1.Resistant hypertension and the risk of major adverse cardiac and cerebrovascular events in outpatients

Objective:To investigate the prevalence and associated risk of cardiovascular event of resistant hypertension in treated outpatients.Methods:This study was a nationwide multi-center prospective cohort study. The participants were treated outpatients enrolled in the China Nationwide Ambulatory and Home Blood Pressure Registry study of 42 hospitals in 19 provinces across the country from August 2009 to October 2017. Apparent resistant hypertension was defined as uncontrolled office blood pressure (≥140/90 mmHg, 1 mmHg=0.133 kPa) in spite of the use of three antihypertensive drugs or controlled office blood pressure (<140/90 mmHg) with four antihypertensive drugs or more. Subjects diagnosed with uncontrolled office blood pressure were further subdivided as pseudo-resistant hypertension and true resistant hypertension based on 24 h ambulatory blood pressure monitoring. The primary endpoint was fatal and non-fatal cardiovascular and cerebrovascular events, which was a composite endpoint consisting of cardiovascular and cerebrovascular death, ischemic and hemorrhagic stroke, myocardial infarction, coronary artery revascularization, unstable angina, heart failure, and coronary artery stenosis≥50% confirmed by coronary angiography. Secondary outcomes included fatal and non-fatal stroke or cardiac events. Patients with controlled office blood pressure after taking only 1 or 2 antihypertensive drugs were included as control. Kaplan-Meier survival curves, log-rank test, and Cox proportional risk model were used to evaluate the risk of apparent refractory hypertension in relation to cardiovascular and cerebrovascular prognosis.Results:A total of 2 782 treated hypertensive patients, aged (58.1±12.3) years were enrolled, including 1 403 (50.4%) men. The prevalence of apparent and true resistant hypertension was 15.1% (420/2 782) and 10.5% (293/2 782), respectively. Among patients with apparent resistant hypertension, during a median of 5 years follow-up, the cumulative incidence rate was 28.2, 11.2 and 19.1 per 1 000 person-years for fatal and non-fatal cardiovascular events ( n=58), stroke ( n=24) and cardiac events ( n=40), respectively. The Kaplan-Meier curve and log-rank test showed that those patients with true resistant hypertension, had the highest cumulative incidence rate of fatal and non-fatal cardiovascular events, stroke, and cardiac events. Multivariable Cox regression analyses showed that true resistant hypertension was associated with a significantly higher risk of fatal and non-fatal cardiovascular events ( HR=1.73, 95% CI 1.17-2.56, P=0.006) and stroke ( HR=2.81, 95% CI 1.53-5.17, P=0.001). Conclusion:Resistant hypertension, especially true resistant hypertension, is associated with a higher risk of fatal and non-fatal cardiac and cerebrovascular events.
2.Advances of structure and mechanisms of bromodomain-containing protein 4 and its related research in tumors.

The bromodomain and extraterminal domain (Bet) family are the regulators of the epigenome and also the pivotal driving factors for the expression of tumor related genes that tumor cells depend on for survival and proliferation. Bromodomain-containing protein 4 (Brd4) is a member of the Bet protein family. Generally, Brd4 identifies acetylated histones and binds to the promoter or enhancer region of target genes to initiate and maintain expression of tumor related genes. Brd4 is closely related to the regulation of multiple transcription factors and chromatin modification and is involved in DNA damage repair and maintenance of telomere function, thus maintaining the survival of tumor cells. This review summarizes the structure and function of Brd4 protein and the application of its inhibitors in tumor research.

