1.Research Progress of Glioma in China in 2024

Glioma is the most common primary malignant tumor of the central nervous system in adults. Despite the standard treatment of surgery combined with radiotherapy and chemotherapy, the prognosis for high-grade glioma patients remains poor, highlighting the urgent need to further explore its pathogenesis and develop new therapeutic strategies. This article reviews the research progress in the field of glioma in China in 2024, covering tumorigenesis mechanisms, tumor immune microenvironment composition, advances in imaging techniques and novel imaging agents, improvements in surgical approaches, mechanisms of radio- and chemoresistance, and explorations of new therapeutic modalities. These studies provide a solid theoretical foundation for advancing clinical diagnosis and treatment of gliomas and may offer new opportunities to improve patient outcomes.
2.Clinical application of detachable string magnetically controlled capsule endoscopy for esophageal diseases in plateau area

To explore the diagnostic value of detachable string magnetically controlled capsule gastroscope (ds-MCE) for esophageal diseases in plateau area, clinical data of 45 patients who underwent ds-MCE at Qinghai Special Hospital of Cardio-Cerebrovascular Diseases from January 2024 to June 2024 were retrospectively analyzed to explore the application characteristics and advantages of ds-MCE. The results showed that ds-MCE examination was successfully completed in all 45 patients. Diagnosis was confirmed in 30 cases (66.67%, 30/45) including 28 cases (93.33%) of esophagitis, 8 cases (26.67%) of esophageal ulcer, 8 cases (26.67%) of esophageal varices, 5 cases (16.67%) of hiatal herniawere, 3 cases (10.00%) of esophageal polyp, 1 case (3.33%) of achalasia, 1 case (3.33%) of esophageal papilloma, and 1 case (3.33%) of lower esophageal and cardiac carcinoma. Compared with traditional endoscopy and capsule endoscopy, ds-MCE has the advantages of no need for anesthesia and high safety, which provides a new and important diagnostic option for esophageal diseases in plateau area.
3.Progress in population-based research of human microbiome and cardiovascular diseases

The human microbiome encompasses a diverse array of microorganisms and their functional interactions within the human body. It exhibits a vast diversity of species and complex roles across various body environments. Advanced sequencing technologies, such as 16S amplicon sequencing and metagenomic sequencing, facilitate in-depth analysis on this microbial community. Recent researches have suggested that characteristics of the human microbiome (such as diversity and composition of microbiome, involving metabolic pathways and metabolites) might be associated with the onset and progression of cardiovascular diseases. These findings provide valuable insights into the etiology of chronic diseases and might aid in the development of novel disease biomarkers and intervention strategies. This paper summarizes the designs, current status and key findings of current population-based research in this field, and introduce the future development and analyze the existing critical problems that need further investigations.
4.Plasma CTRP6 and SIRI levels in patients with heart failure and their clinical significance

Objective To investigate the plasma levels of C1q/tumor necrosis factor-related protein 6(CTRP6)and systemic inflammation response index(SIRI)in patients with heart failure(HF)and their clinical significance.Methods 126 HF patients admitted to the Heart Center,the First Affiliated Hospital of Shihezi University from October 2023 to September 2024 were selected and divided into heart failure with reduced ejection fraction(HFrEF)group(n=43),heart failure with mildly reduced ejection fraction(HFmrEF)group(n=45)and heart failure with preserved ejection fraction(HFpEF)group(n=38)according to left ventricular ejection fraction(LVEF)value.Non-HF patients treated at the same period were selected as the control group(n=38).Blood routine,N-terminal pro-B-type natriuretic peptide(NT-proBNP)and echocardiogram parameters were collected,SIRI and left ventricular mass index(IVMI)were calculated,plasma CTRP6 levels were detected by enzyme-linked immunosorbent assay,and the correlation between CTRP6,SIRI and LVEF,IVMI,NT-proBNP and New York Heart Association grades in HF patients was analyzed.Risk factors of HF were analyzed by Logistic regression analysis and receiver operating characteristic(ROC)curve was drawn.Results Compared with control group,HF patients showed statistically significant increases in creatinine,uric acid,estimate glomerular filtration rate,and SIRI levels,as well as a higher proportion of patients with coronary heart disease and atrial fibrillation.Conversely,CTRP6 levels were decreased,with statistical significance(P<0.05).There were statistical differences in CTRP6,NT-proBNP,LVEF and IVMI of HF patients with different ejection fraction classification(P<0.05).Spearman correlation analysis showed that CTRP6 level was positively correlated with LVEF and negatively correlated with LVMI in HF patients.SIRI was positively correlated with NT-proBNP and negatively correlated with LVEF.Logistic regression analysis indicates that CTRP6 is independent protective factor for HF,and a decrease in CTRP6 is an independent risk factor for HF severity.The area under ROC curve of CTRP6,SIRI and their combined diagnosis HF are 0.762,0.772 and 0.850,respectively.Conclusion The plasma CTRP6 level in HF patients is significantly decreased,and SIRI is significantly increased.Both of them have good diagnostic value for HF patients,and their combined detection can improve the diagnostic efficiency.
5.Effects of Tactile/Motor Stimulation Massage on Feeding Outcomes and Neuromotor Development in Preterm Infants

