1.Preliminary Study on the Relationship between Cancer Testis Antigen ACRBP and Immune Infiltration of Liver Cancer

Objective To explore the expression of cancer-testis antigen ACRBP in liver cancer and its correlation with immune infiltration.Methods Firstly,UALCAN and HPA databases were used to analyze the expression of ACRBP mRNA and protein both in liver cancer and normal liver tissues.The correlation between the expression of ACRBP in liver cancer and immune cell infiltration,and immune regulation genes were analyzed based on TCGA data,TIMER database,CIBERSOFT tool and TISIDB database.Then,human peripheral blood mononuclear cells(PBMC)were isolated and co-cultured with liver cancer cells with down-regulated ACRBP expres-sion.The secretion of immune molecules was measured by ELISA,and the apoptosis of PBMC was detected by flow cytometry.Results Bioinformatics analysis revealed that the expression of ACRBP mRNA and protein in liver cancer tissues was significantly higher than those in normal liver tissues.The expression of ACRBP mRNA was positively correlated with Treg cell infiltration,immunosuppressive mole-cules,chemokines and other immunoregulatory genes.Compared with normal liver tissues,liver cancer tissues with high expression of ACRBP was significantly reduced in the infiltration of Plasma cells,γδT cells,NK cells and M2 macrophages.In vitro,by down-regu-lateing the expression of ACRBP in liver cancer cells,the secretion of TNF-α,IFN-γ and IL-2 was increased,and the apoptosis of PBMC was also increased.Conclusion The expression of ACRBP in live cancer is associated with the infiltration of immune cells.ACRBP participates in the regulation of immune cells and influences the tumor microenvironment,laying the foundation for the future de-velopment of immunotherapy targeting ACRBP.
2.Correlation of SERPINC1 and SERPINE1 gene polymorphisms with venous thromboembolism in patients with malignant tumors

Objective:To investigate the correlation of SERPINC1 and SERPINE1 gene polymorphisms with venous thromboembolism (VTE) in patients with malignant tumors.Methods:A retrospective case-control study was conducted. A total of 227 patients with malignant tumors in the Cancer Hospital Affiliated to Xinjiang Medical University from November 2023 to February 2024 were selected, of which 47 cases developed VTE (VTE group) and 180 cases did not develop VTE (non-VTE group). The patients' venous blood was collected, and D-dimer level was analyzed by using fully automatic coagulation analyzer; fluorescence staining was performed by using digoxin staining solution, and SERPINC1 rs2227589 locus and SERPINE1 rs1799762 locus were used as candidate genes, and fluorescence sequencing was performed by using multichannel fluorescence quantitative analyzer, and the frequencies of C, T, 4G and 5G genes were calculated.Results:There were no statistically significant differences in the age, gender, smoking, alcohol consumption, anticoagulant use, diabetes mellitus, hypertension, coronary artery disease, surgical treatment, intravenous cannulae, radiotherapy, chemotherapy, and targeted therapy between the 2 groups (all P > 0.05). The level of D-dimer in the VTE group was higher than that in the non-VTE group [(8.7±6.9) kg/m 2vs. (2.8±1.0) kg/m 2], and the difference was statistically significant ( t = 5.15, P < 0.001). The differences in C and T genotypes and gene frequencies at the SERPINC1 rs2227589 locus between the VTE group and the non-VTE group were not statistically significant (all P > 0.05). The differences in 4G and 5G genotypes and gene frequencies at the SERPINE1 rs1799762 locus between the VTE group and the non-VTE group were statistically significant (all P < 0.05), and 4G allele frequency in the VTE group was higher than that in the non-VTE group (52.13% vs. 39.72%), and the difference was statistically significant ( χ2 = 4.70, P = 0.030). Conclusions:The elevated expression of 4G allele at the SERPINE1 rs1799762 locus in patients with malignant tumors is associated with development of VTE.

Background and Objectives:
The Fractional Flow Reserve and Intravascular UltrasoundGuided Intervention Strategy for Clinical Outcomes in Patients with Intermediate Stenosis (FLAVOUR) trial demonstrated non-inferiority of fractional flow reserve (FFR)-guided percutaneous coronary intervention (PCI) compared with intravascular ultrasound (IVUS)-guided PCI. We sought to investigate the cost-effectiveness of FFR-guided PCI compared to IVUS-guided PCI in Korea.
Methods:
A 2-part cost-effectiveness model, composed of a short-term decision tree model and a long-term Markov model, was developed for patients who underwent PCI to treat intermediate stenosis (40% to 70% stenosis by visual estimation on coronary angiography).The lifetime healthcare costs and quality-adjusted life-years (QALYs) were estimated from the healthcare system perspective. Transition probabilities were mainly referred from the FLAVOUR trial, and healthcare costs were mainly obtained through analysis of Korean National Health Insurance claims data. Health utilities were mainly obtained from the Seattle Angina Questionnaire responses of FLAVOUR trial participants mapped to EQ-5D.
Results:
From the Korean healthcare system perspective, the base-case analysis showed that FFR-guided PCI was 2,451 U.S. dollar lower in lifetime healthcare costs and 0.178 higher in QALYs compared to IVUS-guided PCI. FFR-guided PCI remained more likely to be cost-effective over a wide range of willingness-to-pay thresholds in the probabilistic sensitivity analysis.
Conclusions
Based on the results from the FLAVOUR trial, FFR-guided PCI is projected to decrease lifetime healthcare costs and increase QALYs compared with IVUS-guided PCI in intermediate coronary lesion, and it is a dominant strategy in Korea.

