Objective To analyze changes in sleep, mood state, and brain function in healthy populations living in near-sea-level environments before and after exposure to high-altitude environment, and to explore the correlations between regional brain functional changes and variations in sleep and mood states. Methods A total of 45 healthy volunteers were enrolled. The participants came from regions of near-sea-level altitudes and were exposed to the high-altitude environment for a short period of time. The Pittsburgh Sleep Quality Index(PSQI),Zung Self-Rating Depression Scale(SDS),Patient Health Questionnaire-9(PHQ-9),Zung Self-Rating Anxiety Scale(SAS),and Generalized Anxiety Disorder-7(GAD-7)were administered to assess sleep quality as well as depressive and anxiety symptoms at 4 time points—prior to high-altitude exposure, immediately after exposure, one month after returning to low-altitude regions, and three months after returning to low-altitude regions.Resting-state functional magnetic resonance imaging(rs-fMRI)data were collected before and after high-altitude exposure, and regional brain functional parameters, including the amplitude of low-frequency fluctuations(ALFF)and functional connectivity strength, were analyzed.Statistical analyses were performed, including a linear mixed-effects model to evaluate longitudinal changes in scale scores,paired-sample t-tests to compare brain function differences before and after exposure, and Pearson correlation analyses to examine the relationship between brain functional changes and alterations in sleep and mood states. Results Compared with the pre-exposure findings, the participants exhibited significantly increased PSQI scores(8.89±4.41 vs.5.08±2.69,P<0.05)and PHQ-9 scores(3.60±4.19 vs.1.54±2.30,P<0.05)immediately after high-altitude exposure. One month after returning to the low-altitude environment, both sleep and depression scores decreased relative to the findings immediately after exposure(PSQI:3.88±2.13 vs.8.89±4.41,P<0.05;PHQ-9:1.50±2.25 vs.3.60±4.19,P<0.05)and showed no statistically significant difference compared with the pre-exposure findings(P>0.05).Three months after returning to near-sea-level environment, sleep, depression, and anxiety scores were all reduced compared with the findings immediately after exposure(PSQI:3.76±2.31 vs.8.89±4.41,P<0.05;PHQ-9:1.24±2.13 vs.3.60±4.19,P<0.05;SAS:23.84±5.93 vs.27.93±7.05,P<0.05),also showing no significant difference compared with the pre-exposure levels(P>0.05).Brain function analysis revealed that, relative to the pre-exposure levels,ALFF in the bilateral superior temporal gyrus, insula, and dorsolateral prefrontal cortex(DLPFC)increased after high-altitude exposure(P<0.05),and that functional connectivity strength in the DLPFC was also elevated(P<0.05).Furthermore,changes in DLPFC functional connectivity strength were positively correlated with changes in sleep and mood scores(P<0.05).Conclusion High-altitude exposure has a significant impact on the sleep, mood states, and brain function of populations from near-sea-level regions, and DLPFC,in particular,is closely associated with changes in sleep and mood states. The findings of this study provide a theoretical basis for health management and intervention strategies in high-altitude environments.

OBJECTIVE To explore the potential mechanism of Yishenqinglihuoxue Formula(YSQLF)in treating chronic kidney disease fibrosis in rats based on UHPLC-Q-TOF-MS technology and network pharmacology.METHODS UHPLC-Q-TOF-MS was used for qualitative analysis of Yishenqinglihuoxue Formula-containing serum.Targets of the plasma constituents and the disease were retrieved from SwissTargetPrediction,OMIM,GeneCards, and other databases. Then the protein-protein interaction(PPI)network was constructed and core targets were screened for GO term enrichment and KEGG pathway enrichment.Cytoscape software was em-ployed to construct the"drug-compound-core target-pathway"network and the targets and signaling pathways of Yishenqinglihuoxue Formula against fibrosis were predicted.A model of renal fibrosis was established to verify the core targets and pathway proteins.RE-SULTS A total of 56 constituents migrating to blood of Yishenqinglihuoxue Formula were identified.97 common targets of the constit-uents and the disease and 33 core targets were screened out.KEGG enrichment and PPI network analysis showed that Yishenqingli-huoxue Formula may play a role in the treatment of fibrosis through PI3K/Akt and other pathways.Furthermore, the results of animal experiments showed that Yishenqinglihuoxue Formula could reduce the levels of Scr and BUN,improve fibrosis areas, inhibit the acti-vation of the PI3K/Akt pathway, and reduce the protein expression of TNF-α.CONCLUSION Yishenqinglihuoxue Formula may play a role in the treatment of fibrosis by inhibiting PI3K/Akt signaling pathway and inhibiting TNF-α expression.

Objective To understand the correlation of caregiver′s burden, psychological state, quality of life, social support and health condition of patients with rheumatism, to provide the basis for improvements in health status and quality of life in patients with rheumatic diseases. Methods Totals of 153 patients and their caregivers were collected form rheumatism outpatient in our hospital, the patients′health condition was assessed by HAQ;Zarit burden scale was used to assess caregivers′burden;CES-D self-report depression scale was used for the assessment of psychological state;patients′health status, quality of life and supportive were assessed by using health assessment questionnaire, social support scale and World Health Organization quality of life assessment. Results Rheumatic patients and caregivers were vulnerable groups and their age gap was large. Caregivers were mainly young and middle aged men. Their relationship were mainly couples or parents and children. Their educational level was low and they usually lived in rural without fixed work. There was a linear relationship between burden and depression degree of caregivers and health status of patients (P<0. 01). There was a linear relationship between caregivers′quality of life and health status of patients (P <0. 05). There was no linear relationship between support of the caregivers and health status of patients (P>0. 05). Conclusions Caregivers have more depression and burden, patients obtain worse health condition. The higher caregivers quality of life, the better health status of patients. It seems no relationship between support of the caregivers and health status of patients.

This article was aimed to study immunomodulatory effect of chemical split fractions ofMori Cortex, in order to initially explain effective parts that played a role in immunomodulatory effect ofMori Cortex. The carbon clearance test, serum hemolysin test, E-rosette test, and lymphocyte transformation test were carried out to explore influence of these chemical split fractions ofMori Cortex on immune organs, nonspecific immunity, humoral immunity and cellular immunity. The results showed that in the carbon clearance test, 50% ethanol fraction markedly reduced the thymus index (P<0.01) and the correction indexα (P<0.05). In hemolysin test, the half value hemolysis (HC50) was improved by 30% ethanol fraction and fatty oil fraction (P<0.05). Besides, in the E-rosette test, the E-rosette ration was increased in the 30% ethanol fraction group (P<0.05). In the lymphocyte transformation test, the 30% ethanol fraction can promote the thymus and spleen lymphocytes proliferation (P<0.05 orP<0.01), while the 50% ethanol fraction inhibited the proliferation (P<0.05 orP<0.01). It was concluded that the 30% ethanol fraction can boost both the humoral immunity and cellular immunity; the 50% ethanol fraction can induce the growth of thymus with a suppressive effect on nonspecific immunity and cellular immunity; the fatty oil fraction can improve humoral immunity.
