This article systematically analyzes the historical evolution of the name, origin, medicinal parts, processing and other aspects of Euphorbiae Pekinensis Radix(EPR) by referring to the herbal medicine, medical books, prescription books and other documents of the past dynasties, combined with the relevant modern research materials, so as to provide a basis for the development and utilization of famous classical formulas containing this herbal medicine. According to research, EPR was first recorded in the Shennong Bencaojing in the name of Daji, and it is the correct name of the herbal medicine in all dynasties, there are also other aliases such as Qiongju, Hongya Daji, and Xiamaxian. The dried roots of Euphorbia pekinensis from Euphorbiaceae was the mainstream of the past dynasties. Before the Ming dynasty, the above ground parts of E. pekinensis were used as Zeqi in herbal works. However, since LI Shizhen in the Ming dynasty proposed that the origin of Zeqi should be E. helioscopia, the aerial part of EPR is no longer used as medicine. Since modern times, the roots of Knoxia valerianoides has been used as EPR, and has become the mainstream of commodities, which should be corrected. Throughout history, it has been recorded that the main producing areas were Jiangsu, Anhui, Zhejiang, Shanxi and other regions, while modern botanical survey have shown that EPR is a widespread species distributed throughout the country. In ancient times, the harvesting time of EPR was mostly the twelfth lunar month, while in modern time, it is more common to harvest in autumn and winter. The main processing methods of EPR in ancient times were vinegar processing, wine processing, and stir frying, while in modern times, it is uniformly vinegar processing. In the medicinal properties and clinical aspects, the records are basically consistent throughout history, mainly characterized by bitter taste, cold and toxic nature. Its main efficacy is expelling water retention and reducing swelling. Based on the textual research, it is suggested to choose the dried roots of E. pekinensis when famous classical formulas containing EPR, processing method can be based on the original specified prescription requirements, if the processing method is not clear, it is recommended to use vinegar-processed products as medicine.

Objective:To construct an infectious clone of a Chinese HIV-1 CRF01_AE epidemic strain with strong replication capacity, and comprehensively identify its viral phenotype and replication capacity. Methods:Using the CRF01_AE clinical isolate GX2005002,which was previously isolated from whole blood of an HIV-1-infected individual in China by our laboratory,as the parental strain, the full-length genome of the virus(9.7 kb)was divided into 5' half fragment(5.1 kb)and 3' half fragment(4.6 kb)for amplification. The proviral DNA was used as a template to amplify the virus genome, which was then ligated into the eukaryotic expression vector pEASY-T1. The consistency of its sequence with the parental strain sequence was identified through full-length genome sequencing and phylogenetic analysis. The replication capacity, syncytium inducibility, and cell tropism were experimentally identified to determine its phenotypic consistency with the parental strain. Results:An infectious clone of the CRF01_AE strain was successfully constructed, and its genome sequence exhibited high consistency with the sequence of the parental strain. By transfecting target cells,a derivative virus with infectious activity and replication capability was successfully rescued. The derived virus maintained phenotypic characteristics consistent with the parental strain, such as cell tropism and syncytium inducibility. Conclusion:This study successfully constructed an infectious clone of a Chinese HIV-1 CRF01_AE epidemic strain with clear background and distinct phenotype. The genomic sequence and viral phenotypic characteristics of the derived virus are basically consistent with the parental strain, providing strong representation of the original isolate and serving as a powerful tool for research on the correlation between the genetic characteristics, viral phenotype, and pathogenicity of HIV-1 CRF01_AE strains.

