1.The Profile of Gut Microbiota in Carcinogenesis Driven by Mutant EGFR in Non–Small Cell Lung Cancer

Purpose:
Accumulating evidence has clarified that gut dysbiosis is involved in lung cancer development and progression. Although the relationship between tumors and gut microbiota has been extensively studied using clinical samples, no studies have examined the association between mutant epidermal growth factor receptor (EGFR)–induced lung carcinogenesis and dysbiosis in gut microbiota. Therefore, we investigated the gut microbiota profiles in stool samples from human lung-specific conditional EGFR-mutant transgenic mice during lung tumor carcinogenesis.
Materials and Methods:
Stool samples were collected before tamoxifen treatment (V1) and at each time point following mutant EGFR expression in lung tissue (V2) and lung tumor appearance (V3). Fecal 16S rRNA taxonomy was analyzed to assess microbial diversity, composition, and dynamic changes at each time point.
Results:
We found that microbiota richness and diversity were significantly elevated when tumors developed and grew in the lung. Phylogenetic analysis of the microbial community revealed that Lachnospiraceae, Ruminococcaceae, Porphyromonadaceae, Rhodospirillaceae, Odoribacteraceae, and Desulfovibrionaceae showed a significant increase at the V3 stage compared to the V1 stage at the family level. In contrast, Lactobacillaceae, Bacteroidaceae, Muribaculaceae, Coriobacteriaceae, and Rikenellaceae significantly decreased at the V3 stage compared to the V1 stage. Furthermore, Lactobacillus species, also known as short chain fatty acid-producing bacteria, were relatively abundant at the V1 stage but were depleted with the occurrence of lung tumors at the V3 stage.
Conclusion
Changes in gut microbiota, such as Lactobacillus species, may be a predictive factor for the emergence and progression of tumors in an animal model of lung adenocarcinoma induced by mutant EGFR.
2.Exploring methylation signatures for high de novo recurrence risk in hepatocellular carcinoma

Background/Aims:
Hepatocellular carcinoma (HCC) exhibits high de novo recurrence rates post-resection. Current post-surgery recurrence prediction methods are limited, emphasizing the need for reliable biomarkers to assess recurrence risk. We aimed to develop methylation-based markers for classifying HCC patients and predicting their risk of de novo recurrence post-surgery.
Methods:
In this retrospective cohort study, we analyzed data from HCC patients who underwent surgical resection in Korea, excluding those with recurrence within one year post-surgery. Using the Infinium Methylation EPIC array on 140 samples in the discovery cohort, we classified patients into low- and high-risk groups based on methylation profiles. Distinctive markers were identified through random forest analysis. These markers were validated in the cancer genome atlas (n=217), Validation cohort 1 (n=63) and experimental Validation using a methylation-sensitive high-resolution melting (MS-HRM) assay in Validation cohort 1 and Validation cohort 2 (n=63).
Results:
The low-risk recurrence group (methylation group 1; MG1) showed a methylation average of 0.73 (95% confidence interval [CI] 0.69–0.77) with a 23.5% recurrence rate, while the high-risk group (MG2) had an average of 0.17 (95% CI 0.14–0.20) with a 44.1% recurrence rate (P<0.03). Validation confirmed the applicability of methylation markers across diverse populations, showing high accuracy in predicting the probability of HCC recurrence risk (area under the curve 96.8%). The MS-HRM assay confirmed its effectiveness in predicting de novo recurrence with 95.5% sensitivity, 89.7% specificity, and 92.2% accuracy.
Conclusions
Methylation markers effectively classified HCC patients by de novo recurrence risk, enhancing prediction accuracy and potentially offering personalized management strategies.

5.Clinical Patterns and Risk Factors of Occupational Contact Dermatitis in Hairdressers: A Questionnaire-based Cross-sectional Study

Background:
Occupational contact dermatitis (OCD) is prevalent among hairdressers due to frequent exposure to chemicals like hair dyes and bleaching agents. Despite the risks, awareness among hairdressers remains low, leading to underreporting and inadequate preventive measures.
Objective:
This study evaluated hairdressers’ awareness of harmful hair dye ingredients, their experiences with OCD, and the association with product usage patterns.
Methods:
A cross-sectional study involving 100 hairdressers in Korea examined the relationship between work experience, product usage, and OCD. Chi-square tests and multivariate regression identified significant correlations.
Results:
Among the participants, 51% reported experiencing adverse skin reactions, with the hands being the most commonly affected area. Longer work experience as a hairdresser was significantly associated with the occurrence of adverse effects ( p=0.046). Notably, shampoo was identified as a suspected causative material significantly more often by the severe group compared to the non-severe group (28.0% vs. 3.8%, p=0.04).
Conclusion
Chemical exposure and frequent wet work contribute to high rates of OCD among hairdressers. Poor glove usage, especially during shampooing due to inconvenience, is a major risk factor. Raising awareness, promoting proper glove use, and improving workplace safety training are essential for reducing these skin conditions.
6.Antler abnormalities with testicular atrophy in a wild roe deer (Capreolus pygargus): a case report

