1.Piezo1 Mediates the Regulation of Substrate Stiffness on Primary Cilia in Chondrocytes

Objective To investigate how substrate stiffness regulates the morphology of primary cilia in chondrocytes and to illustrate how Piezo1 mediates the morphology regulation of primary cilia by substrate stiffness.Methods Polydimethylsiloxane(PDMS)curing agent and the main agent(Dow Corning,Beijing,China)were mixed at the ratio of 1∶10(stiff),1∶50(medium stiffness),and 1∶70(soft),respectively,to prepare substrate films with the thickness of 1 mm at different levels of stiffness,including stiff substrate of(2.21±0.12)MPa,medium-stiffness substrate of(54.47±6.06)kPa,and soft substrate of(2.13±0.10)kPa.Chondrocytes were cultured with the substrates of three different levels of stiffness.Then,the cells were treated with Tubastatin A(Tub A)to inhibit histone deacetylase 6(HDAC6),Piezo1 activator Yoda1,and inhibitor GsMTx4,respectively.The effects of HDAC6,Yoda1,and GsMTx4 on chondrocyte morphology and the length of primary cilia were analyzed through immunofluorescence staining.Results The stiff substrate increased the spread area of the chondrocytes.Immunofluorescence assays showed that the cytoskeleton and the nuclear area of the cells on the stiff substrate were significantly increased(P<0.05)and the primary cilia were significantly extended(P<0.05)compared with those on the medium-stiffness and soft substrates.However,the presence rate of primary cilia was not affected.The HDAC6 activity of chondrocytes increased with the decrease in substrate stiffness.When the activity of HDAC6 was inhibited,the cytoskeletal area,the nuclei area,and the primary cilium length were increased more significantly on the stiff substrate(P<0.05).Further testing showed that Piezo1 activator and inhibitor could regulate the activity of HDAC6 in chondrocytes,and that the length of primary cilia was significantly increased after treatment with the activator Yoda1(P<0.05).On the other hand,the length of primary cilia was significantly shortened on the stiff substrate after treatment with the inhibitor GsMTx4(P<0.05).Conclusion Both substrate stiffness and Piezo1 may affect the morphology of chondrocyte primary cilia by regulating HDAC6 activity.
2.Effect of trunk control training during unstable sitting on patellofemoral pain syndrome

ObjectiveTo explore the effect of trunk control training during unstable sitting on knee pain and function in patients with patellofemoral pain syndrome. MethodsFrom January, 2019 to December, 2021, 41 patients with patellofemoral pain syndrome in Beijing Rehabilitation Hospital were randomly divided into control group (n = 20) and experiment group (n = 21). Both groups accepted routine rehabilitation, and the experiment group accepted trunk control training during unstable sitting in addition, for four weeks. They were assessed with Visual Analogue Scale for pain (VAS) and Anterior Knee Pain Scale (AKPS), and measured stability indexes with Balancer before and after treatment. ResultsAll the VAS score, AKPS score, and the overall, anterior-posterior and left-right stability indexes improved in both groups after treatment (|t| > 12.089, P < 0.001); and improved more in the experiment group than in the control group (|t| > 5.864, P < 0.001). ConclusionTrunk control training during unstable sitting may improve knee pain and function, and motor control.
3.Meta-analysis of the Efficacy of Gongliuqing Capsules Combined with Mifepristone in the Treatment of Uterine Leiomyoma

OBJECTIVE:To systematically evaluate therapeutic efficacy of Gongliuqing capsules combined with mifepristone in the treatment of uterine leiomyoma ,in order to provide evidence-based reference for clinical medication. METHODS :Retrieved from Cochrane Library ,PubMed,Embase,CJFD,VIP,CBM and Wanfang database ,randomized controlled trials (RCTs)about Gongliuqing capsules combined with mifepristone (trial group )versus mifepristone alone (control group )in the treatment of uterine leiomyoma were collected. After literature screening and data extraction ,the quality of included literatures was evaluated with modified Jadad scale. Meta-analysis was conducted by using Stata 14.0 software,and trial sequential analysis (TSA)was performed by using TSA 0.9 software. RESULTS :A total of 12 RCTs were included ,involving 1 210 patients. The results of Meta- analysis showed that the total response rate of trial group [RR =1.12,95%CI(1.00,1.26),P<0.05] was significantly higher than that of control group ;maximum uterine leiomyoma volume after treatment [SMD =-1.08,95%CI(-1.21,-0.95),P<0.05],uterine volume after treatment [SMD =-0.80,95%CI(-1.14,-0.45), P<0.05],follicle stimulating hormone (FSH)level [SMD = - 0.28,95% CI(- 0.45,- 0.19),P<0.05],luteinizing hormone(LH)level [SMD =-0.44,95%CI(-0.52,-0.12), 020-38076311。E-mail:867203217@qq.com P<0.05],E2 level [SMD =-2.69,95%CI(-3.08,-1.49), P<0.05] and progesterone (P)level [SMD =-1.27,95%CI(-1.69,-0.71),P<0.05] of trial group were significantly lower or better than those of control group. Results of subgroup analysis showed that except for the level of FSH in 5 and 10 mg mifepristone groups (P>0.05),maximum uterine leiomyoma volume after treatment ,uterine volume after treatment ,the levels of FSH,LH,E2 and P in trial group were significantly lower than control group. The results of TSA showed that there were definite evidences for total response rate of Gongliuqing capsules combined with mifepristone being better in the treatment of hysteromyoma. CONCLUSIONS :Total response rate of Gongliuqing capsules combined with mifepristone in the treatment of hysteromyoma is better than mifepristone alone ,which can effectively decrease the volume of maximum uterine leiomyoma volume and uterine vilume ,and reduce the level of serum hormone.
4.Factors affecting Sandwich vertebral re-fracture after vertebral augmentation surgery for osteoporotic spine Sandwich fractures

