The glymphatic system(GS)is a unique toxic sub-stance clearance system in brain, which is very important for maintaining the microenvironment stability of the central nervous system. The polarization distribution of aquaporin 4(AQP4)lo-cated in the terminal foot of astrocytes affects the function of GS and participates in the pathological progress of many neurodegen-erative diseases, but the detailed regulation mechanism of AQP4 polarization distribution has not been systematically summarized. Therefore, this paper systematically combs the mechanism of reg-ulating the polarization distribution of AQP4 from the perspective of the composition integrity of dystrophin-glycoprotein complex(DGC)and basement membrane foot complex, and summarizes the potential drug and non-drug therapies for targeted regulation of AQP4 polarization distribution at present, aiming at providing new target reference and theoretical basis for targeted regulation of AQP4 polarization to prevent and treat neurodegenerative dis-eases.

Aim To explore the relation between the protective effect of hyperoside(Hyp)on energy metab-olism and apoptosis in cardiomyocytes after myocardial infarction(MI)and the regulation of peroxisome pro-life rator-activated receptor α(PPARα)pathway. Methods Mouse myocardial infarction injury model was established by ligation of left anterior descending coronary artery. The mice that were successfully liga-ted were randomly divided into the following groups:MI group,Hyp(9,18 and 36 mg·kg-1)group,posi-tive fenofibrate group(120 mg·kg-1)and Hyp(36 mg·kg-1)+PPARα inhibitor GW6471 group. In addition,a sham group was set up,only threading with-out ligature. The mice were administered different kinds of drugs by gavage for seven days, once daily.ECG changes were recorded in mice 5 min before,5 min after surgery, and 1 hour after day 7 using the BL-420F biofunction system. The diagnostic ultrasound in-strument was used to examine the heart structure and function of each group of mice. The influence of myo-cardial histology was observed by hematoxylin and eo-sin (HE) staining and Sirius red(Sirius Red)stai-ning.The changes of creatine kinase isozymin-MB(CK-MB),cardiac troponin Ⅰ(cTnⅠ)and lactate de-hydrogenase(LDH)in mouse serum were assessed by ELISA kit.The expressions of PPARα signaling related proteins and apoptosis-related proteins were detected by immunofluorescence and Western blot. Results Compared with MI mice,Hyp could significantly im-prove the ECG abnormality of MI mice, increase the left ventricular ejection fraction(LVEF)and left ventricu-lar short axis shortening rate(LVFS),reduce serum cTnⅠ content,CK-MB and LDH activity, and reduce myocardial fibrosis, infarct area and cardiomyocyte ap-optosis in the MI area.Meanwhile, this research found that Hyp could activate PPARα signaling pathway and regulate apoptosis-related proteins. However, the car-dioprotective effect of hyperoside was reversed by the combination with the treatment of the PPARα signaling inhibitor,GW6471.Conclusions Hyp has an impro-ving effect on energy metabolism and apoptosis in mice after MI,and the mechanism may be related to its acti-vation of PPARα signaling pathway.

Objective The pathophysiological process of acute high-altitude hypoxia-induced lung injury affects protein expression levels, which are mainly evaluated by Western blot.No systematic study has investigated changes in internal control proteins as calibration loading amounts. Methods Lung injury at an altitude of 6000 m was induced in a low-pressure,low-oxygen chamber for 8,24,and 72 h using C57BL/6J mice.Establishment of the model was confirmed by hematoxylin and eosin staining.Expression levels of various internal control proteins, including vinculin,α-tubulin,eukaryotic translation initiation factor 5(EIF5),β-actin,and glyceraldehyde 3-phosphate dehydrogenase(GAPDH)were detected by Western blot, and total protein expression was detected by Coomassie blue staining.Furthermore, the lung injury model in vitro was establised by using,Bronchial epithelial cell(BZAS-2B)andhunman umbilical vein endothelial cells(HUVECS)confirmed by TUNEL staining.Expression levels of internal control proteins were detected by Western blot, and total protein expression was detected by Coomassie Blue staining. Results Acute 8,24,and 72 h hypoxic models were successfully established in lung tissue, demonstrating consistent total protein expression and stable levels of the internal reference proteins vinculin,α-tubulin,EIF5,andβ-actin.GAPDH expression was elevated in the HH8 h,HH24 h,and HH72 h groups compared with the normoxia(Nor)group, but only the increase at HH72 h groups was significant.Similarly,8,24,and 48 h hypoxic models were successfully established in BEAS-2B cells and HUVECs,with consistent total protein expression. In BEAS-2B cells, expression levels of the internal reference proteins β-actin and GAPDH were consistent with the normoxic control(NC)group, while vinculin,α-tubulin,and EIF5 expression levels were significantly reduced under hypoxic conditions for up to 24 h.In HUVECs,vinculin and α-tubulin expression levels were also consistent with the NC group, while EIF5,β-actin,and GAPDH expression levels were significantly reduced at 8 h and increased at 48 h.Conclusions Acute hypoxia induces lung tissue injury, and protein expression levels of the internal reference proteins vinculin,α-tubulin,EIF5,and β-actin are stable, making them suitable internal references for Western blot.Additionally,Western blot detected differential expression levels of the internal reference proteins vinculin,α-tubulin,EIF5,β-actin,and GAPDH in BEAS-2B cells and HUVECs,as the most important in vitro lung tissue models of hypoxia-induced injury.

