1.The impact of Vibrio vulnificus RpoN on bacterial motility and biofilm formation

Objective:To construct the rpoN gene knockout strain (Δ rpoN) and the complemented strain (CΔ rpoN) of Vibrio vulnificus ( V. vulnificus), and investigate the role of the rpoN gene in regulating bacterial motility and biofilm formation. Methods:The Δ rpoN strain of V. vulnificus was constructed using homologous recombination. Bacterial motility was assessed via swimming assays, and flagellar morphology was observed by transmission electron microscopy. Biofilm formation capacity was evaluated using crystal violet and Congo red staining assays, as well as colony morphology analysis. Real-time quantitative RT-PCR (qRT-PCR) was used to detect mRNA levels of target genes associated with flagellar synthesis and biofilm formation in Δ rpoN and the wild-type strains. Results:The V. vulnificus genome harbored a single rpoN gene, encoding a protein with high amino acid sequence similarity to RpoN homologs in other bacterial species. The Δ rpoN strain was successfully constructed. Compared with the wild-type strain, the Δ rpoN strain exhibited complete loss of motility on soft agar plates, absence of flagellar, and downregulated mRNA levels of flagellar synthesis-related genes. Conclusions:In V. vulnificus, RpoN regulates flagellar assembly by modulating the expression of flagellar synthesis genes, thereby controlling bacterial motility and biofilm formation.
2.Influence of interactive intelligent systems on management and data collection for intensive care equipment of hospital

Objective:To explore the effect of interactive intelligent system on management effect and the influence of that on data collection for intensive care equipment of hospital.Methods:The interactive intelligent system conducted management intervention for equipment from the intelligent system based on quick response(QR)codes,usage registration,maintenance and upkeep of equipment,warranty and preventive maintenance for fault,training for operator,and status display.Seventy-two intensive care equipment in clinical use in the department of intensive care unit(ICU)of Beijing Tongren Hospital Affiliated to Capital Medical University from March 2021 to February 2023 were selected.The management for equipment during March 2021 and February 2022 adopted the conventional management method.From March 2022 to February 2023,the management for equipment adopted the management of interactive intelligent system(intelligent management method).The management quality,data collection and management effect for equipment between the two management methods were compared.A self-made satisfaction questionnaire was adopted to investigate the satisfaction of managers,who used equipment,for the management after the two management methods were used to conduct intervention.Results:After the intervention with intelligent management method for equipment was implemented,the management quality scores of the equipment of infusion and injection,medical monitoring,rehabilitation and treatment,life support,emergency rescue,and disinfection and sterilization were respectively(93.49±4.51),(94.31±4.89),(92.63±4.34),(95.23±4.12),(94.43±4.39)and(94.56±5.12)points,all which were higher than those of conventional management method,and the differences were statistically significant(t=6.253,5.198,6.149,5.673,5.243,6.029,P<0.05),respectively.The scores of completeness,accuracy,overlap,value and rationality of collecting equipment data of the intelligent management method were respectively(95.63±4.12),(96.23±3.61),(96.11±3.29),(95.45±4.39)and(96.21±3.73),all which were higher than those of the conventional management method,and the differences were statistically significant(t=5.149,5.541,6.292,4.784,5.112,P<0.05),respectively.The costs of component purchase,daily maintenance,personnel management,technical consultation and operation of the intelligent management method were all lower than those of the conventional management method,and the differences were statistically significant(t=4.495,5.237,6.112,5.395,5.676,P<0.05),respectively.The satisfaction of managers,who used equipment,for the intervention of adopting intelligent management method in management method,daily maintenance,repair and prevention,skill improvement and data recording for equipment were higher than those of adopting conventional management method,and the differences were statistically significant(x2=6.263,4.096,6.114,5.945,6.535,P<0.05).Conclusion:The application of the interactive intelligent system in the management for equipment of the department of intensive care unit of hospital can improve the quality of management for equipment,and enhance the effect of collecting data,and reduce the operating cost of equipment,which can achieve a higher satisfaction for management.

5.Clinical efficacy of surgery via endoscopic-assisted transcallosal approach in intraventricular hemorrhage

Objective:To investigate the efficacy and safety of surgery via endoscopic-assisted transcallosal approach in intraventricular hemorrhage (IVH).Methods:A retrospective study was performed; the clinical data of 68 IVH patients admitted to our hospital from October 2016 to January 2020 were analyzed. These patients were divided into endoscopic treatment group ( n=34) and drainage group ( n=34) according to the treatment methods. In the endoscopic treatment group, IVH was evacuated via endoscopic-assisted transcallosal approach; in the drainage group, external ventricular drainage and urokinase injection were performed. Gross hematoma clearance rate (clearance rate≥95%) 1, 3, and 7 d after surgery, complication incidence 7 d and 1 month after surgery, and good prognosis rate (ability of daily living [ADL] grading I-III) 3 months after surgery were compared between the two groups. Results:One, 3, and 7 d after surgery, the gross hematoma clearance rate in the endoscopic treatment group was significantly higher than that in the drainage group ( P<0.05). The recurrent hemorrhage rate within 7 d of surgery, intracranial infection rate, and percentage of patients with secondary hydrocephalus requiring shunt within 30 d of surgery in the endoscopic treatment group were significantly lower than those in the drainage group (2.9% vs. 20.6%, 2.9% vs. 23.5%, 5.9% vs. 17.6%, P<0.05). Three months after follow-up, the good prognosis rate of patients in the endoscopic treatment group was significantly higher than that in the drainage group (91.2% vs. 58.9%, P<0.05). Conclusion:Endoscopic-assisted transcallosal approach is recommended in clinical treatment of IVH, which can significantly reduce postoperative complications and improve postoperative prognosis.
6.Effect of three wavelength lasers on mRNA expression of types Ⅰ and Ⅲ procollagen in fibroblasts

