1.Results and analysis of inter-comparison of gross α and β measurement capabilities in Gansu Province, China, 2023

Objective To assess the quality of gross α and β radioactivity measurements conducted by radiological health service institutions and disease prevention and control centers in Gansu Province, China, and regulate their measurement methods. Methods The samples were distributed by Gansu Provincial Center for Disease Control and Prevention as the organizer of the inter-comparison through mail. The institutions participated in the inter-comparison carried out the measurements in accordance with national standards, and submitted the inter-comparison reports in the form required by the inter-comparison scheme. Results A total of 13 institutions participated in the 2023 inter-comparison of gross α and β radioactivity measurement capabilities, and all measurement results met the required standards. The absolute Z-scores for gross α inter-comparison ranged from 0 to 1.21, and the absolute Z-scores for gross β inter-comparison ranged from 0.08 to 1.85. The comprehensive scores ranged from 74.5 to 93.0. Conclusion The measurement capacities of the institutions participated in the 2023 inter-comparison showed improvement compared with the previous year. However, 12 institutions participated in the inter-comparison showed issues in data processing, report formatting, and laboratory quality control. It is necessary to strengthen technical training, standardize the measurement procedures, and improve the measurement capabilities and skills to ensure the quality of services.
2.Mume Fructus Restores Intestinal Mucosal Epithelial Barrier Through MEK/ERK Signaling Pathway in Mouse Model of Inflammatory Bowel Disease

ObjectiveTo clarify the repair effect of Mume Fructus on the intestinal mucosal epithelial barrier in the mouse model of inflammatory bowel disease (IBD) and explore the repair mechanism. MethodsThirty-six male C57BL/6 mice were randomly assigned into six groups: normal, model, low-, medium-, and high-dose (200, 400, and 800 mg·kg-1) Mume Fructus, and sulfasalazine (300 mg·kg-1). Except the normal group, the rest groups had free access to 2% dextran sulfate sodium (DSS) solution for seven days to establish the IBD model, followed by a seven-day drug intervention. The body weight change and disease activity index (DAI) were recorded. After the last administration, spleen and colon tissue samples were collected to analyze the differences in colon length and spleen index. Hematoxylin-eosin staining was used to observe the morphology of the colon tissue. The level of diamine oxidase (DAO) in the serum was measured by the DAO assay kit. Immunohistochemistry was employed to determine the expression of tight junction proteins such as Claudin-1, Occludin, and zonula occludens-1 (ZO-1) in the colon tissue. Real-time PCR was performed to measure the mRNA levels of tumor necrosis factor-α (TNF-α) and interleukin-1β (IL-1β) in the colon tissue. Finally, Western blot was employed to determine the protein levels of mitogen-activated protein kinase kinase (MEK), extracellular signal-regulated kinase (ERK), phosphorylated (p)-MEK, and phosphorylated ERK in the colon tissue. ResultsCompared with the normal group, the model group exhibited decreases in body weight and colon length (P<0.01), increases in DAI, spleen index, and serum DAO level (P<0.01), damaged colonic epithelium and goblet cells, and obvious infiltration of inflammatory cells. In addition, the model group exhibited higher positive expression of Claudin-1, Occludin, and ZO-1 (P<0.01), higher mRNA levels of TNF-α and IL-1β (P<0.01), and higher protein levels of p-MEK and p-ERK (P<0.05, P<0.01) than the normal group. However, sulfasalazine and three doses of Mume Fructus markedly decreased the body weight and DAI (P<0.05), recovered the colon length and spleen index, alleviated colon tissue damage, lowered the level of DAO in the serum (P<0.01), and down-regulated the mRNA levels of TNF-α and IL-1β (P<0.01) and the protein levels of p-MEK and p-ERK (P<0.05). Sulfasalazine and low- and medium-dose Mume Fructus increased the positive expression of Occludin, Claudin-1, and ZO-1 (P<0.05, P<0.01). Furthermore, high-dose Mume Fructus elevated the protein expression of Occludin (P<0.05). ConclusionMume Fructus can restore the expression of intestinal epithelial tight junction proteins by inhibiting the phosphorylation of proteins in the MEK/ERK signaling pathway and down-regulating the levels of TNF-α and IL-1β, thus repairing the intestinal mucosal barrier in the mouse model of IBD.

5.Robot-assisted partial right nephrectomy using a combined retroperitoneal and transperitoneal approach in the treatment of right horseshoe kidney:one case report

Objective To explore the feasibility and safety of robot-assisted partial right nephrectomy using a combined retroperitoneal and transperitoneal approach in the treatment of horseshoe kidney with renal tumor.Methods Clinical and follow-up data of a patient with horseshoe kidney with renal tumor were retrospectively analyzed.Before operation,the diagnosis was confirmed with renal CT scanning and enhancement with angiography to show the blood supply.After that,the patient underwent robot-assisted partial right nephrectomy for horseshoe kidney by robot-assisted partial right nephrectomy using a combined retroperitoneal and transperitoneal approach.After operation,the patient was followed up.Results The operation went smooth with an operation time of 270 min.The intraoperative warm ischemia time was 30 min,intraoperative bleeding was approximately 300 mL,and no intraoperative or postoperative complications occurred.After 2 months of follow-up,the patient recovered well,and no chest and abdominal lymph node enlargement and lung metastasis were observed.Conclusion Robot-assisted partial nephrectomy of horseshoe kidney using a combined retroperitoneal and transperitoneal approach is a safe and feasible minimally invasive treatment option.

10.Causal relationship between immune cells and osteomyelitis through genetic prediction

Objective To predict the causal relationship between immune cells and osteomyelitis(OM)by using Mendelian randomization.Methods Data sets of 731 immune cells and osteomyelitis were obtained from the UK Biobank and the genome wide association study.The inverse variance weighted method,weighted median method,and MR-Egger method were employed sequentially to verify the causal relationship between the 731 immune cells and osteomyelitis.Cochran's Q test,MR-Egger intercept test,mendelian randomization pleiotropy residual sum and outlier(MR-PRESSO),and leave-one-out analysis were evaluatede the stability and reliability of the results.Results A total of 21 immune cells were found to have a causal relationship with osteomyelitis(P<0.05).Among them,T regulatory cells,classical dendritic cells and B cells were exhibited more stable causal relationships with osteomyelitis(P<0.01).Specifically,T regulatory cells(OR=0.925,β=0.078)were identified as protective factors reducing the risk of osteomyelitis,while classical dendritic cells(OR=1.084,β=0.081)and B cells(OR=1.063,β=0.061)were identified as risk factors increasing the risk of osteomyelitis.Conclusion Twenty-one immune cells are genetically associated with the risk of osteomyelitis.
