1.Choline kinase alpha silencing affects proliferation and apoptosis in glioma cells by inducing mitochondrial dysfunction

BACKGROUND:Choline kinase alpha is a key enzyme in phospholipid metabolism,involved in the synthesis of phosphatidylcholine,and plays an important role in maintaining cell membrane integrity and signal transduction.Research has shown that choline kinase alpha is highly expressed in various tumors and is closely related to cell proliferation,metabolic reprogramming,and tumor progression.As a potential therapeutic target,the role of choline kinase alpha in tumor metabolism and mitochondrial function still needs further exploration.OBJECTIVE:To evaluate the effects and the underlying mechanisms of choline kinase alpha on the proliferation and apoptosis of glioma U87MG and U251 cells.METHODS:Short hairpin RNA of choline kinase alpha and its empty vector control were transfected into U87MG and U251 glioma cells.Mitochondrial morphology was observed by transmission electron microscopy.Mitochondrial structure and functional protein levels were assessed by western blot assay.Reactive oxygen species levels in cells were measured using a reactive oxygen species fluorescent probe.Mitochondrial membrane potential was assessed with a JC-1 assay.Intracellular adenosine triphosphate levels were measured by chemiluminescence.Cell proliferation was evaluated using a CCK-8 assay.Apoptosis levels were analyzed by flow cytometry.The mitochondrial fission inhibitor Mdivi-1 was used to protect the mitochondrial function of the choline kinase α-silenced lentiviral cells.Finally,U87MG cells were subcutaneously injected to construct a subcutaneous tumor model in nude mice.The tumor growth in nude mice was observed before and after choline kinase alpha silencing and after the use of the mitochondrial fission inhibitor Mdivi-1.RESULTS AND CONCLUSION:(1)Compared with the empty control group,the mitochondria of U87MG and U251 cells in the choline kinase alpha silencing lentivirus group exhibited significant structural abnormalities in mitochondria,such as vacuolization and cristae disruption.The expressions of mitochondrial structure and function-related proteins TOM20,ACO2,and ATP5A were significantly decreased(P<0.01,P<0.001),the expression of SOD2 was significantly increased(P<0.01,P<0.000 1),the fluorescence intensity of reactive oxygen species was significantly increased(P<0.01),the mitochondrial membrane potential and adenosine triphosphate level were significantly decreased(P<0.01,P<0.001),the cell proliferation ability was reduced(P<0.01),and the apoptosis level was increased(P<0.001).(2)Following Mdivi-1 treatment,the fluorescence intensity of reactive oxygen species in U87MG and U251 cells decreased(P<0.05,P<0.01),mitochondrial membrane potential and adenosine triphosphate levels were significantly restored(P<0.05,P<0.01,P<0.001),cell proliferation ability was improved(P<0.05,P<0.01),and apoptosis level was decreased(P<0.05).(3)In addition,the in vitro subcutaneous tumor formation experiment of nude mice showed that compared with the empty control group,the mass and growth rate of subcutaneous tumors formed by U87MG cells in the choline kinase alpha silencing lentivirus group were significantly reduced(P<0.000 1).After Mdivi-1 treatment,the mass and growth rate of tumors were significantly increased(P<0.000 1).(4)The results show that choline kinase alpha silencing affects the proliferation and apoptosis of glioma cells by inducing mitochondrial dysfunction.

3.Physical activity and its associations with depressive and anxiety symptoms and their co-existence among secondary vocational school students

