1.Protective effect and mechanism of baicalein on hypoxia-induced cortical neuron injury in rats

OBJECTIVE To explore the protective effect and possible mechanism of baicalein on hypoxia-induced cortical neuron injury in rats. METHODS The cortical neurons of rats (RN-C cells) were studied and cultured under hypoxic conditions (5%CO2, 94% N2, 1%O2) for 24 hours; the effects of different concentrations of baicalein (0.01, 0.1, 1, 10, 100 μmol/L) on the survival rate of hypoxic RN-C cells were investigated; the effects of baicalein (0.1 μmol/L) on the activities of lactate dehydrogenase (LDH) and superoxide dismutase (SOD), the content of malondialdehyde (MDA), migration rate, apoptotic rate, cell cycle and the expressions of cleaved caspase-3, B-cell lymphoma-2 (Bcl-2) and Bcl-2 X protein (Bax) were all detected. RESULTS Compared with control group, the survival rate of cells in the hypoxia group was significantly reduced (P<0.01); 0.01, 0.1 and 1 μmol/L baicalein could reverse survival rate of hypoxia-induced cortical neurons (P<0.05 or P<0.01). Scratch experiments showed that baicalein significantly increased the migration rate of hypoxic RN-C cells (P<0.01). Compared with control group, the activity of LDH in the supernatant and the content of MDA in the cells, apoptotic rate and the proportion of cells in G1 phase, were significantly increased in the hypoxia group, while SOD activity and the proportion of cells in G2+S phase was decreased significantly (P<0.01). The protein expressions of cleaved caspase-3 were increased significantly, while the ratio of Bcl-2/Bax in cells was significantly reduced (P<0.05 or P<0.01). Compared with hypoxia group, the above indexes were all reversed significantly in baicalein group (P<0.01). CONCLUSIONS Baicalein can promote the proliferation and migration of cortical neurons, improve hypoxia-induced cell apoptosis and cell cycle distribution, decrease the activity of LDH in supernatant and the level of cellular lipid peroxidation, and improve antioxidant levels. Its mechanism may be related to regulating the caspase- 3/Bax/Bcl-2 pathway.
2.Effect of Xianlian Jiedu Prescription on Proliferation and Glycolysis of Human Colorectal Cancer HCT-116 Cells and Mechanism

ObjectiveTo explore the effect of Xianlian Jiedu prescription (XLJDP) on the proliferation and glycolysis of human colorectal cancer HCT-116 cells and the underlying mechanism. MethodHCT-116 cells were cultured with XLJDP and then the survival rate was examined by methyl thiazolyl tetrazolium (MTT) assay. The effect on the HCT116 cell proliferation was detected by colony formation assay and 5-ethynyl-2′-deoxyuridine (EDU) incorporation assay. The amount of glucose consumed by HCT-116 cells was measured by glucose test kit, and the amount of produced lactic acid was determined by lactic acid test kit 48 h after the treatment with XLJDP. The expression of glycolysis-related proteins mammalian target of rapamycin (mTOR), phosphorylated mTOR (p-mTOR), glucose transporter 1 (GLUT1), and lactate dehydrogenase (LDHA) was detected by Western blot. ResultThe half-maximal inhibitory concentration (IC50) of XLJDP against HCT-116 cells was 6.82 g·L-1. Compared with the blank group, XLJDP (1.625, 3.25, 6.50 g·L-1) inhibited the proliferation of HCT-116 cells (P<0.05, P<0.01). Moreover, compared with the blank group, XLJDP (1.625, 3.25, 6.50 g·L-1) suppressed glucose uptake and lactic acid production in a dose-dependent manner (P<0.05, P<0.01). The expression of p-mTOR/mTOR, LDHA, and GLUT1 was down-regulated by XLJDP (P<0.05, P<0.01). ConclusionXLJDP can significantly inhibit the proliferation and the Warburg effect of glycolysis in colorectal cancer cells by regulating the mTOR signaling pathway and the down-regulating the expression of LDHA, GLUT1, and other key proteins and enzymes in glycolysis.
3.Comparison and enlightenment of undergraduate education in stomatology between China and Russia

