1.Related factors of drug detoxification motivation in methamphetamine-dependent female youth

Objective:To explore drug detoxification motivation in methamphetamine-dependent female youth and its relationship with time perspective,perceived social support and emotion regulation strategies.Methods:To-tally 200 methamphetamine-dependent female youths under compulsory isolation were assessed with the Drug De-toxification Motivation Scale,Zimbardo Time Perspective Inventory-Chinese(ZTPI-C),Emotion Regulation Scale(ERS),and Perceived Social Support Scale(PSSS).Results:Multiple linear regression analysis showed that drug detoxification motivation scores were positively correlated with the scores of ERS reevaluation and expression inhi-bition,PSSS family support scores and level of education(β=0.24,0.16,0.20,0.16).Conclusion:Time perspec-tive,perceived social support and emotion regulation strategies are closely related to drug detoxification motivation in methamphetamine-dependent female youth.
2.Study on the Correlation between Serum miR-424,miR-363-3p Levels and Disease Progression and Prognosis in Patients with Multiple Myeloma

Objective To analyze the relationship between serum levels of miR-424 and miR-363-3p and disease progression in patients with multiple myeloma (MM),and the value of combined prediction of prognosis. Methods A total of 76 patients with MM admitted to Xi'an People's Hospital from January 2022 to September 2023 were included in the study group. There were 19 patients in the International Staging System Phase Ⅰ,27 patients in Phase Ⅰ,and 30 patients in Phase Ⅲ. Immunological classification confirmed 16 cases of IgA type,43 cases of IgG type,11 cases of light chain type,and 6 instances of nonsecretory type. 75 healthy volunteers who underwent physical examinations during the same period were included as the healthy control group. Detected the serum levels of miR-424 and miR-363-3p between groups,perform Logistic factor analysis,and draw receiver operating characteristic (ROC) curves to explore the relationship between miR-424 and miR-363-3p in the progression and prognosis of MM. Results The miR-424 (2.77±0.61) level in the study group was higher than that in the healthy control group (0.83±0.26),and the miR-363-3p level (0.49±0.09) was lower than that in the healthy control group (1.20±0.21),with statistically significant differences (t=25.362,27.064,all P<0.05).The miR-424 level in stage Ⅲ MM patients was significantly higher than that in stage Ⅰ,stage Ⅱ patients (t=11.988,10.366),and miR-363-3p was lower than that in stage Ⅰ,stage Ⅱ MM patients (t=8.077,5.543),with statistically significant differences (all P<0.05).The miR-424 levels in IgG type MM patients were significantly higher than those in IgA type,light chain type,and non secretory type (t=5.967,5.358,3.960),and the miR-363-3p levels were significantly lower than those in IgA type,light chain type,and non secretory type (t=11.594,8.013,6.994),with statistically significant differences (all P<0.05).The levels of Cys-C and miR-424 in MM patients with poor prognosis were significantly higher than those in the good prognosis group,while the levels of miR-363-3p were lower than those in the good prognosis group,with statistically significant differences (t=3.067,6.137,4.011,all P<0.05) .The optimal cut-off values for miR-424 and miR-363-3p in diagnosing MM were 1.73 and 0.40,respectively. The AUC(95%CI) of the combined diagnosis ROC curve[0.911(0.801~0.973)]was higher than that of miR-424[0.792(0.685~0.822)]and miR-363-3p[0.761(0.669~0.798)],and the differences were statistically significant (t=6.026,7.769,all P<0.05). Conclusion High levels of serum miR-424 expression and low levels of miR-363-3p expression suggested MM progression and poor prognosis. The combination of serum miR-424 and miR-363-3p has a high value in predicting the progression and prognosis of MM.
3.Ganoderma Lucidum Extract Nano-emulsion:Preparation,Characterization and Pharmacodynamics Study in Anti-skin Aging

