1.Mining, characterization, and expression of a fructan sucrase for efficient conversion of soybean oligosaccharides.

The high content of sucrose and raffinose reduces the prebiotic value of soybean oligosaccharides. Fructan sucrases can catalyze the conversion of sucrose and raffinose to high-value products such as fructooligosaccharides and melibiose. To obtain a fructan sucrase that can efficiently convert soybean oligosaccharides, we first mined the fructan sucrase gene from microorganisms in the coastal areas of Xisha Islands and Bohai Bay and then characterized the enzymatic and catalytic properties of the enzyme. Finally, recombinant extracellular expression of this gene was carried out in Bacillus subtilis. The results showed that a novel fructan sucrase, BhLS 39, was mined from Bacillus halotolerans. With sucrose and raffinose as substrates, BhLS 39 showed the optimal temperatures of 50 ℃ and 55 ℃, optimal pH 5.5 for both, and Kcat/Km ratio of 3.4 and 6.6 L/(mmol·s), respectively. When 400 g/L raffinose was used as the substrate, the melibiose conversion rate was 84.6% after 30 min treatment with 5 U BhLS 39. Furthermore, BhLS 39 catalyzed the conversion of sucrose to produce levan-type-fructooligosaccharide and levan. Then, the recombinant extracellular expression of BhLS 39 in B. subtilis was achieved. The co-expression of the intracellular chaperone DnaK and the extracellular chaperone PrsA increased the extracellular activity of the recombinant BhLS 39 by 5.2 folds to 17 U/mL compared with that of the control strain. BhLS 39 obtained in this study is conducive to improving the quality and economic benefits of soybean oligosaccharides. At the same time, the strategy used here to enhance the extracellular expression of BhLS 39 will also promote the efficient recombinant expression of other proteins in B. subtilis.

N6-Methyladenosine (m6A) is the most abundant internal modification in eukaryotic mRNA, playing critical role in various bioprocesses. Like other epigenetic modifications, m6A modification can be catalyzed by the methyltransferase complex and erased dynamically to maintain cells homeostasis. Up to now, only two m6A demethylases have been reported, fat mass and obesity-associated protein (FTO) and alkylation protein AlkB homolog 5 (ALKBH5), involving in a wide range of mRNA biological progress, including mRNA shearing, export, metabolism and stability. Furthermore, they participate in many significantly biological signaling pathway, and contribute to the progress and development of cancer along with other diseases. In this review, we focus on the studies about structure, inhibitors development and biological function of FTO and ALKBH5.

Objective To understand and evaluate the knowledge, attitude and practice of female human papillomavirus and HPV vaccine in Chinese mainland. Methods We searched the relevant studies about the knowledge, attitude and behavior factors of HPV infection and HPV vaccine published in PubMed, MEDLINE, CNKI and Wanfang databases about Chinese mainland women from January 1, 1995 to January 31, 2021.Two researchers independently screened the literatures, extracted data and the literatures with quality score≥5 points.Chi square test was used to evaluate its heterogeneity.Begg' s test was used to evaluate publication bias. Results A total of 33 literatures were included, with a total number of 46013 people.The results showed that the total awareness rate of HPV in the population was 65.9%, the lowest rates were 16.0%, 41.2% and 14.4% in Northeast, rural areas and middle school students, respectively; while the highest rates of HPV were 77.4%, 56.3% and 71.0% in Central China, cities and towns and college students, respectively.The main source of HPV knowledge was the network/WeChat official account number (38.9%), and the lowest was family/friend (4.5%).The population's awareness rate of HPV vaccine related knowledge was 41.4%, the highest rates were 51.8% and 69.7% in East China and medical staff, respectively, while the lowest were 23.6% and 12.7% in Southwest and middle school students. Conclusion In order to achieve the total elimination of cervical cancer in China by 2030 as soon as possible, people should strengthen their awareness of HPV and HPV vaccine related knowledge.
5.Determination of lanthanum in whole blood by microwave digestion-inductively coupled plasma optical emission spectrometry

