1.Establishment and evaluation of renal fibrosis model induced by different doses of adenine in mice

Objective To compare the effects of different doses of adenine(ADE)by oral gavage in a mouse model of renal fibrosis(RF),and to provide a more suitable mouse model for further RF research.Methods Thirty C57BL/6J Nifdc mice were divided randomly into a control group,low-dose ADE group(ADE-L group,50 mg/kg),and high-dose ADE group(ADE-H group,100 mg/kg),and the general condition,mortality,and body mass changes of the mice were observed.Serum creatinine(CREA),blood urea nitrogen(BUN),and uric acid(UA)were measured on day 30 to evaluate renal function.Histopathological changes in kidney tissues were observed by hematoxylin-eosin(HE)and Masson staining,and expression levels of fibronectin(FN),collagen Ⅰ(COL Ⅰ),and alpha smooth muscle actin(α-SMA)in kidney tissues were detected by Western Blot.Results The cumulative mortality rates in the ADE-L and ADE-H groups were 91.7%and 58.3%,respectively.The body mass of mice in the ADE-L group was similar to that in the control group(P>0.05),but the body mass of mice in the ADE-H group was significantly lower than that of mice in the control and ADE-L groups from day 3 to day 30(P<0.001).Day 30 CREA and BUN levels in the ADE-L group were similar to those in the blank group,but CREA and BUN levels were significantly increased in the ADE-H group(P<0.001).There were no significant differences in UA levels among all groups.Inflammatory infiltration and tubular dilatation were observed in the ADE-L group on day 30,accompanied by tubular epithelial necrosis,while crystal accumulation in the tubular lumen and interstitium was observed in the ADE-H group,and the degree of interstitial fibrosis was significantly higher than in the control group(P<0.01).Expression levels of interstitial fibrosis-related proteins in the ADE-H group on day 30 were significantly higher than in the control and ADE-L groups(P<0.05,P<0.01 or P<0.001).Conclusions Both 50 and 100 mg/kg of ADE can be used to establish a mouse model of RF,with different doses leading to varying degrees of renal injury.Mice in the ADE-L group developed mild interstitial fibrosis on day 30,while mice in the ADE-H group developed moderate to severe interstitial fibrosis.
2.Prokaryotic expression and polyclonal antibody preparation of N protein of PD-CoV

The synthetic PDCoV N protein gene was optimized and cloned into the pET-30a vector to obtain the pET-30a-N plasmid.Thenthe recombinant plasmid was transformed into three strains of BL21 E.coli using heat-shock to explore protein expression conditions.The expressed proteins was purified using Ni Focurose 6FF(IMAC)and used as antigen to immunize the New Zealand White rabbit to prepare the polyclonal antibody against the PDCoV N protein.The antibody titer was measured by indirect ELISA method.The specificity for the antibody was identified by West-ern blot and indirect immunofluorescence(IFA).The results showed that the pET-30a-N plasmid showed high expression level in BL21 StarTM(DE 3).The optimal expression condition was 37 ℃ 4 h.The purity of the target protein could reach 90.3%after purification.Indirect ELISA showed that the antibody titers was up to 1∶204 800.Western blot and IFA showed that the produced rabbit polyclonal antibody exhibited good specificity.In conclusion,the polyclonal antibody was prepared which specifically recognized the PDCoV N proteins.The results provided some references for the subsequent exploration of PDCoV N protein function and laid a foundation for establishing a diag-nostic method for PDCoV.
3.Research progress on animal models of imiquimod-induced psoriasis

Psoriasis is a chronic inflammatory skin disease with worldwide prevalence,primarily characterized by epidermal hyperplasia,abnormal keratinization,and immune cell infiltration,with a significant negative impact on patients' quality of life and mental well-being.The onset of psoriasis is closely associated with genetic susceptibility,immune dysregulation,and environmental factors.Despite research progress into the pathogenesis of psoriasis,existing treatment method still face problems including limited efficacy and obvious side effects.There is thus an urgent need for an in-depth analysis of its pathological network and the development of novel interventional strategies.The imiquimod-induced psoriasis animal model has accordingly become a crucial tool for studying psoriasis owing to its high reproducibility and excellent pathological simulation.This review systematically summarizes the core mechanism of action of the imiquimod-induced psoriasis model,expounds on the molecular basis of its action via pathways such as the cascade reaction of the core immune-inflammatory axis,the multi-regulatory network of downstream synergistic mechanisms,and the interaction between host and environmental factors.Research based on this model has successfully verified the therapeutic effects of various targeted therapies and natural products on psoriasis,demonstrating its important application value in therapeutic interventional research.We also discuss the limitations of the imiquimod-induced psoriasis model,and indicate future research directions,with the aim of providing references for further in-depth research and the treatment of psoriasis.
4.Influence of Thrombolysis Therapy Before PCI on Long-Term Left Ventricular Global and Regional Function in ST Elevation Myocardial Infarction Patients by Cardiac Magnetic Resonance