Objective To observe the effects of tactile/motor stimulation massage on feeding outcomes and neuromotor development levels of preterm infants.Methods A total of 136 preterm hospitalized infants in the neonatal intensive care unit of a tertiary hospital in Yunnan Province from January 2024 to July 2024 were enrolled and randomly assigned to a control group and an experimental group,with 68 cases in each group.The control group received the routine care for preterm infants,while the experimental group received tactile/motor stimulation massage as an intervention in addition to the routine care.The time taken from gavage to oral feeding(d),the rate of complete oral feeding(%),the incidence of feeding intolerance(%),the weight(g),the length(cm),the head circumference(cm),neonatal behavioral neurometric score(NBNA)and Psychomotor development index(PDI)were compared between the two groups.Results The time from gavage to oral feeding in the experimental group(12.73±1.86)days was significantly shorter than that in the control group(13.66±1.68)days,and the difference was statistically significant(P<0.05);the rate of complete oral feeding in the experimental group(89.7%)was significantly higher than that in the control group(70.6%),P<0.05,and the incidence of feeding intolerance in the experimental group(8.8%)was significantly lower than that in the control group(16.2%),P<0.05;The experimental group showed superior results in weight,length,and head circumference compared to the control group(P<0.05);The neonatal behavioral neurometry score of the experimental group(36.16±1.53)was significantly higher than that of the control group(35.10±2.66),P<0.05,and the motor development index in the experimental group(76.88±6.12)was significantly higher than that in the control group(74.34±5.07),P<0.05.Conclusion Tactile/motor stimulation massage can effectively improve the feeding status and promote the neuromotor development of preterm infants.
6.Effects of Zuogui Jiangtang Qingzhi Formula on the cGAS/STING/TBK1 signaling pathway in mice with type 2 diabetes mellitus combined with non-alcoholic fatty liver disease

Objective:
To investigate how Zuogui Jiangtang Qingzhi Formula (ZGJTQZF), a traditional Chinese medicine compound for lowering blood glucose levels and clearing lipids based on Zuogui Pill, regulates the cyclic GMP-AMP synthase (cGAS)/stimulator of interferon genes (STING)/TANK-binding kinase 1 (TBK1) signaling pathway to improve type 2 diabetes mellitus combined with non-alcoholic fatty liver disease.
Methods:
According to fasting blood glucose (FBG) levels, body weight, and sex, 40 MKR mice, with half being male and the other half female, were randomly divided into four groups: blank, model, ZGJTQZF (29.6 g/kg), metformin + simvastatin ([65.0+ 2.6] mg/kg), 10 mice per group, and a normal group consisting of 10 age-matched FVB mice. Mice in all groups, except in the normal and blank groups, were fed a high-fat diet for 8 weeks to induce the disease model. Each group received daily gavage of the respective treatments for 8 weeks after successful modeling. Various parameters were measured, including FBG, liver function (alanine aminotransferase [ALT] and aspartate transaminase [AST]), and blood lipids (triglyceride [TG] and total cholesterol [TC]). Serum levels of tumor necrosis factor-α (TNF-α) and interleukin-6 (IL-6) were determined using enzyme-linked immunosorbent assays. Histological liver changes were examined using transmission electron and optical microscope. cGAS, STING, and TBK1 protein and mRNA expression levels in liver tissues were detected using Western blotting and real-time PCR, respectively.
Results:
Compared with the normal group, the model group exhibited significant fat accumulation in hepatocytes and liver damage. Additionally, FBG, ALT, AST, serum TNF-α, IL-6, cGAS, STING and TBK1 expression levels in liver tissues were significantly elevated (P<0.05). Treatment with ZGJTQZF and metformin + simvastatin significantly improved FBG, ALT, AST, TG, TC, TNF-α, and IL-6 (P<0.01). Histologically, hepatic steatosis was notably alleviated. Expression of protein and mRNA of cGAS, STING, and TBK1 in liver tissues was significantly reduced (P<0.05).
Conclusion
ZGJTQZF improves lipid and glucose metabolism, mitigates liver injury, and reduces inflammatory markers in MKR mice. These effects may be mediated through the regulation of the cGAS/STING/TBK1 signaling pathway.
7.Comparison of Therapeutic Effect of Different Preparation Processes of Baihe Dihuangtang on Depressed Mice Based on Q-Marker

