1.Preparation of monoclonal antibody against bovine viral diarrhea virus and estab-lishment of double antibody sandwich ELISA method

The purpose of this study was to prepare high affinity monoclonal antibodies(mAbs)a-gainst bovine viral diarrhea virus(BVDV)and establish a double antibody sandwich ELISA detec-tion method.BVDV was purified by differential ultracentrifugation and used to immunize BALB/c mice.Hybridoma cells were prepared by fusing spleen cells from the immunized mice with SP2/0 cells.Positive cells were screened by indirect ELISA.A double-antibody sandwich ELISA method for detecting BVDV was developed using monoclonal antibody 4D11 as the capture antibody and HRP-labeled monoclonal antibody 3F3 as the detection antibody.The results of the ELISA and the determination of the variable region gene sequence of monoclonal antibodies indicated that the two monoclonal antibodies recognize different antigenic epitopes.Specificity tests showed that two monoclonal antibodies specifically recognize BVDV and did not cross-react with other bovine viru-ses associated with diarrhea.Indirect immunofluorescence assay and Western blot assay demonstra-ted that both mAbs exhibited strong reactivity with BVDV.The double antibody sandwich ELISA detection method established in this study had good specificity.The sensitivity test revealed that the method could detect a minimum virus amount of 3.1 × 104 TCID50.The reproducibility test showed that the inter-batch coefficient of variation(Cv)was between 2.47%and 7.44%,and the intra-batch Cv was between 1.71%and 9.89%,indicating good reproducibility.The establishment of this method provides an effective technical tool for the rapid diagnosis and prevention and con-trol of BVDV.
2.Evaluation of immune efficacy of Newcastle disease virus mRNA vaccine

SPF chickens were immunized with HN-mRNA vaccine by intramuscular injection.He-magglutination inhibition(HI)test,lymphocyte proliferation test,RT-qPCR and pathological sec-tions were used to evaluate the humoral and cellular immunity and protection against challenge in-duced by the candidate HN-mRNA vaccine.The results showed that 2.5,5.0,and 7.5 μg HN-mR-NA induced HI antibody antibodies in a dose-dependent manner.Among them,the Hi antibody in-duced by 7.5 μg HN-mRNA was slightly higher than that of the weak toxic vaccine(La Sota strain).In addition,in response to the stimulation of inactivated NDV virus,the proliferation and activation of lymphocytes in 2.5,5.0 and 7.5 μg HN-mRNA immunization groups and commercial vaccine group were observed.To further evaluate the antiviral protection provided by HN mRNA immunization,the 105 ELD50 NDV strong strain NA-1 was used to attack the 7.5 μg HN-mRNA immunised group and the commercial weakened vaccine(La Sota strain)and PBS groups using na-sal and ocular drops.The results showed that the survival rate of 7.5 μg HN-mRNA immunization group and commercial vaccine group was 100%,and these vaccines could protect tissues and or-gans from the damage caused by virus infection.At the same time,7.5 μg HN-mRNA and commer-cial vaccine could shorten the time of virus shedding in vitro and the viral load in vivo.This study provides a foundational framework for the clinical application of NDV HN mRNA candidate vac-cines and offers insights for the development of novel mRNA vaccines for poultry.
3.Study on dietary adherence and influencing factors after bariatric and metabolic surgery in obese of patients

