1.Exploring Mechanisms of Erchentang in Repairing Ileal Immune Barrier and Reducing Weights of Diet-induced Obese Mice Based on Single-cell Transcriptomics

ObjectiveTo investigate the effects of Erchentang (ECD) on the body weight of the mouse model of simple obesity induced by a high-fat diet (HFD) and decipher the underlying mechanisms. MethodsFirstly, single-cell transcriptomics (Sc-RNAseq) was employed to analyze the transcriptional changes in the ileum tissue of mice in the normal group and model group. Then, a mouse model of simple obesity was established with a high-fat diet. The successfully modeled mice were randomly allocated into the following four groups (n=8): model, low-dose (7.5 g·kg-1) ECD, medium-dose (15 g·kg-1) ECD, and high-dose (30 g·kg-1) ECD. Additionally, 8 mice of the same age were selected as the normal group. The body weight was measured at fixed time points during the 4-week gavage period. The overall efficacy of ECD in alleviating obesity was evaluated through glucose tolerance testing, behavioral analysis, hematoxylin-eosin (HE) staining, and biochemical testing. Protein docking was employed to predict the degree of binding between corresponding proteins. Molecular docking was employed to predict the binding degree between key components of ECD and target proteins. Real-time PCR was employed to determine the mRNA levels of tumor necrosis factor-α (TNF-α), inducible nitric oxide synthase (iNOS), interleukin-1β (IL-1β), CD68, CD206, zonula occludens-1 (ZO-1), and Claudin-5 in the ileum. Immunofluorescence staining was used to observe the expression and distribution of Claudin-5 and ZO-1. ResultsThe Sc-RNAseq results indicated that the differentially expressed genes of immune cells in the model group in comparison with the normal group were primarily enriched in biological functions related to lipid metabolism and inflammatory metabolism. Additionally, these genes were associated with the janus kinases(JAK)/signal transducers and activators of transcription (STAT) signaling pathway, an inflammation-related pathway. Compared with the normal group, the model group showed increases in body weight (P<0.01) and blood glucose level (P<0.01), a decrease in limb strength (P<0.01), an increase in liver weight (P<0.05), and elevated serum alanine amino-transferase (ALT) and aspartate transferase (AST) levels (P<0.05, P<0.01). Additionally, the model group exhibited increased hepatic fat vacuoles, notably enlarged adipocytes in the epididymal and inguinal white adipose tissue, and increased inflammation. Compared with the model group, ECD groups showed reduced body weights (P<0.01) and blood glucose levels (P<0.01), increased limb strength (P<0.05, P<0.01), decreased liver weights (P<0.05, P<0.01), and declined serum ALT and AST levels (P<0.05, P<0.01). Additionally, ECD reduced hepatic fat vacuoles and the adipocyte volume in the epididymal and inguinal white adipose tissue, and alleviated inflammation. Potential interactions existed between CD68 and ZO-1/Claudin-5, as well as between CD206 and ZO-1/Claudin-5. The key components of ECD, nobiletin, diosmetin, and naringenin, all demonstrated strong binding affinity with the target proteins ZO-1 and Claudin-5. Compared with the normal group, the model group exhibited up-regulated mRNA levels of the pro-inflammatory cytokines TNF-α, iNOS, IL-1β, and CD68 (P<0.05, P<0.01) and down-regulated mRNA levels of the anti-inflammatory cytokine CD206 (P<0.01) and the tight junction proteins Claudin-5 and ZO-1 (P<0.05, P<0.01). In comparison with the model group, the ECD groups showed down-regulated mRNA levels of TNF-α, iNOS, IL-1β, and CD68 (P<0.05, P<0.01) and up-regulated mRNA levels of CD206, Claudin-5, and ZO-1 (P<0.05, P<0.01). Compared with the normal group, the model group exhibited down-regulated expression of tight junction proteins Claudin-5 and ZO-1 (P<0.01). Compared with the model group, ECD groups showed up-regulated expression of Claudin-5 and ZO-1 (P<0.05, P<0.01). ConclusionECD can significantly ameliorate HFD-induced obesity and excessive body weight gain in mice by improving the inflammatory microenvironment in the ileum and further restoring the integrity of the impaired ileal barrier.
2. Characterization analysis of gM, gL genes of varicella zoster virus in six provinces of China

