1.Correlationship between total proteins SUMOylation and papillary thyroid carcinoma in males

Objective To investigate the relationship between protein SUMOylation level and the prognosis of papillary thyroid carcinoma(PTC)in males.Methods Protein SUMOylation levels in PTC was analyzed by bioinformatics based on GTEx and TCGA databases and validated by immunohistochemical staining and Western blotting in our clinical pairs specimens.The mRNA expression of the protein SUMOylation associated genes were measured by fluorescent quantitative real-time polymerase chain reaction(qRT-PCR)in surgical pairs specimens.Results The expression level of SUMOylation in the tumor tissues of PTC showed an elevated trend(P<0.05),and was associated with poor prognosis of the patients by TCGA and GTEx databases analysis(P=0.021).In the clinical samples of our hospital,it was verified that the level of SUMOylation in tumor tissues was higher than that of the paired non-tumor tissues(P<0.05).However,qRT-PCR showed no significant changes in the transcriptional level of the protein SUMOylation associated genes in most cases.Conclusions Protein SUMOylation in thyroid tumor tissues were higher than that in paired non-tumor tissues,and the higher SUMOylation levels levels in tumor tissues were,the shorter overall survival time of the patients was.
2.Effect of Bobath rehabilitation training on improving upper limb motor function in patients with shoulder-hand syndrome after stroke

Objective To explore the effect of Bobath rehabilitation training on improving upper limb motor function in patients with shoulder-hand syndrome(SHS)after stroke.Methods A total of 110 SHS patients after stroke admitted to Nantong Third People's Hospital(Affiliated Nantong Hospital 3 of Nantong university)from April 2022 to January 2023 were selected as research objects by convenience sampling method.According to the random number method,the patients were divided into research group and control group,with 55 patients in each group.The patients in both groups received basic care.Routine rehabilitation exercise was conducted in the control group.Routine rehabilitation exercise plus Bobath rehabilitation training were given in the research group.Before and after the intervention,the Fugl-Meyer Assessment(FMA)score,the score of motion range of the shoulder,SHS scale(SHSS)score and the Ashworth grade were compared between the 2 groups.Results The FMA score and shoulder flexion,extension and abduction angles of the 2 groups after the intervention were significantly higher than those before the intervention,and the above-mentioned parameters in the research group were significantly higher than those in the control group(P<0.05).The SHSS scores of the 2 groups after intervention was significantly lower than those before intervention,and the SHSS score in the research group was lower than that in the control group(P<0.05).Before intervention,there was no significant difference in the proportion of patients with different Ashworth grades between the 2 groups(P>0.05).After intervention,the proportion of patients with Ashworth grade 0-I+ in the research group was significantly higher than that in the control group(P<0.05).Conclusion Bobath rehabilitation training for SHS patients after stroke can improve the upper limb function and the spasticity of the affected upper limb,and increase the range of motion of shoulder joint.
3.The expression of Rab5 and its effect on invasion, migration and exosome secretion in triple negative breast cancer

Triple-negative breast cancer (TNBC) is the most aggressive subtype of breast cancer and current therapeutic strategies are limited in their effectiveness.The expressions of Rab5 and the M2 tumor-associated macrophage marker CD163 in tissues were detected by Western blot. The migration and invasion of cells were determined using a Transwell assay. The expressions of the exosome markers were evaluated by Western blot. The polarization of human macrophages (THP-1) was determined by incubation of THP-1 cells with conditioned medium or exosomes collected from MDA-MB-231 cells with indicated transfections or by a coculture system of THP-1 and MDA-MB-231 cells. The M1 and M2 macrophage markers were evaluated by qRT-PCR. The expression of Rab5 in TNBC was significantly higher than that in normal breast tissue. Rab5 expressions in triple-negative and luminal A breast cancer were higher than those in other molecular subtypes. Higher CD163 expression was observed in triple-negative breast cancer and in triple-negative and luminal B subtypes. Rab5 knockdown suppressed but Rab5 overexpression promoted the migration and invasion capacity of MDA-MB-231 cells. The levels of CD63 and CD9 in the medium of Rab5 knockdown cells were lower than those in control cells, whereas higher levels of CD63 and CD9 were observed in Rab5 overexpression cells. Rab5 knockdown decreased the excretion but did not alter the diameter of the exosomes. Knockdown of Rab5 facilitated the anti-tumor polarization of macrophages, which was partially reversed by Rab5 overexpression. Therefore, Rab5 is expected to be a potential therapeutic target for triple-negative breast cancer.
4.Expression and evaluation of porcine circovirus type 2 capsid protein mediated by recombinant adenoassociated virus 8

Background:
Porcine circovirus type 2 (PCV2) is an important infectious pathogen implicated in porcine circovirus-associated diseases (PCVAD), which has caused significant economic losses in the pig industry worldwide.
Objectives:
A suitable viral vector-mediated gene transfer platform for the expression of the capsid protein (Cap) is an attractive strategy.
Methods:
In the present study, a recombinant adeno-associated virus 8 (rAAV8) vector was constructed to encode Cap (Cap-rAAV) in vitro and in vitro after gene transfer.
Results:
The obtained results showed that Cap could be expressed in HEK293T cells and BABL/c mice. The results of lymphocytes proliferative, as well as immunoglobulin G (IgG) 2a and interferon-γ showed strong cellular immune responses induced by Cap-rAAV. The enzyme-linked immunosorbent assay titers obtained and the IgG1 and interleukin-4 levels showed that humoral immune responses were also induced by Cap-rAAV. Altogether, these results demonstrated that the rAAV8 vaccine Cap-rAAV can induce strong cellular and humoral immune responses, indicating a potential rAAV8 vaccine against PCV2.
Conclusions
The injection of rAAV8 encoding PCV2 Cap genes into muscle tissue can ensure long-term, continuous, and systemic expression.
5.Applicability of Zebra Fish Thrombosis Model in Antithrombotic Activity Screening of Chinese Materia Medica

Objective To investigate the applicability of zebra fish thrombosis model in antithrombotic activity screening of Chinese materia medica.Methods The living zebra fish thrombosis model was induced by adrenaline hydrochloride. Zebra fish were randomly divided into blank control group, model group, positive medicine group and medication group. Each group was given the corresponding medicine or embryo culture water. O-anisidine staining solution was used to stain and calculate the staining intensity of erythrocytes in zebra fish heart, and quantitative analysis was carried out. The platelet aggregation of transgenic zebra fish was observed and under qualitative analysis. Results Compared with the model group, 100μg/mL salvianolic acid B, 300, 900μg/mL aqueous extract of Salvia miltiorrhiza, 45μg/mL 95% ethanol extract and 400, 1200μg/mL hypothalamus could significantly inhibited the formation of zebra fish thrombosis (P<0.01).ConclusionZebra fish thrombosis model has good applicability in antithrombotic activity screening of Chinese materia medica.