Objective To screen the prescription,preparation,quality evaluation of Ganoderma lucidum extract nano-emulsion(GLEN)and study its anti-skin aging effect.Methods The emulsifier and oil phase of the nano-emulsion were preliminarily screened by solubility method.The pseudo-ternary phase diagram method was used to screen the nano-emulsions,and the prepared GLEN were evaluated for quality in terms of the appearance,particle size,structure,drug loading,stability and skin safety.A mouse skin aging model was constructed by injecting D-galactose combined with ultraviolet radiation.GLEN was applied to the skin at the modeling site and investigated for their anti-skin aging efficacy.Results The optimal GLEN prescription was 30%polyoxymethylene castor oil,14.4%glycerol,11.1%medium chain triglycerides,and 44.5%water.GLEN was a dark brown and transparent liquid at room temperature,O/W type nano-emulsion with an average particle size(32.20±2.89)nm,polydispersity coefficient 0.38±0.03,zeta potential value(-4.05±0.27)mV.The content of Ganoderma lucidum polysaccharides was(36.35±0.55)mg·mL-1,and the content of Ganoderma lucidum triterpenes was(5.99±0.46)mg·mL-1,which possessed good stability.Pharmacodynamic experiments showed compared with the model group,the skin scores of mice in the GLEN group significantly improved(P﹤0.05),the water content increased(P﹤0.01),and the contents of SOD,GSH-PX and Hyp increased(P﹤0.05),while the content of MDA was decreased significantly(P﹤0.01).HE,Masson and Sirius crimson staining further indicated that the pathological changes in mice were alleviated to varying degrees in GLEN group.And GLEN had high safety for skin,could be applied to human body.Conclusion In this study,the prepared GLENs were homogeneous in appearance,stable in nature,and exhibited significant anti-aging effects on mouse skin.

5.Anatomical features and clinical significance of the pelvic segment of the obturator artery

Objective To observe the origin and course of the obturator artery(OA),so as to provide anatomical reference for reducing hemorrhage during pelvic surgery and pubic fracture fixation.Methods A total of 65 human hemi-pelvises specimens with intact structure were dissected to observe the origin,course and other variations of OA.Measure the length of the inner section of OA basin and the outer diameter at the origin,etc.Results OA originated from the internal iliac artery in 57 cases(87.7%),including 3 cases(4.6%)of the superior gluteal artery,5 cases(7.7%)of the inferior gluteal artery,3 cases(4.6%)of the external iliac artery and 5 cases(7.7%)of the inferior epigastric artery.OA participated in the formation of the arterial trunk in 3 cases(4.6%).The length of the pelvic segment of the OA in male and female was(50.87±15.41)mm and(51.71±14.19)mm,respectively,with no statistically significant difference between them(P>0.05).The outer diameters at the origin of the OA in male and female were(2.79±1.05)mm and(2.35±0.86)mm,and there was no statistically significant difference between them(P>0.05).Conclusion OA mainly originated from the anterior trunk of the internal iliac artery,with a few OA originated from the branches of the posterior trunk or the inferior epigastric artery,or participated in the formation of the arterial trunk.In pelvic surgery involving OA area,attention should be paid to the length of its pelvic segment and the outer diameter at the origin of OA,so as to better locate and protect blood vessels during surgery.
6.Clinicopathological,immunophenotypic,and HPV infection status analysis of cer-vical carcinoma with adenoid cystic carcinoma-like features

Purpose To investigate the clinicopathological characteristics,immunophenotype,high-risk human papillomavirus(hrHPV)infection status,treatment,and prognosis of cervical carcinoma exhibiting adenoid cystic car-cinoma(ACC)features.Methods Clinical data from 10 cases of cervical cancer with ACC features were collected.A retrospective analysis was proformed on the patients'clinicopathological data,histological features,and immunopheno-type(EnVision method),along with in situ hybridization detection of hrHPV E6/E7 mRNA covering 18 hrHPV types.Results The median age of the patients was 67.5 years,comprising 4 biopsy specimens and 6 surgical specimens.Except for 2 cases detected incidentally during physical examination,the remaining 8 cases presented with unexplained vaginal bleeding or contact bleeding.The mean tumor diameter was 4.5 cm(range:2.5 to 9.0 cm),and tumor stages were available for 8 patients(5 cases in stage Ⅰ and 1 case each in stages Ⅱ,Ⅲ,and Ⅳ).The follow-up period was 8 to 42 months,excluding 1 case lost to follow-up,2 cases(Ⅲ C1 and ⅣB)died within 1 year after surgery.Histologi-cally,4 cases exhibited pure ACC morphology,while 5 cases were mixed carcinomas(combined with squamous cell carcinoma,adenoid basal cell carcinoma,high-grade neuroendocrine carcinoma,or carcinosarcoma,respectively).The characteristic ACC morphology manifested as cribriform,pseudo-glandular,tubular,trabecular,and/or solid pat-terns with one case mixed carcinoma's lymph node metastasis showing only ACC morphology.Immunohistochemically,all 10 tumors exhibited diffuse strong positivity for p16 and p63,a high Ki67 proliferation index(40%to 90%),and wild-type p53 expression,and in 50%of cases,only a minority of cells were positive for c-MYB.Additionally,SOX10 was positive in 8 of 9 cases and CD117 was positive in 7 of 10 cases,respectively.In situ hybridization for hrHPV E6/E7 mRNA confirmed the presence of transcriptionally active HPV infection in all cases.Conclusion Cervical cancer with ACC features is rare,and predominantly occurs in postmenopausal elderly women.It represents an HPV-associat-ed high-grade carcinoma in which clinical stage is a critical prognostic factor.Immunohistochemical staining for CD117 and SOX10 aids in the pathological diagnosis of this tumor type.