By consulting ancient materia medica, medical books, prescription books and modern literature, this paper systematically combed and reviewed the name, origin, scientific name evolution, producting area, quality evaluation, medicinal parts, harvesting and processing and traditional efficacy of Lasiosphaera Calvatia. The results show that Mabo was first recorded in Mingyi Bielu. Since then, all dynasties have taken Mabo as a legitimate name. Before the Song dynasty, only Calvatia lilacina was used as the original plant of Lasiosphaera Calvatia, which was expanded after the Song dynasty with the appearance of C. gigantea, Lasiosphaera fenzlii, Bovistella radicata and other varieties. Until modern times, there was an addition of Lycoperdon perlatum, L. pyriforme and other original plants of Lasiosphaera Calvatia. Since 1975, the original plant of Lasiosphaera Calvatia in various regulations and academic monographs has been basically uniform for C. lilacina, Lasiosphaera fenzlii and C. gigantea. Resource of the medicinal fungus was widely distributed in China and was mainly wild. From ancient times to the present, the medicinal parts of Lasiosphaera Calvatia are all fruiting body, which is harvested in summer and autumn, and its processing method was to take powder in ancient times, but to cut blocks in modern times. In recent times, its quality has been summarized as large, thin-skinned, intact, full, loose-bubbled and elastic. The medicinal efficacy has been developed from very good for all scores, and after the Ming and Qing dynasties, it is consistent with the 2020 edition of Chinese Pharmacopoeia, with the efficacy of clearing the lung, promoting pharynx, relieving fever and hemostasis, mainly treating cough aphonia, throat obstruction and pharyngeal pain, vomiting blood, epistaxis, hemoptysis, and external treating sores and bleeding from cuts and wounds. Based on the results of herbal textual research, it is suggested that C. lilacina is the first choice for the origin of Lasiosphaera Calvatia involved in famous classical formulas, and it is processed into block or powder for medicine.

By reviewing ancient materia medica, prescription books, medical books and modern literature, this article makes a systematic textual research on the name, origin, commercial specification, producing area, harvesting and processing of Scutellariae Radix used in famous classical formulas according to the historical development, in order to provide a reference for the development and utilization of famous classical formulas. Scutellariae Radix was first named as Qian, Qin and so on, while Huangqin was used as the official name in ancient literature, as well as many aliases such as Fuchang, Huangwen and Dufu. The main origin of Scutellariae Radix in the past dynasties was Scutellaria baicalensis, and other Scutellaria plants were also used for Scutellariae Radix at different times. The medicinal parts of Scutellariae Radix in all dynasties are roots, since the Northern and Southern dynasties, it has gradually differentiated into Ziqin and Kuqin with different efficacy according to their different growth stages and characteristics, and continued to this day. At present, most of Scutellariae Radix are Ziqin in the market, due to the influence of cultivation cost, index component content requirements and other factors. The production area of Scutellariae Radix recorded in ancient literature was wide and gradually expanded from the Yangtze River basin to the northward. Since modern times, Chengde city of Hebei province has been respected as a geo-authentic area, now S. baicalensis is cultivated in a wide area, spreading over north and northeast China. Scutellariae Radix was mostly used as raw products in the early period, and a variety of processed products appeared in the Ming dynasty. Development continues to this day, Scutellariae Radix mainly includes raw products and wine-processed products. According to the research conclusion, it is suggested that S. baicalensis should be used as Scutellariae Radix in famous classical formulas, and the selection of its commercial specifications and processed products can be comprehensively determined according to the requirements of original prescription and the clinical effect.

