1.Illness experience in patients with hepatitis B: a Meta-synthesis

Objective:To systematically evaluate the illness experience of patients with hepatitis B.Methods:A computer-based search was conducted in PubMed, Embase, Web of Science, Cochrane Library, Medline, China National Knowledge Infrastructure, Wanfang Data, VIP, and China Biology Medicine disc for qualitative studies related to the illness experience of patients with hepatitis B, from database inception to April 9, 2024. The Joanna Briggs Institute Critical Appraisal Checklist for Qualitative Research was used to assess the methodological quality of the included studies. A Meta-synthesis method was adopted to summarize and integrate the results.Results:A total of 10 papers were included, from which 43 themes were extracted and summarized into nine categories, ultimately forming four integrated findings: emotional challenges and psychological struggles; changes in social adaptation after experiencing disease trauma, with different coping styles; loss of self-actualization, accompanied by a strong desire for rehabilitation information and social support; a certain degree of illness-related stigma existed among patients.Conclusions:Medical staff should pay attention to the illness experiences of patients with hepatitis B, guide them toward positive coping mechanisms, and provide a basis for developing personalized nursing strategies.

3.Comprehensive quality evaluation of Tianma jiannao granules

OBJECTIVE To establish the fingerprints of Tianma jiannao granules (TJG) and the method for content determination to evaluate the quality of TJG comprehensively combined with chemometric analysis. METHODS High-performance liquid chromatography (HPLC) was used to establish the fingerprints of 13 batches (S1-S3) of TJG and determine the contents of inosine, gastrodin, parishin B and parishin E. Cluster analysis, principal component analysis, and orthogonal partial least squares- discriminant analysis were performed using SPSS 20.0 and SIMCA 18 software; using variable importance projection (VIP) value greater than 1 as a criterion, marker components that affected quality were screened. RESULTS A total of 28 common peaks were identified in the 13 batches of TJG with similarities greater than 0.9, and 7 common peaks were identified, which were gastrodin, p-hydroxybenzyl alcohol, parishin B, parishin E, rhynchophylline, inosine and salidroside. The 13 batches of TJG were clustered into 3 categories, S1-S2, S8-S10 and S12 were clustered into one category; S3 and S7 were clustered into one category; S4-S6, S11 and S13 were clustered into one category. VIP of inosine was greater than 1. The contents of inosine, gastrodin, parishin B and parishin E were 62.637-176.677, 17.821-37.642, 5.748-16.077 and 5.660-13.510 μg/g. CONCLUSIONS The established HPLC fingerprints and content determination method are stable, reliable and highly reproducible, which can be used to evaluate the quality of TJG in combination with chemometric analysis. Inosine may be a marker component that affects the quality of TJG. There are differences in the quality of 13 batches of TJG.
4.Analysis of mini-CEX Scores and influencing factors after teaching"fundamentals of nursing"in the elderly service management program

Objective To investigate the current status of mini-CEX scores among students in the Elderly Service Man-agement program after completing the"Fundamentals of Nursing"course and analyze the influencing factors.Methods A total of 99 students from the Elderly Service Management program at the Wuming campus of Guangxi Medical University were selected as the study subjects.Assessment tools included a general information questionnaire,the Chinese version of the Mini Clinical E-valuation Exercise(mini-CEX),the Self-Rating Scale of Self-Directed Learning(SRSSDL),and a self-learning ability assess-ment scale.Stepwise linear regression analysis was employed to explore the factors affecting mini-CEX scores.Results The total mini-CEX score for the 99 students was 49.00(44.00,55.00).Stepwise linear regression analysis revealed that being a student leader,SRSSDL scores,self-learning ability,and teaching model were significant factors(P<0.05),explaining 56.8%of the total variance.Conclusion The clinical comprehensive ability of students in the Elderly Service Management program requires enhancement,influenced by multiple factors including teaching model,self-learning ability,and self-directed learning capacity.
5.Development and Application Analysis of Administration Standards on Drug Electronic Certificate

This article introduced the policy background and the development of administration standards on drug electronic certificate,analyzed the current status and main problems of electronic certificate standards for drug regulation,elaborated the process of formulating standards,and explained the main contents of the standards.Combined with the application status of the standards,this paper offered suggestions to optimize and improve the standards.It provided guidance for local drug administration agencies to implement electronic certificates,and offered a foundation for the interoperability and mutual recognition of drug administration electronic certificates throughout the country.
6.Effects of host circular RNA circCAMSAP2 on proliferation of porcine deltacoro-navirus

