1.Applications of Vaterite in Drug Loading and Controlled Release

Currently, the drug delivery system (DDS) based on nanomaterials has become a hot interdisciplinary research topic. One of the core issues is drug loading and controlled release, in which the key lever is carriers. Vaterite, as an inorganic porous nano-material, is one metastable structure of calcium carbonate, full of micro or nano porous. Recently, vaterite has attracted more and more attention, due to its significant advantages, such as rich resources, easy preparations, low cost, simple loading procedures, good biocompatibility and many other good points. Vaterite, gained from suitable preparation strategies, can not only possess the good drug carrying performance, like high loading capacity and stable loading efficiency, but also improve the drug release ability, showing the better drug delivery effects, such as targeting release, pH sensitive release, photothermal controlled release, magnetic assistant release, optothermal controlled release. At the same time, the vaterite carriers, with good safety itself, can protect proteins, enzymes, or other drugs from degradation or inactivation, help imaging or visualization with loading fluorescent drugs in vitro and in vivo, and play synergistic effects with other therapy approaches, like photodynamic therapy, sonodynamic therapy, and thermochemotherapy. Latterly, some renewed reports in drug loading and controlled release have led to their widespread applications in diverse fields, from cell level to clinical studies. This review introduces the basic characteristics of vaterite and briefly summarizes its research history, followed by synthesis strategies. We subsequently highlight recent developments in drug loading and controlled release, with an emphasis on the advantages, quantity capacity, and comparations. Furthermore, new opportunities for using vaterite in cell level and animal level are detailed. Finally, the possible problems and development trends are discussed.
2.Chaijin Jieyu Anshen tablets regulating CaMKII and Cofilin dual signal pathway alleviate synaptic remodeling of glutaminergic hippocampal neurons in depression

Aim To reveal the mechanism of Chaijin Jieyu Anshen tablets(CJJYAS)regulating CaMKII and Cofilin dual signal pathway to improve synaptic remod-eling of glutaminergic hippocampal neurons in depres-sion.Methods The cell model of depression was es-tablished by corticosterone and lipopolysaccharide.The cells were randomly divided into the control group,model group,GR blocker group,GR agonist group,CX3CR1 blockergroup,CX3CR1 agonist group,CJJ-YAS group,CJJYAS combined with GR agonist group,and CJJYAS combined with CX3CR1 agonist group.Cell imaging analysis was used to observe the morpho-logical and structural changes of astrocytes,microglia,anterior cingulate cortex(ACC)and hippocampal(HPC)glutaminergic neurons.Immunofluorescence was used to test the activation of ACC and HPC glu-taminergic neurons and synaptic remodeling.Immuno-fluorescence and Western blot were respectively used to detect the expressions of synaptic remodeling-related proteins N-methyl-D-asprtate receptor 2A(GRIN2A),GRIN2B,CaMKII,MK2 and Cofilin in HPC glutamin-ergic neurons.Results CJJYAS could significantly improve the morphological and structural damage of as-trocytes,microglia,ACC and HPC neurons.Further-more,CJJYAS markedly restrained the excessive acti-vation of ACC and HPC glutaminergic neurons and the hippocampal synaptic plasticity damage and synaptic remodeling via down-regulating GRIN2A,GRIN2B and MK2 proteins,and up-regulating CaMKII and Cofilin protein.Conclusions CJJYAS effectively alleviate the synaptic remodeling of glutaminergic hippocampal neurons in depression,and its molecular mechanism might be associated with the regulation of synapse-re-lated NR/CaMKII and MK2/Cofilin signaling path-ways.
3.Codon usage bias of Marburg viruses

The codon usage bias of the Marburg virus(MARV)genome and related influencing factors were investigated to provide a reference for preparation of vaccines and specific antibodies.In total,93 MARV coding sequences were retrieved from the GenBank database and analyzed with EMBOSS,Codon W,and SPSS 26.0,while plots were generated with SigmaPlot14.0 and GraphPad Prism 9.5.The effective number of codons(ENC values)of the MARV genomes for each protein ranged from 49.84 to 60.92,and the codon adaptation index was close to 0.7.Relative synonymous codon usage showed that GCA,AGA,AAU,GA A,GGA,AUU,CCU,and AC A were common high-frequency codons of seven proteins,and the codons with fa-voritism mostly ended with A/U.Neutral,ENC-Plot,and PR2 parity analyses indicated that the MARV codon usage prefer-ence was dominated by natural selection,followed by mutational pressure and various other factors.Comparisons of MARV gene codon usage frequency with four expression systems(Escherichia coli,yeast,human,and baculovirus)found differences in the codon usage frequency between MARV and E.coli.MARV codon usage favoritism was shaped by a combination of fac-tors,with natural selection playing a dominant role.The E.coli expression system was most suitable for in vitro expression of MARV proteins.
4.Application of KIH Structure in Recombinant Expression of Human Interleukin-35 in vitro

