1.Correlation between neutrophil extracellular traps and clinicopathological characteristics and prognosis of EGFR wild-type lung adenocarcinoma

Objective:To investigate the predictive value of neutrophil extracellular traps (NETs) on the prognosis of patients with epidermal growth factor receptor (EGFR) wild-type lung adenocarcinoma.Methods:A total of 132 surgical paraffin specimens of EGFR wild-type lung adenocarcinoma diagnosed at the Tumor Hospital Affiliated to Xinjiang Medical University from January 2016 to December 2023 and 12 pairs of cancer and paracancerous fresh tissues from patients with EGFR wild-type lung adenocarcinoma diagnosed in March-July 2024 were collected with their clinical information. Western blotting and immunofluorescence were used to detect the expression levels of citrullinated histone H3 (CitH3) and myeloperoxidase (MPO) in cancerous and paraneoplastic tissues. Immunohistochemistry was used to detect the infiltration of PD-L1, CD4 + T cells and CD8 + T cells in the tumors. The clinical and prognostic correlations between NETs and EGFR wild-type lung adenocarcinoma patients were analyzed. Results:The expression of MPO ( P<0.001) and CitH3 ( P=0.009) was significantly increased in the tumors compared with the paracancerous tissues. The rate of high expression of NETs in cancer tissues was higher in patients with EGFR wild-type lung adenocarcinoma who were in stage Ⅲ and Ⅳ, with lymph node metastasis, distant metastasis, pleural invasion, high expression of Ki-67, low expression of CD8 + T, and lowered lymphocyte counts when compared to paraneoplastic tissues ( P<0.05). Patients were stratified based on TNM stage Ⅱb for prognostic analysis. Kaplan-Meier univariate analysis showed that the median overall survival (OS) (stage Ⅰ to Ⅱb: 47 vs 87 months; stage Ⅲ to Ⅳ: 27 months vs not reach) and the median disease-free survival (DFS) (stage Ⅰ to Ⅱb: 42 vs 78 months; stage Ⅲ to Ⅳ: 18 vs 39 months) of patients with high expression of NETs in stage Ⅰ to II b and stage Ⅲ to Ⅳ were lower than those with low expression (stage Ⅰ to Ⅱb: OS, P<0.001; DFS, P<0.001; stage Ⅲ to Ⅳ: OS, P=0.001; DFS, P=0.022). The OS (stage Ⅰ to Ⅱb: HR=3.513, 95% CI: 1.966-6.277, P<0.001; stage Ⅲ to Ⅳ: HR=3.215, 95% CI: 1.324-7.806, P=0.010) and the DFS (stage Ⅰ to Ⅱb: HR=2.478 ,95% CI: 1.396-4.400, P=0.002; stage Ⅲ to Ⅳ: HR=2.248, 95% CI: 1.089-4.638, P=0.028) in the group with high expression of NETs in either stage Ⅰ to Ⅱb or stage Ⅲ to Ⅳ were significantly shorter than those in the group with low expression. Conclusion:The EGFR wild-type lung adenocarcinoma patients with high expression of NETs have relatively shorter DFS and OS, which are independent risk factors for the prognosis of patients with EGFR wild-type lung adenocarcinoma, and are likely to be the potential biomarkers for the diagnosis and treatment of EGFR wild-type lung adenocarcinoma.

5.Rapamycin attenuates ioversol-induced acute kidney injury in rat models

Objective To investigate the role of autophagy in contrast-induced acute kidney injury(CI-AKI)in rats and to explore its possible mechanism.Methods SD rats were randomly divided into control group,acute kid-ney injury model group(intravenous injection of contrast medium ioversol via tail vein;model),rapamycin(RAPA)group and hydroxychloroquine(HCQ)group.Blood urea nitrogen(BUN)and serum creatinine(Scr)con-tents were measured and the potential change foun in renal pathology was detected by HE staining and microscopy.Transmission electron microscopy was used to observe auto-phagy-related changes in ultrastructure.Western blot was used to observe the expression of microtubule-associated protein 1 light chain 3(LC3)Ⅱ/LC3Ⅰ,ubiquitin-binding protein p62 and Histone deacetylase 4(HDAC4).The expression of HDAC4 was also observed by RT-qPCR.Results Compared with control group,the level of BUN,Scr and HDAC4 expression in the model and HCQ group was increased(P<0.01),the proximal tubules of the kidney were significantly damaged.In the model group,auto-phagososomes and autolysosomes increased,accompanied by an increase of LC3Ⅱ/LC3Ⅰ and a decrease in the p62 level(P<0.05,P<0.01);Compared with model group,there were more autophagosomes and autolysosomes were found in RAPA group(P<0.01),accompanied by increased LC3Ⅱ/LC3Ⅰratio and decrease in the p62 and HDAC4(P<0.05,P<0.01).In contrast,the number of autophagy related structures decreased in HCQ group(P<0.01),accompanied by the simultaneous increase of LC3Ⅱ/LC3Ⅰ,p62 and the increase of HDAC4(P<0.01).Conclusions Ioversol may induce autophagy activation,while enhancing autophagy by RAPA alleviates CI-AKI induced renal dysfunction.The mechanism is potentially atributed to the regulation of HDAC4.
6.Construction and verification of evaluation indicators for the construction of key specialties in traditional Chinese medicine nursing at the provincial and municipal levels

