1.Characteristic Chromatogram and Index Component Contents of Substance Benchmark of Classic Formula Qingwei Power

OBJECTIVE To establish an HPLC characteristic spectrum and content determination method for the reference mate-rial of the classic formula Qingwei Powder,and control its quality to ensure subsequent preparation.METHODS Following ancient books and combining with the investigation of the previous preparation process,15 batches of Qingwei Powder material standards were prepared.HPLC was used to establish the material standard feature map,and the similarity was calculated using the Chinese medicine chromatographic fingerprint similarity evaluation system(2012 version);seven indicator components from five medicinal herbs were used as the reference content determination indicators for Qingwei Powder,and an HPLC content determination method was established for 15 batches of samples.RESULTS The similarity of the characteristic spectra was≥0.975;13 common peaks were calibrated;and 7 common peaks were identified for chemical composition,including catalpol(peak 1),ferulic acid(peak 5),isoferulic acid(peak 6),berberine hydrochloride(peak 7),palmatine hydrochloride(peak 8),paeonol(peak 12),and ligustilide(peak 13).Two sets of indicator component content determination methods have been established.Firstly,the contents of catalpol,ferulic acid,isoferulic acid,and paeonol were determined,and the four component contents were specified to be 1.48-2.76、0.14-0.26、0.48-0.90、1.72-3.19 mg·g-1,respectively;secondly,the contents of coptisine hydrochloride,palmatine hydrochloride,and ber-berine hydrochloride was determined,and it was specified that the content of the three components should be 0.65-1.21、0.57-1.07、1.86-3.45 mg·g-1,respectively.CONCLUSION A quality control method for the reference material of Qingwei Powder has been established through the characteristic spectrum and content determination method.This method is simple and reliable,providing a basis and foundation for subsequent quality control and formulation development.
2.Inhibition of winter seedling raising in greenhouse of premature bolting in Angelica sinensis.

OBJECTIVETo study the inhibition effect of winter seedling raising in sunlight-greenhouse on premature bolting of Angelica sinensis.
METHODOne factor of sowing date with 4 levels was tested at random design with 3 repeats.
RESULTSowing date of winter seedling raising had an important effect on seedling growth, seedling mature, plant growth, premature bolting, yield, quality, and extreme significant effect on yield. The bolting date of winter raised seedlings started in middle of July, 50 d later than tranditional seedlings, and bolting peck date was in the last ten days of July, 40 d later than the traditional seedlings. The highest ratio of premature bolting for winter raised seedlings was 1%, and the lowest was 0. A. sinensis roots sowed in November 28 had the highest yield, root dry ratio, ethanol extract, essential oil and ferulic acid contents compared to that in other sowing dates. The best sowing time was from end of November to middle of December.
CONCLUSIONPremature bolting of A. sinensis could be greatly inhibited by winter seedling raising, end of November to middle of December would be the best sowing time for winter seedling raising in greenhouse.

3.Effects of TCM Preconditioning of Warming Yang and Supplementing Heart Qi on Myocardial Ultrastructure of Rats with Acute Myocardial Ischemia

Objective To observe the effects of TCM preconditioning of warming Yang and supplementing heart Qi on the myocardial ultrastructure of rats with acute myocardial ischemia, and discuss its protection on the ischemia reperfusion myocardium and relation with myocardial ischemic preconditioning. Method Male Wistar rats were randomly divided into 4 groups, 3 of which were fed normally, one of which was administered with TCM, and myocardial infarction model was established after 10 days. IR group were reperfused 48 hours after 45 min of ischemia. TCM group were treated with prophylactic administration of 0.36 g/100 g body weight for 10 days before the same ischemia and reperfusion as IR group. IP group were also performed the same ischemia and reperfusion as IR group after 5 min of ischemia and 5 min of reperfusion three times. The myocardial ultrastructure of rats was observed by electron microscope. Results In IR group:sarcolemma damaged, myofilament dissolved and fragmented, arranged loosely, mitochondria arranged confused, some mitochondria looked like cavitation or partly cavitation, some also presented coacervation state, glycogen decreased. And the myocardium damaged badly, it proved that part of the myocardium damaged after the ischemia reperfusion. In TCM and IP groups:the state of the cardiocytes were improved relatively and the sarcolemmas were integrated, the cardiocytes’ nucleoli presented to be oval-shape, and the nucleoli looked obviously, the mitochondria were showed relatively obviously, myofilaments were well arranged, the bright and dark bands of the sarcomeres looked apparently, mitochondria arranged in good order between the myofibrils, but still some myofilaments were broke and part of the mitochondria presented cavitation state. Conclusion The myocardial ischemic preconditioning and TCM preconditioning of warming Yang and supplementing heart Qi both can protect the ischemic myocardium through mitigating the damages of the myocardial ultrastructure, and they have the similar protection effects on the damaged myocardial ultrastructure of the rats with ischemia reperfusion.
4.Effect of Acupuncture on the Infracted Area and Apoptosis of Rats with Brain Contusion

Objective To observe the effect of acupuncture on the infracted area and apoptosis of rats with brain contusion.Methods Feeney's method was used to establish brain contusion model.The 40 rats were randomized into sham-operation,model,acupuncture and naloxone (NX) group with 10 in each.Acupuncture was performed for the rats of acupuncture group and NX was injected intraperitoneally for the rats of NX group.The treatment was done once everyday,7 days altogether.Then the brain histotomy was prepared after all the treatment finished.Computer image analysis system was used to detect the infracted area.TUNEL method was adopted to detect the apoptosis.Results The brain contusion in the model group was severe,there were lots of apoptotic cells and the infracted areas were bigger than the injured areas (P0.05).Conclusion Acupuncture can decrease the infracted areas and apoptosis of the rats with brain contusion,being effective in anti-secondary brain injuries.