This study was aimed at investigating the effects and mechanisms of Toxoplasma gondii type Ⅰ(RH strain)ROP16 protein on proliferation and the cell cycle in mouse alveolar macrophage MH-S cells. We constructed a Toxoplasma gondii type Ⅰ(RH)ROP16 overexpression lentivirus, transduced MH-S cells, and then screened cells with puromycin to obtain a cell line stably overexpressing ROP16Ⅰ.RT-qPCR and western blotting were used to verify expression effects,CCK-8 assays were used to detect cell proliferation activity, and flow cytometry was used to detect cell cycle changes.Western blotting and RT-qPCR were used to detect the expression levels of p53,p21,CDK6,Cyclin D1,STAT3,p-STAT3(Y705),and JAK1 proteins or genes, and immunofluorescence was used to detect the expression levels of ROP16Ⅰ and p-STAT3(Y705)and their subcellular co-localization in MH-S cells.ROP16Ⅰ protein and gene expression were detected in MH-S cells transduced with lentivirus for ROP16Ⅰ overexpression.CCK-8 assays revealed that ROP16Ⅰ promoted the proliferation of MH-S cells(P<0.01)and enhanced cell viability.Flow cytometry revealed that ROP16Ⅰ overexpression decreased the G0/G1 phase and elevated the G2 and S phases of the cell cycle in MH-S cells(P<0.01 or P<0.05).Compared with the MH-S cell group and MH-S-empty vector group, the MH-S-ROP16 cell group showed lower expression of p53 and p21 proteins;higher expression of CDK6,Cyclin D1,p-STAT3(Y705),and JAK1 proteins;lower expression of p53 and p21 mRNAs;and higher expression of CDK6 and Cyclin D1 mRNAs(all P<0.01).Immunofluorescence revealed that ROP16Ⅰ co-localized with p-STAT3(Y705)in the nucleus and surrounding cytoplasm. Therefore,Toxoplasma gondii type Ⅰ(RH)ROP16Ⅰ protein activates the JAK-STAT3 pathway;shortens the G0/G1 phase and lengthens the G2/S phase of the cell cycle;and promotes cell proliferation. These findings provide a theoretical basis for revealing the mechanism of immune evasion of Toxoplasma gondii, and lay a foundation for research on the prevention and treatment of Toxoplasma gondii pneumonia.

Chinese Journal of Industrial Hygiene and Occupational Diseases 2025;43(2):91-100
Objective:To investigate the incidence of shoulder work-related musculoskeletal disorders (WMSDs) among occupational population in China, and to explore their intrinsic association with personal and work-related factors. Methods:In April 2024, 73497 valid questionnaires of the Chinese version of the Musculoskeletal Disorders Electronic Questionnaire were retrospectively analyzed from June 2018 to December 2023 in 22 provinces and 29 key industries in China, and the general information, occurrence of WMSDs and related risk factors of key occupational populations in different regions in China were collected. By using Chi-square test and confirmatory factor analysis, the relationship between shoulder fatigue and pain in key occupational groups and individual factors, work type, work posture and work organization was discussed, and the internal relationship was analyzed based on structural equation model. Results:Higher incidence of shoulder fatigue and pain were associated with female, lack of physical exercise, uncomfortable working posture and neck leaning forward ( P<0.05). Structural equation model analysis showed that work type, work posture and work organization were strongly correlated ( r=0.58, 0.55). Work organization and work type were strongly correlated with shoulder fatigue ( r=0.65) and moderately correlated with shoulder fatigue ( r=0.21). Shoulder fatigue was moderately associated with shoulder pain ( r=0.40). Individual factors, work type, work posture and shoulder fatigue could directly affect shoulder pain ( OR=0.07, -0.09, 0.17 and 0.40), and work type and work posture could also indirectly affect shoulder pain through shoulder fatigue ( OR=0.08, 0.03). Work organization only indirectly affected shoulder pain through shoulder fatigue ( OR=0.26) . Conclusion:The main influencing factor of shoulder pain is shoulder fatigue, followed by work posture and individual factors. Structural equation model can better reflect the complex relationship between work type, work posture and work organization and shoulder WMSDs. Improving work posture and work organization may be an effective way to control the influence of shoulder fatigue on shoulder pain.
