1.Systematic review and Metaanalysis of the association between heavy metal exposure and obesity in children and adolescents

Objective:
To systematically evaluate the relationship of exposure to five heavy metals, namely lead, arsenic, cadmium, mercury and aluminum with obesity in children and adolescents, so as to provide a scientific basis for subsequent research in the area.
Methods:
Four Chinese databasesc (CBM, VIP, CNKI and Wanfang) and four foreign databases (OVID, PubMed, Web of Science and EBSCO), were searched to collect relevant studies, and the search period was from the establishment of the database to May 5, 2024. After 2 investigators independently screened the literature, extracted the data and evaluated the risk of bias of the included studies, the results were analyzed quantitatively and summarized qualitatively.
Results:
A total of 5 cohort studies on lead exposure and 17 cross sectional studies involving exposure to lead ( n =13), cadmium ( n =8), mercury ( n =8), arsenic ( n =4), and aluminum ( n =1) were included. Meta analysis of the 2 cohort studies showed that lead exposure was not associated with the risk of overweight and obesity in children ( RR=0.76, 95%CI=0.50-1.16, P >0.05). The cross sectional study Meta-analysis results showed that lead exposure was negatively associated with the risk of childhood overweight ( OR=0.70, 95%CI =0.59-0.84, 2 studies) and obesity ( OR=0.71, 95%CI =0.58-0.87, 3 studies); cadmium exposure was negatively associated with the risk of childhood overweight ( OR=0.83, 95%CI =0.73-0.95, 2 studies) and obesity risk( OR=0.70, 95%CI =0.63-0.78, 3 studies); mercury exposure increased the risk of overweight/obesity ( OR=1.42, 95%CI =1.14-1.76, 2 studies) and abdominal obesity ( OR= 1.99, 95%CI =1.45-2.73, 2 studies) in children; the group with the highest concentration of arsenic in urine had a lower risk of developing obesity compared to the group with the lowest concentration ( OR=0.39, 95%CI =0.23-0.65, 1 study), and the group with the highest concentration of aluminum in urine had a lower risk of obesity compared with the group with the lowest concentration ( OR=0.52, 95%CI =0.31-0.86, 1 study)(all P <0.05).
Conclusion
Heavy metal exposure may be a risk factor for overweight and obesity in children and adolescents, but the conclusions are inconsistent and need to be validated in further high quality prospective cohort studies.
2.Ultrasensitive wash-free quantification of breast cancer-derived small extracellular vesicles via a self-locked DNAzyme nanoprobe

Objective:To develop a self-locked DNAzyme nanoprobe-based fluorescence amplification strategy for wash-free and ultrasensitive detection of breast cancer-derived small extracellular vesicles (sEV).Method:A DNAzyme self-locked probe was designed to recognize the epithelial cell adhesion molecule (EpCAM) specifically expressed on breast cancer-derived sEVs. Upon binding to EpCAM, the DNAzyme-lock structure was opened, restoring the DNAzyme cleavage activity. The activated DNAzyme then cyclically cleaved the RNA site on the substrate strand. Fluorescently labeled substrate strands were used to detect sEVs at varying concentrations, and the detection limit and linear range were determined.Results:The DNAzyme self-locked probe successfully identified breast cancer-derived sEVs and generated a fluorescent signal through cyclic cleavage. The proposed method achieved wash-free detection of sEVs, with the fluorescence intensity showing a strong linear correlation with sEV concentration ( R2=0.98). The linear detection range was 1.0×10 2-1.0×1.0 7 particles/μl, with a detection limit of 59 particles/μl. Conclusion:This study established a wash-free and highly sensitive strategy for quantifying breast cancer-derived sEVs, which provides a promising technical approach for the early diagnosis of cancer.
3.Glioma cell-secreted Prg4 induces the expression of macrophage Dicer,a key reg-ulatory molecule for macrophage alternative activation

Purpose To explore the key molecules mechanisms underlying the selective activation of macrophage and the regulation of Dicer expression induced by glioblastoma(GBM)cells,as well as its prognostic significance.Methods Glioblastoma conditional medium(GCM)was fractionated by molecular weight using ultrafiltration.Specif-ic molecular weight components of GCM that upregulate Dicer expression in mouse bone marrow derived macrophages(BMDMs)were identified.Secreted proteins were identified by mass spectrometry(MS).The correlation between candidate proteins and GBM prognosis was analyzed using the TCGA and CGGA database.In vitro experiments of the candidate proteins on Dicer expression in BMDMs were further carried out.Results GCM components with a molecu-lar weight of>50 kDa significantly upregulated Dicer expression in BMDMs.MS identified five key secreted proteins:Prg4,Psap,Hexa,Aebp1,and Itih2.High expression of Prg4 was significantly positively correlated with poor progno-sis in GBM patients(P<0.001)and was associated with the expression of selective macrophage activation markers.Recombinant Prg4 protein stimulated BMDMs and induced Dicer expression in mouse BMDMs.Conclusion This study reveals that glioma cells induce Dicer expression in macrophages by secreting Prg4,providing a theoretical basis for GBM therapeutic strategies targeting the Prg4-Dicer axis.

