1.Analysis of components absorbed into blood and brain of Lithocarpus litseifolius leaves

OBJECTIVE To analyze the prototype components absorbed into blood and brain of Lithocarpus litseifolius leaves, so as to provide a reference for clarifying the pharmacological material basis of its prevention and treatment of central nervous system dis eases. METHODS The ethanol extract of L. litseifolius leaves, as well as the gastric lavage fluid and perfusion solution were prepared. Using rats as subjects, plasma samples of intestinal wall metabolism, intestinal flora metabolism and hepatic metabolism were prepared via in situ intestinal perfusion and closed intestinal loop method; while comprehensive metabolic plasma samples, brain tissue samples, and cerebrospinal fluid samples were collected after intragastric administration. UPLC-HRMS technology was utilized to analyze and identify chemical components and prototype components absorbed into blood and brain of L. litseifolius leaves. RESULTS A total of 66 chemical constituents were identified in L. litseifolius leaves, primarily consisting of flavonoids, organic acids, and others. A total of 16, 13, 11, and 5 prototype components were identified in intestinal wall metabolism, intestinal flora metabolism, hepatic metabolism, and comprehensive metabolic plasma samples, respectively. Additionally, 4 prototype components were detected in brain tissue and 9 in cerebrospinal fluid. Phloridzin, trilobatin, phloretin-2- O -malonyl hexoside, and phloretin were identified as common components across all sample types. CONCLUSIONS Prototype components absorbed into blood and brain of L. litseifolius leaves, such as phloridzin, trilobatin, phloretin, and other components may serve as the pharmacological material basis for their therapeutic effects on central nervous system diseases.
2.Manganese porphyrin metal-organic framework nanoparticles loaded with DMXAA combined with sonodynamic therapy for the treatment of triple-negative breast cancer mouse xenografts

[摘 要] 目的:构建负载STING激动剂DMXAA的锰卟啉金属有机框架纳米颗粒(DPM),探讨其对三阴性乳腺癌(TNBC)细胞4T1及其小鼠移植瘤的治疗效果。方法:通过物理吸附法制备 DPM 纳米颗粒,利用透射电镜、扫描电镜及纳米粒度电位仪表征其形貌与理化性质。常规培养4T1细胞,细胞实验分为对照组、超声辐照组(US组)、DPM治疗组(DPM组)和DPM治疗联合超声辐照组(DPM + US组),用CCK-8法检测细胞活性,免疫荧光法检测高迁移率族蛋白B1(HMGB1)和钙网蛋白(CRT)的表达,WB法检测STING通路相关蛋白的表达。构建4T1细胞移植瘤小鼠模型,分为四组,处理同细胞实验,测量肿瘤体积,免疫荧光法检测移植瘤组织中Ki-67、HMGB1、CRT和缺氧诱导因子-1ɑ(HIF-1ɑ)蛋白的表达,TUNEL法检测细胞凋亡,流式细胞术检测免疫细胞活化情况,对主要器官进行H-E染色,以评估纳米材料的体内安全性。结果:DPM呈梭形,平均粒径(268 ± 3.302)nm,电位(33.1 ± 0.87)mV。细胞实验中,DPM联合超声辐照可明显抑制4T1细胞的增殖(P < 0.001),提高4T1细胞中ROS水平(P < 0.001),诱导4T1细胞CRT表达上调(P < 0.001),HMGB1从细胞核中移至细胞质,激活STING信号通路[p-STING、p-TBK1、p-IRF3蛋白表达均显著增加(均P < 0.001)]。体内实验中,DPM联合超声辐照可显著抑制4T1细胞移植瘤生长(P < 0.001)并促进免疫细胞表型转化(P < 0.001),抑制移植瘤组织中Ki-67、HIF-1α蛋白表达(均P < 0.01),谷胱甘肽(GSH)产生(P < 0.01),促进CRT、HMGB1蛋白表达、ROS产生(P < 0.001),对主要器官结构无明显影响。结论: DPM联合超声辐照可通过激活STING通路显著抑制4T1细胞及其移植瘤的生长,诱导抗肿瘤免疫应答,且对主要器官无明显毒性。
3.PET/CT imaging of PD-1 receptor probe targeting S180 sarcoma in mice