Objective To evaluate the transdermal safety of lanthanum (La) in sunscreen and jellyfish sting protective lotion, establish a microwave digestion-inductively coupled plasma opticalemission spectroscopy (ICP-OES) method for determination of lanthanum (La) in rat’s whole blood. Methods The whole blood samples were digested by microwave and analyzed by inductively coupled plasma emission spectrometer (ICP-OES). Using 333.749 nm as the analysis line, the content of La in rat whole blood was determined. Results The correlation linearity of the standard curve of this method was good (r>0.9994), the detection limit of the method was 0.0025 μg/ml, the limit of quantification was 0.0077 μg/ml, the precision was less than 3%, and the recovery rate was between 94.9% and 102.0%. Conclusion The ICP-OES method based on microwave digestion is stable and reliable, and can provide an important basis for the study of the transdermal safety of lanthanum.
6.Clinical efficacy of magnetic compression anastomosis for congenital esophageal atresia and stenosis

Objective To investigate the clinical efficacy of magnetic compression anastomosis for congenital esophageal atresia and stenosis.Methods The retrospective and descriptive study was conducted.The clinical data of 4 children who underwent magnetic compression anastomosis for congenital esophageal atresia and stenosis in the Northwest Women and Children's Hospital from December 2017 and February 2019 were collected.There were 2 males and 2 females.The children were aged 11 days,7 days,5 days,and 3 years,respectively.The children underwent magnetic compression anastomosis.Observation indicators:(1) surgical and postoperative situations;(2) follow-up.Follow-up using outpatient examination and telephone interview was performed to detect food intake and complications of children up to May 2019.Measurement data with normal distribution were represented as Mean±SD,and measurement data with skewed distribution were represented as M (range).Results (1) Surgical and postoperative situations:four children underwent magnetic compression anastomosis successfully.Of the 4 children,3 with esophageal atresia underwent open tracheoesophageal fistula repair and endoscopeassisted magnetic compression anastomosis,and 1 with congenital esophageal stenosis underwent endoscopic gastrostomy combined with magnetic compression anastomosis.The operation time of 4 children was (2.3±0.9) hours.The length of esophageal blind ending in the 3 children with esophageal atresia and length of esophageal stenosis were in the children with esophageal stenosis 30-35 mm and 8 mm.Four children has good magnet apposition,and time of postoperative magnet removal was (29± 10)days.Three children with esophageal atresia had oral removal of magnet,and 1 with esophageal stenosis had magnet removed by gastrostomy.One child complicated with postoperative fistula and anastomotic stenosis was cured by unobstructed drainage and nutritional support treatment.The duration of postoperative hospital stay was (39± 10)days.(2) Follow-up:4 patients were followed up for 3-17 months,with a median time of 10 months,and restored to oral intake after oral removal of magnet and removal of magnet by gastrostomy on the days 14-36 postoperatively.One child was detected anastomotic stenosis by esophagography at the postoperative 3 months,and was improved after esophageal dilatation.The other 3 children recovered to normal connectivity of esophagus postoperatively and maintain unobstructed.Four children had normal eating,without dysphagia or other serious complications.Conclusion Magnetic compression anastomosis is safe and feasible for congenital esophageal atresia and stenosis,with good short-term efficacy.
7.The new approach in the location of the fetal conus medullaris and its application in tethered cord syndrome

Objective To evaluate the position of the fetal conus medullaris during pregnancy and its value in detecting tethered cord syndrome(TCS). Methods Nine hundred and seventy-four normal fetuses and 46 fetuses with TCS between 15 and 41 weeks gestation were involved in the study.Parameters D 1 (the distance between the end of the conus medullaris and the caudal edge of last vertebral body ossification center) and D2 (the distance from the end of the conus medullaris to the caudal skin namely the intersection point of the extending line of D1 and the skin) were measured in the caudal midsagittal plane of the spine. Sixty normal fetuses were chosed randomly for interobserver variability.Correlation analysis between these two parameters and gestational age(GA) were conducted and the normal reference value of these parameters were calculated in normal group. The ratios of growth parameters ( Biparietal diameter, Head circumference,Abdominal circumference,Femur length) to D1 and D2 were calculated separately to observe the difference of the ratios between two groups. All the parameters and ratios of normal fetuses were compared with that of TCS cases.Results There was no significant difference in D1 and D2 between two observers.A significant linear correlation between the parameters and GA was found in normal group,linear regression equations were D1=0.251 GA -2.265 cm (R2=0.926,P <0.01) and D2=0.267 GA -1.812 cm(R2=0.928,P <0.01),respectively.D1 and D2 were much lower in normal group than in abnormal group (all P <0.01). The ratios of the growth parameters to D1 and D2 were relatively stable and had statistically differences between two groups in different gestational age. Conclusions The methods that determination of D1 and D2 are simple and feasible,and could help to the prenatal diagnosis of TCS.
8.Correlational Study on Traditional Chinese Medicine Syndrome Distribution Characteristics of HIV Infection in Xinjiang Region Based on Information Sharing System

