Targeted radiation therapy using radionuclides is a favored approach for treating tumors.This procedure involves the delivery of drugs to the lesion site via carriers or interventional methods,followed by the emission of radiation energy that selectively irradiates the lesion tissue.This approach minimizes damage to normal tissue and achieves the desired therapeutic effect.Factors such as the type of therapeutic radionuclide,radiation energy,physical half-life,method of preparation,and toxicity determine their clinical application.In this paper,the characteristics and clinical application of therapeutic radionuclides were reviewed to providing reference for the clinical application of targeted therapeutic radionuclides.
2.Regular whole blood donaion in Tianjin from 2013 to 2022

【Objective】 To analyze the situation of regular whole blood donation in Tianjin between 2013 and 2022, in order to provide data support for improving the recruitment and retention measures of regular blood donors and ensuring safe clinical blood supply. 【Methods】 From 2013 to 2022, 185 639 regular whole blood donors in Tianjin were selected as the study group and 1 015 312 other whole blood donors in the same period were selected as the control group. The demographic data, blood collection volume and blood retest screening of blood donors in the two groups were statistically analyzed. 【Results】 The number of regular blood donors and the volume of blood donated in Tianjin increased year by year from 2013 to 2022, with an average annual growth rate of 6.22% and 6.18%, respectively. From 2013 to 2021, the retention rate of regular blood donors increased first and then decreased. The proportion of male blood donors in the study and control groups showed a decreasing trend but the proportion of female donors showed an increasing trend, with the proportion of male donors in the study group higher than that in the control group and the proportion of female donors lower than that in the control group (both P<0.05). In the study group, the majority of blood donors were in the age of 26-35 years old, followed by those of 36-45 years old; in the control group, the majority of blood donors were in the age of 18-25 years old, followed by those of 26-35 years old; the proportion of blood donors in the study group in the age of 18-25 years old was lower than that in the control group, while the proportion of blood donors of other age group was higher than that in the control group, and the difference was statistically significant (P<0.05). The rates of 200 mL and 300 mL blood donations and insufficient blood donations in the study group were lower than those in the control group, while the rate of 400 mL blood donations was higher than that in the control group, and the differences were statistically significant. Among the blood donors in the study group, the proportion of students, civil servants, medical workers, military personnel, teachers and others was lower than that of the control group, while the proportion of the rest occupations was higher in the study group than that of the control group, and the differences were all statistically significant. There was a significant difference in the proportion of regular blood donors among blood donors of different professions. The re-test deferral rates of ALT and anti-TP in the study group showed a decreasing trend followed by an increasing trend, and the re-test deferral rates of HBV, HCV and HIV showed an increasing trend followed by a decreasing trend, and all the re-test deferral rates in the study group were lower than those in the control group, and the differences were statistically significant (P<0.05) . 【Conclusion】 From 2013 to 2022, the situation of regular blood donors in Tianjin has a certain regularity, and there is certain room for growth. Precise recruitment strategies targeting different populations should be adopted to have more regular blood donors.
3.Effect of Modified Huangqi Guizhi Wuwutang on Stress-related IRE1α/CHOP Pathway of Sciatic Nerve Endoplasmic Reticulum in Diabetes Rats

ObjectiveTo investigate the protective effect of modified Huangqi Guizhi Wuwutang (MHGW) on endoplasmic reticulum stress in the sciatic nerve of diabetes rats based on the pathways of inositol-requiring enzyme 1α (IRE1α) and CCAAT/enhancer-binding protein homologous protein (CHOP). MethodSixty rats were fed on a high-sugar and high-fat diet for six weeks, followed by intraperitoneal injection of streptozotocin at a dose of 35 mg·kg-1. Random blood glucose levels were measured three days later and rats with a sustained blood glucose level ≥ 16.7 mmol·L-1 were included in study (n=48). The rats were randomly divided into a model group, an α-lipoic acid group (0.026 8 g·kg-1·d-1), a high-dose MHGW group (2.5 g·kg-1·d-1), and a low-dose MHGW group (1.25 g·kg-1·d-1). Another 10 rats were assigned to the normal group. The intervention lasted for 16 weeks. After 16 weeks, the sciatic nerve structure of the rats in each group was observed under light microscopy using Luxol fast blue (LFB) staining. Transmission electron microscopy was used to observe the ultrastructure of the sciatic nerve. Chemiluminescence method was employed to measure the serum reactive oxygen species (ROS) levels. Western blot and real-time fluorescence quantitative polymerase chain reaction (Real-time PCR) were used to evaluate the expression of p-IRE1α protein, IRE1α mRNA, CHOP protein, and CHOP mRNA in the sciatic nerve of the rats. ResultCompared with the normal group, the model group showed elevated serum ROS levels (P<0.01). In contrast, the serum ROS levels were significantly reduced in the treatment groups compared with those in the model group (P<0.01). The sciatic nerve of the model group showed pathological changes compared with that in the normal group, while the treatment groups exhibited improvement in sciatic nerve pathology compared with the model group. The protein expression of p-IRE1α and CHOP in the sciatic nerve significantly increased in the model group as compared with that in the normal group (P<0.01). However, the treatment groups showed a significant decrease in the protein expression of p-IRE1α and CHOP in the sciatic nerve compared with the model group (P<0.05, P<0.01). Furthermore, compared with the normal group, the model group showed upregulated mRNA expression of IRE1α and CHOP in the sciatic nerve (P<0.01), while the treatment groups exhibited a significant decrease in the mRNA expression of IRE1α and CHOP compared with the model group (P<0.01). ConclusionMHGW can alleviate endoplasmic reticulum stress-induced cell apoptosis and improve the structure and function of the sciatic nerve in diabetes rats by inhibiting the expression of IRE1α/CHOP pathway-related proteins and mRNA, thereby preventing and treating peripheral neuropathy in diabetes.
4.Effect of Modified Huangqi Guizhi Wuwutang on Apoptosis-related Bax and Caspase-12 of Sciatic Nerve Cells in Diabetes Rats