Interleukin-35(IL-35)is an important immunosuppressive cytokine that has been shown to play a role in the immune response of various diseases.In this study,we cloned the coding sequence of human IL-35 gene,constructed single subunit expression vectors pXC17.4-p35 and pcDNA3.1(+)-EBI3,and co-transfected CHO-K1 cells to express IL-35 in vitro.No binding was found between subunits of p35 and EBI3.Knobs-into-Holes(KIH)can solve the problem of heavy chain mismatch of heterolo-gous antibodies.Therefore,expression vectors pXC17.4-p35-Fch and pcDNA3.1(+)-EBI3-Fck were constructed by fusing KIH structures on the basis of the original sequences to express the recombinant fu-sion protein of KIH-IL-35.The expression vectors of two subunits were exchanged at the same time to verify the influence of different vectors on the expression level of KIH-IL-35.The analysis of various pro-tein detection methods showed that the correct expression rate of KIH-IL-35 structure was significantly im-proved.Affinity purification of KIH-IL-35 was performed after large amount of expression,and the bind-ing activity of KIH-IL-35 to glycoprotein 130(gp130)was detected by ELISA.The results showed that the binding of KIH-IL-35 to gp130 was concentration dependent.The indirect activity of KIH-IL-35 and M1 cells was detected by cell activity assay.Further results showed that the inhibition rate of M1 cells in-creased in a dose-dependent manner with the concentration of KIH-IL-35.In addition,a method for de-termining IL-35 activity by activated human peripheral blood mononuclear cells was successfully estab-lished.Activated PBMCs increased in a dose-dependent manner with KIH-IL-35 concentration.In sum-mary,this study utilized the KIH-IL-35 model to enhance the expression of recombinant human IL-35 and validated its high activity in vitro,providing new ideas for the study of IL-35 and the recombinant expres-sion of similar heterodimeric cytokines.
5.Development of microsatellite markers and analysis of genetic characteristics of laboratory Apodemus peninsulae population

Objective Polymorphic microsatellite markers developed for Apodemus peninsulae can enrich its genetic data and lay a foundation for genetic quality control and gene mapping.Methods Microsatellite loci were screened based on the genome sequence of Apodemus peninsulae,and microsatellite primers were identified.The genetic diversity of the population was analyzed by multiplex PCR.Results Thirty microsatellite markers were successfully developed and evaluated using 60 samples of Apodemus peninsulae.A total of 152 alleles were detected,with an average of 5.067 alleles per locus.The average observed heterozygosity was 0.592.The average Shannon index was 1.265.The average polymorphism information content was 0.598.Conclusions Based on the microsatellite loci developed in this study,the genetic diversity of Apodemus peninsulae can be effectively analyzed,laying a foundation for establishing genetic quality standards and detection method.
6.Genetic Analysis of Thalassemia in Children in Liuzhou of Guangxi Zhuang Autonomous Region

Objective:To investigate the characteristics of thalassemia gene types in children in Liuzhou,Guangxi.Methods:A total of 822 children suspected thalassemia aged from 1 day to 14 years who were admitted to our hospital from January 2019 to April 2022 were collected.Gap-PCR and PCR combined with reverse dot blot hybridization were used to detect α-and β-thalassemia genes.Results:Among 822 children,561 thalassemia carriers were detected,with a detection rate of 68.25%.Among them,303 cases were detected with α-thalassemia,and the most common genotype was--SEA/αα(163 cases),followed by-α3.7/αα(37 cases)and αcsα/αα(26 cases),44 cases with HbH disease.240 cases were detected with β-thalassemia,with a detection rate of 29.20%,and the most common genotype was βCD41-42/β N(112 cases),followed by βCD17/βN(75 cases)and βIVS-Ⅱ-654/β N(11 cases),11 cases with moderate to severe β-thalassemia.18 cases were detected with α β-thalassemia,with a detection rate of 2.19%,and--SEA/αα complex βCD41-42/βN was the most common genotype(4 cases).In Zhuang and Han populations,the detection ratio of-α3.7α/αα in α-thalassemia was the same(both 12.50%).While,the other main types such as--SEA/αα,αCSα/αα and-α4.2α/αα had certain differences.In β-thalassemia,CD41-42 and CD17 were the main genotypes detected in Han and Zhuang.Conclusion:In Liuzhou of Guangxi autonomous region,α-thalassemia is the main type in children,with a detection rate of 68.25%,and--SEA/αα is the most common genotype in mild thalassemia,followed by βCD41-42/βN.The detection rate of moderate to severe α-and β-thalassemia is relatively high.There are certain differences in the distribution of thalassemia among different ethnic groups.
7.Genetic and clinical characteristics of 46,XX testicular disorders of sex development

