1.Application of laser microporous decellularized scaffolds in tissue regeneration

BACKGROUND:In the field of tissue engineering,decellularized scaffolds are widely used owing to their biocompatibility.Nonetheless,these scaffolds frequently encounter the limitation of inadequate cell infiltration due to their densely structured composition.Using laser micro-patterning technology for structural modification can improve cell adhesion and proliferation.OBJECTIVE:To summarize the research progress of laser microporous decellularized scaffolds in tissue regeneration.METHODS:Related literature was searched in PubMed and CNKI databases.The Chinese and English search terms were"decellularized scaffold,laser micro-patterning technology,laser microporous decellularized scaffold,extracellular matrix,laser drilling,tissue regeneration."References were screened according to inclusion and exclusion criteria,and 65 articles were finally included for analysis and discussion.RESULTS AND CONCLUSION:(1)Laser micro-patterning technology can selectively remove tissues to achieve scaffold modification.Laser treatment can have controllable effects on the pore structure,mechanical properties and cell permeability of the scaffold.(2)To obtain the optimal cell integration and remodeling effect,factors such as scaffold pretreatment,laser and decellularization process sequence,laser parameter setting,decellularization process and pore structure should be considered.(3)At present,the application of laser microporous decellularized scaffolds has shown that it can promote tissue repair,and it is also necessary to solve the problem of tissue thermal damage.Depending on the absorption of different wavelengths of laser light by the tissue,a reasonable selection of laser type is a feasible solution.(4)Tissue damage repair by laser microporous decellularized scaffold is expected to achieve clinical transformation,and optimizing laser technology and carrying out larger scale biosafety testing are future research directions.

3.Clinical efficacy and mechanism of Zishen Shujing Decoction in treating premature ovarian failure with kidney deficiency and liver depression

Objective To observe the clinical efficacy of Zishen Shujing decoction in the treatment of kidney deficiency and liver depression type premature ovarian failure(POF).Methods A total of 70 female POF patients who visited the Fourth Clinical Medical College of Guangzhou University of Traditional Chinese Medicine from July 2023 to April 2024 were selected as the research subjects,they were divided into traditional Chinese medicine(TCM)treatment group(n=35)and Western medicine treatment group(n=35)by using a random number table method.Western medicine treatment group was treated with femostone,and TCM treatment group was treated with Zishen Shujing decoction.Differences of TCM syndrome scores and clinical efficacy were compared between two groups before and after treatment,and changes of peripheral blood follicle-simulating hormone(FSH),anti-Müllerian hormone(AMH),anal follicle count(AFC)and interleukin(IL)-4,IL-6 before and after treatment were compared.Results The effective rate of TCM treatment group was higher than that of Western medicine treatment group,the difference was statistically significant(P<0.05).The TCM syndrome scores after treatment were lower than before treatment,and TCM treatment group was lower than Western medicine treatment group,the difference was statistically significant(P<0.05).After treatment,FSH levels in two groups were lower than before treatment,AMH and AFC levels were higher than before treatment,and improvement of hormone levels,AMH and AFC in TCM treatment group was better than that in Western medicine treatment group,with statistical significance(P<0.05).After treatment,the anti-inflammatory factors IL-4 were increased,and the pro-inflammatory factors IL-6 were decreased,and the increase effect of anti-inflammatory factors and the decrease effect of pro-inflammatory factors in TCM treatment group were better than those in Western medicine treatment group with statistical significance(P<0.05).Conclusion Zishen Shujing decoction can improve the clinical symptoms,serum hormone levels of POF patients with kidney deficiency and liver depression,improve ovarian function,reduce immune inflammation,reduce oxidative stress,and effectively improve the quality of life of patients.
4.Effects of treadmill running on hippocampal sigma-1 receptor and endoplasmic reticulum stress pathway in chronically stressed rats

Objective:To investigate the possible mechanism of treadmill exercise(TR)in improving cognitive function of rats with chronic unpredictable mild stress(CUMS)by detecting pathological changes and the expression of sigma-1 receptor(SIR),endoplasmic reticulum stress-related proteins in hippocampus.Methods:30 SD rats were ran-domly allocated into control group(n=10),CUMS group(n=10)and CUMS exercise group(CUMS+TR,n=10).Subsequently,depressive model was established in rats of CUMS group and CUMS+TR group,meanwhile,CUMS+TR group rats were subjected to 4 weeks of moderate intensity treadmill exercise(20 m/min).After the CUMS and tread-mill exercise,the eight-arm radial maze test(RAM)was carried out to evaluate the learning-memory functions.Subse-quently,the pathological changes of neurons in the hippocampus of rats were measured by HE staining,the protein expression of the GRP78,IRE1,XBP1,CHOP,caspase-3 and SIR in the hippocampus were measured by Western blot.Results:Compared with the control group,the time of finishing the eight-arm radial maze prolonged and the num-ber of total memory errors increased(P<0.05)in the RAM of the CUMS group rats.The morphology and structure of neurons in the hippocampus of CUMS group rats showed obvious pathological changes,the protein expression levels of GRP78,IRE1,XBP1,CHOP and caspase-3 significantly increased and SIR protein expression decreased(P<0.05)in hippocampus in the CUMS group compared with the control group rats.4-week treadmill exercise significantly im-prove the learning and memory ability and pathological changes of neurons in hippocampus of CUMS+TR group rats.Compared with CUMS group rats,the protein expression levels of GRP78,IRE1,XBP1,CHOP and caspase-3 signifi-cantly decreased,the SIR expression in hippocampus significantly increased in the CUMS+TR group rats(P<0.05).Conclusion:Treadmill exercise can improve cognitive impairment in CUMS rats,and its mechanism may be related to the up-regulation of SIR expression and the inhibition of ERS-mediated apoptosis pathway in hippocampus.
5.Effects of treadmill running on hippocampal sigma-1 receptor and endoplasmic reticulum stress pathway in chronically stressed rats