Neurodegenerative diseases are a group of disorders characterized by progressive and chronic degeneration of neurons in the central or peripheral nervous system,primarily including conditions such as amyotrophic lateral sclerosis,frontotemporal dementia,Alzheimer's disease,Parkinson's disease,multiple sclerosis,Huntington's disease and age-related macular degeneration.The condition worsens over time,ultimately leading to neuronal loss and synaptic dysfunction.Despite significant advances in medical research,the diagnosis and treatment of neurodegenerative diseases still face major challenges.There is an urgent need for effective and innovative treatment strategies as well as early monitoring methods.This involves not only new drug targets but also research into disease mechanisms.Neuronal pentraxin 2(NPTX2)is a protein expressed in the nervous system,highly expressed in synaptic regions of the brain,where it plays a critical role in synaptic plasticity and the formation and maintenance of neural networks.This paper reviews the role of NPTX2 in neurodegenerative diseases and summarizes recent research progress.
8.Ratio of right ventricular free wall longitudinal strain to pulmonary artery systolic pressure for evaluating right ventricular systolic function in patients with hypertrophic cardiomyopathy

Objective To observe the value of right ventricular free wall longitudinal strain(RVFWLS)to pulmonary artery systolic pressure(PASP)ratio(RVFWLS/PASP)for evaluating systolic function of right ventricle(RV)in patients with hypertrophic cardiomyopathy(HCM).Methods Fifty-two HCM patients were retrospectively collected and divided into group A(RVFWLS/PASP≤0.75,n=26)and B(RVFWLS/PASP>0.75,n=26)according to median RVFWLS/PASP.Meanwhile,26 healthy individuals were included as control group.Parameters of echocardiography and speckle tracking imaging(STI)were obtained and compared among 3 groups.The correlations of RVFWLS/PASP and other parameters were analyzed with linear regression.Results In group A and B,left ventricular ejection fraction(LVEF),RV septal longitudinal strain(RVSepLS),RVFWLS,RV end-diastolic volume index(RVEDVi),RV stroke volume(RVSV),RV fractional area change(RVFAC)and left ventricular global longitudinal strain(LVGLS)were all lower,while the ratio of peak E blood flow velocity at mitral orifice diastole to early mitral intervalal annulus diastole velocity(E/e′)were higher than those in control group(all adjusted P<0.05).Compared with group B and control group,group A had higher PASP but lower RV ejection fraction(RVEF)(all adjusted P<0.05).Furthermore,RVFWLS and LVGLS were both lower in group A than in group B(both adjusted P<0.05).Linear regression analysis showed that RVSepLS and RVEF were independently linearly related to RVFWLS/PASP(both P<0.05).Conclusion RVFWLS/PASP could be used to monitor changes of RV systoic function in HCM patients at early stage.
9.Summary of best evidence for early maternal-neonatal skin-to-skin contact during cesarean section

Objective:To summarize the best evidence regarding early maternal-neonatal skin-to-skin contact during cesarean section, with the aim of providing an evidence-based foundation for clinical nursing practice.Methods:Developing a literature search strategy to systematically retrieve relevant literature on early maternal-neonatal skin-to-skin contact during cesarean sections from UpToDate, BMJ Best Practice, PubMed, Cochrane Library, CINAHL, Web of Science, Embase, China National Knowledge Infrastructure, VIP, China Biology Medicine disc, and Wanfang Database. The search focus on clinical decision-making, guidelines, expert consensus, evidence summaries, and systematic reviews. The search timeframe should cover from the inception of each database to June 2024. Literature was subjected to quality appraisal, and only high-quality sources were included for evidence extraction and synthesis.Results:A total of eight documents were included, consisting of one clinical decision-making, five guidelines, one expert consensus, and one systematic review. A total of 23 best practice recommendations were extracted and categorized into six domains: team composition and training, indications and contraindications, pre-implementation preparation, implementation protocols, implementation recommendations, and precautions.Conclusions:This study provides a comprehensive summary of current evidence related to early maternal-neonatal skin-to-skin contact during cesarean section. The findings offer evidence-based guidance for clinical nursing practice and contribute to improving the safety of maternal-neonatal skin-to-skin contact implementation during cesarean section.
