1.Preliminary study on the characterization of exosomes in cryoprecipitate

Objective: To explore the types and levels of exosomes in cryoprecipitate from donors of different blood types, genders, and age groups, and to preliminarily characterize their specific proteins. Methods: Exosomes were extracted from cryoprecipitate using the polymer precipitation method. The exosomes were identified by particle size analysis, transmission electron microscopy, and Western blot. Finally, the specific proteins CD63, CD81, CD9, Cytochrome C, Syntenin-1, and VLA-4 of exosomes were characterized using Luminex. Results: The purified exosomes exhibited a double-membrane vesicle structure under microscopy, appearing saucer-shaped or as hemispheres with a concave side. They specifically expressed the CD63 and TSG101 proteins, while the endoplasmic reticulum protein Calnexin was not expressed. The particle sizes(nm) of exosomes from blood types A, B, O, and AB were 119.6±47.5, 120.5±47.4, 133.3±54.1 and 103.1±35.3, respectively. Taking the exosomal specific protein CD63 as an example, its expression levels(pg/mL) in cryoprecipitate from female and male donors were 128.22±63.52 and 117.39±40.84, respectively; the expression(pg/mL) levels in the 20-40 and 40-60 age groups were 114.20±37.28 and 131.41±64.98, respectively; and the expression levels(pg/mL) in the four blood types A, B, O, and AB were 109.58±37.79, 127.45±44.46, 131.89±40.59, and 122.29±78.68, respectively. The expression levels of CD63 in exosomes from different blood types, genders, and age groups were similar, with no statistically significant differences (all P>0.05). The distribution of the other five exosomal-specific proteins was also comparable among the normal blood donors. Conclusion: The expression levels of exosomal-specific proteins in cryoprecipitate from normal blood donors are not associated with blood type, gender, and age group.

3.Enhanced MR texture analysis in the differential diagnosis of solitary brain metastasis and high-grade glioma

Objective To investigate the value of enhanced MR texture analysis in the differential diagnosis of single brain metastasis (SBM) and high grade glioma (HGG). Methods The preoperative MRI images of 21 SBM and 23 HGG patients confirmed by pathology were retrospectively analyzed. The T1WI enhanced images were used for texture analysis to obtain the mean, median, entropy, skewness, kurtosis, heterogeneity and other parameters. An independent sample t-test or Mann-Whitney U test was used to statistically analyze the values of the parameters between the two groups, and ROC evaluation parameters were used to identify the diagnostic efficacy of SBM and HGG. Results There were significant differences in entropy and heterogeneity parameters between SBM and HGG: (4.838 ± 0.523 vs. 5.358 ± 0.449, 0.276 ± 0.066 vs. 0.331 ± 0.101), P<0.05. When the cut-off value of entropy was 4.88, the sensitivity of differential diagnosis between SBM and HGG was 47.6%, specificity was 100.0%, and the area under ROC curve was the largest (0.745). Conclusions The entropy value and heterogeneity have certain value in distinguishing SBM and HGG in MRI texture analysis.
4.Analysis of risk factors for carotid atherosclerosis in patients with renal glomerular diseases

Objective To study the relationship between intrarenal arterial lesions(IAL)and carotid atherosclerosis(AS)in patients with renal glomerular disease.Methods 251 cases with IAL were selected and 129 age-,pressure-and eGFR-matched renal glomerular disease cases without IAL were randomly selected.The carotid intima-media thickness(IMT)was detected.Clinical and laboratory examination and renal histological characteristics were compared the two guoups.Results ①The detection rate of carotid AS was higher in patients with IAL than those without IAL(38.2%vs.20.2%),and higher in patients≥40 years than in patients<40 years(51.3%vs.13.1%)(P<0.05 for each). ②The carotid AS group was older and had higher frequencey of fasting blood glucose,body mass index,smoking,and family history of hypertension,longer duration of established hypertension and renal glomerular disease,higher prevalence of hypertension,IAL and renal lesions,and had lower eGFR than the carotid normal group(all P<0.05).Binary logistic regression analysis revealed that IAL and age emerged as an independent risk factor for carotid AS(OR=1.826 and 1.129,P=0.001 and 0.003).Conclusion The intrarenal arterial lesion is an independent risk factor for carotid atherosclerosis in patients with renal glomerular disease.Controlling blood glucose,blood pressure,weight,smoking quit and delaying progression of kidney disease have important significance in relieving or preventing atherosclerosis and intrarenal arterial lesions of patients with renal glomerular diseases.
5.Effect of tumor suppressor phosphatase and tensin homology deleted in chromosome 10 on renal interstitial fibrosis in IgA nephropathy

AIM:To explore the potential correlation between the expression of phosphatase and tensin homology deleted on chromosome 10(PTEN) and RIF in IgA nephropathy.METHODS:Forty-seven patients diagnosed as primary IgA nephropathy by renal histology were involved.10 specimens from normal renal tissue of renal carcinoma served as control group.Tubulointerstitial lesion(TIL) was classified by using Katafuchi scale,including no TIL(group I),mild TIL(group II),moderate TIL(group III),severe TIL(group IV).The expressions of PTEN,TGF-?1,?-SMA and ColⅢ in renal tissue were detected by immunohistochemistry.PTEN mRNA was detected by in situ hybridization.RESULTS:Renal tissues from renal biopsy showed abundant expressions of PTEN and PTEN mRNA in endochylema of renal tubular epithelial cells,and negligible expression in glomeruli.With the progress of TIF in IgA nephropathy,the expressions of PTEN and PTEN mRNA decreased gradually(P