Purpose To explore the influence of thrombolysis therapy before percutaneous coronary intervention(PCI)on long-term left ventricular global and regional function in ST segment elevation myocardial infarction(STEMI)patients by cardiac magnetic resonance(CMR).Materials and Methods A retrospective analysis was conducted on 67 STEMI patients who were enrolled in a prospective study from November 2021 to August 2022 in the First Affiliated Hospital with Nanjing Medical University,the First People's Hospital of Lianyungang,the Affiliated Hospital of Jiangnan University,Changzhou No.2 People's Hospital and Huai'an Second People's Hospital and underwent CMR examination one year later.STEMI patients were divided into thrombolysis group and non-thrombolysis group according to whether a single half-dose of 5 mg recombinant staphylokinase(r-SAK)was given within two hours before the first medical contact and PCI.Based on CMR cine images,the traditional left ventricular function,global and segmental functional parameters were measured,and the differences between the two groups were compared.According to the degree of late gadolinium enhancement involvement,myocardial segments were divided into the following four types:transmural infarcted segments,non-transmural infarcted segments,locally infarcted segments,and non-infarcted segments.The parameters of the two groups were compared across these different segments.Results At the patient level,the cardiac index,left ventricular wall thickening and left ventricular wall motion in r-SAK thrombolysis group were higher than those in non-thrombolysis group(t/Z=-2.426,-4.307,-2.735,all P<0.05).At the segment level,compared with non-thrombolysis group,patients received r-SAK before primary PCI showed greater segmental radial strain in non-transmural infarcted segments(Z=-2.117,P=0.034);larger segmental wall motion in locally infarcted segments(Z=-2.235,P=0.025),and better segmental circumference strain in the non-infarcted segments(Z=-3.869,P<0.001).Conclusion The thrombolytic therapy before PCI in STEMI patients retain better long-term left ventricular global and regional function evaluating by CMR.

6.Evaluation of the Quality Management Effectiveness of a Digital Risk Prevention and Control Platform for Venous Thromboembolism

Objective To evaluate the efficacy of the venous thromboembolism(VTE)digital risk prevention and control platform based on whole-process closed-loop management for the quality management of VTE in Peking University Shenzhen Hospital.Methods The data of 360 718 hospitalized patients from 2021 to 2024 were collected to analyze the rate of VTE risk assessment,the proportion of VTE intermediate and high-risk patients,the rate of VTE prophylaxis implementation(physical/pharmacological),and the VTE outcome indexes,and the differences between groups were verified using the Cochran-Armitage χ2 trend test.Results The VTE initial risk assessment rate,the implementation rate of VTE physical prophylaxis,and the rate of VTE pharmacological prophylaxis all showed an upward trend;the incidence rate of hospital correlated VTE,and the incidence rate of VTE in hospitalized surgical patients all showed a downward trend.Conclusion The establishment of the VTE digital risk prevention and control intelligent platform can effectively improve the level of standardized prevention and treatment of in-hospital VTE and the efficacy of quality management,and provide a digital management paradigm for the implementation of the national healthcare quality and safety improvement goals and the monitoring indicators of surgical quality and safety improvement actions.

8.Efficacy Mechanism of Xianlian Jiedu Prescription Against Colorectal Cancer Recurrence vias Regulating Angiogenesis