For adapting to the change of modern medical model and suiting social demand of high quality dentists, this paper compares the differences of undergraduate education in stomatology between China and Russia by using the method of Bereday comparative education, trying to provide some reference for the reform of Chinese stomatology education. Curriculum arrangement in China is being constantly explored based on the Russian model. In China, courses are divided into basic medicine, clinical medicine and stomatology in three stages, making students relatively passive in learning. And the ratio of practice to lecture is far more higher in Russia than that in China. Thus, it's necessary to optimize structure and balance the distribution of stomatological curriculum in China, improve and popularize learning models such as problem -based learning (PBL) and so on, so as to promote the development of stomatology education of China to a higher level.
4.Comparison of stomatology education systems between two medical universities in China and Russia

By comparing the similarities and differences in the higher education system and degree levels of stomatology,especially in the undergraduate-level courses,between I.M.Sechenov First Moscow State Medical University and Harbin Medical University,we found that:the class hours for undergraduates majoring in stomatology in Russia are three times as many as those in China,and the former's class hours for PBL accounts for 80% of its total class hours,which can greatly develop the medical students' competence of life-long learning and innovation.Russia focuses more on the early exposure to oral medical and clinical education.Moreover,they make stomatology penetrate the whole education process.The system of combing postgraduate medical education with residency training in Russia has been developed early and relatively mature,which can provide example for Chinese standardized training of oral residents.The study for the current education system of stomatology in Russia is of great significance in strengthening the cooperation between China and Russia on stomatology education and promoting the reform of Chinese stomatology education.
5.The effect of hyperbaric oxygen therapy on cognitive dysfunction caused by traumatic brain injury : A diffusion tensor imaging study

Objective To observe any effect of hyperbaric oxygen therapy on cognitive dysfunction caused by traumatic brain injury,and to explore possible neural mechanisms.Methods Sixty-four patients with cognitive impairment after a traumatic brain injury were randomly divided into a hyperbaric oxygen group (n =32) and a control group (n=32) using a random number table.Both groups accepted routine medical therapy and cognitive rehabilitation training,but the hyperbaric oxygen group additionally received hyperbaric oxygen treatment.Both groups' cognitive functioning was assessed using the Mini Mental State Examination (MMSE) and the Montreal Cognitive Assessment (MoCA) before and after the treatment.Fifteen patients were randomly selected from both groups to receive T1WI and diffusion tensor imaging scans.The correlation between the two evaluation resuhs was analyzed.Results After the intervention,improvement was observed in the average MMSE and MoCA scores of both groups,with the improvement in both average scores in the hyperbaric oxygen group significantly greater than among the control group.There was no significant correlation between the fractional anisotropy (FA) values of damaged white matter and the average MMSE or MoCA score in the controi group,but in the hyperbaric oxygen group there were significant positive correlations between the FA values of the corpus callosum,the anterior limb of the internal capsule and the left superior longitudinal fasciculus and the average MMSE and MoCA scores.Conclusions Hyperbaric oxygen therapy combined with rehabilitation training can further improve cognition after a traumatic brain injury.This is probably due to its adjusting the structure and function of the corpus callosum,of the anterior limb of the internal capsule and of the left superior longitudinal fasciculus.
6.Left ventricular flow energy loss at diastolic phase in patients after video-assisted thoracic lung resection by ultrasonic vector flow mapping

Objective To evaluate the changes of left ventricular structure and cardiac function in patients after video-assisted thoracic pneumonectomy by ultrasonic vector flow mapping (VFM) and quantitatively evaluate the diagnostic value of left ventricular flow energy loss(EL) for pneumonectomy patients with impaired cardiac function.Methods Thirty-six pneumonectomy patients were selected as case group and 30 health cases as control group.The echocardiographic parameters were acquired and left ventricular volume,left atrial volume and parameters of left diastolic founction were routinely measured.The EL of the left ventricularat early diastole and late diastole under the VFM mode were acquired.Results ①Left ventricular volume and left atrial volume comparsion:there was no significant difference in left ventricular end diastolic volume(LVEDV),left ventricular end systolic volume (LVESV),left ventricular ejective fraction(LVEF),and left atrial end diastolic volume(LAVd) between the two groups (all P > 0.05);left atrial end systolic volume(LAVs) and left atrial emptying volume(LAVe) in case group were mild lower than control group(P <0.05).②Comparsion of diastolic function parameters:E,A,E/e and HR of case group were significant higher than those in control group (all P <0.05),EDT and E/A ratio in case group were significant lower than those in control group (all P < 0.05),there was no significant difference of IVRT between two groups (P >0.05).③Comparison of EL at early diastole:only ELTB and ELAM had mild difference between case group and control group (all P <0.05).④Comparison of EL at late diastolic:EL of entirety and each segment of left ventricle of case group increased (all P < 0.05).Conclusions VFM can quantitatively evaluate the left ventricular flow characteristics after video assisted thoracic pneumonectomy,and the increase in EL may reflect the left ventricular diastolic dysfunction in patients after pneumonectomy.
7.Resting state function magnetic resonance imaging study on the effect of hyperbaric oxygen on the improvement of executive function of the patients with traumatic brain injury