3.Application of Modified FuMai Decoction (复脉汤加减方) based on Sanjiao (三焦) Theory

Review of historical literature showed that the understanding of the indicated disease location of Modified FuMai Decoction (复脉汤加减方) has evolved from the upper jiao (焦) to the middle and lower jiao. Initially, it is used for the treatment of yin deficiency of both yin and yang in the upper jiao, changes to supplement stomach and produce fluids in the middle jiao, and is used to protect yin, clear the pathogens, conslidate yin and subdue yang so as to store the true yin of lower jiao. The unchanging principle of Fumai Decoction modifications is nourishing yin, while the changing aspects are determining the secondary treatment methods based on disease location of sanjiao, concomitant disease natures, internal injury or external contraction, warm disease or cold damage, thereby choosing the corresponding added or subtracted herbs, and providing reference for the application of classical formulas.
4.Psychological status and correlation between anxiety and science literacy of medical staff under the public health emergency

BackgroundIn public health emergencies, medical staff undertake many important tasks. Having a good mental health status and capability in popularization of science can improve the work efficiency of medical staff, and thus enhancing the public's trust and support for medical care. ObjectiveTo investigated the anxiety, insomnia and security level of medical staff under the public health emergency, and to explore the correlation between anxiety and capability in popularization of science of them. MethodsFrom June 1 to September 1, 2020, a total of 588 medical staff in Sichuan Province were investigated through the Questionnaire Star platform. The survey included a self-designed general information questionnaire, the Psychological Security-Insecurity Questionnaire (S-I), Self-rating Anxiety Scale (SAS), Athens Insomnia Scale (AIS) and science popularization questionnaire during the COVID-19 epidemic. Pearson correlation analysis was used to examine the correlation among SAS, S-I and AIS scores, as well as the correlation between SAS and the science popularization questionnaire during the COVID-19 epidemic. ResultsDuring the public health emergency, the S-I score of the medical staff was (27.46±9.20), with 21 cases (3.57%) having a low level of security. The SAS score was (44.21±9.57), with 208 cases (35.37%) having anxiety symptoms. The AIS score was (11.40±5.25), with 450 cases (76.53%) suffering from insomnia. Correlation analysis showed that SAS score was negatively correlated with S-I score (r=-0.640, P<0.01), while positively correlated with AIS score (r=0.618, P<0.01). There was a negative correlation between the degree of usage about medical staff's science popularization tool and SAS score (r=-0.501~-0.185, P<0.01). The use of science popularization channels was negatively correlated with anxiety (r=-0.510~-0.232, P<0.05 or 0.01). There was a negative correlation between the level of trust to popularization of science and medical staff anxiety (r=-0.548~-0.338, P<0.01). ConclusionUnder the public health emergency, the detection rates of anxiety symptoms and sleep disorders among medical staff are high, and anxiety is negatively correlated with medical staff's capability in popularization of science. 5.GranatumX: A Community-engaging, Modularized, and Flexible Webtool for Single-cell Data Analysis.

We present GranatumX, a next-generation software environment for single-cell RNA sequencing (scRNA-seq) data analysis. GranatumX is inspired by the interactive webtool Granatum. GranatumX enables biologists to access the latest scRNA-seq bioinformatics methods in a web-based graphical environment. It also offers software developers the opportunity to rapidly promote their own tools with others in customizable pipelines. The architecture of GranatumX allows for easy inclusion of plugin modules, named Gboxes, which wrap around bioinformatics tools written in various programming languages and on various platforms. GranatumX can be run on the cloud or private servers and generate reproducible results. It is a community-engaging, flexible, and evolving software ecosystem for scRNA-seq analysis, connecting developers with bench scientists. 6.Evaluation of Cell Type Annotation R Packages on Single-cell RNA-seq Data

