1.Consensus statement on research and application of Chinese herbal medicine derived extracellular vesicles-like particles (2023 edition).

To promote the development of extracellular vesicles of herbal medicine especially the establishment of standardization, led by the National Expert Committee on Research and Application of Chinese Herbal Vesicles, research experts in the field of herbal medicine and extracellular vesicles were invited nationwide with the support of the Expert Committee on Research and Application of Chinese Herbal Vesicles, Professional Committee on Extracellular Vesicle Research and Application, Chinese Society of Research Hospitals and the Guangdong Engineering Research Center of Chinese Herbal Vesicles. Based on the collation of relevant literature, we have adopted the Delphi method, the consensus meeting method combined with the nominal group method to form a discussion draft of "Consensus statement on research and application of Chinese herbal medicine derived extracellular vesicles-like particles (2023)". The first draft was discussed in online and offline meetings on October 12, 14, November 2, 2022 and April and May 2023 on the current status of research, nomenclature, isolation methods, quality standards and research applications of extracellular vesicles of Chinese herbal medicines, and 13 consensus opinions were finally formed. At the Third Academic Conference on Research and Application of Chinese Herbal Vesicles, held on May 26, 2023, Kewei Zhao, convenor of the consensus, presented and read the consensus to the experts of the Expert Committee on Research and Application of Chinese Herbal Vesicles. The consensus highlights the characteristics and advantages of Chinese medicine, inherits the essence, and keeps the righteousness and innovation, aiming to provide a reference for colleagues engaged in research and application of Chinese herbal vesicles at home and abroad, decode the mystery behind Chinese herbal vesicles together, establish a safe, effective and controllable accurate Chinese herbal vesicle prevention and treatment system, and build a bridge for Chinese medicine to the world.
2.Study on the Preparation Proec s of Zhe nwutang Granule

Objective:To optimize the extraction process of the water extract of Zhenwutang and study on the preparation of gran -ule of Zhenwu decoction to provide reference for the development and utilization of Zhenwutang granule .Methods: Heating refluxing was used, and the effects of the ratio of solid to liquid , extraction time and times were investigated by orthogonal test .As the synthetic indices of evaluation , the yield of extraction and the contents of paeoniflorin and benzoylmesaconine measured by HPLC were deter -mined to confirm the optimal water extraction process of Zhenwutang granule .Besides, granularity pass rate, moisture, loss on drying, solubility and angle of repose of the granule were regarded as the indices to evaluate the best ratio of the excipients in the preparation of the granule by single factor test.Results:Paeoniflorin and benzoyl mesaconitine had good linearity within the range of 5.45-32.70μg (r=0.9996) and 3.24-16.80 μg(r=0.9997), respectively.The average recovery was 99.62% (RSD =1.34% , n=6) and 1017.2 %(RSD=1.74%, n=6), respectively.The optimum extraction process was as follows :the ratio of solid to liquid was 1:12 with twice refluxing extraction ( 2h for each time) .The optimum granule forming process was as follows:the pharmaceutical excipients were a mixture of dextrin and soluble starch with the best ratio of 1:3. The granularity pass rate , moisture, loss on drying, solubility and angle of repose of the granule was94 .12% ,4.87 %, 0.93%,89 .23% and 36.18°, respectively.Conclusion:The optimized re-fluxing extraction process is stable , reliable and feasible , and the prepared granule is in good formability and melting .
3.Experimental Study on Kidney-yang Reinforcing Effect of Bama Nectar