Objective:To screen the HIV standard strains with typical biological characteristics of HIV strains circulating in China through the isolation, culture, genotype and phenotype identification of HIV from the whole blood samples of HIV-infected persons, confirm genetic characteristics, traceability, and in line with the Standard Strains of Pathogenic Microorganism-technical Specifications for Establishment of HIV Strains (T/CPMA 027—2023).Methods:Whole blood samples were collected from 48 HIV infected patients. Peripheral blood mononuclear cells (PBMCs) were isolated from the samples and co-cultured with PBMCs isolated from healthy persons′ whole blood samples to isolate and culture HIV from infected persons. We determined concentration of p24 antigen and the virus titer in the culture supernatant. The viral RNA was extracted from the successfully isolated strains, and the gag, pol genes and env C2V3 fragments of the viral genome were amplified and sequenced. The genotype, gene recombination and drug resistance sites were determined according to the viral gene sequences. Virus infection and replication were monitored by inoculating the virus culture supernatant into Ghost cells expressing CCR5 or CXCR4 to determine the viral tropism. The formation of syncytium was observed by inoculating the virus culture supernatant into MT-2 cells to determine whether was a syncytium-induced phenotype. Results:Fourteen strains with p24 antigen concentration > 1 ng/ml in culture supernatant were isolated and cultured from 48 fresh EDTA anticoagulated whole blood samples of HIV infected persons. Of the 14 strains, only one strain with a titer≥10 5 TCID 50/ml, 8 strains with titers ≥10 4 TCID 50/ml, and the other 5 strains with titers≥10 3 TCID 50/ml. Phylogenetic analysis showed that the genotypes of the strains were 9 strains of subtype B, 3 strains of CRF01_AE and 2 strains of CRF07_BC recombinant. Genotypic resistance analysis showed that 11 strains contained drug resistance sites. Ghost cells were used to verify the tropism of the strains, and it was found that 8 strains were CCR5 tropism, 6 strains were CXCR4 & CCR5 dual tropism. Only 2 of the 14 strains could induce MT-2 cytopathic effect, which was syncytium-inducing phenotype. Conclusions:Fourteen HIV strains with typical biological and genetic characteristics were isolated to screen the standard HIV strains. Among which, 1 strain was evaluated as a standard HIV strain that meets the Standard Strains of Pathogenic Microorganism-technical Specifications for Establishment of HIV Strains (T/CPMA 027—2023). This study can also provide technical guidance for the screening of the HIV standard strains. Next step is to complete the application and reserve database construction according to the sharing mechanism of the HIV standard strains, to provide resources for the researches of HIV vaccines and drugs.

Chinese Journal of Microbiology and Immunology 2022;42(2):81-87
Objective:To analyze the effects of the main drug resistance mutations in the integrase (IN) region on the resistance of HIV-1 CRF01_AE strains, and compare the differences with subtype B strains. Methods:Seven IN region mutations or combined mutations (T66K, F121Y, Q148K, N155H, G118R, R263K, Q148K/N155H) were selected from the HIV drug resistance database of Stanford University in the United States, and introduced to the IN region of HIV-1 B subtype infectious clone pNL4-3 and CRF01_AE infectious clone pGX002 by seamless cloning, homologous recombination and point mutation. The mutant plasmids were transfected into 293T cells for virus packaging. The culture was expanded in MT2 cells and infectious titers were detected. Half maximal inhibitory concentrations (IC 50) of four integrase inhibitors (INSTIs), raltegravir (RAL), elvitegravir (EVG), dolutegravir (DTG) and bictegravir (BIC), against 14 mutant viruses were detected and compared with the IC 50 against the wild-type viruses. Results:B subtype and CRF01_AE plasmids carrying seven IN region mutations or combined mutations were successfully constructed, and 14 recombinant viruses were packaged with an infectious titer of 10 4-10 6 median tissue culture infective dose (TCID 50)/ml. The recombinant viruses replicated efficiently in MT2 cells. The concentrations of HIV-1 p24 antigen contained in the supernatants of cell culture reached 830-2 700 ng/ml. Five mutations or combined mutations (T66K, F121Y, Q148K, N155H, Q148K/N155H) caused CRF01_AE and B subtype strains to be highly resistant to RAL and EVG, resulting in an increase in the IC 50 by 200 times and 2 000 times or more as compared with the IC 50 against the wild-type viruses. The same mutation-caused fold changes of IC 50 of RAL and EVG against CRF01_AE were significantly lower than that of subtype B ( P<0.01). Q148K/N155H mutation caused B subtype and CRF01_AE to be highly resistant to DTG and BIC, with IC 50 increased by more than 50 times. Other mutations had little effects on the sensitivity to DTG and BIC. Conclusions:Fourteen HIV-1 strains carrying seven INSTI resistance mutations based on B subtype and CRF01_AE were constructed. Five mutations resulted in high resistance to RAL and EVG, and there was a high level of cross-resistance. Resistance to RAL and EVG caused by the same mutation was higher in B subtype than in CRF01_AE. The combined mutation of Q148K and N155H was associated with greater resistance to DTG and BIC, indicating that the genetic barrier of DTG and BIC resistance was high. DTG and BIC could effectively inhibit the strains carrying INSTI resistance mutations without obvious subtype difference.
