1.Analysis of Influencing factors on the therapeutic effect of endoscopic retrograde cholangiopancreatography stent implantation for Malignant obstructive jaundice

Objective To investigate the influencing factors of ERCP stent implantation for patients with malignant obstructive jaundice.Methods 130 patients with malignant obstructive jaundice who received ERCP stent implantation in our hospital from January 2017 to January 2024 were retrospectively included,and grouped according to the jaundice control effect 4 weeks after surgery.Univariate and multivariate analysis of the efficacy of ERCP stent placement in malignant obstructive jaundice after 4 weeks.Construction of a predictive model for the efficacy of ERCP stent placement in malignant obstructive jaundice after 4 weeks and clinical efficacy analysis.Results There were 64 cases for jaundice resolved in 4 weeks after surgery among all 130 patients with the regression rate for 49.23%.The results of univariate analysis showed that the type of biliary obstruction,stent type,and preoperative Child Pugh grading may all be related to the efficacy of ERCP stent placement in malignant obstructive jaundice after 4 weeks(P<0.05).The results of Logistic multivariate analysis showed that the type of biliary obstruction,stent type,and preoperative Child Pugh grade were all independent influencing factors on the efficacy of ERCP stent placement for malignant obstructive jaundice at 4 weeks(P<0.05).Using the independent influencing factors and P-value prediction probability of Logistic regression model to predict the prognosis of patients,the ROC curve was used,with areas under the curve of 0.713,0.823,0.907,and 0.971,respectively.Conclusion The clinical effects of ERCP stent implantation in malignant obstructive jaundice was closely related to the type of biliary obstruction,stent type and preoperative Child-Pugh grade.The data model constructed using the above three factors has shown good performance in predicting the prognosis of patients.

3.The application of whole genome amplification in DNA typing of trace samples

Objective To explore the effectiveness of whole genome amplification technology in DNA typing of trace samples.Methods Simulated trace samples which contain 1~20 cells were prepared by micromanipulation.Whole genome amplification was added before conventional PCR-STR typing step,to compare the effectiveness of PEP and MDA in DNA typing of trace samples from four aspects i.e.allele imbalance,allele drop-out,locus drop-out and pseudo allele (which contains the stutter peak).Results Amplification efficiency of MDA was higher than PEP method,but allele drop-out and pseudo allele were more frequently detected.Correct DNA typing rate of PEP is higher than MDA method,however,advantaged amplification of small fiagments DNA is more obvious.Conclusion MDA method is not suitable for the current STR typing.When the absolute amount of trace samples is quite small,we couldconsider using the PEP method to enhancethe sample quantity to meet the requirement of repeat testing.At the same time it could encounterthe failure of the large DNA fragments.
4.Rosiglitazone improves learning and memory impairment of 3 x Tg mice.

This study is to investigate the protective effect of rosiglitazone (RSG) against learning and memory impairment of APP/PS1/tau transgenic mice. AD mice model was replicated by using 6-month APP/PS1/tau transgenic mice. The learning and memory ability of mice was evaluated by Morris water maze and Western blotting assays was applied to measure the phosphorylation and O-glycosylation of Tau and neurofilaments (NFs) protein. The results demonstrated that RSG could reverse the learning and memory deficits of 3 x Tg mice significantly. It was also found that RSG could suppress the hyperphosphorylation of Tau and NFs protein levels and increase the glycosylation expression of Tau and NFs proteins in 3 x Tg mice brain. Together, RSG ameliorates cognitive impairments of 3 x Tg mice via the alleviation of the hyperphosphorylated Tau and NFs proteins burden in the brain.