Objective:To observe the effects of bama nectar in kidney-yang deficienct mice and on the mating ability and sperm counts of normal mice. Methods:The weight, body temperature, locomotor activity, cold-resistant,genital organ coefficient and lipid peroxidation of the hydrocortisone-induced kidney-yang deficient mice were detected. The genital organ ccoefficient, immune organ co-efficient and erectile function of the ovariectomized mice were detected. The mating ability and the sperm counts of normal mice were measured. Results:Compared with the model group, bama nectar obviously increased the weight, body temperature and locomotor ac-tivity of the hydrocortisone-induced kidney-yang deficient mice, and improved the locomotor activity, the organ coefficient of seminal vesicle, prostate gland, preputial gland and epididymis. Furthermore, bama nectar significantly increased the SOD activity in serum and reduced the level of MDA in serum when compared with the model group (P<0.05 or P<0.01). Compared with the model group, bama nectar could increase the organ coefficient of foreskin and thymus gland in the ovariectomized mice, and shorten the erec-tile latency (P<0. 05). Compared with the normal group, bama nectar improved the mating ability and the sperm counts of the male mice (P<0. 05 or P<0. 01). Conclusion:Bama nectar has beneficial effects on invigorating kidney and strengthening yang.
4.Counteractive Mechanism of Cyclovirobuxinum D for Myocardial Ischemia Induced by Pituitrin

[Objective] To observe the counteractive action of cyclovirobuxinum D (Cvb-D) for myocardial ischemia and to explore its possible mechanism. [Methods] Sixty SD rats were randomized into blank control group, Cvb-D groups in the dosage of 2.2, 1.1 and 0.55 mg? kg-1 ? d-1 respectively, isosorbide dinitrate group (25 mg? kg-1 ?d-1). The blank control group was treated with distilled water and the other groups were treated with the corresponding drugs by gavage according to the experimental design for 30 days. One hour after the last medication, pituitrin (1.5 U/kg) was injected into the caudal vein of the rats except the blank control to induce coronary artery spasticity myocardial ischemia. After that, electrocardiogram (ECG) was recorded, plasma superoxide dismutase (SOD) activity, malonyldialdehyde (MDA), lactate dehydrogenase (LDH) and phosphocreatine kinase ( CPK) levels were detected. [ Results ] The incidence of ischemic changes in ECG was decreased, plasma SOD activity increased and MDA, LDH and CPK levels lowered in Cvb-D groups as compared with those in the model group (P
5.Pharmacological Action of Shenfukang on Rat Model with Nephritis Induced by Cationized Bovine Serum Albumin

Shenfukang is an effective formula for glomerulonephritis applied in the first affiliated hospital of Guangzhou University of TCM. It is composed of Radix Folium seu Flos Tripterygii wilfordii, Radix Astragali seu Hedysari, Radix Notoginseng, Herba Leonuri, etc.. Glomerulonephritis rat model was induced by cationized bovine serum albumin (C-BSA) according to Border's method and was used to observe the pharmacological action of Shenfukang. The results showed that Shenfukang can eliminate urine protein, increase the levels of serum protein, albumin (A), globulin (G) and A/G ratio, and decrease the contents of serum cholesterol, creatinine and urea nitrogen. It can also counteract the increase of plasma TXB 2 and the decrease of plasma 6-keto-PGF 1? . LD 50 of Shenfukang for mice is 174.34?36.74?g/kg which is much larger than that of Tripterygium wilfordii (29.73?6.46?g/kg for female mice and 101.74?10.93?g/kg for male mice). It is indicated that Shenfukang exerts a protective effect on renal function and reduces toxicity of Tripterygium wilfordii.
6.Therapeutic Effect of Kummerowia Striata on IgA Nephropathy Rats