Objective To investigate the risk factors affecting Sandwich vertebral re-fracture after vertebral augmentation surgery for osteoporotic spine Sandwich fractures.Methods A retrospective case control study was conducted on the clinical data of 90 patients with osteoporotic spine Sandwich fractures treated with vertebral augmentation surgery from January 2004 to December 2015.There were 26 males and 64 females,aged (69.9 ± 7.4) years.The patients were divided into fracture group (n =15) and non fracture group (n =75) according to the incidence of re-fracture after operation during follow-up.Gender,age,height,weight,bone mineral density,cumulative number of cemented vertebrae,location of the Sandwich vertebrae in the thoracolumbar segment,the preoperative kyphosis angle of the Sandwich fracture segment,surgical approach,cement injection approach (unilateral or bilateral),cement injection volume,cement leakage between the Sandwich intervertebral spaces,the postoperative kyphosis angle of the Sandwich fracture segment,and follow-up duration were used as independent variables.Chi-square test and multivariate logistic regression analysis were used to analyze the risk factors of vertebral Sandwich fracture.Results There were 97 Sandwich vertebrae in 90 patients,and 226 fractured vertebrae were treated with initial intensive treatment.The mean follow-up was 18.1 months.There were no significant differences in gender,age,height,weight,cumulative number of cemented vertebrae,whether the Sandwich vertebrae was located at the thoracolumbar segment,the preoperative kyphosis angle of the Sandwich fracture segment,the cement injection approach (unilateral or bilateral),the volume of cement injection,and the cement leakage between the Sandwich intervertebral spaces (P > 0.05).There were significant differences in bone mineral density,surgical approach,and postoperative kyphosis angle of the Sandwich fracture segment between the two groups (P < 0.05).Further logistic regression analysis showed that bone mineral density ≤ 3.5 SD and segmental kyphosis (≥ 10°) were risk factors for Sandwich fracture (P < 0.05).Conclusions The low bone mineral density (T value ≤-3.5 SD) and postoperative kyphotic angle (≥ 10°) are risk factors for re-fracture of Sandwich vertebrae.Vertebroplasty itself does not increase the risk of recurrent vertebral fractures.

Objective To compare the effect of arthroscopic hollow screw reset and modified 10# Ethicon silk thread internal fixation on treatment of patients with tibial eminence fracture.Methods A total of 21 patients with tibial eminence fracture were selected and randomly divided into the arthroscopic hollow screw reset fixation group (n =11) and modified 10# Ethicon silk suture fixation group (n =10).The operation time,fracture healing time,knee joint mobility,complications,Lysholm knee score were observed between the two groups.Results In the modified 10# Ethicon silk suture fixation group,the operation time was significantly higher than the arthroscopic hollow screw fixation group (P < 0.05),and there were no significant differences in the amount of bleeding,hospitalization time,fracture healing time,knee flexion and extension activity,Lysholm knee score and complications between two groups (P > 0.05).Conclusion Arthroscopic hollow screw reset and modified 10# Ethicon silk thread internal fixation are good methods for the treatment of patients with tibial eminence fracture.

7.Clearing, amplification and activity detection of the recombinant adeno-associated virus vector2/1with adiponectin

Objective To clear, amplify and detect the activity of the recombinant adeno-assoeiated virus vector with adiponectin( rAAV2/1-Aerp30 ). Methods Recombinant plasmid pSNAV2.0-Acrp30 was obtained. The recombinant plasmid was then transfected into BHK21 cells using LipofectAMINETM 2000. The G418 resistant cells were obtained consequently. These cells were infected with HSVI-rc/△UL2 which has the function of packaging and copying recombinant AAV. After purification, the construction of recombinant rAAV2/1-Aerp30 was collected. Results The construction of recombinant pSNAV2.0-Acrp30 was confirmed by PCR electrophoresis and digestion with restriction enzyme. The gene sequencing showed that the recombinant pSNAV2.0-Acrp30 was correct. The virus titer was about 1.0×1012 μg/ml. The purity f the recombinant AAV2/1 was fairly high using the SDS-PAGE method. Conclusion With this method, rAAV2/-Aerp30 with high virus titers and purity can be acquired successfully and it can meet the demands of the experimental study of Acrp30 gene therapy of GK rats.
