1.Flos Sophorae improves psoriasis in mice by inhibiting the PI3K/AKT pathway.

OBJECTIVES:
To explore the therapeutic mechanism of Flos Sophorae (FS) for treatment of psoriasis.
METHODS:
The active ingredients, targets and psoriasis-related disease targets of FS were obtained from TCMSP, GeneCards, OMIM, DisGeNET and String databases, and Cytoscape 3.8.0 software was used to construct the "FS -active ingredient-key target-signaling pathway-psoriasis" network. GO and KEGG enrichment analyses of the key targets were conducted, and molecular docking was performed using Discovery Studio 2019. In a BALB/c mouse model of imiquimod-induced psoriasis, the effects of vaseline, FS at high, medium and low doses (3.00, 1.50 and 0.75 g/kg, respectively) and a positive drug, given 1 week before and during modeling, were evaluated on body weight changes, spleen coefficient, psoriasis area and severity index (PASI) score and skin pathological changes. Phosphorylation levels of PI3K and AKT proteins were detected using immunohistochemistry and Western blotting.
RESULTS:
A total of 10 active components and 110 key targets were screened. GO and KEGG pathway enrichment analysis suggested that FS improved psoriasis primarily through the PI3K/AKT, TNF, and IL-17 signaling pathways. Molecular docking showed that both quercetin and kaempferol could spontaneously bind to AKT1, TNF and other sites. In the mouse model of psoriasis, treatment with low-dose FS significantly improved epidermal thickening, increased body weight, lowered PASI score, and reduced phosphorylation levels of PI3K and AKT proteins.
CONCLUSIONS
The therapeutic mechanism of FS for psoriasis involves multiple components, targets, and pathways that mediate the inhibition of the phosphorylation levels of PI3K and AKT proteins to suppress the activation of the PI3K/AKT signaling pathway.

2.Impact of ambient air pollution on hospital visits for mental and behavioral disorders among residents in an industrial area in Henan Province from 2016 to 2021

Objective:To explore the impact of air pollution on hospital visits for mental and behavioral disorders among residents in an industrial area in Henan Province from 2016 to 2021.Methods:Daily outpatient visits data for mental and behavioral disorders were collected from Angang General Hospital in Angang Industrial Area at Anyang City between January 2016 and December 2021. And air pollutants and meteorological data during the same period were also collected. A generalized additive model was used for time-series analysis to examine the relationship between daily average concentrations of nitrogen dioxide (NO 2), sulfur dioxide (SO 2), fine particulate matter (PM 2.5), inhalable particulate matter (PM 10), carbon monoxide (CO), and ozone (O 3) with a lag of 0 to 7 days on the number of visits for mental and behavioral disorders among residents. The single-day lag effect (lag0-lag7 d) and cumulative lag effect (lag01-lag07 d) were analyzed. The smooth cubic spline function was used to fit the exposure-response relationship, and subgroup analysis was performed according to different genders, seasons and ages. Results:A total of 26 268 hospital visits for mental and behavioral disorders were collected from the industrial area between 2016 and 2021. The daily average concentrations of SO 2, NO 2, PM 2.5, PM 10, and CO were (27.50±27.33), (43.11±18.33), (73.87±60.30), (134.01±83.81) μg/m 3, and (1.72±1.03) mg/m 3, respectively. The daily maximum 8-hour average concentration of O 3 was (82.18±53.70) μg/m 3. After controlling for long-term trends, temperature, relative humidity, day of the week effects, and holiday effects, the generalized additive model analysis showed that NO 2 had a statistically significant impact on the hospital visits for mental and behavioral disorders at lag0 d, lag2 d and lag01-lag05 d and CO had a statistically significant impact at lag0-lag3 d and lag01-lag06 d (all P<0.05). NO 2 at lag02-lag04 d and CO at lag0-lag2 d and lag01-lag04 d had statistically significant effects on the visits for neurasthenia (both P<0.05). The impacts of NO 2 at lag03-lag04 d, PM 2.5 at lag3 d and lag03-lag04 d, PM 10 at lag3 d and lag03 d, and CO at lag3 d and lag01-lag05 d on visits for generalized anxiety disorder were also statistically significant (all P<0.05). After false discovery rate (FDR) correction, it was shown that for every 10 μg/m 3 increase in NO 2 and every 0.1 mg/m 3 increase in CO, the percentage increase in visits for mental and behavioral disorders and its 95% confidence interval (95% CI) were 3.38% (0.95%-5.87%) and 0.78% (0.38%-1.17%), respectively. For every 0.1 mg/m 3 increase in CO, the visits for neurasthenia increased by 0.78% (0.27%-1.29%). For every 10 μg/m 3 increase in PM 2.5 and every 0.1 mg/m 3 increase in CO, the visits for generalized anxiety disorder increased by 1.07% (0.46%-1.68%) and 1.17% (0.37%-1.97%), respectively (adjusted P<0.05). There was a linear exposure-response relationship between NO 2 and CO and the hospital visits for mental and behavioral disorders, CO and the hospital visits for neurasthenia, and CO and PM 2.5 and the hospital visits for generalized anxiety disorder ( P<0.05 for the overall association test and P>0.05 for the non-linearity test). Stratified analysis showed that air pollutants had an impact on male patients with neurasthenia, female patients with generalized anxiety disorder, individuals aged <45 years with mental and behavioral disorders, and individuals aged ≥65 years with generalized anxiety disorder. The impact of air pollutants was greater during the cold season or winter. Conclusion:Exposure to air pollution can increase hospital visits for mental and behavioral disorders among residents in industrial areas, with a higher risk among those aged<45 years old and during the cold season.