Objective:
To examine the characteristics of physical activity among secondary vocational school students and its associations with depressive symptoms, anxiety symptoms, and their co-existence, so as to provide evidence for promoting their mental health.
Methods:
From September to December 2024, a questionnaire survey was conducted on 5 141 secondary vocational school students using convenience sampling method across 16 secondary vocational schools in 8 provinces/municipalities, including Fujian, Chongqing, Guangdong, Guangxi, Hubei, Jiangxi, Shanghai and Zhejiang. A self designed questionnaire, International Physical Activity Questionnaire Short Form, Patient Health Questionnaire-9 and Generalized Anxiety Disorder Scale-7 were used to assess basic information, physical activity characteristics, and depressive and anxiety symptoms. Chi-square test and Logistic regression model were used to analyze the associations of physical activity characteristics with depressive symptoms, anxiety symptoms, and their co-existence.
Results:
Among secondary vocational school students, 22.2% had depressive symptoms, 12.2% had anxiety symptoms, and 10.5% had both conditions. After adjustment for confounding factors such as sex, grade, and family structure, Logistic regression analyses showed that a high physical activity level, meeting the recommendation for daily moderate to vigorous physical activity (MVPA), and a concentrated physical activity pattern were associated with lower odds of depressive symptoms, anxiety symptoms, and their co-existence ( OR =0.70-0.81, all P <0.05). Regular weekly MVPA and a regular physical activity pattern were associated only with lower odds of depressive symptoms ( OR =0.79, 0.70, both P <0.05). Sex stratified analyses showed that meeting the recommendation for daily MVPA and a concentrated physical activity pattern were associated with lower odds of depressive symptoms, anxiety symptoms, and their co-existence among boys ( OR =0.60-0.71); a high physical activity level, regular weekly MVPA, and a regular physical activity pattern were associated with lower odds of depressive symptoms among boys ( OR =0.65-0.78)(all P <0.05). Meanwhile, a high physical activity level, meeting the recommendation for daily MVPA, regular weekly MVPA, and regular and concentrated physical activity patterns were associated with lower odds of depressive symptoms in female students ( OR =0.72-0.79) (all P <0.05).
Conclusions
Physical activity characteristics are associated with depressive symptoms, anxiety symptoms, and their co-existence among secondary vocational school students. Schools should optimize physical activity promotion strategies according to sex specific characteristics to reduce the occurrence of depressive and anxiety symptoms among secondary vocational school students.
4.Diffusion tensor imaging analysis index along the perivascular space for assessing age-related functional changes in glymphatic system

Objective To explore the value of diffusion tensor imaging analysis index along the perivascular space(DTI-ALPS)for assessing age-related functional changes in glymphatic system(GS).Methods Totally 27 healthy subjects from Meizhou People's Hospital and 100 healthy subjects from neuroimaging informatics tools and resources collaborator database who underwent T1-weighted magnetization-prepared rapid gradient echo(T1-MPRAGE)and DTI scanning were retrospectively enrolled and divided into youth group(n=38),middle-aged group(n=57)and elderly group(n=32).Automated DTI-ALPS index analysis procedure was used to minimize manual errors and derive DTI-ALPS index.The general data,neuropsychological assessment results and DTI-ALPS indices were compared among groups.Spearman correlation analysis was performed to observe the relationships of DTI-ALPS index and age,gender,as well as neuropsychological scores.Results The average age in youth group,middle-aged group and elderly group was(28.5±5.8),(53.7±6.8)and(73.8±2.3)years,respectively.No significant difference of DTI-ALPS index was found between middle-aged group and elderly group(P>0.05),which were both lower than that in youth group(both P<0.05).DTI-ALPS index was weakly negatively correlated with age(rs=-0.340,P<0.001),but not significantly correlated with gender nor neuropsychological assessment results(both P>0.05).Conclusion DTI-ALPS index was negatively correlated with age in healthy individuals,hence having potential utility for assessing age-related functional changes in GS.
5.Community resilience evaluation index system based on Delphi method for emergent major infectious diseases

Objective To establish a scientific, comprehensive, and operable community resilience evaluation index system for emergent major infectious diseases. Methods Based on the social ecosystem theory, a preliminary evaluation index system was formed by using content analysis and boundary analysis. The index system was then supplemented and revised through panel discussions. The final index system and index weights were clarified by two rounds of Delphi method. Results The expert positive coefficient, expert authority coefficient, and expert coordination coefficient of the two rounds of expert consultations were examined. According to the screening principle of the “threshold method”, the indicators were screened, and the weights of each indicator were determined in the second round of Delphi expert consultation. The analysis of the reliability of the indicator system showed Cronbach's α= 0.399 , indicating that the indicator system had a relatively high reliability. Factor analysis was carried out on 7 primary indicators, and the measure of sampling adequacy (MSA) values were all greater than 0.5, which passed the validity test. Conclusion A set of evaluation index system that can accurately reflect the resilience level of communities with emergent major infectious diseases has been constructed, including 7 primary indicators, 21 secondary indicators, 54 tertiary indicators, and 108 tertiary indicators, which has realized the quantitative evaluation of the hidden resilience level of communities.
6.Survey of dietary iodine intake of one university students in Mudanjiang City, Heilongjiang Province

