"smart hospital"were the predominant hotspots in public hospital management research in China.The research themes included high-quality development of public hospitals,big data ap-plications,and digital transformation and upgrading.Conclusion Smart hospital development remains to be a key research fo-cus.Enhanced interdisciplinary collaboration,methodology innovation,and multidimensional perspectives are recommended to advance hospital digitalization.
3.Association between the overuse of short-form video,inattention and school refusal behavior among junior high school students

Objective:To explore the prominent manifestations and internal associations of the overuse of short-form videos,inattention,and school refusal behavior in junior high school students.Methods:A survey was conducted among 1 106 junior high school students,utilizing the Short-form Video APP Overuse Scale,the Inatten-tion subscale of the Swanson,Nolan,and Pelham Rating Scale(SNAP-Ⅳ),and the School Refusal Behavior Ques-tionnaire to assess the extent of short-form video overuse,inattention,and school refusal behavior.By using the R language for network analysis,the core nodes of individual networks and the bridge nodes connecting the combined networks were analyzed,and the structural differences in networks across genders and grades were compared.Re-sults:The core nodes of the network pertaining to overuse of short-form video,inattention,and school refusal behav-ior were"Using short videos enhances connection with others"(expected influence value=1.03),"easily distract-ed"(expected influence value=1.99),and"school alienation"(expected influence value=0.83).The bridge nodes in the combined network were"defiant behavior"(bridge expected influence value=0.69)and"difficulty sustaining mental activities"(bridge expected influence value=0.47).Analysis of network structure differences i-dentified that the core nodes within the combined networks of the first,second,and third grades of junior high school students were respectively"prioritizing short videos over other activities","defiant behavior",and"academic aptitude".Conclusion:Junior high school students exhibiting overuse of short videos,inattention,or school refusal behavior are characterized by enhanced social connectivity through platforms such as short video apps,a propensity for distraction,and feelings of school alienation,respectively.When these three behaviors coexist,the most salient related factor is oppositional behavior,coupled with difficulties in sustaining cognitive tasks and maintaining con-centration.

6.The effect of self-worth on depression in the elderly: the chain mediating effect of aging attitude and anxiety

Objective:To explore the mediating role of aging attitude and anxiety in the relationship between self-worth and depression among the elderly.Methods:From July to August 2022, a total of 536 elderly people in Shijiazhuang University for the Elderly were surveyed by the elderly self-worth scale, generalized anxiety disorder-7(GAD-7), attitudes to aging questionnaire(AAQ) and patient health questionnaire(PHQ-9). SPSS 26.0 software was used for descriptive statistics and correlation analysis.AMOS 24.0 was used to construct an intermediation model, and Bootstrap method was used for mediating effect testing.Results:(1)The score of self-worth was 101.00 (92.00, 112.00), the score of aging attitude was 92.00 (83.00, 101.75), the score of anxiety was 2.00(0, 6.00), and the score of depression was 2.00 (0, 5.00). (2)Self-worth was positively correlated with aging attitude ( r=0.67, P<0.01), while negatively correlated with anxiety ( r=-0.45, P<0.01) and depression ( r=-0.48, P<0.01). The aging attitude was negatively correlated with anxiety ( r=-0.41, P<0.01) and depression ( r=-0.41, P<0.01). Anxiety was positively correlated with depression ( r=0.71, P<0.01). (3)The indirect effect of aging attitude between self-worth and depression in the elderly was -0.016, accounting for 19.75% (-0.016/-0.081)of the total indirect effect.The indirect effect of anxiety between self-worth and depression in the elderly was -0.045, accounting for 55.56%(-0.045/-0.081) of the total indirect effect.The chain mediating effect of aging attitude and anxiety between self-worth and depression in the elderly was -0.020, accounting for 24.69%(-0.020/-0.081) of the total indirect effect. Conclusion:Aging attitude and anxiety play a chain mediating role between self-worth and depression in the elderly.
7.Research on Configuration Paths of Digital Literacy Empowering Medical Students'Innovative Behaviors:Based on Fuzzy-Set Qualitative Comparative Analysis