ObjectiveTo explore the feasibility of constructing a programmed death receptor-1(PD-1) molecular probe for non-invasive micro-positron emission tomography/computed tomography (Micro-PET/CT) imaging of PD-1 protein in mouse S180 sarcoma. MethodsA transgenic PD-1 C57 S180 sarcoma mouse model was established using the S180 sarcoma cell injection. Furthermore, 124I-anti-PD-1 monoclonal antibody probe was synthesized. 18.5 MBq of the 124I-anti-PD-1 probe was injected into the tail vein of transgenic PD-1 C57 mice. Subsequently, S180 sarcoma was imaged using Micro-PET/CT. ResultsStudy successfully established a transgenic PD-1 C57 S180 sarcoma mouse model. Immunohistochemical (IHC) results showed PD-1 protein expression in S180 sarcoma. Micro-PET/CT imaging successfully visualized the PD-1 protein receptor in S180 sarcoma at different time points (20, 48, 72, and 120 h) after probe injection. ConclusionThe 124I-anti-PD-1 monoclonal antibody molecular probe successfully targets the PD-1 receptor in S180 sarcoma of transgenic PD-1 C57 mice, and presents clear Micro-PET/CT immunoassay results, thus it potentially enables the non-invasive screening of patients with PD-1 positive malignant tumors.
4.Statistical analysis of vector monitoring in port areas of Shandong Province from 2017 to 2023

Objective To provide a scientific basis for vector prevention and control in port areas, vector monitoring has been conducted continuously to characterize the composition, density, and seasonal fluctuations of vectors across 21 port-areas of Shandong province from 2017 to 2023. Methods In accordance with the Regional Vector Monitoring Plan for Frontier Ports, vector surveillance of mosquitoes, flies, cockroaches, rodents, and other disease vectors was performed. Data were analyzed to describe vector community composition and identify seasonal fluctuations. Results A total of 601 rodents belonging to 1 family, 4 genera, and 4 species were captured. Three dominant rodent species inhabit port areas. In addition, 88 104 mosquitoes were captured and classified into 3 subfamilies, 4 genera, and 12 species; 38 959 flies were trapped and classified into 8 families,31 genera, and 51 species, including 9 dominant fly species in port-areas; and 54 672 captured cockroaches were classified into 2 families,4 genera, and 4 species, with Blattella germanica as the dominant species. Conclusions Significant vector community changes(P<0.005)have been identified after the epidemic. To improve the pertinence and efficacy of port health quarantine work, continuous and strong attention should be paid to vector compositions and population densities at Shandong ports.
5.Genetic diversity analysis of Aedes albopictus populations in Shandong Province using mitochondrial mtDNA-COⅠ gene sequences

Objective This study aimed to investigate the genetic diversity, differentiation, and population structure of Aedes albopictus across different geographical regions in Shandong Province, and to explore the relationship between genetic diversity and geographical distribution. Methods Between July and August 2024, ten Ae. albopictus populations were sampled from seven cities in Shandong Province. Genomic DNA was extracted from individual mosquitoes, and the mitochondrial cytochrome c oxidase subunit I(COⅠ)gene was amplified using PCR and sequenced. The obtained sequences were verified using BLAST and analyzed with MAFFT, MEGA 11, DnaSP v6.12, Arlequin 3.5, PopART 1.7, STRUCTURE 2.3.4 and the R packages adegenet and vegan to assess genetic diversity and population structure. Results A total of 229 COⅠ sequences(662 bp) was obtained, revealing ten variable sites with no insertions or deletions. The overall base composition showed an AT bias of 67.7%. Haplotype analysis identified 11 haplotypes, with Hap2 being the dominant and most widely distributed haplotype across all populations. Neutrality tests showed significant population expansion only in the Rizhao population. Mantel testing result revealed that geographical distance does not significantly impede gene flow. Overall, genetic differentiation among populations was low, indicating frequent gene flow. When combined with additional samples from Shanghai, Fujian, Guangdong, Yunnan, Hainan and Guangxi, STRUCTURE, UPGMA and DAPC analyses revealed two primary genetic clusters of Ae. albopictus. Conclusions Ae. albopictus populations in Shandong Province exhibit frequent gene flow, low genetic differentiation, and relatively low overall genetic diversity. However, the Rizhao population showed signs of recent expansion, highlighting the need for enhanced surveillance and targeted control measures in this area.
6.Relationship between MLR,SII,MPR and prognosis of maintenance hemodialysis patients with chronic kidney disease