Objective To observe the effect of hyperbaric oxygen therapy (HBOT) on the improvement of executive function of the patients with traumatic brain injury (TBI) by using resting state functional magnetic resonance (rs-fMRI).Methods Fifty patients with executive disorder after traumatic brain injury were randomly divided into the HBO group (n =25) and the control group (n =25).The patients of the control group received routine treatment and executive function rehabilitation training,while the patients of the HBO group were supplemented with HBOT in addition to the treatment received by the control group.The applied HBO pressure was 2.0 MPa,one session a day with a duration of 105 minutes,five days a week,for a total of 20 sessions.Both before and after treatment,all the subjects received executive function assessment and rs-fMRI examination by using the drawing of clock test,the Chinese version Stroop color word test and the Wechsler adult intelligence scale similarity subtest.In the study,we closely observed the effects of HBOT on the intensity of metabolic activities in the brain,changes in neuron cell synchronization and coordination in TBI patients through the approach of rs-fMRI technique combined with ReHo and ALFF analysis methods.At the same time,the correlation between imaging data and neuropsychological assessment results was also analyzed in the study.Results (1) After intervention,Stroop-A time-consumption and Stroop-C time-consumption,correct reading scores,as well as interference scores of the HBO group were all significantly better than those of the control group,and statistical significance could be found when comparisons were made between the 2 groups (P < 0.05).(2) Following intervention with different methods,obvious differences in the ALFF value of superior parietal lobe in the common brain regions of the TBI patients in the 2 groups could be seen,with statistical significance (P < 0.05).Correlation analysis revealed that there was a positive correlation between the superior parietal lobe and interference time in the HBO group (P < 0.05).(3) There was also a positive correlation between correct readings of Stroop-A in the middle frontal gyrus of the HBO group (P < 0.05).Conclusion Executive function rehabilitation training,HBO therapy combined with executive function rehabilitation training could improve the executive function of TBI patients to various degrees.HBO therapy combined with executive function rehabilitation training was more beneficial to further improvement of executive function in patients with TBI.The improvement of executive function in TBI patients by HBO therapy might be achieved through the regulation of spontaneous activity and regional homogeneity of neurons in the middle frontal gyms,and superior parietal lobe.

9.Survey on the Cognition and Attitude towards Dignity Therapy in College Students

Objective:To investigate the cognition of terminal stage patients'dignity therapy,attitude towards its concept and scheme,willingness of its promotion in college students and to gain some recommendations for improvement.Methods:Combined quantitative and qualitative methods,an anonymous questionnaire survey was conducted in a convenience sample of 220 college students,of which 20 students were selected for in-depth interviews.Results:Only 17.73% of students had a correct understanding of dignity therapy,94.98% of them accepted its concept,92.96% of them held a positive attitude,74.38% of them were willing to promote dignity therapy,in which female students were dominated (P =0.041).Interview results showed that the contents of dignity therapy were to some extent stiff and not fit for Chinese patients,which might impede its promotion.Conclusion:College students lack the cognition of dignity therapy.But they hold a positive attitude and are willing to promote dignity therapy when they get to know its concept and contents.As seen,dignity therapy has a good future but there are still some obstructions in the promotion,and need to be localized.
10.The research advance on the effect of several common used general anesthetics on the intraocular pressure

General anesthetics were widely used in ophthalmology surgical and other surgical anesthesia, sedation and animal experiments. The current study found that anesthesia drugs such as ketamine, sevoflurane and propofol had a great influence on eyes. Elevated intraocular pressure was a risk factor for glaucoma occurrence and progress and it can change the intraocular structure, and damage the visual function. So in this paper, the effects of several kinds of general anesthetics commonly used were systematically described, in order to hold the attention of the clinicians and the researchers.
