随着铜绿假单胞菌（铜绿）的耐药性逐年增强，铜绿感染已经成为公共医疗卫生的重点关注问题。线粒体自噬及其介导的线粒体功能障碍在多种细菌感染中已被报道，但线粒体功能障碍在宿主调控铜绿感染中的作用尚不明确。因此，本研究建立铜绿刺激小鼠巨噬细胞感染模型和小鼠急性铜绿感染模型，探讨铜绿是否通过诱导线粒体自噬改变线粒体功能，进而影响宿主免疫炎症反应和细胞毒性，并通过监测生存率和肺组织病理学变化进一步确定线粒体自噬在小鼠铜绿体内感染模型中的作用。结果表明，铜绿引起小鼠腹腔巨噬细胞线粒体功能障碍，并通过线粒体自噬途径清除铜绿刺激引起的活性氧（ROS）累积，从而抑制铜绿引起的促炎性细胞因子分泌并增强细胞毒性。体内实验进一步确认线粒体自噬在铜绿体内感染中的作用。

Drinking culture has high significance in both China and the world, whether in the entertainment sector or in social occasions; according to the World Health Organization's 2018 Global Alcohol and Health Report, about 3 million people died from excessive drinking in 2016, accounting for 5.3% of the total global deaths that year. Oxidative stress and inflammation are the most common pathological phenomena caused by alcohol abuse (Snyder et al., 2017). Scutellarin, a kind of flavonoid, is one of the main active ingredients extracted from breviscapine. It exerts anti-inflammatory, antioxidant, and vasodilation effects, and has been used to treat cardiovascular diseases and alcoholic liver injury. Although scutellarin can effectively alleviate multi-target organ injury induced by different forms of stimulation, its protective effect on alcoholic brain injury has not been well-defined. Therefore, the present study established an acute alcohol mice brain injury model to explore the effect of scutellarin on acute alcoholic brain injury. The study was carried out based on the targets of oxidative stress and inflammation, which is of great significance for the targeted therapy of clinical alcohol diseases.

Objective:To investigate the clinical effect of the medial perforator flap of the first plantar artery for repairing the soft tissue defect near the first metatarsophalangeal joint.Methods:From January 2016 to April 2018, 10 patients of soft tissue defect near the first metatarsophalangeal joint were treated in the Department of Orthopedics, Shandong Police General Hospital, including 6 males and 4 females, aged 22-61 years old, with an average age of 40 years. The defects of the feet were recovered by the medial perforator flap of the first plantar artery, and the flap donor defects were covered by split-thickness skin grafts.Results:All flaps and skin grafts of the patients were survived.All patients were followed up for 12-22 months postoperationly/after operation. The flaps were not swollen or ruptured, and the patients walked normally with full weight-bearing.Conclusions:The perforator artery of this flap is consistant, the harvesting of the flap is simple, and the texture of skin is similarwith surrounding tissue, there is no deficit to the flood supply of the foot after harvesting the flap. So, it is an ideal choice for repairing the defect of the soft tissue near the first metatarsophalangeal joint.

Objective To explore and analyze the effect and value of combined detection of urine neutrophil gelatin-associated apolipoprotein (NGAL) and kidney injury molecule 1 (KIM-1) in the early diagnosis of contrast-induced nephropathy.Methods The clinical data of 116 patients with coronary heart disease who received coronary intervention treatment in our hospital in 2017 were collected with forward-looking research methods.The patients were divided into the non-contrast-induced nephropathy group (control group, n=90) and the contrast-induced nephropathy group (observation group, n=26) according to the occurrence of contrast-induced nephropathy.The levels of serum creatinine, serum urea nitrogen, urine NGAL and KIM-1 were compared at different time points between the two groups.Results From 2 days after surgery, the serum creatinine levels were increased significantly (P＜0.05).The serum creatinine levels at 2 days after surgery (102.43±20.31) μmol/L and 3 days after surgery (107.22±25.13) μmol/L in the observation group were significantly higher than those in the control group[ (92.89±16.74) μmol/L, (91.97±15.38) μmol/L];The serum urea nitrogen levels in the observation group were increased significantly from 12 hafter surgery (P＜0.05);the serum urea nitrogen levels of the observation group at 12 h, 1 d, 2 dand 3 dafter surgery were significantly higher than those of the control group (P＜0.05);The urine NGAL levels at 4 and 12 hour and 1 and 2 days after surgery in the observation group were significantly higher than those in the control group;The KIM-1 levels at 1 day after surgery (5.14±0.96) μg/L and 2 days after surgery (5.58±1.33) μg/L in the observation group were significantly higher than those in the control group [ (3.58±1.23) μg/L, (3.64±1.15) μg/L], and the differences were statistically significant (P＜0.05).Pearson correlation analysis showed that there was a positive correlation between urinary NGAL at 4 hours postoperatively and serum creatinine at2 days postoperatively (r=0.784, P=0.000), and positively correlated with serum urea nitrogen level at 1 day postoperatively (r=0.811, P=0.000).The KIM-1 level at 1 day postoperatively was positively correlated with the serum creatinine level at 2 days postoperatively (r=0.596, P=0.000), and positively correlated with the serum urea nitrogen level at 2 days postoperatively (r=0.644, P=0.000).ROC curve analysis showed that the area under curve (AUC) of urine NGAL was 0.917[95％confidence interval (CI) :0.884-0.951], the sensitivity was 86.74％, and the specificity was 93.92％;AUC of KIM-1 was 0.842 (95％CI:0.755-0.901), the sensitivity was 81.16％, and the specificity was 83.47％.Conclusion Urine NGAL and KIM-1 are biochemical markers that can early react to the impairment of renal function, and have positive value in the early diagnosis of contrast-induced nephropathy.