Objective:To investigate and analyze the current status of dietary behavior compliance among patients following bariatric metabolic surgery, as well as its associated influencing factors.Methods:A convenience sampling approach was employed to select 149 discharged patients who underwent bariatric metabolic surgery at the Gastric Surgery Department of a tertiary hospital in Fuzhou between August 2021 and February 2024. A WeChat communication group was established for survey participants, and a questionnaire survey was conducted via the mobile app "Wenjuanxing." Patients′ data were collected using the General Information Questionnaire and the Dietary Behavior Compliance Scale for Patients After Bariatric Metabolic Surgery. Multiple stepwise linear regression analysis was used to evaluate the influencing factors of dietary behavior compliance in postoperative patients.Results:A total of 165 questionnaires were distributed, with 149 valid responses retrieved. Among the 149 post-bariatric surgery patients, 37 were males and 112 were females, with an average age of (31.10 ± 7.26) years. The overall level of dietary behavior compliance was (56.91 ± 9.70). Significant differences in dietary behavior compliance were observed among patients of different genders, educational levels, drinking habits, and postoperative durations ( t = 2.17, 1.93, F = 4.27, 3.50, all P<0.05). Specifically, postoperative duration emerged as the primary factor influencing dietary behavior compliance following bariatric metabolic surgery ( t = -2.22, P<0.05). Conclusions:Female patients, those with a college degree or higher, and individuals within 0 to 6 months post-surgery demonstrated better dietary behavior compliance. Medical staff should comprehensively consider patient individual differences, develop, and implement personalized health education plans to enhance patient dietary behavior compliance.
4.Relationship between serum prolactin and mild cognitive impairment in postmenopausal women with type 2 diabetes mellitus

Objective To investigate the relationship between prolactin and mild cognitive impairment(MCI)in postmenopausal women with type 2 diabetes mellitus(T2DM).Methods A total of 319 postmenopausal women with T2DM who were hospitalized in the Department of Endocrinology,Drum Tower Hospital,Affiliated Hospital of Medical School,Nanjing University were enrolled in this study from August 2016 to October 2023.All the patients were divided into two groups according whether they had MCI:T2DM group(n=161)and MCI group(n=158).Differences in clinical characteristics were compared between the two groups.Pearson correlation was used to analyze the correlation between sex hormones and cognitive domains,and Logistic regression analysis was used to evaluate the influencing factors for MCI development.Results Serum prolactin levels were significantly lower in the MCI group than in the T2DM group[(5.5±2.1)vs(7.2±2.9)μg/L,P<0.05].Serum prolactin level was positively correlated with mini-mental state examination score,Montreal cognitive assessment score,immediate memory score,visuopatial constructional score,attention score and hippocampal volume(P<0.05),and negatively correlated with processing speed test(time)and executive function test(time)(P<0.05).Logistic regression analysis demonstrated that serum prolactin level was an influencing factor for the risk of MCI in postmenopausal women with T2DM(OR 0.715,95%CI 0.605~0.845,P<0.01).Conclusions The decrease of serum prolactin level is associated with an increased risk of MCI in postmenopausal women with T2DM.

9.Advances of metabolomics in ocular diseases

Ocular diseases pose a significant challenge to global health. The field of metabolomics, which involves the systematic identification and quantification of metabolites within a biological system, has emerged as a promising research approach for unraveling disease mechanisms and discovering novel biomarkers. Through its application, metabolomics has yielded valuable knowledge pertaining to the initiation and advancement of various ocular diseases. This review presents an overview of metabolomics and examines recent research progess in four ocular diseases, specifically diabetic retinopathy, age-related macular degeneration, glaucoma, and dry eye, summarizing potential biomarkers and metabolic pathways associated with these diseases. Additionally, this review offers insights into the future prospects of utilizing metabolomics for the management and treatment of ocular diseases.
10.Construction and identification of bivalent chimeric virus-like particles of Newcas-tle disease virus,serotype 4 and serotype 8 avian adenovirus

A bivalent chimeric virus-like particle(ND-FAdV-4/8a/8b cVLPs)displaying NDV HN protein,FAdV4 fiber-2 protein,FAdV-8a Fiber protein and FAdV-8b Fiber protein was construc-ted based on insect baculovirus expression system and Newcastle disease virus-like particle vector platform and was identified using indirect immunofluorescence,Western blot and transmission e-lectron microscopy.The results showed that recombinant baculoviruses rBV-c8aFiber and rBV-c8bFiber expressing FAdV-8a Fiber and FAdV-8b Fiber were successfully constructed.In addition,all components of ND-FAdV-4/8a/8b cVLPs were correctly expressed,and ND-FAdV-4/8a/8b cVLPs was a nanoparticle about 100 nm in size,with a capsule membrane and fibers,providing a green and safe virus-like particle vaccine candidate for the control of Newcastle disease and avian adenovirus disease.