6.The treatment of Dong qi beside the navel in Shanghan Lun

Dong qi,referring to beating beside the navel,was proposed for the first time by Nan Jing."Beating above the navel""beating below the navel",and"beating below the heart"in Shanghan Lun all belong to the category of Dong qi.The pathogenesis of Dong qi can be learned from Shanghan Lun.Yang deficiency of the heart,spleen,and kidney,with qi failing to transform body fluids,results in fluid retention.Fluid retention is the key etiological factor of Dong qi.Although there is an exterior pattern with Dong qi,the exterior-releasing and purgative method cannot be used for treatment,or the patient will be impaired of yang and exhausted of yin,fluid circulation will be disturbed,and so on.Reinforcing yang qi to drain water retention is the treatment principle of Dong qi.The coupled medicines of Fuling and Guizhi were commonly used by ZHANG Zhongjing to treat Dong qi.This reflects the treatment principle of strengthening the spleen and warming yang for transforming qi to circulate body fluids.ZHANG Bi,a physician from the Yuan Dynasty who learned from ZHANG Zhongjing,thought that the treatment of Dong qi was based on the method of cultivating earth to restrain water and taking into account the ascending,descending,entering,and exiting of qi of the liver,heart,lung,and kidney while tonifying the spleen,and then created Baoming Siqi Powder(Zhiqiao,Fuling,Fangkui,and Qianhu Powder)for the treatment of Dong qi,which is of great significance to the clinic.
7.Preparation of Young Erythrocytes and Evaluation of Their Mean Cell Age

Objective To establish a simple and feasible method of preparing young erythrocytes and test some indexes of evaluating the mean cell age.Methods We collected 34 bags of whole blood from healthy blood donors,400ml/each bag,then eliminated WBCs by leukoreduction filter.After twice centrifuging and manual speration,we finally obtained young erythrocytes from the upper section of the red cell layer,the remains were old erythrocytes.These different age erythrocytes were separated by blood bags.Some indexes as pyruvate kinase,reticulocyte count,mean corpuscular volume(MCV),mean corpuscular hemoglobin concentraion(MCHC),erythrocytes deformation index EI were tested on the non-separation erythrocytes,young erythrocytes and old erythrocytes,simultaneouosly,the detecting hemoglobin recovery rate.Results There was significant difference of all indexes among the three kinds of erythrocyte products.The PK activity ratio of young erythrocytes' to non-preparating erythrocytes was 1 175?0 13,showed the mean cell age was about 40 days.The hemoglobin recovery rate was(48 37?3 82)%.Conclusions The method is effective for preparing young erythrocytes,PK,REI,MCV,MCHC and EI can be used to evaluate the young erythrocytes' mean cell age.
8.The Technical Method for Noninvasive Magnetic Stimulation of Nerve Center and Its Clinical Application

The increasing use of non-invasive magnetic stimulation technique for nerve center function examination and functional neuromuscular rehabilitation is a new cynosure in biomedical engineering and neuro-electrophysiologic researches. This paper analyzes theoretically the magnetic stimulation of first demonstration extending the technique model to human body, and the magnetic principile of physics. The characteristics of magnetic stimulation can be highlighted by comparing it to conventional electrical stimulation. Also presented are the peripheral multichannel magnetic stimulation to mimic electrical stimulation and the clinical functions toward neuromuscular rehabilitation.
9.EFFECTS OF MUSICAL MEDITATION TRAINING ON AUDITORY MISMATCH NEGATIVITY AND P300 IN NORMAL CHILDREN

The auditory mismatch negativity(MMN)and P300 of event-related potentials were compared in normal children either with or without musical meditation training.The experimental group consisted of 11 subjects who had been trained with musical meditation for six months and the control group consisted of 12 subjects(matched for age,sex and grade)who had not received musical meditation.MMN amplitudes in the trained children were larger than those in the control group.In addition,the MMN amplitudes were identical in attend and ignore conditions for both groups.This evidence suggests that auditory brain function has been affected by musical meditation training.lt thus suggests that the MMN is capable of assessing changes to the brain function in normal subjects.There were no significant differences in the P300 latencies and amplitudes between the two groups.This result suggests that MMN and P300 may reflect different aspects of the brain function.
