1.Effect of circHIPK2 on angiotensin Ⅱ-induced apoptosis of vascular endothelial cells through regulation of the miR-7-5p/TCF4 axis

Objective To investigate the effect of circRNA-homeodomain-interacting protein kinase 2(circHIPK2)on angiotensinⅡ(AngⅡ)-induced apoptosis of vascular endothelial cells through the regulation of the miR-7-5p/transcription factor 4(TCF4)axis.Methods Human umbilical vein endothelial cells(HUVECs)were randomly divided into the control,model,negative control cotrans-fection,circHIPK2 knockdown,miR-7-5p overexpression,and circHIPK2 knockdown+miR-7-5p knockdown groups.Except for the control group,all other groups were administered 10 nmol/L Ang Ⅱ to establish a hypertensive injury model.The circHIPK2,miR-7-5p,and TCF4 mRNA expression levels were detected after transfection.Apoptosis,proliferation,mitochondrial membrane potential,reactive oxygen species(ROS),antioxidant enzymes,pro-inflammatory factors,and TCF4 protein expression were assessed.Results Compared with the control group,the expressions of circHIPK2 and TCF4 mRNA,cell apoptosis rate,relative expression of ROS,levels of IL-6,IL-1β,and IL-18,and expressions of Bax and TCF4 protein increased,and cell viability,miR-7-5p mRNA expression,mitochondrial mem-brane potential,activities of superoxide dismutase(SOD)and catalase(CAT),and Bcl-2 protein expression decreased in the model group(P<0.05).Both circHIPK2 knockdown and miR-7-5p overexpression reversed Ang Ⅱ-induced pathological changes in vascular endothelial cells.miR-7-5p knockdown reduced the effect of circHIPK2 knockdown on pathological cellular changes in the model group.Conclusion circHIPK2 knockdown can weaken TCF4 expression by upregulating miR-7-5p,thereby reducing Ang Ⅱ-induced inflam-mation and oxidative stress in vascular endothelial cells and ultimately inhibiting cell apoptosis.

3.Effect of clopidogrel bisulfate tablets combined with hyperbaric oxygen on patients with coronary heart disease after TRA-pPCI

Objective:To investigate the effect of clopidogrel bisulfate tablets combined with hyperbaric oxygen on patients with coronary heart disease after trans-radial access primary percutaneous coronary intervention (TRA-pPCI), and to investigate its influence on the levels of sE-selectin and Adiponectin (ADPN).Methods:A total of 108 patients with coronary heart disease after TRA-pPCI admitted in the Health Management Center of the First Hospital of Shanxi Medical University from July 2016 to October 2018 were randomly divided into control group and observation group, with 54 patients in each group. Both groups were treated with clopidogrel sulfate tablets before and after the operation. The patients in the observation group were additionally treated with hyperbaric oxygen after operation. Clinical efficacy, incidence of major adverse cardiovascular and cerebrovascular events during one-year follow-up after the discharge, blood lipid levels before and after treatment, carotid plaque area, carotid intima-media thickness (IMT), serum inflammatory factors, sE-selectin and ADPN levels were compared between the two groups.Results:The total effective rate of the observation group (96.30%) was significantly higher than that of the control group (81.48%) with statistically significant difference ( P<0.05); the incidence of major adverse cardiovascular and cerebrovascular events during one-year follow-up in the observation group (11.11%) was significantly lower than that of the control group (44.44%) with statistically significant difference ( P<0.01). Compared with those before treatment, the levels of TC, TG, LDL-C, hs CRP, TNF-α, IFN-γ, IL-10, carotid plaque area, and IMT of the two groups after the treatment decreased significantly, while HDL-C increased significantly; and the differences of blood lipid level, carotid plaque area, and IMT after treatment between the two groups were statistically significant ( P<0.05 or P<0.01). The serum levels of sE-selectin of the two groups were significantly lower than those before the treatment, while ADPN levels were significantly higher. And the differences in serum sE-selectin and APN levels between the two groups were statistically significant ( P<0.05 or P<0.01). Conclusion:Clopidogrel bisulfate tablets combined with hyperbaric oxygen therapy can effectively regulate the levels of blood lipid, serum inflammatory factors, serum sE-selectin and ADPN levels in patients with coronary heart disease after TRA-pPCI, and inhibit the formation of atherosclerotic plaque, thereby effectively reducing the incidence of adverse cardiovascular and cerebrovascular events after operation and improving the therapeutic effect on coronary heart disease.
4.Therapeutic effect of hyperbaric oxygen therapy on acute ischemic stroke and its effects on lipid metabolism, serum IL-1β, IL-6, and TNF-α levels