ObjectiveBased on modern analytical techniques and a depressed mouse model established by chronic unpredictable mild stress(CUMS), to evaluate the quality marker(Q-Marker) and pharmacodynamic difference of Baihe Dihuangtang prepared by different processes. MethodsHigh performance liquid chromatography(HPLC) was used to establish the characteristic profiles of Baihe Dihuangtang, and determine the content of Q-Marker in the samples prepared by ancient and modern processes. Seventy C57BL/6J mice were randomly divided into the normal group, model group, fluoxetine group(3 mg·kg-1), low and high dose groups of ancient process(6.5, 26 g·kg-1), and low and high dose groups of modern process(6.5, 26 g·kg-1), with 10 mice in each group. Except for the normal group, CUMS was used to induce depression in mice from the other groups for 28 d. After successful modeling, administration groups were given the corresponding drugs by gavage every day, and the normal and model groups were given an equal volume of pure water by gavage for 21 consecutive days. Change in body mass of mice was recorded, tail suspension test and open field test were used to evaluate the depressive behavior of mice, and enzyme-linked immunosorbent assay(ELISA) was employed to determine the contents of tumor necrosis factor-α(TNF-α), interleukin(IL)-1β and IL-6 in serum. ResultsCharacteristic profiles of Baihe Dihuangtang prepared by the two processes were established, the similarity between the two was 0.951, and 8 characteristic peaks were recognized with the reference peak of regaloside A. The results of quantitative analysis showed that the Q-Marker content was similar in Baihe Dihuangtang prepared by ancient and modern processes. The results of pharmacodynamics showed that, compared with the normal group, the model group showed increased immobility time in the tail suspension test, reduced total movement distance in the open field test, and elevated IL-1β, IL-6 and TNF-α levels in the serum(P<0.01). Compared with the model group, the behavioral indicators of mice in the Baihe Dihuangtang treatment group were significantly improved in terms of tail suspension time and open field exercise, and the levels of IL-1β, IL-6 and TNF-α in serum were significantly reduced(P<0.05, P<0.01). Baihe Dihuangtang prepared by the two processes both had antidepressant effects, and the difference between the two was not statistically significant in improving depressive symptoms. ConclusionQ-Marker of Baihe Dihuangtang prepared by modern and ancient methods are equivalent in content, and the pharmacological effects are consistent, indicating that dried Lilii Bulbus can replace fresh products in the preparation of Baihe Dihuangtang. This study provides a scientific basis for the development of new drugs of Baihe Dihuangtang and a reference for its rational application and clinical use.

9.Clinical characteristics of adult patients with eosinophilic esophagitis in China

