Objective:To analyze the clinical characteristics, imaging, myopathology and outcomes of patients with COVID-19 related autoimmune myopathy. Methods:The clinical features, serum creatine kinase (CK), myositis antibodies, muscle magnetic resonance imaging, myopathology and therapy of 5 patients with COVID-19 related autoimmune myopathy diagnosed in Peking University First Hospital from December 2022 to April 2023 were collected. The effects of the therapy after a short term follow up were analyzed. Results:Among the 5 patients, there were 3 males and 2 females, with onset age of 42-86 years. All patients presented with proximal muscle weakness in the recovery term of COVID-19. Myalgia was noted in 3 cases, dysphagia in 1, skin damage in 2, interstitial lung disease in 1. The serum CK of the 5 patients was 1 663-16 000 IU/L, 1 patient had anti-3-hydroxy-3-methylglutaryl-CoA reductase autoantibodies and 1 patient had anti-signal recognition particle autoantibodies. The electromyography showed myogenic lesions in all patients. Muscle magnetic resonance imaging showed diffuse muscle edema in all patients, myofascial edema in 3 and subcutaneous-tissue edema in 3. The muscle biopsies in 4 patients revealed necrotic myopathy, with high P62 expression in muscle fibers. The electromicroscopy of 2 patients revealed vacuolated mitochondria and intranuclear tubulofilamentous inclusions in muscle fibers. Four patients were treated with glucocorticoids, of whom 2 patients combined with intravenous immunoglobulin, tacrolimus or cyclophosphamide. One case had close monitoring without drug therapy. They showed significant improvement, but the CK was still abnormal in 4 patients.Conclusions:COVID-19 leads to immune mediated myopathy. The manifestation of patients is characterized by proximal predominant weakness and high creatine kinase level. Muscle magnetic resonance imaging shows diffuse muscle edema. The muscle biopsies reveal necrotic myopathy. The effectiveness of immunosuppression needs to be further studied.

Objective The purpose of this study was to investigate the bacterial communities of biting midges and ticks collected from three sites in the Poyang Lake area, namely,Qunlu Practice Base,Peach Blossom Garden, and Huangtong Animal Husbandry, and whether vectors carry any bacterial pathogens that may cause diseases to humans,to provide scientific basis for prospective pathogen discovery and disease prevention and control. Methods Using a metataxonomics approach in concert with full-length 16S rRNA gene sequencing and operational phylogenetic unit(OPU)analysis,we characterized the species-level microbial community structure of two important vector species, biting midges and ticks, including 33 arthropod samples comprising 3,885 individuals, collected around Poyang Lake. Results A total of 662 OPUs were classified in biting midges, including 195 known species and 373 potentially new species, and 618 OPUs were classified in ticks, including 217 known species and 326 potentially new species.Surprisingly,OPUs with potentially pathogenicity were detected in both arthropod vectors, with 66 known species of biting midges reported to carry potential pathogens, including Asaia lannensis and Rickettsia bellii, compared to 50 in ticks, such as Acinetobacter lwoffii and Staphylococcus sciuri. We found that Proteobacteria was the most dominant group in both midges and ticks.Furthermore, the outcomes demonstrated that the microbiota of midges and ticks tend to be governed by a few highly abundant bacteria.Pantoea sp7 was predominant in biting midges, while Coxiella sp1 was enriched in ticks.Meanwhile,Coxiella spp.,which may be essential for the survival of Haemaphysalis longicornis Neumann, were detected in all tick samples. The identification of dominant species and pathogens of biting midges and ticks in this study serves to broaden our knowledge associated to microbes of arthropod vectors. Conclusion Biting midges and ticks carry large numbers of known and potentially novel bacteria, and carry a wide range of potentially pathogenic bacteria, which may pose a risk of infection to humans and animals. The microbial communities of midges and ticks tend to be dominated by a few highly abundant bacteria.

Objective To investigate the protective effect of Toll-like receptor (TLR)2/TLR9 agonists,Pam2 CSK4(Pam)and CpG ODN (CpG)on mice infected with Acinetobacter baumannii (Ab)in the lungs. Methods Female C57 mice (6~8 weeks old)were randomly divided into PBS,Pam,CpG and Pam+CpG groups. In 24 h after intranasal immunization with different doses of the corresponding agonists, the mice were given a lethal dose of Ab infection in the lungs, and the survival rates of the mice were observed.A sublethal dose lung infection model of Ab was then established, and the bacterial colonization in the blood, lungs, liver, kidneys and spleen was measured respectively in the mice after infection.HE staining was used to observe the pathological damages in the lungs and kidneys. The protective effect of the agonists in the immunized mice against Ab was examined at 1,3 and 7 d after immunization to explore the protective time window.Pam+CpG was used to stimulate A549 cells and RAW264.7 cells to investigate the killing or phagocytic effects on Ab.Results Compared to PBS,Pam+CpG treatment significantly improved the survival rate of the mice after a lethal dose of Ab lung infection (P<0.05,P<0.01 ),reduced bacterial colonization in the blood (P<0.01 ),lungs (P<0.01 ),liver (P<0.01 ),kidneys (P<0.01 )and spleen (P<0.01 )in the mice after sublethal challenge, and alleviated pathological damage caused by infection. Immunization at 1 or 3 d before infection significantly improved the survival rate (P<0.05 ),and the protective effect was the best in 3 d after immunization.Furthermore, compared to single PBS,Pam and CpG immunization,Pam+CpG significantly promoted the killing and phagocytic effects of A549 epithelial cells and RAW264.7 cells, respectively, against Ab (P<0.01 ).Conclusion Combined application of TLR2/TLR9 agonists exerts a significant protective effect on both lethal and sublethal infections of Ab,which might be by its promoting the killing or phagocytic effect of lung epithelial cells and macrophages against Ab.