Objective:To investigate the therapeutic effect of hyperbaric oxygen therapy (HBOT) on acute ischemic stroke (AIS) and its effects on lipid metabolism, serum levels of interleukin-1β (IL-1β), interleukin-6 (IL-6), and tumor necrosis factor-α (TNF-α).Methods:A total of 84 AIS patients treated in the First Hospital of Shanxi Medical University from January 2017 to January 2020 were enrolled and divided into observation group ( n=42) and control group ( n=42) by random number table method. The control group received conventional treatment, while the observation group received conventional treatment combined with HBOT. The indicators related to lipid metabolism, the levels of serum inflammatory medicators, brain microcirculation, and prognosis were compared between the two groups. Results:After treatment, the levels of the total cholesterol (TC), triglyceride (TG), and low-density lipoprotein cholesterol (LDL-C) were significantly decreased, and the level of high-density lipoprotein cholesterol (HDL-C) was significantly increased in both groups ( P<0.05); and compared with the control group, the observation group had significantly lower LDL-C level and significantly higher HDL-C level ( P<0.05). After treatment, the levels of serum IL-1β, IL-6, and TNF-α were significantly decreased in both groups, and the three indicators in the observation group were all significantly lower than those in the control group ( P<0.05). The systolic velocity (Vs), diastolic velocity (Vd), and mean velocity (Vm) on the healthy side of carotid artery of the patients in both groups had no significant changes after treatment ( P>0.05), while Vs, Vd, and Vm on the affected side of carotid artery of the patients were significantly increased ( P<0.05), and all of them were significantly higher in the observation group than those in the control group ( P<0.05). Before treatment, the activity of daily living (ADL) score and clinical neurological function deficit (CNFD) score showed no significant difference between the two groups (all P>0.05). After treatment, there was an increase in the ADL scores increased and a decrease in CNFD scores decreased in both groups, and the observation group had significant higher ADL scores and CNFD scores than those of the control group [(73.99±11.15) vs. (54.69±12.03), (8.64±2.25) vs. (12.31±2.03); all P<0.05]. Conclusion:HBOT can effectively reduce the lipid metabolism disorders in AIS patients, alleviate their inflammatory response, improve their brain microcirculation, neurological function deficits, and the activity of daily life, so as to ensure the therapeutic effect.

7.Research advances in the clinical features of liver disease during pregnancy

Severe liver disease during pregnancy is uncommon in clinical practice. The most common cause of liver disease during pregnancy is liver dysfunction, with an overall prevalence rate of approximately 3%. Liver disease during pregnancy is classified into the liver diseases directly caused by pregnancy and those co-existing with pregnancy, i.e., pre-existing liver disease or occasional liver disease during pregnancy. A differential diagnosis of pre-existing and co-existing liver diseases may help to improve maternal and fetal outcome. During clinical diagnosis and selection of treatment and intervention measures, priority should be given to the potential impact on mother and fetus. This article introduces the latest research advances in the general information, pathogenesis, treatment, and pregnancy outcome of major liver diseases during pregnancy and elaborates on the risk of pregnancy and related coping measures for patients with pre-existing liver disease, so as to guide clinical diagnosis and treatment and patient management.
8.Studies on Susceptibility of A. Minimus to P. Yoelii Yoelii

Anopheles minimus was infected with Plasmodium yoelii yoelii under adequate condition. The malaria parasite could develop normally and produce mature oocysts containing sporozoites. Large numbers of free active sporozoites were found in the haemocele but most of them failed to penetrate into the mosquito's salivary glands. Thus the sporogonic cycle of the parasite could not be completed.But in one mosquito, a few sporozoites were found in its salivary glands. It seems that there is an individual difference in the susceptibility of the mosquitoes and this throws some light on the possibility of selecting a susceptible strain of A. minimus to establish the A. minimus-P. yoelii system for malaria research.