Objective To examine the therapeutic effect of Kummerowia striata (KS) on IgA nephropathy (IgAN) rats.Methods Sixty SD rats were equally randomized into 6 groups after adaptive feeding for 3 days:blank control group,model control group,tripterygium glycosides (TG) group (in the dose of 0.0165 g/kg),and high-,middle-and low-dose KS groups (in the dose of 16.0,8.0,4.0 g/kg respectively).IgAN rat model was established with bovine serum albumin (BSA),lipopolysaccharide (LPS) and carbon tetrachloride.After modeling,rats except those in blank control group received gastric gavage of corresponding drugs for 10 continuous weeks.Levels of 24-hour proteinuria and hematuria in each group were measured,serum levels of total protein (TP),serum albumin (ALB),aspartate aminotransferase (AST),and alanine aminotransferase (ALT) were tested.The pathological changes of renal tissue were observed under light microscope by HE staining.Results Water decoction of KS could significantly decrease the proteinuria and hematuria levels,and reduce renal pathological changes of IgAN model rats.However,water decoction had no effect on indicators of liver function such as TP,ALB,AST,and ALT in IgAN rats.Conclusion KS has certain therapeutic effect on IgAN model rats.
7.Effect of Ganoderma Lucidum Brewage in Decreasing Blood Lipid

[Objective] To observe the pharmacological action of Ganoderma Lucidum Brewage (GLB) in decreasing blood lipid. [ Methods ] Eighty rats were randimized into 8 groups: blank control (Group A), model ( Group B) high-dose GLB ( Group C), moderate-dose GLB ( Group D), low-dose GLB ( Group E), Ganoderma Lucidum Decoction (Group F), Ganoderma Lucidum Capsule (Group G) and Xuezhi Kang (Group H). So did the mice. Hyperlipidemia mouse models and rat models were induced by yolk and high-fat diet respectively. Effect of GBL in various doses to decrease blood lipid was observed in the two kinds of hyperlipidemia models and was compared with that of Xuezhi Kang, Ganoderma Lucidum Decoction and Ganoderma Lucidum Capsule. Objective To observe the effects of cyclovirobuxinum D(Cvb-D)on the myocardium cell apoptosis of myocardial ischemia rats.Methods The rats were given Cvb-D 0.55,1.1 and 2.2 mg/kg orally per day for 21 days respectively.The myocardial ischemia model was induced by isoprenaline intraperitoneal injection.Myocardium tissue was observed under light microscope and end labelling TUNEL method was used to determine the cell apoptosis.Then the average percentage of apoptosis cell was figured out to evaluate the effect of Cyb-D.Results Cvb-D obviously lightened isoprenaline-induced myocardial pathological changes such as turbulence of myocardial fiber,plasmolysis of necrotic cell,inflammatory cell infiltration of necrotic interstitial region.Cvb-D remarkably inhibited myocardium cell apoptosis of myocardial ischemia rats caused by isoprenaline.Conclusion Cvb-D has the action of ameliorating myocardium cell apoptosis caused by myocardial ischemia.
9.A Study on the Sedative and Hypnotic Effects of Ganoderma lucidum Brewage

Objective To investigate the sedative and hypnotic effects of Ganoderma lucidum brewage(GLB).Methods The NIH mice were given with gavage of GLB 0.06,0.12,and 0.24g/kg per day respectively for 15 days.The spontaneous motion,the sleeping time induced by pentobarbital sodium in subthreshold dosage and threshold dosage,and the number of mice subjected to pentylenetetrazol-induced convulsion were observed.Results GLB can reduce the spontaneous motion,significantly longthen the mice sleeping time of pentobarbital sodium in threshold dosage and increase the number of sleeping mice under subthreshold dosage of pentobarbital sodium.GLB was synergic with pentobarbital sodium,and antagonized pentylenetetrazol-induced convulsion in a dose-dependent manner.Conclusion GLB possesses obvious sedative,hypnotic and anticonvulsive effects.
10.Study on Purification of Sini Decoction with Macroporous Resin D_(101)

Objective: To study the purification of Sini Decoction (SND) with macroporous resin D 101 . Methods: SND was isolated and purified by macroporous resin D 101 . The purification of SND was evaluated by chemical and pharmacological methods. Results: The thin layer chromatograms, high performance liquid chromatograms of the SND purification and those of SND were almost the same; also the SND purification can significantly promote the content of nitric oxide (NO) in serum of mice treated by Metaraminol. Conclusion: The SND purification can keep the active ingredients of SND.
