1.Patent analysis of key technologies for mRNA vaccines
Chinese Journal of Biologicals 2025;38(07):882-889
As a major breakthrough in biomedical field, mRNA vaccine technology has broad application prospects in the prevention and treatment of infectious diseases and tumor treatment. However, mRNA vaccines are a patent-intensive industry, and related core technology patents are mainly controlled by foreign companies. By analyzing the global patent application situation of key technology branches of mRNA vaccines and the differences between domestic and foreign enterprises' products and technology platforms, this paper constructs a patent barrier index model covering patent layout density and claim intensity, and systematically combs the technology competition pattern combined with patent dispute cases of leading enterprises, in order to provide ideas for domestic enterprises to break through patent barriers, optimize research and development paths and risk prevention and control strategies.

BACKGROUND:
To evaluate the efficacy and safety of the first cohort of people in China treated with a responsive neurostimulation system (Epilcure TM , GenLight MedTech, Hangzhou, China) for focal drug-resistant epilepsy in this study.
METHODS:
This multicenter, before-and-after self-controlled study was conducted across 8 centers from March 2022 to June 2023, involving patients with drug-resistant epilepsy who were undergoing responsive neurostimulation (RNS). The study was based on an ongoing multi-center, single-blind, randomized controlled study. Efficacy was assessed through metrics including median seizure count, seizure frequency reduction (SFR), and response rate. Multivariable linear regression analysis was conducted to explore the relationships of basic clinical factors and intracranial electrophysiological characteristics with SFR. The postoperative quality of life, cognitive function, depression, and anxiety were evaluated as well.
RESULTS:
The follow-up period for the 19 participants was 10.7 ± 3.4 months. Seizure counts decreased significantly 6 months after device activation, with median SFR of 48% at the 6th month (M6) and 58% at M12 ( P <0.05). The average response rate after 13 months of treatment was 42%, with 21% ( n = 4) of the participants achieving seizure freedom. Patients who have previously undergone resective surgery appear to achieve better therapeutic outcomes at M11, M12 and M13 ( β <0, P <0.05). No statistically significant differences were observed in patients' scores of quality of life, cognition, depression and anxiety following stimulation when compared to baseline measurements. No serious adverse events related to the devices were observed.
CONCLUSIONS:
The preliminary findings suggest that Epilcure TM exhibits promising therapeutic potential in reducing the frequency of epileptic seizures. However, to further validate its efficacy, larger-scale randomized controlled trials are required.

Objective To explore the relationship between triglyceride-glucose index(TyG)and acute ischemic stroke with large vessel occlusion(AIS-LVO)of anterior circulation. Methods A retrospective study was conducted on patients with anterior circulation AIS-LVO who underwent emergency endovascular thrombectomy at Neurovascular Center of The First Affiliated Hospital of Naval Medical University from Jan.2018 to Dec.2019.According to modified Rankin scale(mRS)score 90 d after operation, the patients were assigned to favorable outcome group(mRS score 0-2)or unfavorable outcome group(mRS score 3-6),and the TyG was compared.According to the median of TyG,the patients were assigned to low-TyG group(TyG<8.57)or high-TyG group(TyG ≥8.57),and the clinical data, laboratory indexes, and imaging characteristics were compared.Receiver operating characteristic curve was used to evaluate the predictive value of TyG for poor prognosis. Results A total of 135 patients were enrolled, with 72 in the favorable outcome group and 63 in the unfavorable outcome group. The TyG of the unfavorable outcome group was significantly higher than that of the favorable outcome group(8.82+0.63 vs 8.43+0.60,P<0.001).There were 67 patients in the low-TyG group and 68 in the high-TyG group.Compared with the low-TyG group, the proportion of patients with hyperlipidemia history(P=0.003),systolic blood pressure at admission(P=0.018),fasting blood glucose level(P<0.001),and triglyceride level(P<0.001)were significantly higher in the high-TyG group, the infarct core volume was significantly larger(P=0.025),the high density lipoprotein-cholesterol level was significantly lower(P=0.013),and the mRS score 90 d after operation was significantly higher(3[1,5]vs 1[0,5],P=0.049).The TyG had certain predictive value for poor prognosis in anterior circulation AIS-LVO patients(area under curve value=0.662,95%confidence interval 0.571-0.753).Conclusion TyG is elevated in anterior circulation AIS-LVO patients with poor prognosis, and may be a potential prognostic indicator for anterior circulation AIS-LVO patients.

With the development of medical technology and interventional equipment, neurointervention has become an important treatment method for cerebrovascular diseases.Intravascular stenting plays a crucial role in the treatment of cerebral artery stenosis and intracranial aneurysms, and its clinical efficacy and safety are constantly improving. However,as the neurointervention enters the mature stage, the long-term effect of the interventional materials placed within the brain and the problem of the non-degradable nature of the stent material have got obvious concern in clinical practice. The application of biodegradable materials holds promise for solving this dilemma,as their degradation products can be absorbed or excreted by the body. The development history of the biodegradable stents shows that biodegradable stents began to be studied as early as in the 1980s.Due to the features of intracranial arteries, the stents used for intracranial stenosis have to meet higher clinical requirements in terms of flexibility, precision and the influence of degradation products, which are still studied by animal experiment and preliminary clinical research.Flow-diverter devises have already achieved definite clinical effect.Due to the disadvantages of conventional metal stents, such as artifacts, which have a long-term influence on the imaging quality, the degradable flow-diverter devices have become a current research hot spot, the materials include metal,metal-polymer mixture, polymer, etc.Researchers are still exploring the material selection, control of degradation time and optimization of pore density. In conclusion, biodegradable stents used for intracranial stenosis and biodegradable flow-diverter devices have broad application prospects, although more studies are need to be conducted before biodegradable stents can be widely used in clinical practice.

γδ T cells are a kind of innate immune T cell. They have not attracted sufficient attention because they account for only a small proportion of all immune cells, and many basic factors related to these cells remain unclear. However, in recent years, with the rapid development of tumor immunotherapy, γδ T cells have attracted increasing attention because of their ability to exert cytotoxic effects on most tumor cells without major histocompatibility complex (MHC) restriction. An increasing number of basic studies have focused on the development, antigen recognition, activation, and antitumor immune response of γδ T cells. Additionally, γδ T cell-based immunotherapeutic strategies are being developed, and the number of clinical trials investigating such strategies is increasing. This review mainly summarizes the progress of basic research and the clinical application of γδ T cells in tumor immunotherapy to provide a theoretical basis for further the development of γδ T cell-based strategies in the future.
