Objective To elucidate the molecular mechanism of sirtuin-3 (SIRT3) in regulating mitochondrial biogenesis in human renal tubular epithelial cells. Methods Cells were stimulated with different concentrations of H₂O₂ and divided into four groups: control (NC), 50 μmol/L H₂O₂, 110 μmol/L H₂O₂ and 150 μmol/L H₂O₂. SIRT3 protein expression was then measured. SIRT3 was knocked down with siRNA, and cells were further assigned to five groups: control (NC), negative-control siRNA (NCsi), SIRT3-siRNA (siSIRT3), NCsi+H₂O₂, and siSIRT3+H₂O₂. After 24 h, cellular adenosine triphosphate (ATP) and mitochondrial superoxide anion (O₂•⁻) levels were determined, together with mitochondrial expression of SIRT3, peroxisome proliferator-activated receptor γ coactivator-1α (PGC-1α), nuclear respiratory factor 1 (NRF1), mitochondrial transcription factor A (TFAM), superoxide dismutase 2 (SOD2), acetylated-SOD2 and adenosine monophosphate activated protein kinase α1 (AMPKα1). Results The 110 and 150 μmol/L H₂O₂ decreased SIRT3 protein (both P<0.05). ATP and mitochondrial O₂•⁻ did not differ between NC and NCsi groups (both P>0.05). Compared to the NCsi group, the siSIRT3 group exhibited elevated O₂•⁻ level, decreased SIRT3 protein and increased expression levels of SOD2 and acetylated SOD2 protein (all P<0.05). Compared to the NCsi group, the NCsi+H₂O₂ group exhibited decreased cellular ATP levels, elevated mitochondrial O₂•⁻ levels, and reduced protein expression levels of SIRT3, SOD2, TFAM, AMPKα1, PGC-1α and NRF1 (all P<0.05). Compared with the siSIRT3 group, the siSIRT3+H₂O₂ group showed a decrease in cellular ATP levels, an increase in mitochondrial O₂•⁻ levels, a decrease in SIRT3, SOD2, TFAM, AMPKα1, PGC-1α and NRF1 protein expression levels and a decrease in acetylated SOD2 protein expression levels (all P<0.05). Compared with the NCsi+H₂O₂ group, the siSIRT3+H₂O₂ group showed a decrease in cellular ATP levels, an increase in mitochondrial O₂•⁻ levels, a decrease in SIRT3, AMPKα1, PGC-1α and NRF1, TFAM protein expression levels, and an increase in SOD2 and acetylated SOD2 protein expression levels (all P<0.05). Conclusions SIRT3 promotes mitochondrial biogenesis in tubular epithelial cells via the AMPK/PGC-1α/NRF1/TFAM axis, representing a key mechanism through which SIRT3 ameliorates oxidative stress-induced mitochondrial dysfunction.

ObjectiveTo explore the medication patterns of Professor Zhang Farong in treating type 2 diabetes mellitus （T2DM） and the clinical efficacy of his core prescription. MethodsClinical case records of T2DM treated by Professor Zhang Farong were collected to establish a prescription database. Frequency statistics， visual analysis， and factor analysis were employed to investigate the characteristics and principle of the prescriptions， and a core prescription was derived. A randomized controlled trial was conducted， enrolling 60 T2DM patients with the dampness-heat syndrome. The patients were allocated into an observation group （core prescription + metformin） and a control group （metformin alone）， with both groups undergoing a 12-week treatment course. Changes in TCM symptom scores， glucose metabolism indicators ［fasting plasma glucose （FPG）， 2-hour postprandial blood glucose （2 hPG）， and glycated hemoglobin （HbA1c）］， pancreatic function indicators ［fasting C-peptide （FCP）， 2-hour postprandial C-peptide （2 hCP）， and area under the C-peptide curve （AUCcp）］， and lipid profiles were measured before and after treatment. The adverse reactions were observed and recorded. ResultsA core prescription named modified Gegen Qinlian Decoction was formulated， comprising Puerariae Lobatae Radix， Coptidis Rhizoma， Scutellariae Radix， Astragali Radix， Lycii Cortex， Mori Cortex， Jineijin Endothelium Corneum Gigeriae Galli， Rehmanniae Radix Praeparata， Atractylodis Rhizoma， Polygonati Rhizoma， and Pogostemonis Herba. The clinical trial results showed that both groups had significantly decreased FPG， 2 hPG， and HbA1c （P0.05）， and the observation group outperformed the control group in recovering the level of HbA1c （P0.05）. After treatment， both groups had declined TCM symptoms scores （P0.05）， and the declines in the observation group were larger than those in the control group （P0.05）. After treatment， the TC and LDL-C levels declined in the observation group （P 0.05）， while the lipid levels showed a decreasing trend with no statistically significant difference in the control group. After treatment， both groups showed increases in FCP and AUCcp （P0.05）， and the 2 hCP in both groups presented a recovering trend with no statistically significant difference. There was no statistically significant difference in the incidence of adverse reactions between the two groups. ConclusionModified Gegen Qinlian Decoction embodies Professor Zhang Farong's academic philosophy of treating consumptive thirst by tonifying the spleen and kidney， replenishing Qi and Yin， clearing deficiency and heat， unblocking stasis in collaterals， and addressing both deficiency and stasis. The combination of the core prescription with metformin alleviates clinical symptoms in T2DM patients with the dampness-heat syndrome， demonstrating potential effects in restoring pancreatic islet function， regulating blood glucose， and improving lipid profiles. It serves as a therapeutic option for T2DM in the patients with the dampness-heat syndrome under syndrome differentiation， meriting broader clinical application.