4.Diagnostic value of miRNA-133b and miRNA-150 for non-small cell lung cancer

Objective:To investigate the expressions of miRNA-133b (miR-133b) and miRNA-150 (miR-150) in non-small cell lung cancer (NSCLC) and their clinical significances.Methods:Thirty patients with NSCLC who underwent surgery and were pathologically diagnosed in the First People's Hospital of Lianyungang from June 2018 to June 2019 were selected. Before surgery, 10 ml fasting peripheral blood of patients was collected in the morning. The fresh cancer tissues, adjacent tissues (distance from cancer tissues ≤ 3 cm), normal lung tissues (distance from cancer tissues > 5 cm) were collected after surgery. Thirty people who had a physical examination during the same period were served as the healthy control group, and 10 ml fasting peripheral blood was collected in the morning. Real-time fluorescence quantitative polymerase chain reaction (qRT-PCR) was used to detect the relative expressions of miR-133b and miR-150 in the serum of the two groups of subjects and the tissues of NSCLC patients. The relationships between serum miR-133b and miR-150 levels and the clinical features in NSCLC patients were analyzed. The pathological diagnosis was used as the gold standard, and the receiver operating characteristic (ROC) curve was used to determine the efficacy of miR-133b and miR-150 in diagnosing NSCLC.Results:In NSCLC patients, the relative expressions of miR-133b in cancer tissues, adjacent tissues and normal lung tissues were 0.55±0.17, 0.68±0.11 and 0.69±0.12, and the expression of miR-133b in cancer tissues was lower than that in adjacent tissues and normal lung tissues (both P < 0.01). The relative expressions of miR-150 in cancer tissues, adjacent tissues and normal lung tissues were 1.19±0.14, 1.13±0.13 and 1.09±0.12, and there was no significant difference in the expression of miR-150 between cancer tissues and adjacent tissues ( t = 1.98, P = 0.051), and the expression of miR-150 in cancer tissues was higher than that in normal lung tissues ( t = 3.06, P = 0.003). The relative expression of plasma miR-133b in NSCLC patients was lower than that in healthy controls (0.43±0.11 vs. 0.55±0.16, t = 3.68, P<0.05); the relative expression of plasma miR-150 was higher than that in healthy controls (1.14±0.13 vs. 1.04±0.12, t = -3.18, P < 0.05). The expression level of miR-133b in the serum of NSCLC patients was related to lymph node metastasis ( P = 0.003), but not related to the patients' gender, age, pathological type, and TNM stage (all P > 0.05). The expression level of miR-150 in serum of NSCLC patients was not related to the clinicopathological characteristics (all P > 0.05). According to the ROC curve, the area under the curve of miR-133b and miR-150 in serum of NSCLC patients were 0.732 and 0.726, and the area under the curve of the combined detection of the two was 0.811. Conclusions:The expression levels of miR-133b in cancer tissues and serum of NSCLC patients decrease, and the expression levels of miR-150 increase. The expression trends of the two in cancer tissues and serum of NSCLC patients are consistent. miR-133b may be related to the malignant degree, invasion and metastasis of NSCLC. miR-133b and miR-150 may become molecular markers for the diagnosis of NSCLC, and the combined detection of the two has higher diagnostic efficiency.
5.Expressions of cyclin D2 and bcl-2 in diffuse large B-cell lymphoma and their clinical significances