Objective:Eosinophilic esophagitis (EoE), an allergen/immune-mediated chronic disorder of the esophagus, is witnessing an incremental rise in its global incidence. However, data in China are very limited. This study aimed to clarify the characteristics of EoE in Chinese adults at our medical center.Methods:We collected information, including clinical characteristics and endoscopic findings, relative to all cases of adult patients who met the pathological diagnostic criteria for EoE at our institution from January 2019 to May 2024. Patients diagnosed with reflux esophagitis (RE) were matched in a 1∶1 ratio based on age and sex, and comparisons were made between the two groups.Results:The overall detection rate of EoE in our hospital was approximately 0.05%, with a trend increasing yearly. In total, 61 patients met the diagnostic criteria for EoE, of whom 42 (68.9%) were male. The median age was 53 years. The clinical manifestations of EoE patients were diverse, with common symptoms including dysphagia, acid reflux, and upper abdominal pain. Serological examination revealed that 23.1% (9/39) of patients had increased absolute eosinophil counts. Allergic diseases were observed in 73.8% (45/61) patients. Most patients exhibited typical mucosal changes in the esophagus during endoscopy, the median EREFS score was 3 points. The most commonly administered treatment included proton pump inhibitors, and 13 patients received steroid treatment. During a follow-up period, 42 patients (68.9%) exhibited clinical symptom improvement or pathological remission. In comparison to Western countries, patients with EoE in China typically presented with milder disease severity and a more favorable prognosis. No significant differences were observed between younger and older adult groups regarding clinical symptoms, the proportion of comorbid allergic diseases, medications, or the rate of improvement ( P for all>0.05). Additionally, patients with EoE more commonly presented with dysphagia (29.5% vs. 1.6%, χ2=15.96, P<0.001), comorbid allergic diseases (73.8% vs. 13.1%, χ2=45.67, P<0.001), increased eosinophil counts (23.1% vs. 3.3%, χ2=7.61, P<0.01), and a positive response to corticosteroids when compared to patients with RE. Conclusions:This study systematically determined the clinical characteristics of adult EoE patients in China. Compared to Western countries, patients from China typically presented with milder disease severity and a more favorable prognosis. No significant clinical manifestations or prognosis differences were observed between younger and older patients. EoE can be differentiated from RE based on symptoms, the presence of comorbid allergic diseases, and the degree of serum eosinophil infiltration. For patients suspected of EoE, endoscopic esophageal biopsy will facilitate early diagnosis.
10.Effects of Zuogui Jiangtang Qingzhi Prescription on CD9,CD63 and Apoptosis of Liver Tissue in Type 2 Diabetes Mellitus Mice Combined with Non-alcoholic Fatty Liver Disease

Objective To study the molecular mechanism of Zuogui Jiangtang Qingzhi Prescription in improving type 2 diabetes mellitus(T2DM)with non-alcoholic fatty liver disease(NAFLD).Methods Totally 40 8-week-old MKR mice were selected and randomly divided into blank group,model group,Chinese medicine group and positive drug group,with 10 mice in each group.Additionally,10 FVB mice of the same age were set as the normal group.Except for the normal group and the blank group,all other groups were fed with a high-fat diet for 8 weeks.The Chinese medicine group and the positive drug group were given corresponding drugs by gavage for 8 weeks after modeling.Fasting blood glucose(FBG)was measured using a blood glucose meter,and the serum contents of alanine aminotransferase(ALT),aspartate aminotransferase(AST),triglycerides(TG)and total cholesterol(TC)were measured using biochemical methods,HE staining and transmission electron microscopy were used to observe the morphology of liver tissue,PCR was used to detect the mRNA expressions of CD9,CD63,Bax and Bcl-2 in liver tissue,immunohistochemistry and Western blot were used to detect the protein expressions of CD9,CD63,Bax and Bcl-2 in liver tissue.Results Compared with the normal group and the blank group,the contents of FBG,ALT,AST,TG and TC in the model group mice significantly increased(P<0.01);the volume of liver cells increased,the structure was disordered,and a large number of lipid droplets were visible,the cytoplasm showed vacuolar degeneration,mitochondria were severely swollen,and there were a large number of fat particles around lysosomes,the expressions of CD9,CD63,Bax mRNA and protein in liver tissue significantly increased(P<0.01),while the expression of Bcl-2 mRNA and protein significantly decreased(P<0.01).Compared with the model group,the contents of FBG,ALT,AST,TG and TC in the Chinese medicine group mice decreased(P<0.01);the degree of liver cell swelling was reduced,lipid droplets and vacuolar degeneration were significantly improved,and the expressions of CD9,CD63,Bax mRNA and protein in liver tissue were significantly reduced(P<0.01),while the expression of Bcl-2 mRNA and protein significantly increased(P<0.01).Conclusion Zuogui Jiangtang Qingzhi Prescription may inhibit liver cell apoptosis,improve liver injury,and exert therapeutic effects on T2DM combined with NAFLD by reducing the expression of CD9 and CD63,down-regulating Bax,and increasing Bcl-2 expression.