ObjectiveTo investigate the effect of modified Huangqi Guizhi Wuwutang (MHGW) on the protein and mRNA expression of B-cell lymphoma-2-associated X protein (Bax) and cysteinyl aspartate specific proteinase-12 (Caspase-12) related to the apoptosis of sciatic nerve cells in diabetes rats to explore the mechanism of MHGW in the treatment of peripheral neuropathy in diabetes. MethodAnimal experiments were conducted. A diabetes model was induced in sixty male sprague-dawley (SD) rats by feeding on a high-sugar and high-fat diet combined with streptozotocin (STZ) intraperitoneal injection. Rats with random blood glucose levels ≥ 16.7 mmol·L-1 for three consecutive days were considered to have successfully developed diabetes. Forty-eight rats that successfully developed diabetes were randomly divided into a model group, an α-lipoic acid group (0.026 8 g·kg-1·d-1), a high-dose MHGW group (2.5 g·kg-1·d-1), and a low-dose MHGW group (1.25 g·kg-1·d-1), with 12 rats in each group. Another 10 rats were assigned to the normal group. Body weight and random blood glucose levels of the rats were monitored. At the end of a 16-week intervention period, the sciatic nerve conduction velocity of the rats was measured using the Key point electromyography collection system. The protein and mRNA expression of Bax and Caspase-12 in the sciatic nerve cells was detected by Western blot analysis and real-time quantitative polymerase chain reaction (Real-time PCR), respectively. ResultCompared with the normal group, the model group showed a significant decrease in body weight (P<0.01) and a significant increase in random blood glucose levels (P<0.01). After a 16-week intervention, compared with the model group, the high-dose MHGW group exhibited a significant increase in body weight (P<0.05), while there were no statistically significant differences in body weight changes among the other treatment groups. Random blood glucose levels significantly decreased in all treatment groups (P<0.01). After 16 weeks of intervention, compared with the normal group, the model group had significantly reduced motor and sensory nerve conduction velocities (P<0.01). Compared with the model group, all treatment groups showed significant increases in motor and sensory nerve conduction velocities (P<0.05, P<0.01). The expression of Bax and Caspase-12 proteins in the sciatic nerve cells was significantly elevated in the model group compared with that in the normal group (P<0.01). In contrast, all treatment groups showed significant reductions in the expression of Bax and Caspase-12 proteins in the sciatic nerve cells as compared with that in the model group (P<0.01). The expression of Bax and Caspase-12 mRNA in the sciatic nerve cells significantly increased in the model group compared with that in the normal group (P<0.01). Compared with the model group, the α-lipoic acid group and the high-dose MHGW group showed significant reductions in the expression of Bax mRNA in the sciatic nerve cells (P<0.05, P<0.01), while the low-dose MHGW group showed a decreasing trend in the expression of Bax mRNA. The expression of Caspase-12 mRNA in the sciatic nerve cells significantly decreased in all treatment groups (P<0.01). ConclusionMHGW may improve and repair sciatic nerve damage in diabetes rats by inhibiting sciatic nerve cell apoptosis.
5.Study on the Quality Standard of Lixieling Tablets Based on Multicomponent Analysis