Objective To construct evaluation indicators for the construction of key specialties in traditional Chinese medicine nursing at the provincial and municipal levels and conduct preliminary verification,providing references for evaluating the level of construction of key specialties in traditional Chinese medicine nursing.Methods From December 2022 to March 2023,based on the Wuli-Shili-Renli system methodology,the content and weight of the evaluation indicators for the construction of key specialties in traditional Chinese medicine nursing at the provincial and municipal levels were determined through literature review,semi-structured interviews,Delphi method,and analytic hierarchy process.Opinion consultation and satisfaction survey were conducted on 46 nursing managers of secondary and above traditional Chinese medicine hospitals in Chongqing in April 2023 to preliminarily verify the application effect of the indicators.Results After 2 rounds of consultation with 16 experts,the questionnaire response rates were 100%and 100%,respectively.The expert authority coefficients were 0.963 and 0.950,respectively,and the coordination coefficients were 0.216~0.307,0.134~0.144,respectively(P<0.05).Ultimately,an evaluation index for the construction of key specialties in traditional Chinese medicine nursing at the provincial and municipal levels was formed,which includes 3 primary indicators,17 secondary indicators,and 76 tertiary indicators.97.83%nursing managers of hospitals are satisfied with the constructed indicators,believing that the indicator content is clear,comprehensive,professional,and has good practicality.Conclusion The evaluation indicators for the construction of key specialties in traditional Chinese medicine nursing at the provincial and municipal levels are scientific,reliable,which can provide references for the construction of key specialties in traditional Chinese medicine nursing and promote the development of traditional Chinese medicine nursing.
7.Analysis on risk factors for retinal detachment occurrence in fellow eye of patients with monocular rhegmatogenous retinal detachment

Objective To analyze the risk factors for the occurrence of rhegmatogenous retinal detach-ment(RRD)in the fellow eye of the patients with monocular RRD.Methods A single-center retrospective cohort study was conducted.The information such as the gender,age,laterality,bilateral best corrected visual acuity(BCVA),bilateral refractive error,whether the fellow eye implanting an intraocular lens,and location distribution of retinal tears or degenerative areas(such as lattice,cystic,and snail track degeneration)in the fellow eye of 331 patients with unilateral RRD surgery in this hospital from January 2018 to December 2020 was analyzed.The patients were followed up for one year,during this period,the prophylactic laser photocoag-ulation was performed if retinal tears or degenerative areas occurred in the fellow eye.The risk factors for RRD occurrence in the fellow eye were analyzed by the logistic regression.The Kappa value was calculated.The location distribution of retinal tears or degenerative areas in the affected eye and the fellow eye conducted the consistency analysis.Results The influencing factors for RRD occurrence of the fellow eye in the two groups mainly included the age,preoperative BCVA and preoperative refractive error(P<0.05).Logistic re-gression analysis revealed that the age 40 to<60 years old(OR=6.906,P<0.001),preoperative BCVA<0.05(OR=3.015,P<0.001)and refractive error-3.00 to<-6.00 D(OR=5.511,P<0.001)were sig-nificant independent risk factors for the occurrence of retinal tears or degenerative areas in the fellow eye.The age<40 years old(OR=0.101,P<0.001)and refractive error+0.50 D-<-0.50 D(OR=0.160,P=0.001)were the protective factors.The numbers of cases in the fellow eye with retinal tears or degenerative areas in the 4 quadrants were 59,14,27,and 8,respectively.In the affected eye,the numbers of cases with reti-nal tears or degenerative areas in the same quadrants were 43,6,10,and 3,respectively.The Kappa value was 0.296,P<0.001.All patients with RRD or lattice degeneration in the fellow eye received prophylactic laser photocoagulation,and no further retinal detachment occurred during the one-year follow-up period.Conclusion Ai-ming at the patients with unilateral RRD,the contralateral eye should be examined in detail,especially in mid-dle-aged and elderly patients with BCVA<0.50 and diopter-3.00-<-6.00 D,focusing on the retinal quadrant of the contralateral eye that is the same as the affected eye hole,and the incidence of contralateral RRD should be effectively reduced through regular and comprehensive eye examination.
8.Protective effect of racanisodamine on radiation-induced lung epithelial cell injury