Objective:To analyze the application effect of condition scale in nursing management of children bronchial asthma in acute attack stage.Methods:A total of 136 children with acute attack of bronchial asthma admitted to Shanxi Children's Hospital from January 2020 to December 2021 were selected by convenience sampling method, and randomly divided into observation group and control group according to random number table method, 68 cases in each group. Children in the observation group received graded nursing intervention based on disease scale, while children in the control group received routine nursing intervention. The first second forced expiratory volume (FEV 1), forced vital capacity (FVC), ratio of FEV l/FVC, peak expiratory flow rate (PEF), medical intervention frequency, the number of ICU transfer and length of stay were compared between the two groups. Results:The results of repeated measurement ANOVA showed that there were intergroup, time and interaction effects on the levels of FEV 1, FVC, FEV 1/FVC and PEF in the two groups, and the differences were statistically significant ( P<0.05). The frequency of medical intervention in observation group was less than that in control group, and the length of hospitalization was shorter, with statistical significance ( P<0.05). There was no significant difference in ICU patients transferred between the two groups ( P>0.05) . Conclusions:Application of Condition Scale in nursing management of children with acute attack of bronchial asthma can improve the lung function of children with acute attack bronchial asthma more effectively, reduce the frequency of medical intervention, shorten the length of hospital stay, and significantly improve the nursing effect.

Objective:To explore the current situation and effective strategies of clinical full-time scientific research team construction by introducing the experiences and lessons learned during the process of research hospital construction at Peking University Cancer Hospital.Methods:To sort out the clinical full-time scientific research personnel team in the hospital, analyze the status quo of the talent team and the achievements, identify and illustrate problems and solutions in the process of clinical full-time scientific research team construction in a research hospital.Results:The hospital clinical full-time scientific research team was developed and played its roles in clinical research to some extent. Administration should provide appropriate incentives and optimize the training strategies for scientific researchers.Conclusions:Full-time clinical research teams at hospitals can effectively improve the scientific research ability of clinical departments, more attention from the administration perspective should be paid to problems and appropriate strategies identified during the construction of such teams.

Objective To analyze the standardization and management experience of the clinical research projects,which were supported by Beijing Municipal Science & Technology Commission.Methods Based on recent data of clinical research projects in one hospital,orientation and management experience of the projects were analyzed.Moreover,shortcomings and possible countermeasures about this issue were explored.Results Since 2010,the bid-winning rate of this kind of projects has been basically stable,but the age structure has tended to be younger,which may be benefited from the hospital scientific research culture and advantage training strategy for youth talents.Scientific research management department also took an important role in this improvement,but did have some problems in real practice.Conclusions We hope to share the management experience of clinical research projects in the hospital,which may improve the management process and serve the researchers better.