Objective:
To analyze the genetic characterization of glycoprotein M(gM.),glycoprotein L(gL) of varicella zoster virus.
Methods:
According to the program of "Ministry of Science and Technology of China" , Based on the 12 suspected VZV patients monitored in Beijing (1 case), Shanghai (5 cases), Jilin (2 cases), Qinghai (1 case), Guangdong (2 case) and Sichuan (case) in 2007-2015. A total of 12 Vesicle fluid and throat swab samples were collected. Positive samples were identified by Agarose gel electrophoresis and two glycoprotein genes were amplified by polymerase chain reaction (PCR). Nucleotide sequences were determined and analyzed by PCR amplification of VZV positive specimens V-OKA-BK of the domestic varicella attenuated live vaccine and the Varilrix-1 of the imported attenuated live vaccine. Nucleotide sequences of VZV positive specimens, vaccine strains (V-OKA-BK, varilrix-1) and GenBank foreign wild strains (41 strains), parent strains (P-oka), vaccine strains (V-oka, Varilrix, Varivax) were compared using BioEdit and MEGA 5.0.
Results:
12 specimens were VZV positive. Compared with the vaccine strains and the parent strains, the GM gene of 1 positive specimen had radical mutation at 86686 sites, which resulted in amino acid mutation, 5 positive specimens had base mutation at 87844 sites, and 30 strains of foreign wild strains had the same variation at 87 844 sites. 1 positive specimens of gL gene in 101245 sites had base mutation, and led to amino acid mutation, 6 positive specimens at 101624, 101625, 101626 sites had base of loss and the foreign wild strains in these 3 sites had the same variation. Compared with the vaccine strains, the nucleotide and amino acid homology of gM of 12 VZV positive specimens were 99.2%-100% and 98.2%-100%, respectively, and gL of those were 99.3%-100% and 98.6%-100%, respectively. Compared with 41 strains of foreign wild strains, homology of gM's nucleotides and amino acid were 99.3%-100% and 98.5%-100%, respectively; 99.1%-100% and 98.6%-100% for gL. The results of phylogenetic analysis showed that 7 VZV positive samples were on the same branch with 4 vaccine strains and p-oka strain. Based on gL, 12 VZV positive samples were on the same branch as the vaccine strains and p-oka strain.
Conclusion
This study demonstrates that the genes of gM, gL are highly conserved and remain stable immunogen, which may be involved in the attenuation of VZV and need to be further researched.
3.Effect of different dentin cleaning agents on the bond strength of self-adhesive resin cement to dentin.

OBJECTIVEThis study aims to evaluate the bond strength of a self-adhesive resin cement to dentin by ethylene diamine tetraacetic acid (EDTA) and NaClO.
METHODSTwenty-seven freshly extracted non-carious human premolars were prepared to expose the buccal dentin and randomly divided into three groups: control group (A group), EDTA group (B group) and NaClO group (C group). All teeth were bonded to dentin using a self-adhesive resin cement after the teeth in the A group were processed with distilled water. The B and C group were processed with 3%EDTA and 1%NaClO, respectively. After 24 hours at 37 °C water, the shear bond strengths of the twenty-four specimens were measured. All statistical analysis was performed using SPSS 17.0 software package. Each fractured specimen was examined under dental microscope. Three new specimens were cut, and the morphologies of the cement-dentin interface were observed under scanning electron microscope (SEM).
RESULTSThe shear bond strength in the A group, B group and C group was (8.55±0.63), (8.47±0.56) and (12.97± 0.59) MPa, respectively. The difference between A group and B group was no statistically significant (P>0.05), whereas the difference between C group and B group (or A group) was statistically significant (P<0.05). SEM observation of the cement-dentin interface in the C group showed good adaptation, but resin tags were not observed. The other two groups showed poor bonding interface. Most of the fractured adhesive dentin surfaces exhibited cohesive failure in the A group and B group. All the fractured adhesive dentin surfaces exhibited cohesive failure in the C group.
CONCLUSION1% NaClO can increase the bond strength of self-adhesive resin cement to dentin, but 3%EDTA has no effect.