Objective:To investigate the dietary iodine intake of students at Mudanjiang Medical University, Heilongjiang Province.Methods:From September 2023 to June 2024, a two-stage sampling method was used to investigate students at Mudanjiang Medical University. In stage one, 250 students were selected and investigated on their dietary iodine intake using the Iodine Specific Food Frequency Questionnaire (I-FFQ). In stage two, 55 students were selected and the double-meal method was used to investigate the dietary iodine intake of students. Iodine content in food samples was determined by inductively coupled plasma mass spectrometry. Curve estimation (power function, S-shaped curve, cubic equation, growth curve) was applied to analyze the dietary iodine intake of I-FFQ and double-meal method, the regression equation was used to adjust I-FFQ.Results:A total of 237 students were enrolled in I-FFQ, with a dietary iodine intake [ M( Q1, Q3)] of 342.61 (203.77, 399.26) μg/d. The dietary iodine intake of males and females was 361.82 (235.56, 428.98) and 330.87 (190.47, 366.11) μg/d, respectively, and the difference between the two was statistically significant ( Z = - 3.06, P = 0.002). The proportions of insufficient iodine intake, appropriate iodine intake, and excessive iodine intake were 13.92% (33/237), 82.70% (196/237), and 3.38% (8/237), respectively. A total of 52 students were enrolled in the double-meal method, with an average dietary iodine intake of 645.22 (435.91, 960.50) μg/d over 3 days. The proportion of appropriate iodine intake and excessive iodine intake was 44.23% (23/52) and 55.77% (29/52), respectively. After adjusting for I-FFQ, the dietary iodine intake was 650.13 (441.68, 728.53) μg/d, with the proportions of appropriate iodine intake and excessive iodine intake being 35.02% (83/237) and 64.98% (154/237), respectively. Conclusion:More than half of the students at Mudanjiang Medical University in Heilongjiang Province are in a state of excessive iodine intake, and dietary iodine intake should be appropriately controlled.
7.Effects of polygonatum sibiricum polysaccharides on the growth performance,in-testinal morphology,antioxidant capacity,and intestinal function of chicks

Polygonatum sibiricum polysaccharide(PSP)is a polysaccharide with multiple pharma-cological activities that has been widely studied and used in the human body.However,there is cur-rently a lack of research investigating the potential advantages of PSP in poultry farming.This study investigated the effects of adding PSP to drinking water on the growth performance,antioxi-dant status,serum biochemical indicators,ileal tissue morphology,immune organs,and intestinal function of chicks.88 Hailan brown laying hens were randomly divided into 4 groups,with 22 hens in each group,namely the blank control group(CON),and fed with basic feed;The low-dose PSP group(250 mg/L),the medium dose PSP group(500 mg/L),and the high-dose PSP group(1 000 mg/L)were fed with corresponding doses of PSP through drinking water on the basis of basic feed,and the experimental period was 21 d.The initial and final body weight and immune or-gan relative quality of chicks,serum biochemical indicators,the activities of SOD,CAT,and GSH-Px,as well as the contents of T-AOC and MDA in the serum of chicks were measured;HE stai-ning method was used to observe the pathological changes of intestinal tissue slices in the ileum;Real time fluorescence quantitative PCR technology was used to detect the mRNA expression lev-els of cytokines ZO-1,Claudin-1,Occludin,Mucin-2,IL-1β,TNF-a,IFN-γ,IL-4,IL-8,and IL-10 in the ileum.The results showed that compared with the blank control group,the addition of medium dose PSP significantly increased the final relative quality(P<0.01),the final body weight and ADG of PSP500 group chicks significantly increased(P<0.01),and the F/G of PSP250 and PSP500 groups significantly decreased(P<0.05 or P<0.01).The villus height of the jejunum in the 200,500,and 1 000 mg/L PSP groups of chicks significantly increased(P<0.05).The SOD ac-tivity significantly increased(P<0.01),and the CAT activity in the PSP1000 group significantly increased(P<0.01).The PSP500 and PSP1000 groups significantly reduced the mRNA expression of cytokines IL-1β,IL-4,and IFN-γ in the ileum(P>0.05);PSP did not show significant changes in serum total protein(TP),albumin(ALB),glucose(GLU),cholesterol(T-CHO)content,and immune organ index(P<0.05).In summary,PSP can improve growth performance,enhance an-tioxidant capacity,improve ileal morphology and epithelial barrier function,and regulate mucosal immune status.Considering the overall economic benefits,the recommended level of PSP addition is 500 mg/L.
8.Protective effect of lycium barbarum polysaccharide on testicular injury induced by perfluorooctanoic acid in offspring mice