The presence of 4-aminophenol(4-AP)in wastewater from the pharmaceutical industry is a common occurrence due to its role as a byproduct or intermediate during the hydrolysis process of paracetamol metabolism, resulting in significant water pollution. Therefore,it is crucial to employ a straightforward and reliable analytical approach for detecting 4-AP in the environment. In this study,a specific type of metal-organic framework(MOF)material called[Cd4(ABTC)2(H2O)12]n(SXNU-4-Cd,H4ABTC=3,3′,5,5′-azobenzene tetracarboxylic acid)was successfully synthesized, which exhibited a unique two-dimensional layered structure consisting of three intertwined spiral chains forming a distinctive″twist braid″.These layers underwent π-π stacking, creating three-dimensional channels with azo bonds decorating the channel walls. This p-π interaction significantly enhanced the adsorption capacity of SXNU-4-Cd towards 4-AP,thereby improving its recognition sensitivity. The fabricated SXNU-4-Cd/GCE sensor showed high sensitivity towards 4-AP in the linear concentration range of 0.1-130 μmol/L,with a detection limit of 8.6 nmol/L,and also exhibited good anti-interference capability, reproducibility and stability. The SXNU-4-Cd/GCE sensor was successfully used for detecting 4-AP in lake water sample, with spiked recoveries of 95.9%-102.8%.This study introduced a novel technique that utilized pure Cd-MOFs to develop electrochemical sensor capable of effectively detecting 4-AP in water samples.

Recent Advances in Ophthalmology 2024;44(7):526-530
Objective To explore the effects of the reactive oxygen species(ROS)inhibitor Dendrimer and N-acetyl cysteine(NAC)polymer(D-NAC)on retinal revascularization and its mechanism by inhibiting the expression of ROS in mice with oxygen-induced retinopathy(OIR).Methods The C57BL/6J mice were randomly divided into three groups:normal control group(NC group),OIR+Dendrimer group(OIR models+intravitreal injection of Dendrimer),and OIR+D-NAC group(OIR models+intravitreal injection of D-NAC).The mice were exposed to 75%oxygen from postnatal day 7(P7)to P1 2 and then to 21%oxygen from P1 2 to P1 7 to establish OIR models. The concentration of glutathione(GSH)was detected by GSH assay. The areas of retinal vessel occlusion(VO)and pathological neovascularization(NV)were quantitatively compared between the two OIR groups after fluorescence staining of the retina. The mRNA levels of retinal NV-related factors in mice between the two OIR groups were detected by quantitative reverse transcription polymerase chain reaction(qRT-PCR).The protein expression of retinal vascular endothelial growth factor(VEGF)in mice between the two OIR groups was detected by enzyme-linked immunosorbent assay(ELISA).The mRNA levels of Delta-like ligand 4(DLL4)/Notch pathway-associated factors in mice between the two OIR groups were detected by qRT-PCR.Results GSH assay results showed that there was no statistically significant difference in the retinal GSH level of mice between the NC group and the OIR+Dendrimer group on P12,P13,P14,P16,and P17(all P>0.05).Compared with the OIR+Den-drimer group, the retinal GSH level of mice in the OIR+D-NAC group increased on P16 and P17(both P<0.05).The reti-nal flat-mount staining results showed that compared with the OIR+Dendrimer group, the areas of VO and pathological NV in the OIR+D-NAC group decreased on P17(both P<0.05).qRT-PCR results showed that on P16,compared with the OIR+Dendrimer group, the mRNA levels of fibroblast growth factor, angiogenin,VEGF,VEGF receptor 2 and erythropoie-tin decreased in the OIR+D-NAC group(all P<0.05);ELISA results showed that compared with the OIR+Dendrimer group, the protein expression of VEGF decreased in the OIR+D-NAC group(P<0.01).qRT-PCR results showed that compared with the OIR+Dendrimer group, the mRNA levels of DLL4/Notch pathway-associated factors such as Hey1,NRARP,DLL4,Notch1,Hes1 and Hey2 decreased on P16 in the OIR+D-NAC group(all P<0.01).Conclusion ROS inhibitor D-NAC promotes physiological NV formation in OIR by inhibiting the DLL4/Notch signaling pathway, and restrains the pathological NV formation.