Objective To investigate the relationship between monocyte-to-lymphocyte ratio(MLR),sys-temic immune inflammation index(SII),and mean platelet volume to platelet count ratio(MPR)and the prognosis of maintenance hemodialysis patients with chronic kidney disease(CKD).Methods The medical re-cords of totally 152 patients with CKD who received maintenance hemodialysis in the hospital from July 2022 to September 2023 were retrospectively analyzed.The patients were divided into the death group and the sur-vival group according to the prognosis.General data such as age,gender,height,weight,and dialysis age,as well as clinical and laboratory indicators such as blood cell count,hemoglobin,albumin,blood creatinine,blood urea,and blood uric acid were collected and analyzed.MLR,SII and MPR were calculated for each patient,and multivariate Logistic regression analysis was used to identify risk factors affecting the survival prognosis of CKD patients after maintenance hemodialysis treatment,and receiver operating characteristic(ROC)curve was used to evaluate the predictive value of the above indicators for patients'prognosis.Results Among 152 CKD patients,11.8%died,and the main causes of death include cardiovascular events,infections,multiple or-gan failure,and so on.Compared with the survival group,the levels of blood urea,blood uric acid,MLR,SII and MPR in the death group were significantly increased(P<0.05).Multivariate Logistic regression analysis revealed that MLR,SII and MPR were independent risk factors affecting the survival prognosis of CKD pa-tients after receiving maintenance hemodialysis treatment(P<0.05).ROC curve analysis showed that the are-a under the curve(AUC)of MLR for predicting the survival prognosis of CKD patients after maintenance he-modialysis treatment was 0.783,the cut off value was 0.58,the sensitivity was 72.22%,and the specificity was 76.87%.The AUC of SII for predicting the survival prognosis of CKD patients after maintenance hemodi-alysis treatment was 0.714,the cut off value was 480.50,the sensitivity was 66.67%,and the specificity was 70.15%.The AUC of MPR for predicting the survival prognosis of CKD patients after maintenance hemodial-ysis treatment was 0.706,the cut off value was 0.095,the sensitivity was 61.11%,and the specificity was 75.37%.The AUC of the combined detection was 0.843,which was better than the individual predictions(P<0.05).Conclusion MLR,SII and MPR are closely related to the prognosis of maintenance hemodialysis patients with CKD,and these indexes could be used as important reference for assessing the prognosis of pa-tients,which could help in the clinical decision-making and the development of individualized treatment plans.
7.Intergenerational Associations of Hypertensive Disorders of Pregnancy With Offspring Metabolomics: A Systematic Review

Objective::To examine the impact of hypertensive disorders of pregnancy (HDP) on offspring metabolomics.Methods::We searched five databases: PubMed, Ovid Embase, MEDLINE, Web of Science, and China National Knowledge Infrastructure, and included studies that reported metabolomics among human offspring born to HDP-complicated pregnancies.Results::Database search yielded 4054 articles, and after full-text screening, ten observational studies met inclusion criteria. Half of the studies had a sample size of less than 100 and were all observational studies in preeclampsia (PE) and gestational hypertension.Neonates were the most focused group in all included studies. Offspring born to HDP-complicated pregnancies exhibited statistically significant variations in blood metabolomics compared to their counterparts, characterized by amino acids, lipids, carnitine, and others (e.g., 1α,25-(OH) 2-D). Most studies reported a significant increase in differential metabolites of offspring born to HDP-complicated pregnancies. Four studies ( n = 1109) measured lipids-related metabolites, and all consistently showed that offspring born to PE-complicated pregnancies had significantly higher concentrations than non-PE exposed offspring. Conclusion::The existing evidence suggests an intergenerational effect of HDP on offspring metabolomics. Long-term follow-up studies are needed to advance the health effects of related adverse health outcomes and inform the prevention of offspring’s health.
8.Changpu Yujin Tang alleviates neuroinflammation in rats with Tourette syndrome by inhibiting the NLRP3/Caspase-1/GSDMD signaling pathway

Objective To explore the effects and mechanisms of Changpu Yujin Tang(CPYJT)on the NOD-like receptor thermal protein domain associated protein 3/cysteinyl aspartate specific proteinase-1/Gasdermin D(NLRP3/Caspase-1/GSDMD)signaling pathway-mediated neuroinflammation in rats with Tourette syndrome(TS).Methods SPF-grade male SD rats were randomly divided into the Control and TS groups.After successful modeling in the TS group,the rats were randomly divided into the Model,Tiapride,and CPYJT groups,and were treated with the corresponding drugs for 4 weeks.After the treatment,the rats' behavior was scored,H & E staining was used to observe pathological changes in the striatum,ELISA was used to measure the content of IL-1β and IL-18,RT-qPCR was used to detect the expression of NLRP3 and ASC mRNA,and Western blot was used to detect the expression of NLRP3,ASC,Caspase-1,Cleaved-Caspase-1,GSDMD,and GSDMD-NT proteins.Results Compared with the Control group,the Model group showed increased scores of stereotyped and motor behaviors(P<0.01),pathological changes in the striatal tissue,increased content of IL-1β and IL-18(P<0.01),and upregulated expression of NLRP3,ASC mRNA,and NLRP3,ASC,Caspase-1,Cleaved-Caspase-1,GSDMD,and GSDMD-NT proteins(P<0.01).Compared with the Model group,the Tiapride group and the CPYJT group showed decreased scores of stereotyped and motor behaviors(P<0.01),improved pathological damage in the striatal tissue,reduced content of IL-1β and IL-18(P<0.01),and inhibited expression of NLRP3,ASC mRNA,and NLRP3,ASC,Caspase-1,Cleaved-Caspase-1,GSDMD,and GSDMD-NT proteins(P<0.01).Conclusion The therapeutic effect of CPYJT on TS is related to the inhibition of the neuroinflammatory response mediated by the NLRP3/Caspase-1/GSDMD signaling pathway.
9.Correlation between brain gray matter volume changes and neurotransmitter receptors/transporters in patients with first-episode schizophrenia