To examine how Lycium barbarum polysaccharide(LBP)impacts reproductive harm caused by perfluorooctanoic acid(PFOA)in male offspring mice.Sixty expectant female rats were randomly allocated into 6 groups,with each group consisting of 10 rats.From 1 to 17 d of gesta-tion,the blank control group was given 0.2 mL deionized water;The PFOA group was given 3.5 mg/kg PFOA;the LBP intervention group were given different doses(40,80,120 mg/kg)of LBP by gavage based on the PFOA group;the control group was given 80mg/kg.At 21 d of age,the offspring mice were weaned,and the offspring male mice were fed normally until 35 d of age,and testicular tissue was taken.HE staining was used to observe the pathological changes of testic-ular tissue sections.Body mass and testicular index were measured.The levels of SOD,MDA,LDH,SHD,GSH-PX and AR in testicular tissue were determined.Serum levels of LH,FSH,Gn-RH and T were determined by ELISA.Differential genes and differential metabolites of testis were analyzed in combination with transcriptome and metabolome.Real-time fluorescence quantitative PCR was used to detect the expression of key genes in fatty acid metabolic pathway.The results showed that LBP could alleviate PFOA-induced decline in body mass and increase testicular index(P<0.01).LBP could alleviate the necrosis,edema of testicular cells caused by PFOA.LBP can in-crease the contents of SOD,GSH-Px,LDH,SDH and AR in testicular tissue of offspring male mice induced by PFOA,increase the contents of FSH,LH,GnRH and T in serum of offspring male mice,and decrease the content of MDA.Analysis of the transcriptome revealed that 1 388 genes exhibited differential expression between the groups exposed to PFOA and 80 mg/kg LBP,with 696 genes being up-regulated and 692 genes being down-regulated.The differentially ex-pressed genes were predominantly involved in pathways such as fatty acid metabolism,unsaturated fatty acid biosynthesis,cholesterol metabolism,the PPAR signaling pathway,and protein digestion and absorption,with fatty acid metabolism being the most prominent among them.Examination of the metabolome revealed 34 metabolites that were expressed differently between the PFOA group and the 80 mg/kg LBP group.Key metabolic pathways such as fatty acid biosynthesis,unsaturated fatty acid biosynthesis,and steroid hormone biosynthesis were found to be significant.In a joint transcriptome and metabolome analysis,the Differentially expressed genes(DEGs)and Differential expression of metabolites(DEMs)are highly correlated in signaling pathways such as fatty acid bi-osynthesis and unsaturated fatty acid biosynthesis.This study shows that PFOA can cause testicu-lar injury in offspring male mice,and LBP can effectively alleviate the testicular injury caused by PFOA in offspring male mice.Meanwhile,transcriptomic metabolism shows that LBP can effec-tively improve the testicular injury caused by PFOA in offspring male mice through fatty acid bio-synthesis and unsaturated fatty acid biosynthesis signaling pathways.Among them,80 mg/kg LBP has better effect.
9.Prognostic value of 18F-NaF PET/CT coronary plaque imaging in patients with coronary heart disease