Objective:To investigate the genetic and clinical characteristics of 46,XX testicular disorders of sex development(DSD).Methods:We collected the clinical data on the patients with 46,XX testicular DSD diagnosed in the Center of Reproductive Medicine of the First Affiliated Hospital of Nanjing Medical University from January 2017 to January 2023,and analyzed their genetic and clinical characteristics and the SRY gene chromosomal location for those with SRY-positive.Results:A total of 26 patients were included in this study,all with 46,XX and deletion of the AZFa,b and c regions,with a mean height of(168.3±5.9)cm,body weight of(64.0±7.5)kg,BMI of(22.66±2.79)kg/m2,left testis volume of(2.53±1.16)ml and right testis volume of(2.74 ±1.34)ml.The semen volume of the patients averaged 1.35(0.18-2.78)ml,FSH(36.85±18.01)IU/L,LH(19.71± 9.71)IU/L,and T(6.08±2.71)nmol/L.The SRY-negative patients had a higher incidence rate of development disorders in the re-productive system than the SRY-positive ones(5/6 vs 3/20,P=0.004),but no statistically significant differences were observed in the other parameters.The SRY gene was localized at the end of Xp in 13 of the 14SRY-positive cases,and at chromosome 15 in the other 1.Conclusion:46,XX testicular DSD has some similarity and heterogeneity in genetics and clinical characteristics.
8.Clinical efficacy of repeated transcranial magnetic stimulation combined with acupuncture for chronic insomnia

Objective To explore the clinical efficacy of repeated transcranial magnetic stimulation(rTMS)and acupuncture therapy in the treatment of chronic insomnia disorder(CID)patients.Methods A total of 80 patients with CID,who were treated at Nanchang First Hospital from January 2022 to December 2023,were selected for the study.The patients were randomly divided into control group and treatment group,with 40 cases in each group.The control group patients were treated with dexmedetomidine,while the treatment group patients received rTMS and acupuncture therapy in addition to control group.The treatment course was 4 weeks,and the sleep quality,sleep related indicators,and psychological condition improvement of both groups of patients were observed before and after treatment.Results After treatment,the Pittsburgh sleep quality index scores of both groups of patients decreased(P<0.05);The sleep latency and number of awakenings were lower than before treatment(P<0.05),and the total sleep time,sleep efficiency,and proportion of rapid eye movement sleep were higher than before treatment,treatment group showed more significant improvement than control group(P<0.05).After treatment,the Hamilton anxiety and depression scale scores of both groups of patients decreased compared to before treatment,but there was no statistically significant difference in control group before and after treatment(P>0.05).However,there was a statistically significant difference in treatment group before and after treatment(P<0.05).Conclusion The combination of rTMS and acupuncture treatment can significantly improve the sleep quality of CID patients,while also reducing the accompanying symptoms of anxiety and depression.
9.Independent risk factors related to unintended initial dissection of the poste-rior plane in small incision lenticule extraction performed by surgeons with different qualifications

Objective To investigate independent risk factors for unintended initial dissection of the posterior plane(UIDPP)during small incision lenticule extraction(SMILE)by surgeons with different qualifications.Methods A total of 1 600 patients(3 003 eyes)who underwent SMILE performed by three surgeons with different qualifications from April to September 2021 were selected as the research subjects,including 911 males and 689 females,with an average age of(21.82±3.55)years.The incidence of UIDPP in patients was recorded,along with a comprehensive summary of the age,gender,eyes,surgeons,surgical proficiency,subjective refraction,central corneal thickness,lenticule thickness,side cut angle,and peripheral lenticule thickness of the patients.Independent risk factors of UIDPP were analyzed using the binary logistic regression.Test level:α=0.05.Results In this study,the incidence of UIDPP was 6.56%(197/3 003),and the lenticule was successfully removed in all eyes.Multivariate logistic regression analysis demonstrated that surgeon(P=0.035),surgical proficiency(P=0.026),eyes(P=0.007),lenticule thickness(P<0.001),and peripheral lenticule thick-ness(P<0.001)were independent risk factors associated with the UIDPP during SMILE surgery.No significant differences were observed in other parameters(all P>0.05).Conclusion Surgeons,surgical proficiency,eyes,lenticule thick-ness,and peripheral lenticule thickness are independent risk factors of UIDPP.
10.Electromagnetic compatibility evaluation of brain-computer interface rehabilitation robots and improvement suggestions

The determinations of the operation mode and basic performance conformity for evaluating the electromagnetic compatibility of brain-computer interface rehabilitation robots were described based on relevant standards of the electromag-netic compatibility of medical electrical equipment.The performance deviations and their causes during the electromagnetic compatibility evaluation of brain-computer interface rehabilitation robots were analyzed with case studies,and some improve-ment suggestions were proposed accordingly.Technical references were provided for the manufacturers of brain-computer interface rehabilitation robots.[Chinese Medical Equipment Journal,2024,45(9):84-88]