ObjectiveTo explore effect of Xianlian Jiedu prescription on the recurrence of colorectal cancer (CRC) and investigate the related mechanisms. MethodsA postoperative recurrence model was established in 25 Balb/c mice by injecting CT26 cells subcutaneously into the armpit, followed by surgical removal of 99% of the subcutaneous tumor. The mice were randomly divided into model group, low-dose Xianlian Jiedu prescription (XLJDP-L) group (6.45 g·kg-1·d-1), medium-dose Xianlian Jiedu prescription (XLJDP-M) group (12.9 g·kg-1·d-1), high-dose Xianlian Jiedu prescription (XLJDP-H) group (25.8 g·kg-1·d-1), and 5-fluorouracil (5-FU) group (1×10-3 g·kg-1·d-1). The mice were euthanized after 14 days of continuous intervention, and recurrent tumor tissue was harvested. Hematoxylin and eosin (HE) staining was used to observe pathological and morphological changes in the recurrent tumor tissue. Immunohistochemistry (IHC) was employed to assess the expression of proliferating cell nuclear antigen (Ki67), vascular endothelial growth factor (VEGF), and platelet-endothelial cell adhesion molecule (CD31) in recurrent tumor tissue. The Western blot was used to detect the protein expression levels of angiopoietin-2 (ANG-2), VEGF, phosphorylated-protein kinase B (p-Akt), protein kinase B (Akt), phosphorylated-phosphatidylinositol 3-kinase (p-PI3K), and phosphatidylinositol 3-kinase (PI3K) in recurrent tumor tissue. ResultsBefore treatment, there were no statistical differences in tumor volume, tumor weight, and body mass among the XLJDP-L, XLJDP-M, and XLJDP-H groups and the 5-FU group compared to the model group, indicating model stability. After treatment, compared with those in the model group, the tumor volume and tumor weight in the XLJDP-L, XLJDP-M, and XLJDP-H groups and the 5-FU group were significantly reduced (P<0.01), showing dose dependency. Meanwhile, there were no significant differences in body weight among the XLJDP-L, XLJDP-M, and XLJDP-H groups and the 5-FU group compared to the model group. HE staining showed that compared with that in the model group, tumor tissue in the XLJDP-L, XLJDP-M, and XLJDP-H groups and the 5-FU group had loosely arranged cells, increased intercellular spaces, small and shriveled nuclei, light staining, fewer mitotic figures and atypical nuclei, and increased necrotic areas. IHC showed that compared with those of the model group, the positive rates of Ki67, VEGF, and CD31 in the recurrent tumor tissue of the XLJDP-L, XLJDP-M, and XLJDP-H groups and the 5-FU group were significantly reduced (P<0.01) in a dose-dependent manner. Western blot results showed that compared with those of the model group, the protein expression levels of ANG-2 and VEGF in the recurrent tumor tissue of the XLJDP-L, XLJDP-M, and XLJDP-H groups and the 5-FU group were significantly downregulated (P<0.05, P<0.01), and the p-Akt/Akt and p-PI3K/PI3K ratios were significantly decreased in a dose-dependent manner (P<0.05, P<0.01). ConclusionXianlian Jiedu prescription significantly inhibits the recurrence of CRC in mice after subcutaneous tumor surgery. The mechanism may involve regulating the PI3K/Akt pathway and downregulating key angiogenic proteins such as ANG-2, VEGF, and CD31.
9.Longitudinal analysis of the functional development of elderly Chinese using CHARLS data with the ICF-RS classification structure

Objective:Based on the functional classification structure of the International Classification of Functioning, Disability and Health′s rehabilitation set (ICF-RS), the data available in the open database of the China Health and Retirement Longitudinal Study (CHARLS) were analyzed longitudinally to construct the development trajectories of different functional dimensions among China′s elderly population.Methods:The CHARLS data for 2013, 2015 and 2018 describing persons aged 60 and older were distributed using the ICF-RS′s classification structure. Group-based multi-trajectory modeling (GBMTM) was then applied to analyze the characteristics of the development trajectories of physical functioning, daily activities and social participation.Results:Different development trajectory subtypes emerged from the data. A gradual decline in physical functioning was evident throughout the study period, while daily activities first improved and then declined. Social participation was in decline throughout the period. There were, however, significant differences in the initial functional level, the degree of decline and the degree of decline reversal among the different subtype groups.Conclusions:The overall functional performance of the elderly in China shows a downward trend, but some functional dimensions can still improve with time, at least during the specific period covered in this open database. It is recommended that the ICF-RS be integrated into the CHARLS to comprehensively assess the classification of function and developmental changes among China′s elderly.
10.Building a "six-in-one" community cough and asthma center under a "dual-contract physician" model: a preliminary evaluation

This report evaluates the preliminary outcomes of a "six-in-one" integrated cough and asthma center developed under a dual-contract physician model at the Changfeng Community Health Service Center in Putuo District, Shanghai. By combining the efforts of family doctors and medical specialists, the center integrated six core functions-clinical treatment, prevention, nursing, rehabilitation, pharmacy, and nutrition-into a seamless management system covering screening, diagnosis, therapy, and follow-up. Supported by specialist guidance and teaching clinics, the model significantly enhanced comprehensive respiratory disease management capabilities within the community setting. The initiative not only improved patient health outcomes but also strengthened multidisciplinary collaboration and resource efficiency, offering a replicable example for improving chronic disease management in primary care through integrated and coordinated service delivery.