Objective:To investigate the clinical value of 18F-NaF PET/CT coronary plague imaging in evaluating the long-term prognosis of patients with coronary artery disease (CAD). Methods:A retrospective cohort study was conducted among 54 patients (37 males and 17 females, aged (57.2±9.8) years) diagnosed with CAD from a multicenter study between September 2015 and October 2022. All patients underwent 18F-NaF PET/CT and coronary angiography (CAG) within 1 week, and the PET/CT imaging was performed at the First Hospital of Shanxi Medical University. Major adverse cardiovascular events (MACE) were followed up. ROC curves were established to obtain the optimal thresholds of SUV max and accumulated SUV max of all lesions of main coronary artery branches (S-SUV max) for predicting MACE. Cox proportional risk model and Kaplan-Meier method (log-rank test) were used to analyze the predictive value of PET parameters for MACE. Differences in metabolic parameters between 2 groups were compared by Mann-Whitney U test. Results:The median follow-up time of the 54 patients was 6.0(1.8, 6.6) years, and 13(24.1%) patients developed MACE, including 7 deaths, 5 myocardial infarction and 1 severe arrhythmia. S-SUV max in MACE group was significantly higher than that in the non-MACE group (2.64(2.08, 4.49) vs 1.83(0.95, 2.90); Z=-2.04, P=0.041). ROC curve showed that the optimal threshold of S-SUV max for MACE prediction was 2.05 (AUC=0.690). Multivariate Cox analysis showed that S-SUV max was a strong predictor of MACE (hazard ratio ( HR)=2.434(95% CI: 1.547-3.828), P<0.001). ROC curve showed that the optimal threshold of SUV max to predict MACE was 0.55 (AUC=0.659), and univariate Cox analysis showed that SUV max was a factor to predict MACE ( HR=10.192 (95% CI: 2.667-38.953), P=0.001). In 25 patients with incomplete revascularization (ICR), Kaplan-Meier analysis showed that the incidence of MACE in patients with positive 18F-NaF uptake (single medium stenosis (40%-70%) with SUV max≥0.55) was significantly higher than that in patients with negative 18F-NaF uptake (5/14 vs 0/11; χ2=6.07, P=0.014). Conclusions:18F-NaF PET/CT can be used as an independent predictor of MACE in patients with CAD and can quantitatively assess the long-term progression of moderate coronary artery stenosis. In the future, it is expected to be a new non-invasive way to guide the revascularization treatment decision of multi-vessel CAD.

Objective To establish a rat model of venous thrombosis in a plateau hypobaric hypoxic environment and to investigate the effect of this environment on venous thrombosis.Methods A total of 144 healthy male SD rats were assigned randomly to four groups(n=36 rats per group):a plains sham operation(A)group,plains operation(B)group,plateau altitude 6000 m+sham operation(C)group,and plateau altitude 6000 m+surgery(D)group.Rats in A and B groups were maintained in a plains normoxic environment,while rats in C and D groups C and D were subjected to a plateau environment.Rats in the surgical groups underwent quantitative constriction to incompletely obstruct the inferior vena cava blood flow.Each group was further divided into subgroups based on time:1,3,5,7,14,and 21 d(n=6 rats per group).Regular vascular ultrasound monitoring was conducted,and blood samples were taken for whole blood viscosity testing and the assessment of inflammatory indicators,including endothelin-1(ET-1),interleukin-6(IL-6)and tissue factor(TF).Coagulation function was evaluated through the activated partial thromboplastin time(APTT),prothrombin time(PT),thrombin time(TT),fibrinogen(FIB)and D-dimer.After the observation period,the experimental animals were sacrificed and the limbs were removed.Thrombus samples were stained with hematoxylin/eosin(HE),and the thrombus wet mass was measured.Results The thrombosis incidence was significantly higher in the plateau D group than in B group,accompanied by a marked increase in blood viscosity and hematocrit(P<0.01).Additionally,levels of ET-1,IL-6,and TF were significantly elevated(P<0.05),indicating a coagulation disorder.Conclusions A plateau hypoxic environment model can be successfully simulated by quantitative coarctation of the inferior vena cava,combined with a specialized environmental chamber.The findings of this study suggest that a plateau hypoxic environment promotes venous thrombosis.