The purpose of this study is to investigate the impact of host circular RNA(circRNA)on the proliferation of porcine deltacoronavirus(PDCoV).Among the significantly downregulated cir-cRNAs identified during PDCoV infection from circRNA expression profiles in ST cells,circCAM-SAP2 was selected for further analysis.The gene origin and junction sequence of circCAMSAP2 were validated using the online Ensembl database and Sanger sequencing.The molecular character-istics of circCAMSAP2 were determined through nucleoplasmic separation assays and RNase R tests.Additionally,the differential expression of circCAMSAP2 during PDCoV infection was con-firmed using RT-qPCR.Overexpression and knockdown experiments were conducted to investigate the regulatory role of circCAMSAP2 in PDCoV replication.The results indicated that circCAM-SAP2 is a circRNA derived from exon 2 of the CAMSAP2 gene through reverse splicing.It is re-sistant to RNase R,has a length of 260 nucleotides,and is predominantly located in the cytoplasm.circCAMSAP2 expression is downregulated during PDCoV replication.Overexpression of circ-CAMSAP2 inhibited PDCoV proliferation,while knockdown promoted it in ST cells.These find-ings suggest that porcine circCAMSAP2 acts as a negative regulator of PDCoV replication and could serve as a potential target for the development of anti-PDCoV therapeutics.
7.Qualitative study on residents' cognition of commercial long term insurance in Tianjin

Objective:To understand the residents' cognition of commercial long-term insurance in Tianjin so as to provide a reference for improving service effect and quality of long-term insurance.Methods:A total of 31 residents from six districts of Tianjin were selected for semi-structured interviews from December 2022 to February 2023 by the convenient sampling method, and the data were analyzed by Colaizzi 7-step analysis method.Results:Three themes were extracted, namely, inadequate interpretation and promotion of commercial long-term insurance, immature development of commercial long-term insurance, and the ability of commercial long-term insurance to help caregivers reduce their family burden.Conclusions:The residents of Tianjin have a low awareness of commercial long-term insurance, which cannot meet the needs of most residents. It is urgent to strengthen their publicity further and improve their system.

9.Inhibition of caspase-1-dependent apoptosis suppresses peste des petits ruminants virus replication

Background:
Peste des petits ruminants (PPR), caused by the PPR virus (PPRV), is an acute and fatal contagious disease that mainly infects goats, sheep, and other artiodactyls.Peripheral blood mononuclear cells (PBMCs) are considered the primary innate immune cells.
Objectives:
PBMCs derived from goats were infected with PPRV and analyzed to detect the relationship between PPRV replication and apoptosis or the inflammatory response.
Methods:
Quantitative real-time polymerase chain reaction was used to identify PPRV replication and cytokines expression. Flow cytometry was conducted to detect apoptosis and the differentiation of CD4+ and CD8+T cells after PPRV infection.
Results:
PPRV stimulated the differentiation of CD4+ and CD8+ T cells. In addition, PPRV induced apoptosis in goat PBMCs. Furthermore, apoptosis and the inflammatory response induced by PPRV could be suppressed by Z-VAD-FMK and Z-YVAD-FMK, respectively.Moreover, the virus titer of PPRV was attenuated by inhibiting caspase-1-dependent apoptosis and inflammation.
Conclusions
This study showed that apoptosis and the inflammatory response play an essential role in PPR viral replication in vitro, providing a new mechanism related to the cell host response.
10.Selection and identification of singledomain antibody against Peste des Petits Ruminants virus

Background:
Peste des petits ruminants (PPR) is an infectious disease caused by the peste des petits ruminants virus (PPRV) that mainly produces respiratory symptoms in affected animals, resulting in great losses in the world's agriculture industry every year. Singledomain variable heavy chain (VHH) antibody fragments, also referred to as nanobodies, have high expression yields and other advantages including ease of purification and high solubility.
Objectives:
The purpose of this study is to obtain a single-domain antibody with good reactivity and high specificity against PPRV.
Methods:
A VHH cDNA library was established by immunizing camels with PPRV vaccine, and the capacity and diversity of the library were examined. Four PPRV VHHs were selected, and the biological activity and antigen-binding capacity of the four VHHs were identified by western blot, indirect immunofluorescence, and enzyme-linked immunosorbent assay (ELISA) analyses. ELISA was used to identify whether the four VHHs were specific for PPRV, and VHH neutralization tests were carried out. ELISA and western blot analyses were used to identify which PPRV protein was targeted by VHH2.
Results:
The PPRV cDNA library was constructed successfully. The library capacity was greater than 2.0 × 106 cfu/mL, and the inserted fragment size was approximately 400 bp to 2000 bp. The average length of the cDNA library fragment was about 1000 bp, and the recombination rate was approximately 100%. Four single-domain antibody sequences were selected, and proteins expressed in the supernatant were obtained. The four VHHs were shown to have biological activity, close affinity to PPRV, and no cross-reaction with common sheep diseases. All four VHHs had neutralization activity, and VHH2 was specific to the PPRV M protein.
Conclusions
The results of this preliminary research of PPRV VHHs showed that four screened VHH antibodies could be useful in future applications. This study provided new materials for inclusion in PPRV research.
