1.Analyses of the appropriate cutoff age for detecting fasting hyperglycemia in a hospital-based physical examination population

ObjectiveTo investigate the relationship between fasting plasma glucose (FPG) and age/sex, to further analyze the appropriate cutoff age for detecting fasting hyperglycemia (FHG), and to provide a reference for blood glucose screening and health management. MethodsIn this cross-sectional study, a total of 50 607 examinees from the Health Management Center of a Shanghai-based Medical Consortium in 2023 were included. The age and sex distributions of FPG levels and the detection rate of FHG (FPG≥6.1 mmol·L⁻¹) were analyzed. A non-parametric method was used to plot the receiver operating characteristic (ROC) curve of age for predicting hyperglycemia. The area under the ROC curve (AUC) reflected the predictive value of age for FHG, and the appropriate cutoff age for hyperglycemia detection was determined by the maximum Youden index. ResultsThe median and the 25th and 75th percentiles of FPG in the study population was 5.00 (4.68, 5.41) mmol·L⁻¹, with levels systematically higher in males [5.06 (4.73, 5.53) mmol·L⁻1] than in females [4.93 (4.64, 5.28) mmol·L⁻1]. Both FPG levels and the abnormal detection rate increased non-linearly with age, plateauing approximately at 80 years of age. ROC curve analyses showed that age had good discriminatory value for hyperglycemia status (AUC=0.778 for the total population), with the appropriate predictive age threshold being 48 years old (48 years old for males, 49 years old for females). ConclusionThe present study found that both FPG levels and the risk of abnormal glucose regulation were increased at 48‒49 years old among the hospital-based physical examination population, with risk remaining consistently higher in males than in females. The appropriate cutoff age for FPG was 48 years old. Specifically, the period of 45‒50 years of age should be set as an intensified screening phase for blood glucose screening, with particular emphasis on earlier intervention for males. Furthermore, glycemic management targets for the elderly population require individualized consideration.
2.Regional differences in brain activation after music intervention for Alzheimer's disease:a meta-analysis of func-tional magnetic resonance imaging

Objective To systematically investigate the impact of music intervention on neuroplasticity in patients with Alzheimer's disease(AD).Methods PubMed,EBSCOhost,Web of Science,Embase,CNKI,Wanfang data and VIP were searched for studies pub-lished up to February,2025.They were analysed using GingerALE 3.0.2 for meta-analysis.Results Five fMRI studies were included,involving 134 participants(99 with AD).A total of 49 activation coordinates were extracted.The main activation regions in the healthy group were concentrated in the left precuneus,the right inferior frontal gyrus and the right superior temporal gyrus,while the primary activated regions in AD group shifted to the right operculum and adjacent right insula,left superior frontal gyrus,right medial frontal gyrus,and left superior temporal gyrus,indicating that during music processing,neural activation patterns in AD patients shifted from higher-order associative cortices to subcortical structures,accompanied by compensatory changes in frontal lobe activation and a loss of activation in the parietal lobe,particularly in the precuneus.Conclusion Music intervention can activate the relatively preserved subcortical-limbic system in patients with AD and promote compensatory reorganization of the frontal lobes.

4.Npac Is A Co-factor of Histone H3K36me3 and Regulates Transcriptional Elongation in Mouse Embryonic Stem Cells

Chromatin modification contributes to pluripotency maintenance in embryonic stem cells(ESCs).However,the related mechanisms remain obscure.Here,we show that Npac,a"reader"of histone H3 lysine 36 trimethylation(H3K36me3),is required to maintain mouse ESC(mESC)pluripotency since knockdown of Npac causes mESC differentiation.Depletion of Npac in mouse embryonic fibroblasts(MEFs)inhibits reprogramming efficiency.Furthermore,our chromatin immunoprecipitation followed by sequencing(ChIP-seq)results of Npac reveal that Npac co-localizes with histone H3K36me3 in gene bodies of actively transcribed genes in mESCs.Interestingly,we find that Npac interacts with positive transcription elongation factor b(p-TEFb),Ser2-phosphorylated RNA Pol Ⅱ(RNA Pol Ⅱ Ser2P),and Ser5-phosphorylated RNA Pol Ⅱ(RNA Pol Ⅱ Ser5P).Furthermore,depletion of Npac disrupts transcriptional elongation of the pluripotency genes Nanog and Rifl.Taken together,we propose that Npac is essential for the transcriptional elongation of pluripotency genes by recruiting p-TEFb and interact-ing with RNA Pol Ⅱ Ser2P and Ser5P.
5.Screening of tomato cultivars in cadmium-polluted areas and study on their antioxidant capacity.

To screen the available tomato pollution-safe cultivar varieties and reduce the potential food safety risks in Cd-polluted areas, the differences of Cd accumulation in different tomato (Solanum lycopersicum) varieties in southern China were studied by soil culture and hydroponic experiments. Firstly, the high and low accumulation varieties were selected from 25 tomato varieties under 2.94 mg/kg Cd stress by soil culture test, and then the responses of high and low accumulation tomato varieties to Cd stress were determined by hydroponic experiments. The results of soil culture test show that under 2.94 mg/kg Cd stress, there were significant differences in plant height, total biomass and yield among 25 tomato cultivars, and the Cd contents of fruits of all 25 tomato cultivars exceeded the highest limit value (0.05 mg/kg) of CAC (Codex alimentarius commission). Through cluster analysis, 7, 4 and 14 varieties accumulating relatively high, medium, and low concentrations of Cd in the fruits were screened, among which the highest, the lowest, and the average Cd contents in the fruits were 3.06 mg/kg DW, 1.47 mg/kg DW, and 2.21 mg/kg DW, respectively. The results of hydroponic experiment show that under the same concentration of Cd stress, Qiantangxuri F1, a high Cd accumulating variety, absorbed Cd faster, accumulated more Cd, used shorter oxidative stress response time and had stronger tolerance to Cd than Zhefen 3053, a low Cd accumulating variety. The typical high and low Cd accumulating varieties can provide a reference for agricultural production in heavy metal polluted areas and the development of molecular-assisted breeding methods of PSC. At present, cultivating low Cd accumulating PSC varieties and dynamic monitoring of Cd contents in tomato fruits are feasible methods in medium and light Cd-polluted areas.