Annotating cell types is a critical step in single-cell RNA sequencing (scRNA-seq) data analysis.Some supervised or semi-supervised classification methods have recently emerged to enable automated cell type identification.However,comprehensive evaluations of these methods are lacking.Moreover,it is not clear whether some classification methods originally designed for ana-lyzing other bulk omics data are adaptable to scRNA-seq analysis.In this study,we evaluated ten cell type annotation methods publicly available as R packages.Eight of them are popular methods developed specifically for single-cell research,including Seurat,scmap,SingleR,CHETAH,Sin-gleCellNet,scID,Garnett,and SCINA.The other two methods were repurposed from deconvolut-ing DNA methylation data,i.e.,linear constrained projection (CP) and robust partial correlations(RPC).We conducted systematic comparisons on a wide variety of public scRNA-seq datasets as well as simulation data.We assessed the accuracy through intra-dataset and inter-dataset predic-tions;the robustness over practical challenges such as gene filtering,high similarity among cell types,and increased cell type classes;as well as the detection of rare and unknown cell types.Over-all,methods such as Seurat,SingleR,CP,RPC,and SingleCellNet performed well,with Seurat being the best at annotating major cell types.Additionally,Seurat,SingleR,CP,and RPC were more robust against downsampling.However,Seurat did have a major drawback at predicting rare cell populations,and it was suboptimal at differentiating cell types highly similar to each other,compared to SingleR and RPC.All the code and data are available from https://github.com/qian-huiSenn/scRNA_ cell_ deconv_benchmark.
7.Investigation on the mental health condition and the styles of help seeking of college students during the COVID-19 outbreak

ObjectiveTo investigate the mental health condition of college students during the Corona Virus Disease 2019 (COVID-19) , and to provide accurate basis for their psychological intervention. MethodsThrough the way of electronic questionnaire, questionnaire star as tools, random investigation of college students in a university in Sichuan. Self-rating Anxiety Scale (SAS), Self-rating Depression Scale (SDS) and Somatization Symptom Scale (SSS) were used to evaluate their anxiety, depression and somatization symptoms. At the same time, a self-made questionnaire was used to investigate their styles of help seeking, and the relationships between them. ResultsA total of 551 valid questionnaires were collected, including 4.90% of anxiety symptoms, 28.68% of depression and 17.97% of somatization symptoms. In terms of somatization, the detection rate was higher in urban college students than those in rural (23.35% vs. 13.27%, P<0.05), non-medical students was higher than medical students (21.40% vs. 13.89%, P<0.05). Women's SSS score was higher than men's [(26.51 ± 5.44) vs. (25.27 ± 5.78)], urban college students' SSS score were higher than rural [(26.69 ± 5.98) vs. (25.38 ± 5.12)], and non-medical students’ SSS score were higher than medical students [(26.65 ± 5.59) vs. (25.21 ± 5.45)], and the differences were statistically significant (P<0.05). Partial correlation analysis showed that SSS score was positively correlated with SAS score (r=0.110, P=0.010) and negatively correlated with SDS score (r=-0.087, P=0.042). The top three common ways of help-seeking pattern among college students were self-regulation (81.85%), talking to family or friends (70.78%) and offline psychological counseling (28.31%). ConclusionUnder the COVID-19 epidemic, the detection rate of depressive symptoms and somatization symptoms of college students is high. The detection rate of somatization symptoms of female, urban and non-medical college students is high. Self-regulation and talking to family or friends are the main psychological styles of help seeking for college students.
8.Application of dialectical behavior therapy in adolescents with non-suicidal self-injury behaviors

The purpose of this article is to analyze the effectiveness of dialectical behavior therapy (DBT) in adolescent non-suicidal self-injury (NSSI) behavioral therapy, in order to provide a reference for the intervention of NSSI behavior in adolescents in China. NSSI behavior is a common mental health threat for adolescents. In recent years, it has become a mental health problem that cannot be ignored worldwide. At present, psychotherapy is mainly used for NSSI behavior. Increasing evidence shows that DBT is effective in reducing NSSI behavior. This article summarizes the effectiveness of DBT in the treatment of NSSI behavior by describing the risk factors of NSSI behavior, an overview of DBT and the efficacy of DBT in the application of NSSI.
9.Identification and Characterization of Two New S-Adenosylmethionine-Dependent Methyltransferase Encoding Genes Suggested Their Involvement in Stipe Elongation of Flammulina velutipes