Objective To explore the causal complexity between individual digital literacy and innovative behaviors by focusing on medical students,and to provide scientific references for empowering their innovative behaviors.Methods A questionnaire survey was conducted to collect relevant data from students currently enrolled in a medical school in Anhui Province.Fuzzy-set qualitative comparative analysis(fsQCA)was performed to examine the different combination paths for empowering innovative behaviors in medical students.Results A total of 922 valid questionnaires were collected.Based on six conditional variables of digital literacy,namely information and data literacy,communication and collaboration literacy,digital content creation literacy,security literacy,problem solving literacy,and career-related literacy,there were five configurations for high-level innovation behaviors of medical students,with the overall consistency being 0.816 and the overall coverage being 0.664.On the other hand,there were three configurations for their low-level innovation behaviors,with the overall consistency being 0.901 and the total coverage being 0.585.Conclusion There is a causal complexity between medical students'digital literacy and their innovative behaviors.Different dimensions of digital literacy act synergistically to produce multiple paths to empower medical students'innovative behaviors.Among them,a high level of competence in digital content creation is the core condition that empowers innovative behaviors in medical students,while a low level of problem-solving competence is the key barrier to their innovative behaviors.
8.Effects of dexmedetomidine combined with propofol on hemorheology and hemodynamics during anesthesia for hypertensive intracerebral hemorrhage

Objective:To investigate the effects of dexmedetomidine combined with propofol on hemorheology and hemodynamics in patients with hypertensive intracerebral hemorrhage.Methods:126 patients with hypertensive intracerebral hemorrhage undergoing emergency surgery in Tengzhou Central People's Hospital from June 2017 to June 2019 were prospectively selected and randomly divided into control group (63 cases) and observation group (63 cases). The control group was given propofol induction anesthesia, and the observation group was given dexmedetomidine combined with propofol induction anesthesia. Hemodynamics, hemorheology before and after operation, clinical efficacy and postoperative complications were compared between the two groups.Results:24 hours after operation, the whole blood viscosity and plasma viscosity levels of the two groups were higher than those of the same group before operation, and the levels of erythrocyte aggregation index was lower than those of the same group before operation ( P>0.05); 24 hours after operation, the whole blood viscosity, plasma viscosity levels and the erythrocyte deformation index of the observation group were lower than those of the control group, and the erythrocyte aggregation index was higher than those of the control group, with statistically significant difference ( P<0.01). The levels of heart rate (HR) and systolic pressure (SP) in the observation group at T 1, T 2 and T 3 were normal and lower than those in the control group at the same time ( P<0.01). There was no significant difference in operation time and extubation time between the two groups ( P>0.05); the awakening time of the observation group was faster than that of the control group ( P<0.01). The incidence of restlessness, pharyngalgia, nausea, vomiting and rebleeding in the control group was 25.4%, higher than that in the observation group (9.5%), with statistically significant difference ( P<0.05). Conclusions:Dexmedetomidine combined with propofol induced anesthesia can stabilize intraoperative hemodynamics, improve postoperative hemorheology, promote postoperative awakening and reduce postoperative complications in patients with hypertensive intracerebral hemorrhage.
9.Impacts of Different Doses of Alcohol on Rat Endothelial Microparticles

Objective To discuss the impacts of different doses of alcohol on the level of microparticles (EMPs) of rat endothelial cells. Methods Sixty male SD rats were randomly allocated into four groups:high dose group (group A), me-dium dose group (group B), low dose group (group C) and the blank contrast group (group D). There were15 rats in each group. Rats were fed alcohol for 8 weeks. Flow cytometer was used to measure the level of circulating CD31+/CD42-EMPs in four groups, and which was compared with the alcohol dosage. Results Compared with group D, the level of circulating EMPs was significantly increased in group A and B (P<0.05). There was no significant change in the level of circulating EMPs in group C (P>0.05). Conclusion The moderate and high doses of alcohol are harmful to the function of vascular en-dothelial cells in rats, which show a significant dose-effect relationship. The low dose of alcohol shows no effect on the func-tion of vascular endothelial cells in rats. The protective effect of alcohol needs further investigation.
10.Relationship between serum uric acid and nonalcoholic fatty liver in patients with type 2 diabetes

A total of 1 007 inpatients with type 2 diabetes mellitus (T2DM) were divided into 2 groups,according to whether there was accompanied with nonalcoholic fatty liver(NAFLD) ; and the data of the two groups were analysed.Serum uric acid levels were positively correlated with body mass index and blood urea nitrogen and creatinine,while negatively correlated with HbA1c.T2DM patients complicated with NAFLD were younger,had shorter duration of T2DM,and showed impaired uric acid clearance with raised serum uric acid.The prevalence of NAFLD was higher with increasing serum uric acid levels.
