1.Partial knockout of NtPDK1a/1b/1c/1d enhances the disease resistance of Nicotiana tabacum.

The protein kinase A/protein kinase G/protein kinase C-family (AGC kinase family) of eukaryotes is involved in regulating numerous biological processes. The 3-phosphoinositide- dependent protein kinase 1 (PDK1), is a conserved serine/threonine kinase in eukaryotes. To understand the roles of PDK1 homologous genes in cell death and immunity in tetraploid Nicotiana tabacum, the previuosly generated transgenic CRISPR/Cas9 lines, in which 5-7 alleles of the 4 homologous PDK1 genes (NtPDK1a/1b/1c/1d homologs) simultaneously knocked out, were used in this study. Our results showed that the hypersensitive response (HR) triggered by transient overexpression of active Pto (PtoY207D) or soybean GmMEKK1 was significantly delayed, whereas the resistance to Pseudomonas syrangae pv. tomato DC3000 (Pst DC3000) and tobacco mosaic virus (TMV) was significantly elevated in these partial knockout lines. The elevated resistance to Pst DC3000 and TMV was correlated with the elevated activation of NtMPK6, NtMPK3, and NtMPK4. Taken together, our results indicated that NtPDK1s play a positive role in cell death but a positive role in disease resistance, likely through negative regulation of the MAPK signaling cascade.

2.Clinical Distribution Characteristics and Correlation of Cat and Dog Dander Allergens and Mold Allergens

ObjectiveTo explore the distribution patterns and correlations of pet-related cat/dog dander allergens and mold allergens in patients with allergic diseases, providing evidence for individualized diagnosis, treatment, and prevention strategies. MethodsA retrospective analysis was conducted on 798 patients diagnosed with allergic diseases at the Third Affiliated Hospital of Sun Yat-sen University between April 2021 and October 2023. All patients underwent UniCAP platform testing for specific immunoglobulin E (sIgE) levels against cat dander, dog dander, and mold mix (mx1/mx2), alongside total IgE (tIgE) quantification. Descriptive statistics, Mann-Whitney U tests, and chi-square analyses were employed to evaluate allergen distribution and interrelationships. ResultsAmong the 798 patients (395 males, 403 females, ratio 1∶1.02), their ages ranged from 0.67 to 69 years (median 14 years, IQR 6-29). A total of 63.2% (504/798) had a single allergic disease, with allergic rhinitis (AR, 49.2%) being the most common. The remaining 36.8% (294/798) had ≥2 allergic diseases, with AR combined with atopic dermatitis (AD, 10.7%) as the predominant comorbidity. The positivity rate for cat/dog dander sIgE was 24.1% (192/798), with a significantly higher prevalence in females (30.8%) than males (16.7%, P<0.05). Cat dander sensitization increased with age in patients under 18 years. Among positive cases, cat dander sIgE level 2 was most frequent (25.9%), while dog dander sIgE level 1 predominated (55%). Patients with cat dander sIgE levels 4-6 had significantly higher tIgE than those with levels 1-3 (P<0.05). The positivity rate for mold mix (mx1/mx2) sIgE was 7.4% (59/798), with mx2 as the primary sensitizer and level 2 being the most common. In mx2-positive patients, the cat/dog dander sIgE positivity rate (44.8%) was significantly higher than that in mx2-negative patients (17.9%, P<0.05), and tIgE levels were also higher (P<0.05). ConclusionCat dander sensitization increases with age in children. Cat/dog dander and mold allergens are closely linked to AR and AR combined with AD. Synergistic correlations exist between cat/dog dander sIgE and mold mx2 sIgE. Combined detection of these allergens is critical for precision diagnosis and management of pet-related allergic diseases.
3.Item analysis on the general module in a multidimensional health measurement scale for elderly patients with chronic diseases

Objective:To analyze and evaluate the items of the general module of multidimensional health measurement instruments system for elderly patients with chronic disease (MHIEC-GM)(V1.0) based on classical test theory (CTT) and item response theory (IRT).Methods:A self-administered survey of 2 375 elderly patients with chronic diseases was conducted using the MHIEC-GM(V1.0) scale. The quality of items was analyzed using the variability method, correlation coefficient method, factor analysis method, and Cronbach's α coefficient method based on CTT. The difficulty coefficient, differentiation coefficient, and information content of items were analyzed using the IRT analysis software MULTILOG 7.03.Results:The CTT results showed that the standard deviations of all nine items were greater than 0.9. The results of the correlation coefficient method, the factor analysis method and the Cronbach′s α coefficient method showed that except for the GMI9 item, the correlation coefficients between the other items and the total scale were all greater than 0.5, and the factor loads were all greater than 0.5, and the Cronbach′s α coefficients after the deletion of the items were all less than 0.763. Based on the results of the four analysis methods, except for the GMI9 item, all the other items had good characteristics. The IRT results showed that the information contents of all items were >0.333 (5/15) except for the GMI4, GMI6, and GMI8, and the differentiation coefficient of each item was 1.44. The items that difficulty coefficient met the [-4, 4] inclusion criteria were the GMI2, GMI3, GMI5, GMI7 and GMI9, showing a monotonically increasing trend with increasing difficulty level. The items of GMI1, GMI4, GMI6, and GMI8 needed further improvement.Conclusion:Most of items of the MHIEC-GM(V1.0) scale have good psychometric properties, but some items need to be further revised and validated according to the characteristics of chronic diseases in the elderly.