Objective:To investigate the expressions and significance of cyclin D2 and bcl-2 in diffuse large B-cell lymphoma (DLBCL) and their clinical significances.Methods:The tissues of 87 DLBCL patients undergoing resection and 23 patients with lymphoid tissue reactive hyperplasia (RLH) in the First Central Hospital of Baoding from January 2015 to March 2020 were collected. Immunohistochemistry method was used to detect the expressions of cyclin D2 and bcl-2 in tissues of DLBCL and RLH, and the relationship between the expressions of cyclin D2 and bcl-2 as well as the association with the clinicopathological features of DLBCL patients.Results:The positive rates of cyclin D2 protein in DLBCL and RLH were 33.3% (29/87) and 2.0% (1/23), the positive rates of bcl-2 protein in DLBCL and RLH were 60.9% (54/87) and 7.0% (3/23); the positive rates of cyclin D2 and bcl-2 in DLBCL were higher than those in RLH, and the differences were statistically significant( χ2=7.566, P=0.006; χ2=17.512, P < 0.01). The expressions of cyclin D2 and bcl-2 proteins were related to the Ann Arbor staging and immunophenotype of DLBCL patients (all P < 0.05), while not related to age, gender, cancer location, tissue type (all P > 0.05). There was a positive correlation between the expressions of cyclin D2 and bcl-2 protein in DLBCL ( r=1.000, P < 0.01). Conclusions:cyclin D2 and bcl-2 may be related to the development and progression of DLBCL, and both may have some synergies.

7.Expressions of HIF-1α and CA9 in breast carcinoma and their relationships with basal-like subtype

Objective To study the expressions of hypoxia inducible factor-1 α (HIF-1 α) and carbonic anhydrase 9 (CA9) in different molecular subtypes of breast carcinoma and their relationships with basallike subtype.Methods The expressions of HIF-1α and CA9 were detected by immunohistochemistry in five subtypes of breast carcinoma (n =803).The relationships of the expressions of HIF-1α and CA9 with the clinicopathologic parameters of basal-like subtype and with the prognosis of patients were analyzed.Results The positive expression rates of HIF-1α and CA9 protein in breast carcinoma were much higher than those in paratumor breast tissues (43.96% vs.3.86%,x2 =354.858,P < 0.001;19.80% vs.4.61%,x2 =86.495,P < 0.001).The positive expression rates of HIF-1α in Luminal A,Luminal B,epidermal growth factor receptor-2 (HER-2) over-expression,basal-like and normal breast-like subtypes were 31.97%,25.00%,56.06%,77.89% and 55.76% respectively,and the positive expression rates of CA9 were 7.48%,5.56%,60.61%,40.00% and 27.88% respectively,with significant differences (x2 =88.628,P < 0.001;x2 =147.128,P <0.001).The positive expression rates of HIF-1α and CA9 in estrogen receptor (ER)-negative breast carcinomas(HER-2 over-expression,basal-like and normal breast-like) were significantly higher than those in ER-positive subtypes (Luminal A and Luminal B,all P < 0.005).There was a significant positive correlation between HIF-1 α and CA9 (C =0.315,P =0.001),and both were correlated with the lymph node metastasis (x2 =4.542,P =0.033;x2 =7.025,P =0.008),pTNM stage (x2 =5.390,P =0.020;x2 =10.179,P =0.001) and poor prognosis (x2 =4.353,P =0.037;x2 =6.056,P =0.014) of basal-like subtype.Conclusion Over-expressions of HIF-1α and CA9 may be related to the ER-negative breast cancer more closely,and both may play important roles in the occurrence and development of basal-like subtype.Overexpressions of HIF-1α and CA9 may suggest the poorer prognosis of patients with basal-like breast carcinoma.
8.Expression of HIF-1αand Glut-1 in molecular subtypes of breast carcinoma and their rela-tionship with basal-like breast carcinoma