This paper was aimed to study traditional Chinese medicine (TCM) syndrome distribution characteristics of HIV infection in Xinjiang region based on the Clinical Medical Research Information Sharing System (CMRISS).CMRISS was used to establish a database (data were from 1151 hospital electronic medical records from May 2011 to March 2012).Oracle and ETL software were used in the data processing.Visualization analysis was used in the data mining.The results showed that syndrome distribution of HIV infection in Xinjiang region was concentrated in four categories with the blockade of dampness due to qi deficiency,deficiency of both qi and yin,yin deficiency of liver and kidney,and the stagnation of liver qi.The syndrome of blockade of dampness due to qi deficiency was relatively concentrated in the range of 20-30 years old,with the majority of female population.The proportion of sexually transmitted infection was more.The syndrome of deficiency of both qi and yin was relatively concentrated in the age range of 30-40 years old,with the majority of male population.The large proportion of infection was due to intravenous drug use.Among different TCM syndromes,Uyghur population occupied relatively large part.Stratified analysis on disease course due to the change of TCM syndrome according to 1 year,3 years and 5 years revealed the disease development rule from excess syndrome,deficiency combined with excess,to deficiency syndrome.It was concluded that the application of CMRISS was able to process a large amount of clinical data.The data mining results can be used to guide clinical practice.It provided a better platform for the scientific research of TCM clinical practice.

ObjectiveTo evaluate the vitro anti-HIV-1 activity ofTaiqi Peiyuan Granules.MethodsMTT was used to detect cytotoxicity ofTaiqi Peiyuan Granules; cytopathy method was used to detect the inhibitory activity of Taiqi Peiyuan Granules on acute infection of HIV-1; HIV-1 p24 antigen ELISA detection was used to detect inhibitory activity ofTaiqi Peiyuan Granules for virus replication of HIV-1 acute infection cells, and count the medical therapeutic indexes.ResultsCC50ofTaiqi Peiyuan Granules in cytotoxicity test was 3.761±0.370 mg/mL; the EC50 of inhibition syncytial test was 0.454 5±0.204 6 mg/mL; the therapeutic index was between 5.84 and 12.97; p24 ofTaiqi Peiyuan Granules in inhibition experiments was 0.56±0.27 mg/mL, and the therapeutic index was between 5.30 and 8.74.ConclusionAnti-HIV-1 activity ofTaiqi Peiyuan Granules is relatively weak.
10.Influences of self-efficacy intervention on compliance of liver transplantation recipients during hospitalization

Objective To explore the influences of self-efficacy intervention on compliance and self efficacy of liver transplantation recipients during hospitalization.Methods A total of 64 liver transplantation recipients in liver transplantation center of 302 Military Hospital of China from October 1 st 201 2 to September 30th 201 4 were recruited and divided into intervention group (targeted measures to enhance self efficacy)and control group (routine nursing and health education)according to the serial number designed by sequence of operation.General information of patients was collected by transplantation registration system.Besides,self-efficacy level before operation,self-efficacy and compliance level at 1 ,6 months after operation was evaluated by questionnaire.And then,self-efficacy and compliance were compared between two groups.Results There were statistically significant differences in the score of self efficacy of patients between two groups at 1 st and 6th month (P <0.01 ).The scores of self-efficacy of patients in two groups before and after intervention were analyzed with repeated measurement analysis of variance.The results showed that it was an obvious difference between two groups.At the same time,there was a statistical difference between two groups with the time changing (P <0.05).The scores of compliance,self-monitoring nursing and life habits of patients in intervention group at 1 st and 6th month were higher than those in control group with significant differences (P <0.01 );however,there was no significant difference in medication compliance and scores of two sub-scales (P >0.05).Besides,there was no significant difference on whether complications occurred (P >0.05 ).Conclusions Self-efficacy intervention can enhance self-efficacy of liver transplantation recipients after operation,improve patients′ability of self-care,promote their health behavior and improve patients′compliance so as to improve the quality of life of patients after operation.