8.Investigation of effects of petroleum ether fraction from Derris eriocarpa on glucose and lipid metabolism in a mouse model of metabolic syndrome via ATF3/HNF4ɑ/CYP7A1 pathway

AIM To investigate effects of petroleum ether fraction from Derris eriocarpa How on glucose and lipid metabolism in a mouse model of metabolic syndrome(MS).METHODS KM mice were fed a high-fat diet and administered streptozotocin intraperitoneally to establish MS models.The MS mice were then randomly assigned to the model group,the metformin hydrochloride group,the lovastatin group,the ursolic acid group,and the high-,medium-and low-dose D.eriocarpa petroleum ether fraction groups,with 10 mice in each group.Ten additional mice maitained on a normal diet served as the normal control group.After 4 weeks of intragastric administration,glucose and lipid metabolism indicators were measured.Hepatic pathological changes were assessed using HE staining and oil red O staining.Liver tissue mRNA expressions of ATF3,PEPCK,FXR,CYP7A1,HNF4ɑ,CYP8B1 and SRB1 were quantified by RT-qPCR.Hepatic protein expressions of ATF3,HNF4ɑ,PEPCK,FXR and CYP7A1 was analyzed by Western blot in MS mice.RESULTS Compared to the model group,the high-dose D.eriocarpa petroleum ether fraction group exhibited significant glucose tolerance improvement(reduced OGTT-AUC,P<0.01);favorable serum lipid modulation in terms of increased HDL-C levels(P<0.01)and decreased TG,TC,LDL-C(P<0.01);reduced renal biomarkers(BUN,SCR)and hepatotoxic indicators of TBA,AST and ALT activities(P<0.01);alleviated hepatic lipid accumulation and histopathological damage;downregulated mRNA and protein expressions of ATF3,HNF4ɑ and PEPCK,as well as CYP8B1 mRNA expression(P<0.01);and upregulated mRNA and protein expressions of FXR and CYP7A1,along with SRB1 mRNA expression(P<0.01).CONCLUSION D.eriocarpa petroleum ether fraction ameliorates glucose and lipid metabolism dysregulation in MS mice by modulating the ATF3/HNF4ɑ/CYP7A1 signaling pathway,consequently eliciting hypoglycemic,hypolipidemic,hepatoprotective and nephroprotective effects.

10.Treatment of Globus Hystericus Based on the Theory of "Regulating the Five Zang Organs and Harmonizing the Spleen and Stomach"

Based on the concept of "regulating the five zang organs and harmonizing the spleen and stomach", globus hystericus is believed to originate from dysfunction of the five zang organs and disharmony of the spleen and stomach. Treatment primarily focuses on regulating the spleen and stomach while also considering other affected organs, with a self-prescribed Anpiwei Jingyan Formula (安脾胃经验方) for harmonizing the spleen and stomach as the foundational treatment. Additionally, syndrome-based modifications are applied according to imbalances in the heart, lung, kidney, or liver. For heart-yang deficiency, modified Linggui Zhugan Decoction (苓桂术甘汤) could be combined; for heart-yin deficiency, modified Tianwang Buxin Pill (天王补心丹) could be combined. For lung failing to disperse and descend and fluid retention, modified Sanao Decoction (三拗汤) could be combined; for lung and stomach yin deficiency, modified Shashen Maidong Decoction (沙参麦冬汤) could be combined. For kidney-yang deficiency with ascending counterflow of cold water, modified Jingui Shensi Pill (金匮肾气丸) could be combined; for kidney-yin deficiency, modified Liuwei Dihuang Pill (六味地黄丸) could be combined. For liver constraint and spleen deficiency, modified Sini Powder (四逆散) could be combined; for liver-yin deficiency or liver stagnation transforming into fire and attacking the stomach, modified Yiguan Decoction (一贯煎) could be combined.