Purpose To study the expression of HIF-1αand Glut-1 in the molecular subtypes of breast carcinoma and their correlation with basal-like breast carcinoma. Methods 803 cases of invasive breast carcinoma from our database were identified. The clinicopath-ologic findings and the biologic markers including estrogen receptor ( ER) , progesterone receptor ( PR) , and human epidermal growth factor receptor-2 (HER-2) status were reviewed. Immunohistochemical MaxVision stains for cytokeratin 5/6 (CK5/6) and epidermal growth factor receptor ( EGFR) were performed. All breast carcinomas were subclassified into Luminal A, Lumincal B, HER-2 over-expression, normal-like, and basal-like subtypes according to Nielsen criteria. Immunohistochemical stain was also used to detect the expression of HIF-1αand Glut-1. Results Positive expression rates of HIF-1αprotein in basal-like, HER-2 over-expression, normal-like, Luminal A and Luminal B substypes were 77. 89% (74/95), 56. 06% (37/66), 55. 76% (92/165), 31. 97% (141/441), 25. 00% (9/36), respectively. The positive expression rates of Glut-1 protein were 80. 00% (76/95), 57. 58% (38/66), 58. 18%(96/165), 34. 01% (150/441), 25. 00% (9/36), respectively. The positive expression rates of HIF-1α and Glut-1 in the basal-like, HER-2 over-expressing and normal-like subtypes were remarkably higher than that in Luminal A and Luminal B subtypes ( P<0. 004 5) and the expression of HIF-1a and Glut-1 was negatively correlated with the expression of ER (P<0. 01). In the ER-negative breast cancers, the positive expression rates of HIF-1a and Glut-1 in basal-like substype were much higher than that in the other sub-types (P<0. 004 5), and the expression of HIF-1α was positively correlated with expression of Glut-1 in basal-like breast carcinoma (P<0. 01). Conclusion The overexpression of HIF-1αand Glut-1 may be closely related to the ER-negative breast cancer and HIF-1α and Glut-1 might play an important role in the development of basal-like breast carcinoma.
9.Expression of S100A7 in basal-like breast carcinoma and its clinical significance

Objective To detect the protein expressions of S100A7 and CD34 in basal-like breast carcinoma and analyze the clinical relationship between S100A7 and basal-like breast cancer and the connection with CD34 markers microvessel denisity (MVD).To study the function of S100A7 in basal-like breast cancer angiogenesis,invasion and metastasis.Methods Immunohistochemistry (IHC) was used to detect the expressions of S100A7 and CD34 in 60 cases of basal-like breast cancer,40 cases of non basal-like breast cancer and 20 cases of normal breast tissues.Results The expressions of S100A7 in basal-like breast cancer,non basallike breast cancer and normal breast tissues were 83.33%,62.50% and 35.00% respectively.There were significant differences among them (x2 =5.556,P < 0.051; x2 =17.107,P < 0.05; x2 =4.051,P < 0.05).There was positive correlation between S100A7 and lymph node metastasis,TNM staging and MVD of basal-like breast cancer patients.But there was no significant correlation between S100A7 and patient's age and tumor sizes.Conclusion The up-regulation of S100A7 expressions may be associated with the occurrence and development,invasion and metastasis,angiogenesis of basal-like breast cancer.The effects of S100A7 promoting angiogenesis may be an important factor in the acceleration of the local invasion and distant metastasis in basallike breast cancer.
10.Klippel-Trenaunay syndrome associated with bladder hemangioma: a case report and literature analysis

ObjectiveTo discuss the treatment of a rare case of Klippel-Trenaunay syndrome associated with bladder hemangioma and conduct a brief literature review. MethodsA 23-year-old female manifested episodes of painless hematuria for 1 month.A biopsy by cystoscopy diagnosed angiomatosis.The patient had a history of left lower limb hemangioma for 23 years,and underwent 3 surgical operations at 3 months old,3 years old,and 13 years old.The left leg was longer than the right leg.There was some soft tissue hypertrophy in the left lower extremity.Ultrasonography showed a bladder mass.Cystoscopy revealed a 3 cm x 5 cm sized hemangiomatous on the left lateral and anterior wall of the bladder. ResultsThe patient was diagnosed with bladder hemangioma and Klippel-Trenaunay syndrome.Partial cystectomy was performed.The pathological diagnosis was bladder intramuscular hemangioma. ConclusionKlippel-Trenaunay syndrome associated with bladder hemangioma can be treated by partial cystectomy.