6.Clinical efficacy and mechanism of Zishen Shujing Decoction in treating premature ovarian failure with kidney deficiency and liver depression

7.The design and application of a bleeding alarm bandage for maintenance hemodialysis patients

Objective To design and produce a bleeding alert bandage and investigate its effectiveness in patients undergoing hemodialysis.Methods The bleeding alert bandage consists of elastic bandage,monitoring element and monitoring box.A convenience sampling method was used to select patients who received regular hemodialysis treatment in the blood purification center of a tertiary A comprehensive hospital in Anhui Province from January 2021 to June 2021.These patients were randomly assigned into an experimental group and a control group,with 40 cases in each group.Patients in the control group were inspected every 30 min by traditional method.Patients in the experimental group were monitored in real-time with a bleeding alert bandage in addition to the 30 min rounds.The detection rate,incidence,and volume of bleeding at the puncture site were compared in 2 groups of patients.Results The incidence of bleeding from the puncture sites was 17.00%in the experimental group and 25.25%in the control group,with statistical significance(x2=8.170,P=0.004).The incidence of bleeding for severe bleeding from puncture sites was 1.50%in the experimental group and 11.50%in the control group,with statistical significance(x2=32.910,P<0.001).The bleeding detection rate from puncture sites was 94.12%in the experimental group and 89.11%in the control group,with no statistical difference(x2=1.261,P=0.262).The detection rate for moderate bleeding from puncture sites was 100%in the experimental group and 85.37%in the control group,with statistical significance(x2=6.170,P=0.013).Additionally,the bleeding volume from the puncture site was 1.29(0.88,1.74)g in the experimental group and 2.77(1.60,4.77)g in the control group,with statistical significance(Z=-5.825,P<0.001).The moderate bleeding volume from the puncture site was 1.49(1.27,1.81)g in the experimental group and 1.92(1.52,2.44)g in the control group,with statistical significance(Z=-2.835,P=0.005).Conclusion The bleeding alert bandage is effective in real-time monitoring and alerting of bleeding at the puncture sites during hemodialysis,which can help reduce the incidence of blood seepage,improve the detection rate of blood seepage,reduce the amount of bleeding and improve the safety of dialysis treatment.
8.Trends of Incidence and Age at Onset of Uterine Corpus Cancer in Jiangsu Cancer Registration Areas from 2009 to 2019

[Purpose]To investigate the trends of incidence and age at onset of uterine corpus can-cer in Jiangsu cancer registration areas from 2009 to 2019.[Methods]The continuous monitoring data of uterine corpus cancer from 2009 to 2019 were collected from 16 cancer registries in Jiang-su Province.The crude incidence rate,the age-standardized incidence rate by Chinese standard population(ASIRC),crude and adjusted mean age,and standardized age-specific incidence composition were calculated.The average annual percentage change(AAPC)were analyzed by the Joinpoint regression model.The linear regression model was used to analyze the relationship be-tween mean age at onset and year.The standardized age-specific incidence composition in 2009 and 2019 were compared.[Results]The ASIRC of uterine corpus cancer in all registration areas and in rural areas of Jiangsu Province showed upward trends with AAPC of 1.78%and 2.38%,re-spectively(P<0.05),but not showed in the urban areas(AAPC=1.30%,P>0.05).The crude mean age at onset increased from 56.48 years old in 2009 to 58.26 years old in 2019 with an average annual growth of 0.173 years old(P=0.001).After the population structure standardized,the trends disappeared in all registration areas.[Conclusion]From 2009 to 2019,the standardized incidence rates of uterine corpus cancer were on rise in Jiangsu cancer registration areas,especially in the age group of 50 to 59 years old.

10.Clinical and genetic analysis of a child with early-onset severe obesity.

OBJECTIVE:
To explore the clinical phenotype and genetic etiology of a child with early-onset severe obesity.
METHODS:
A child who presented at the Department of Endocrinology, Hangzhou Children's Hospital on August 5, 2020 was selected as the study subject. Clinical data of the child were reviewed. Genomic DNA was extracted from peripheral blood samples of the child and her parents. Whole exome sequencing (WES) was carried out on the child. Candidate variants were verified by Sanger sequencing and bioinformatic analysis.
RESULTS:
This child was a 2-year-and-9-month girl featuring severe obesity with hyperpigmentation on the neck and armpit skin. WES revealed that she has harbored compound heterozygous variants of the MC4R gene, namely c.831T>A (p.Cys277*) and c.184A>G (p.Asn62Asp). Sanger sequencing confirmed that they were respectively inherited from her father and mother. The c.831T>A (p.Cys277*) has been recorded by the ClinVar database. Its carrier frequency among normal East Asians was 0.000 4 according to the 1000 Genomes, ExAC, and gnomAD databases. Based on the guidelines from the American College of Medical Genetics and Genomics (ACMG), it was rated as pathogenic. The c.184A>G (p.Asn62Asp) has not been recorded in the ClinVar, 1000 Genomes, ExAC and gnomAD databases. Prediction using IFT and PolyPhen-2 online software suggested it to be deleterious. Based on the guidelines from the ACMG, it was determined as likely pathogenic.
CONCLUSION
The c.831T>A (p.Cys277*) and c.184A>G (p.Asn62Asp) compound heterozygous variants of the MC4R gene probably underlay the early-onset severe obesity in this child. Above finding has further expanded the spectrum of MC4R gene variants and provided a reference for the diagnosis and genetic counseling for this family.