Objective:To investigate the protective effect of racanisodamine (654-2) on lung epithelial cell injury induced by X-ray in mice and unravel the underlying mechanism.Methods:Mouse alveolar epithelial cells MLE-12 were used to establish radiation-induced lung injury (RILI) model in vitro and divided into 4 groups as follows: control (no irradiation), radiation (16 Gy radiation), treatment 1 (16 Gy radiation + 2 μmol/L 654-2), treatment 2 (16 Gy radiation + 10 μmol/L 654-2), and inhibitor (16 Gy radiation + 10 μmol/L 654-2 + 2 μmol/L ML385), respectively. Cells were sampled at different time points after radiation. Cell senescence was detected with senescence-associated β-galactosidase (SA-β-Gal) staining. Cell colony-forming ability was detected to observe the recovery capability of cells after treatment. The expression levels of p21, p16, phosphorylated histone H2AX(γH2AX), nuclear factor erythroid-2 related factor 2 (Nrf2), Nrf2 Ser40 site phosphorylation (p-Nrf2), p62, heme oxygenase-1 (HO-1) and NAD(P)H quinone oxidoreductase 1 (NQO1) proteins were measured by Western blot. Cell apoptosis and the production of intracellular reactive oxygen species (ROS) were determined by flow cytometry. The expression levels of glutathione (GSH) and superoxide dismutase (SOD) were detected according to the manufectuer instructions. The expression levels of glutamate-cysteine ligase catalytic subunit (GCLC) and glutamate-cysteine ligase modifier subunit (GCLM) mRNA were determined by real time reverse transcription PCR. Measurement data were expressed as Mean ±SD. Comparison between two groups was conducted by independent sample t-test, and comparison among multiple groups was performed by one-way ANOVA. Results:Compared with the radiation group, the proportion of cells with positive staining of SA-β-Gal was significantly lower and cell senescence were alleviated in the treatment 1 and 2 groups (all P<0.001). Compared with the radiation group, the expression level of γH2AX protein was significantly down-regulated ( P=0.037), cell apoptosis rate was significantly decreased ( P=0.026), the proliferation capacity of MLE-12 was enhanced ( P=0.004), GSH ( P=0.002) and SOD ( P<0.001) activity was enhanced and ROS production was declined ( P=0.001) in the treatment 2 group. The expression levels of Nrf2 and p-Nrf2 in total protein were up-regulated over the time of 654-2 intervention. Meanwhile, the expression levels of antioxidant proteins of NQO1 and HO-1 were up-regulated and that of GCLC and GCLM mRNA was also up-regulated. There were no significant differences in the number of cells with positive staining of SA-β-Gal ( P=0.145) and ROS production ( P=0.317) between the inhibitor and radiation groups after supplement of ML385, small-molecule inhibitor of Nrf2. Conclusion:654-2 can activate the Nrf2 pathway, enhance cell antioxidant capacity and inhibit cell senescence, thereby playing a protective role on radiation- induced lung injury.
9.Application of single-cell RNA sequencing in pulmonary infection by pathogenic microorganisms

Pulmonary infection by pathogenic microorganisms is a common disease of the respiratory system with high mortality and poor prognosis,but its pathogenesis remains unclear.In recent years,single-cell RNA sequencing(scRNA-seq)has been widely used in the study of lung infection to provide an atlas of immune cells of patients,identify new immune cell subsets,analyze differential expression genes,reveal pathways related to inflammatory immune pathways in pathogenic microbial infection,monitor the levels of cytokines,and identify novel biomarkers of lung infection.There-fore,this paper presents a review of the application of scRNA-seq in pulmonary infection by pathogenic microorganisms.
10.Research progresses of Qa-1 restricted CD8+ regulatory T cells in the pathogenesis of infectious diseases.

The Qa-1 in mice is homologous to human leukocyte antigen E(HLA-E), and both of them belong to the non-classical major histocompatibility complex I b(MHC-I b) molecules. Qa-1 is capable of presenting self or exogenous antigen peptides to interact with two distinct receptors, namely T cell receptor (TCR) and natural killer cell group 2 member A (or C) (NKG2A/C), thus playing an important role in immune response and regulation. Qa-1-restricted regulatory CD8+ T cell (CD8+ Treg) is one of the most studied CD8+ Treg subgroups, which can maintain immune homeostasis and autoimmune tolerance by exerting immunosuppressive effects. Consequently, Qa-1-restricted CD8+Treg cells are closely associated with the occurrence and development of various clinical diseases, such as tumors, infections, autoimmune diseases, and transplant rejections. This paper provides a comprehensive review of the phenotypic characteristics, functional effects, regulatory mechanisms of Qa-1-restricted CD8+ Treg cells, as well as the latest research progresses of Qa-1-restricted CD8+ Treg cells involved in the pathogenesis of infectious diseases.