Objective To analyze the extension mode and existing problems of clinical science and technology achievements in order to promote the popularization of clinical scientific and technological achievements in practice.Methods Conducting literature review to better understand the basic concepts and promotion patterns of clinical medical science and technology achievements,identifying problems and discussing possible solutions by taking into account of the practical work.Results The clinical science and technology achievements usually include suitable technology,industry standard,diagnosis and treatment standard or guide,policy document,science popularization knowledge and so on.In the way of popularization,we can communicate through academic exchange,training courses,digital multimedia network and so on.However,in practice,due to the insufficient awareness of promotion,management mechanism,funds and so on,the promotion is negatively affected,which is worth the attention of the administrators.Conclusions It is expected that through the discussion of the concept of clinical science and technology achievements and the mode of popularization,it is possible to arouse the thinking of the managers and promote the transformation of clinical medical scientific and technological achievements.

Objective To investigate the clinical application of self-made snare in laparoscopic appendectomy. Methods The clinical datas of 197 cases who were acute appendicitis under laparoscopic appendectomy in the first people's hospital of Fuyang in Hangzhou from June 2015 to June 2017 were collected, then they were divided into self-made snare group (85) and Hem-o-lok group (112). Variations of operation time, operation bleeding, first postoperative anal exhaust time and length of stay above two groups were analyzed. Variations of incision infection, abdominal abscess and intestinal obstructionabove two groups were analyzed. Results Two groups of patients were compared with operation time [self-made snare group(43. 71±5. 54)min: Hem-o-lok group(42. 29±5. 34)min], operation bleeding[self-made snare group(l 1. 60±2. 44)mL: Hem-o-lok group(l 1. 00±2. 56)mL],first postoperative anal exhaust time [self-made snare group (10. 01±2. 27)h: Hem-o-lok group(9. 60±2. 32)h] and length of stay[self-made snare group(5. 19±0. 95)d: Hem-o-lok group (4. 99±0. 85)d], the differences were not statistically significant(P＞0. 05). Two groups of patients were compared with incision infection[self-made snare group 2 cases(2. 35％): Hem-o-lok group 3 cases(2. 68％)], abdominal abscess [self-made snare group 1 case(1. 18％): Hem-o-lok group 2 cases (1. 79％)] and intestinal obstructionabove [self-made snare group 3 cases(3. 53％): Hem-o-lok group 5 cases(4. 46％)], the differences were not statistically significant(P＞0. 05). Conclusion Self-made snare in laparoscopic appendectomy is safe and feasible.

Objective To study the influencing factors and clinical significance of ultrasonic quantitative analysis of the hip-flexion coronal section in the normal infants.Methods Totally 100 normal infants were enrolled.Angle α,angle β and FHC on different positions(mild-flexion and flexion of the hip) were mearured.And angle β on different points(the labrum central and the acetabular tips) were measured.The variation of the measurement index between different positions were analyzed.Results ①There was no significant difference in angle α between the neutral position and hip-flexion position (P ＞0.05).② FHC decreased in the hip-flexion position,and there was significant difference compared with the neutral position (P ＜0.05).③βc were greater than βt in the two positions (P ＜0.05);βc and βt were all greater in hip-flexion position than those in neutral position,the difference was statistically significant (P ＜0.05).Conclusions The results obtained from angle α is stable under the coronal flexion view of the normal infants,and does not vary with changing position.The change of the angle β and FHC with the hip flexion could be used to evaluate the stability of the hip.Measured angle β on the labrum tip has good repeatability.So this point should be selected to measure the angle β.

Objective To investigate the hospital laboratory management mode.Methods We used literature review and comparative analysis to understand the different laboratory management mode,explore suitable operation mechanisms for hospital research and the development of hospital la boratory.Through a variety of measures to improve the efficiency and level of management,we tried to ensure the high efficiency and high quality operation of clinical laboratory,hospital science research,personnel training,discipline construction to provide quality services.Results There were two kinds of hospital laboratory management modes,which were centralized and decentralized management mode.Both management modes had advantages and disadvantages.We thought the centralized-decentralized binding mode was an ideal combination.Based of this mode,we implemented a number of initiatives and achieved significant results.Conclusions From the scale of hospital and its long-term development,centralized decentralized binding mode is a kind of ideal mode.

Based on the project data and SCI output data of 5431 projects supported by National Natural Science Foundation of China (NSFC) in oncology from 2008 to 2012,this paper took advantage of the Web of Science database,studied the number of papers published and the citations per paper of different research fields in oncology,different subcategories of projects,top 20 institutions funded by NSFC.Based on the study findings,we tried to make some suggestions for the funding agency,researchers,as well as research management personnel for increase the effectiveness of funding use,improve the administration work and policy-making concerns.