Post-traumatic stress disorder in parents of children in neonatal intensive care unit ( NICU) directly influences children′s remedy and development, to which has been paid increasing attention by nurses. The review introduces the basic concept of post-traumatic stress disorder in parents of children in NICU, symptoms, research tools and influencing factors. Based on the results and experience of foreign research, the review provides references for domestic nurses to study post-traumatic stress disorder in parents of children in NICU.

The positive pressure powered air-filter protective hood can prevent physicians and nurses from being infected by serious respiratory infections. The contaminated air is filtered by the high efficiency specific air-filter (HESA) and then transported into the hood by a blower to form about 140 +/- 10 Pa positive pressure environment, which makes the air in hood overflow out from the lower part of the hood, by which carbon dioxide and vapor can be removed, and excreted. The HESA's filters of simulative virus up to 99.9999% with the structure of double layers filtering. With going at 4 km/h pace on a walking vehicle wearing the hood, the testees' heart rate is 90-105/min oxygen saturation of the blood(SPO2) is 97%-98%. When the air flow is from 75 L/min to 125 L/min, this can meet the testees' physiological demand under heavy workload. Clinical trial results show that the hood can effectively protect the physicians and nurses from infection.

OBJECTIVETo explore the effect of LPS on the expression of CD14 and the activation of Kupffer cells (KCs).

METHODSRat KCs were isolated and cultured with LPS. Immunohistochemistry and RT-PCR methods were employed to determine the changes in the CD14 expression and the concentration of TNFalpha, IL-6 and NO in the supernatant of the cultured KCs with LPS.

RESULTS(1) The expression of CD14mRNA and the synthesis of CD14 protein in the KCs increased evidently when stimulated by various concentrations of LPS, and the CD14mRNA expression was correlated in dose-dependent manner with LPS levels. (2) The expression of CD14mRNA and the synthesis of CD14 protein in KCs induced by LPS (10 micro g/ml) increased significantly and peaked at 3 approximately 6 hours. (3) The expression of CD14mRNA and the synthesis of CD14 protein in freshly cultured KCs were obviously up-regulated by the active mediators produced by KCs after being stimulated by LPS. (4) The release of TNFalpha, IL-6 and NO from cultured KCs was evidently down-regulated by the addition of anti-CD14McAb in the presence of serum or by the addition of LPS in the absence of serum, but up-regulated by the concomitant addition of LPS and LBP.

CONCLUSION(1) The CD14mRNA expression and the protein synthesis in cultured KCs were closely related to LPS and the active mediators produced from the KCs.The increased CD14 expression was possibly caused by LPS, and the further increase of the expression might be closely correlated to the cytokines released from the KCs. (2) The KC activation by low concentration of LPS was CD14 dependent.

OBJECTIVETo determine the feasibility and accuracy of high-frequency ultrasound in evaluating the left ventricular (LV) structure and function in normal and pressure overload rats and to examine the changes of the left ventricle during its transition from hypertrophy to heart failure.

METHODSThirty-eight female rats were randomly assigned to normal (n = 10), operated (n = 16) and sham-operated (n = 12) groups. Parasternal long axis and short axis images were acquired by a 7.5 mHz linear ultrasound probe at 12 weeks and 20 weeks after the operation respectively.

RESULTSLeft ventricular structure and function could be satisfactorily imaged for dimensions and mass. Compared to the sham-operated groups, at 12 weeks after the operation, the operated rats had increased LV wall thickness and mass (P < 0.01) with normal cavity and FS% (P > 0.05). At 20 weeks after the operation, the LV wall thickness showed no further progressive change and the LV mass increased greatly with slightly dilated LV cavity and decreased FS% (P < 0.05).

CONCLUSIONSHigh-frequency echocardiography provides a useful means to noninvasively evaluate LV dimensions, mass and function in rats. It will have great value for evaluating LV remodeling during the transition from LV hypertrophy to heart failure, as well as the effects of intervening drugs.

Objective Study and determine the suitable negative pressure condition for a negative pressure isolator. Method The filter efficiency, pressure, capabilities of inner environment and physiological parameters of the user were tested in different negative pressure conditions. Results The different negative pressure conditions influence above mentioned capabilities of the isolator considerably except for the filter efficiency Conclusion Under the conditions of -30Pa～-50Pa,those are optimized the capabilities of inner environment and physiological parameters of the user.