5.Assessment of occlusal vertical dimension using angles:a pilot study

Objective This study aims to identify highly accurate quantitative methods for evaluating the occlusal vertical dimension by using cephalometric radiographs to analyze the craniofacial hard tissue landmarks in 30 cases with normal occlusions. Methods A total of 30 subjects with normal occlusions and aged between 20 and 30 years old were included in the study. Cephalometric radiographs were obtained and analyzed using the Winceph 8.0 measurement software. Landmark points were identified and measured. Results The mean distribution difference of the center angle (CA) and lower angle (LA) of the face was not statistically significant (t=-1.490, P=0.147). The CA and LA had a significantly linear correlation (r=0.976, P=0.000), and the regression equation was y=0.298+0.997x. Conclusion The CA can be used to assess and quan-tify the occlusal vertical dimension in clinical practice.
6.Correlation between memory function of patients of generalized seizures and magnetic resonance spectroscopy of hippocampal in patients with generalized seizures

Objective To investigate the features of the memory function disturbance in patients of generalized seizures and the correlation between the memory function and proton magnetic resonance spectroscopy(1H-MRS).Methods The clinical memory scale of 45 general seizures and 20 normal age and sex-matched controls wss measured and a1 H-MRs was performed in the bilateral hippoeampal.The levels of NAA.Cho.Cr and the ratios of NAA/Cr+Cho were compared between these two groups.Meanwhile,tHe correlation coefficients was calculated and the results of clinical memory scale scoreB and the level of NAA,Cho.Cr and the ratios of NAA/Cr+Cho in groups were compared.Results As compared with the controls.the scores of pointing memory(17.19±5.86),associational learning(20.00±6.77),image free memory(18.44±6.62),association memory of portrait(13.19±6.62)and memory quotient (MQ,79.07±20.20)excepts insignificant figure recognition wcrc significandy decreased in patients of seizures(t=4.794-10.224,all P<0.01);The scores of scale and MQ of both the group of patients who wok antiepileptic drugs(AEDs)and the group of patients whose frequency exceeded 3 per month were significantly lower than the group who didn't take AEDs and the group whose frequency was lower than 3 per month(t=3.267-6.537,all P<0.01).The volumes of hippocampal formations(2.45 ±0.25 on left and 2.56 ±0.31 on right),the level of NAA(12.93 ±1.73 on left and 11.88 ±1.69 on right),Cho(15.02 ±0.86 on left and 14.94 ±0.96 on right),Cr(11.86 ±0.71 on left and 10.71 ±0.42 on right)and the ratios of NAA/Cr+Cho(0.48 ±0.08 on left and 0.39 ±0.07 on riht)in the group of pailents of generalized seizures were significantly difference than those in the controls(t=4.103-5.768,all P<0.01).Patients with seizures showed significant correlation between tlle scores of all items.MQ in clinical memory scale and NAA,NAA/Cr+Cho ratio(r=0.489-0.727,all P<0.01),and inverse correlation between the scores and MQ and Cho of in the bilateral hippocampal(r=-0.604-0.484,all P<0.01).However,there was no correlation between the scores and MQ and Cr of in the bilateral hippocampal.Condusions The patients of the general seizures do have the problem of the disturbance of short-term memory including acoustic memory and some visual memory.Both the high frequency of seizure and AEDs could affect the short-term memory function.The correlation between the scores of all the items and MQ in clinical memory scale proved that lack of nerve cell and hyperplasia of gliocyte was one of the reasons of the short-term memory disorder in epileptic.