Deer antlers are regenerate annually under hormonal control, particularly testosterone. Here, we report a case of antler deformity associated with testicular atrophy in a wild roe deer (Capreolus pygargus) in South Korea. Both antlers were abnormally enlarged and entangled in agricultural netting. Gross and histopathological examination revealed severe testicular atrophy, interstitial fibrosis, and hindlimb muscle necrosis. Serum testosterone was extremely low (0.032 ng/mL). These findings explain the abnormal antler growth and suggest susceptibility to capture myopathy due to hormonal imbalance. This is the first documented case in South Korea linking antler abnormalities to testicular atrophy in wild roe deer, highlighting the impact of endocrine disruption on cervid health.
7.Tumors in pet hamsters in Korea: a retrospective study of 80 cases (2017–2023)

This study analyzed 80 biopsy specimens from pet hamsters in South Korea, focusing on naturally occurring tumors, particularly integumentary tumors. Tumors were most commonly found in the integumentary system (71.3%), followed by the reproductive (16.3%) and hematopoietic systems. In Djungarian hamsters, cutaneous atypical fibroma (27.8%) was the most frequent integumentary tumor, with the abdomen being the most affected site (42.6%). These findings highlight the importance of monitoring common neoplastic sites for better clinical management and preventative care, ultimately improving the health and longevity of pet hamsters.

Objective: To evaluate the effects of Capsosiphon fulvescens (C. fulvescens) ethanolic extract on inflammation in lipopolysaccharide (LPS)-induced RAW296.7 macrophages. Methods: The protective effects of C. fulvescens ethanolic extract on LPS-induced inflammation in RAW264.7 macrophages were assessed using biochemical analysis, including enzyme-linked immunosorbent assay, quantitative reverse transcription-polymerase chain reaction, and Western blot analysis. To examine reactive oxygen species (ROS) production, flow cytometry analysis, and immunofluorescence staining were used. Furthermore, the modulatory effect of C. fulvescens ethanolic extract on NF-κB activation was investigated. Results: C. fulvescens ethanolic extract significantly attenuated LPS-induced levels of pro-inflammatory cytokines and notably reduced the secretion and mRNA levels of LPS-mediated matrix metalloproteinases. In addition, C. fulvescens ethanolic extract decreased ROS production and suppressed the TLR4/NF-κB signaling pathway. Conclusions: C. fulvescens ethanolic extract alleviates inflammation as well as oxidative stress by modulating the TLR4/NF-κB signaling in LPS-induced RAW264.7 macrophages. C. fulvescens can be used as a potential therapeutic agent to suppress inflammation and oxidative stress-associated diseases.

Recently, herbal medicines have gained attention for the treatment of benign prostatic hyperplasia (BPH), a common disease in elderly men. In this study, we aimed to determine the effect of ethanol extract of Asparagi radix (EAR), which is traditionally used to treat various diseases, on BPH development using a testosteroneinduced BPH model.MATERIALS/METHODS: Testosterone propionate (TP)-treated Sprague–Dawley rats were used to establish a BPH model in vivo. EAR was orally administered along with TP, and finasteride was used as a positive control. All rats were sacrificed at the end of the experiment, and pathological changes in the prostate tissue and levels of key biomarkers associated with BPH pathogenesis were assessed.
RESULTS:
Oral administration of EAR significantly inhibited TP-induced BPH by reducing the prostate weight, lumen size, and epithelial thickness in a concentration-dependent manner. EAR also significantly abrogated the expression of 5α-reductase type 2 (SRD5A2), proliferating cell nuclear antigen, and prostate-specific antigen (PSA) induced by TP.Additionally, serum levels of testosterone, dihydrotestosterone, and PSA were elevated in the TP-induced group but decreased in the EAR-treated group. EAR also decreased the expression levels of the androgen receptor (AR) and its coactivators in TP-induced BPH model rats.
CONCLUSION
Our findings revealed that EAR protected against BPH by inhibiting 5α-reductase activity and AR signaling pathway, suggesting its potential for BPH treatment.