Objective To investigate the effects of different wavelength (595 nm,755 nm and 1064 nm) lasers on the mRNA expression of types Ⅰ and Ⅲ procollagen in cultured fibroblasts.Methods Fibroblasts from Kunming mice were cultured in vitro.They were divided into 595 nm laser group,755 nm laser group,1064 nm laser group and no laser irradiating group.The mRNA expression of the types Ⅰ and Ⅲ procollagen was detected by RT-PCR.Results The mRNA expression level of type Ⅰ procollagen in 1064 nm group was higher than that of 755 nm group,595 nm group and control group (P<0.05).The expression levels of 755 nm group and 595 nm group were higher than that of control group (P<0.05).But the difference between 755 nm group and 595 nm group was not statistically significant (P>0.05).The mRNA expression level of type Ⅲ procollagen in 1064 nm group was higher than that in 755 nm group,595 nm group and control group (P<0.05).755 nm group had higher expression than 595 nm group and control group (P<0.05).But the difference between 595 nm group and the control group was not statistically significant (P > 0.05).Conclusions Three wavelength lasers can directly promote mRNA expression of types Ⅰ and Ⅲ procollagen in fibroblasts.595 nm laser mainly promotes mRNA expression of type Ⅰ procollagen,and 755 nm laser promotes more mRNA expression of type Ⅲ procollagen than 595 nm laser.The most mRNA expression of types Ⅰ and Ⅲ procollagen is promoted by 1064 nm laser.
7.Analysis of polymorphism at sites -597 and -572 of interleukin-6 promoter in Tibetan population from Tibet autonomous region

BACKGROUND: Polymorphism in promoter region can change the expression of genes, which may be associated with susceptivity of diseases. Gene polymorphism of interleukin-6 (IL-6) promoter is associated with nationality and many diseases. Different nationalities often display different characteristics of gene polymorphism.OBJECTIVE: To observe the distribution of polymorphism at sites -597 and -572 in IL-6 promoter region in Tibet Tibetan population and to provide the theoretical data for Tibetan population genetics and background of immunity.DESIGN: Randomized investigation.SETTING: Institute of Anthropology, Jinzhou Medical College.PARTICIPANTS: Totally 108 healthy Tibetan teenagers were selected from Lasa and Naqu region in Tibet autonomous region from October 2003 to July 2004, including 60 males and 48 females, aged from 14-21 years. Inclusive criteria:The parents of the volunteers were healthy Tibetans after body examination. The volunteers knew the fact, agreed to participate into the trail and signed the informed consent.METHODS: 5 mL peripheral vein blood was collected from 108 Tibetan teenagers. DNA from human leucocytes was extracted by salt fractionation. IL-6 promoter including -597 and -572 fragments was amplified by polymerase chain reaction (PCR). Representative fragments were cloned then sequenced after restriction fragment length polymorphism (RFLP).MAIN OUTCOME MEASURES: Distribution of polymorphism in Tibet Tibetan population; Results after comparison with those of other nationalities including Han population.RESULTS: Data of 108 Tibetan teenagers were involved in the result analysis. ①Distribution of polymorphism on -572C/G site of IL-6 promoters in population of either sex: There were no GA and AA genotypes at site -597, but only GG genotype appeared. There were CC, CG and GG genotypes at site -572, and the frequencies were 0.63, 0.35 and 0.02 in order. Distribution of genotype with representativeness met the Hardy-Weinberg equilibrium. Allele frequencies were 0.81 and 0.19, respectively. There was no significant difference of either sex in genotype and allele frequency (P > 0.05). ②Distribution of polymorphism on -597 G/A and -572 C/G in different nationalities: GG, GA, AA genotypes appeared on -597 site in England and France, and G and A allele frequencies were 0.60 and 0.40,respectively. It was significantly different from that of Tibetan in Tibet. Furthermore, Japanese had no polymorphism,which was similar to that of Hans in China (P > 0.05). ③Genotype of different straps and results of DNA sequencing:Only GG genotype was found on -597 site (without the restriction site, one fragment after restriction, PCR amplification products), no GA and AA genotypes. CC, CG and GG genotypes appeared at site -572, and frequencies were 0.64,0.35 and 0.01, respectively. Distribution of genotype with representativeness met the Hardy-Weinberg equilibrium (P >0.05). Allele frequencies were 0.81 and 0.19, respectively. There was no significant difference of either sex in genotype and allele frequency. Distribution of gene frequency and allele frequency in IL-6 were similar between Tibetan and that of Hans, but it was significantly different from that of population in England, France and America.CONCLUSION: There are nationality differences of IL-6 gene polymorphism at sites -597 and -572. No polymorphism is found at site -597 in Tibetan. Race differences are seen at site -572, having CC, CG and GG genotypes and G allele is rate. Compared with white population, there is significant difference in genotype and allele frequency at site -572. Their characteristics are close to Hah population and Japanese, which may be associated with genetic gene of persons living in plateau.