Two new SAM-dependent methyltransferase encoding genes (fvsmt1 and fvsmt2) were identified from the genome of Flammulina velutipes. In order to make a comprehensive characterization of both genes, we performed in silico analysis of both genes and used qRT-PCR to reveal their expression patterns during the development of F. velutipes. There are 4 and 6 exons with total length of 693 and 978 bp in fvsmt2 and fvsmt1, respectively. The deduced proteins, i.e., FVSMT1 and FVSMT2 contained 325 and 230 amino acids with molecular weight 36297 and 24894 Da, respectively. Both proteins contained a SAM-dependent catalytic domain with signature motifs (I, p-I, II, and III) defining the SAM fold. SAM-dependent catalytic domain is located either in the middle or at the N-terminal of FVSMT2 and FVSMT1, respectively. Alignment and phylogenic analysis showed that FVSMT1 is a homolog to a protein–arginine omega-N-methyltransferase, while FVSMT2 is of cinnamoyl CoA O-methyltransferase type and predicted subcellular locations of these proteins are mitochondria and cytoplasm, respectively. qRT-PCR showed that fvsmt1 and fvsmt2 expression was regulated in different developmental stages. The maximum expression levels of fvsmt1 and fvsmt2 were observed in stipe elongation, while no difference was found in mycelium and pileus. These results positively demonstrate that both the methyltransferase encoding genes are involved in the stipe elongation of F. velutipes.
10.Comparision of the pharmacodynamics between different batches of recombinant decoy receptor innovative drug RC28-E in retinal vascular endothelial cells

Objective To compare the protective effects of pharmacological batch RC28-E1 and pilot batch RC28-E2 on retinal vascular endothelial cells (RF/6A) under the stimulation of vascular endothelial growth factor (VEGF) and fibroblast growth factor (FGF).Methods RF/6A cells were divided into normal control group,VEGF + FGF group and RC28-E1 groups with different concentrations.The optimal concentration of RC28-E1 was determined by cell counting kit-8 (CCK-8) method.Cells were divided into normal control group,VEGF+FGF group,RC28-E1 group,RC28-E2 group,conbercept group and FGF trap group,and cultured with serum-free culture medium,serum-free culture medium containing VEGF+FGF,serum-free culture medium containing VEGF+FGF+RC28-E1,serum-free culture medium containing VEGF+FGF+RC28-E2,and serum-free culture medium containing VEGF+FGF+ conbercept,serum-free medium containing VEGF+FGF+FGF trap,respectively.Cell proliferation rate was measured by CCK-8 method,cell migration ability was detected by Transwell test,and tube formation ability was detected by Matrigel assay.Results The cell proliferation rate of 0.080 mg/ml RC28-E1 group was significantly lower than that of VEGF+FGF group (P<0.05).The cell proliferation rate of RC28-E1 group,RC28-E2 group and FGF trap group were significantly lower than that of VEGF+FGF group (P<0.05).The number of migrated cells in RC28-E1 group,RC28-E2 group,conbercept group and FGF trap group were significantly lower than that in VEGF+FGF group (P=0.000).The numbers of meshes formed by retinal vascular endothelial cells in RC28-E1 group,RC28-E2 group,conbercept group and FGF trap group were significantly lower than that in VEGF+FGF group (P =0.003,0.001,0.009,0.018).The number of tube formation in FGF trap group was significantly higher than those in RC28-E1 group,RC28-E2 group,conbercept group and normal control group (P =0.014,0.000,0.008,0.014).Conclusions Under the stimulation of VEGF + FGF,the inhibitory effect of RC28-E on the proliferation of retinal vascular endothelial cells is greater than that of conbercept,and its inhibitory effect on the tube formation is superior to that of FGF trap.There is no significant difference in the effects of different batches of recombinant decoy receptor innovative drugs on retinal vascular endothelial cells.
