ObjectiveTo investigate the trend and influencing factors of newly reported human immunodeficiency virus (HIV) positivity rate among men who had sex with men (MSM) in Shanghai from 2021 to 2024, and to provide evidence for formulating scientific prevention and control measures of AIDS. MethodsMultiple rounds of cross-sectional questionnaire surveys were conducted among MSM by Shanghai Qing’ai Health Promotion Center. Pearson and Cochran-Armitage trend χ2 tests were used to analyze the differences and changes in population characteristics and newly reported HIV positivity rates. A logistic regression model was applied for multivariate analyses of factors associated with newly reported HIV positivity. ResultsA total of 1 653 MSM who had not been previously diagnosed with HIV infection were surveyed. The newly reported HIV positivity rates in 2021, 2023, and 2024 were 7.87%, 3.91%, and 3.06%, respectively, showing a decreasing trend (χ2trend=13.460, Ptrend<0.001). Multivariate analyses revealed that MSM aged 18‒<25 years, residing locally for <1 year, identifying as bisexual, lacking HIV knowledge, and having ≥10 same-sex partners in the past 6 months exhibited higher newly reported HIV positivity rates. Conversely, MSM knowledgeable about HIV prevention, residing locally for 1‒5 years, and engaging in oral sex with male partners in the past 6 months demonstrated lower HIV positivity rates. Annual analyses revealed that MSM with HIV knowledge had lower newly reported HIV positivity rates in 2023 and 2024 (aOR=0.300, 95%CI: 0.811‒0.111; aOR=0.202, 95%CI: 0.085‒0.483). ConclusionThe newly reported HIV positivity rate among MSM in Shanghai from 2021 to 2024 showed a decline. Future interventions should focus on young and mobile MSM, strengthen HIV knowledge education through platforms such as the internet, promote safe sexual behaviors and regular testing, and further expand the coverage of pre-exposure prophylaxis (PrEP) and post-exposure prophylaxis (PEP) to control HIV transmission within this population.

Ulcerative colitis(UC)is a common chronic non-specific intestinal inflammatory disease clinically. Its pathogenesis is complex, prolonged the disease course, and it is frequent clinical recurrence. In recent years, the incidence of UC has shown an upward trend and tends to younger onset, which seriously affects the quality of life of patients and increases the social medical burden.Mitogen-activated protein kinase(MAPK)plays an important role in the occurrence and development of UC,participating in inflammatory response, oxidative stress, autophagy, apoptosis, pyroptosis, and intestinal barrier.MAPK may be a new target for the treatment of UC.Traditional Chinese medicine has a long history and remarkable curative effect in treating UC,with the advantages of multiple pathways, multiple targets, and multiple components. In recent years, many studies have shown that single Chinese herbal medicines and compound prescriptions can mediate MAPK signaling pathway to inhibit inflammatory response and oxidative stress, regulate autophagy, inhibit apoptosis and pyroptosis, repair the intestinal barrier, and treat UC in many aspects. This article reviews the MAPK signaling pathway, the mechanism by which the MAPK signaling pathway regulates UC,and the research progress of traditional Chinese medicine in treating UC based on the MAPK signaling pathway, hoping to provide theoretical support for the treatment of UC by traditional Chinese medicine and new ideas and references for the research of related new drugs.

IGF-1 is involved in the growth and development of mammals, but its role in sebaceous gland excretion,B16 cell proliferation, and migration in the skin has not been reported yet. This study aims to reveal the function of IGF-1 by detecting its expression in animal skin.Using nano-body screening and purification methods,IGF-1 nanobodies(IGF-1-VHH)were obtained.Using IGF-1-VHH as the primary antibody,Western blot and immunohistochemistry were used to detect the expression of IGF-1 in bovine skin and alpaca acne.IGF-1-VHH was added to melanoma B16 cell culture medium, and CCK-8 and scratch healing methods were used to detect the effects of IGF-1 nanobodies on B16 cell proliferation and migration,as well as their possible molecular mech-anisms.The results showed that the obtained IGF-1-VHH could be applied in immunohistochemis-try and Western blot detection methods, with strong IGF-1 positive expression signals in bovine sebaceous glands and alpaca acne.IGF-1-VHH binds to IGF-1 in cells and has a certain inhibitory effect on the proliferation and migration of B16 cells by regulating the expression of RAS,ERK,and RAF.In summary,IGF-1 maybe involved in the secretion and excretion of sebum in animal skin.IGF-1-VHH inhibits the proliferation and migration of B16 cells by binding to IGF-1,provi-ding a new theoretical basis for ensuring normal physiological function of the skin and clinical di-agnosis and treatment of melanoma.