Objective:To explore the spatial correlation between gray matter volume (GMV) changes and neurotransmitter receptors/transporters in patients with first-episode schizophrenia (FES) .Methods:Fifty-four FES patients(FES group) and fifty-nine healthy controls (HC group) were selected from June 2014 to May 2020 in the Psychiatry Department of Renmin Hospital of Wuhan University. Structural magnetic resonance imaging (sMRI) was conducted on all subjects. Differences of GMV were compared across 400 cortical regions and 32 subcortical regions. Based on the positron emission tomography(PET) data from Neuromaps, which provides the density of 19 different neurotransmitter receptors and transporters, Spearman correlation analysis was performed to evaluate the spatial correlation between GMV changes and neurotransmitter systems.Results:Compared to the HC group, FES group exhibited significant GMV reductions in widespread cortical (90/400) and subcortical (6/32) regions (all FDR-corrected P<0.05). The effect size of GMV reduction (Cohen’s d) showed significant positive correlations with the density of 5-hydroxytryptamine 1a(5HT1a) ( r=0.400, Pspin=0.002), γ-aminobutyric acid type A receptor(GABA A)( r=0.307, Pspin=0.002), and metabotropic glutamate receptor 5(mGluR5) ( r=0.275, Pspin=0.020) receptors (all FDR-corrected P<0.05). Conclusion:GMV reductions in a wide range of brain regions existed in patients with FES. There are significant correlations between 5HT1a, GABA A and mGluR5 receptors and gray matter reduction in patients with FES. The disorder of these neurotransmitter receptors may be the potential neurobiological mechanism of gray matter structural abnormalities in the early stage of schizophrenia.
10.Evaluation of the Effect of Pelvic Floor Reconstruction Using Transvaginal Ti-LOOP Total 6 Mesh in Patients with Pelvic Organ Prolapse

Objective:To evaluate the efficacy and safety of pelvic floor reconstruction using TiLOOP Total 6 mesh combined with perineal body reconstruction and posterior vaginal wall repair in patients with pelvic organ prolapse(POP).Methods:A retrospective analysis was conducted on 52 patients diagnosed with stage Ⅲ-ⅣPOP who underwent surgical intervention at Central Hospital of Wuhan,Tongji Medical College,Huazhong Univer-sity of Science and Technology between January 2022 and March 2023.The perioperative indicators and surgical complications were evaluated.The patient global impression of improvement(PGI-I),subjective and objective cure rates were evaluated.The POP-Q points before and 12 months after surgery were compared.Postoperative quali-ty of life was also assessed.Results:All procedures were successfully completed without intraoperative complica-tions.No intraoperative hematomas or injuries to the bladder and rectum were documented.Mean operative dura-tion was120±18 min,with an intraoperative blood loss of 141±44 ml.Postoperative complications included ve-nous thromboembolism(5.7%,3/52)and moderate-to-severe pain(53.8%,28/52).Until the follow-up period of 12 months,the surgical success rate was 100%.Mesh exposure rate and new stress urinary incontinence rate were 3.8%(2/52).The subjective and objective cure rate were 100%.Furthermore,POP-Q index points at the time of 12 months after operation were significantly improved compared with preoperative(P<0.0001).PFDI-20,PFIQ-7 and PISQ-12 scales were also significantly improved after the operation(P<0.0001).Patient satisfaction,as reflected by PGI-I scores,was unanimously positive.Conclusions:Pelvic floor reconstruction using TiLOOP Total 6 mesh combined with perineal body reconstruction and posterior vaginal wall repair is a safe and effective procedure for patients with advanced-stage POP(Ⅲ-Ⅳ),achieving high patient satisfaction.