OBJECTIVE To establish the quality standard of Lixieling tablets. METHODS Thin layer chromatography was used to identify Bistortae Rhizoma, Andrographis Herba and Sophorae Flavescentis Radix in Lixieling tablets. The components of andrographolium and matrine were determined by HPLC. RESULTS The thin layer spots of Bistortae Rhizoma, Andrographis Herba and Sophorae Flavescentis Radix were clear, with good separation effect and repeatability. The linear relationship between andrographolide, neoandrographolide, 14-deoxygenated andrographolide, dehydrated andrographolide, sophorine, oxymatrine, sophorine and oxysophorine and their respective concentration ranges was good. The RSDs of instrument precision, repeatability, stability and recovery all met the requirements. CONCLUSION The method is simple and reproducible, and can be used for the quality control of Lixieling tablets.
6.Whole blood donation related adverse reactions and predisposing factors: a study of blood donors interviewed within 2 weeks after donation

【Objective】 To study the adverse reactions(ADRs) in blood donors after whole-blood donation in terms of frequency and types, so as to explore the predisposing factors of adverse reactions. 【Methods】 A questionnaire was designed to investigate ADRs within 2 weeks after whole blood donation. A total of 812 whole blood donors were randomly selected from September to October 2021. They were interviewed by trained medical staff before leaving the site after blood donation, then followed up by telephone 24 hours and 2 weeks after blood donation. The frequency and type of ADRs were surveyed, and the physical examination data such as gender, age, blood pressure, mean arterial pressure (MAP) and weight were collected to, analyze the predisposing factors that correlated with ADRs. 【Results】 Finally, 734 people [90.39% (734/812)], with valid demographic and physical examination information, were included in this study after on-site interviews and two follow-ups within two weeks after blood donation. The incidence of ADRs was 13.22% (97/734), of which the incidence of on-site vasovagal reactions during blood donation was 10.49% (77/734) (with the constituent ratio as 79.38%, 77/97)and the incidence of ADRs ≤ 2 weeks after leaving the blood collection site was 2.73% (20/734, including 12 donors had (1.63%, 12/734) local symptoms)(with the constituent ratio as 20.62%, 20/97). Multivariate logistic regression analysis showed that the factors influencing the occurrence of ADRs were as follows: 400mL-blood donation (OR 6.312, 95% CI 3.336-11.944), 45~60kg weight (OR 3.744, 95% CI 2.017-6.949), MAP ≤ 90 mmHg (OR 3.101, 95% CI 1.416-6.788), novel blood donor (OR 2.237, 95% CI 1.199-4.174), age of 18~30 years (OR 2.197, 95% CI1.107-4.361), female (OR 1.871, 95% CI 1.026-3.413); blood pressure, occupation, education background, body mass index (BMI) and Hb of blood donors were not the influencing factors and risk predictors of ADRs. 【Conclusion】 The application of on-site interview and two follow-ups after donation can effectively obtain the true information of blood donors′ ADRs, which can timely grasp and predict the relevant factors affecting the occurrence of ADRs, so as to take targeted preventive measures to ensure the safety of blood donors.

Postzygotic mutations are acquired in normal tissues throughout an individual's lifetime and hold clues for identifying mutagenic factors.Here,we investigated postzygotic mutation spectra of healthy individuals using optimized ultra-deep exome sequencing of the time-series samples from the same volunteer as well as the samples from different individuals.In blood,sperm,and muscle cells,we resolved three common types of mutational signatures.Signatures A and B represent clock-like mutational processes,and the polymorphisms of epigenetic regulation genes influence the pro-portion of signature B in mutation profiles.Notably,signature C,characterized by C>T transitions at GpCpN sites,tends to be a feature of diverse normal tissues.Mutations of this type are likely to occur early during embryonic development,supported by their relatively high allelic frequencies,presence in multiple tissues,and decrease in occurrence with age.Almost none of the public datasets for tumors feature this signature,except for 19.6%of samples of clear cell renal cell carcinoma with increased activation of the hypoxia-inducible factor 1(HIF-1)signaling pathway.Moreover,the accumulation of signature C in the mutation profile was accelerated in a human embryonic stem cell line with drug-induced activation of HIF-1α.Thus,embryonic hypoxia may explain this novel signature across multiple normal tissues.Our study suggests that hypoxic condition in an early stage of embryonic development is a crucial factor inducing C>T transitions at GpCpN sites;and indi-viduals'genetic background may also influence their postzygotic mutation profiles.

9.Characteristics of blood donors under COVID-19 epidemic in Tianjin

【Objective】 To study and analyze the basic characteristics of blood donors under the COVID-19 epidemic in Tianjin area, so as to provide data support for improving the blood collection for emergency incidents. 【Methods】 The blood collection data of Tianjin Blood Center under the COVID-19 epidemic in 2020 were collected in terms of the number of blood donations, changes in blood donation units over time, group blood donation, donations given by regular blood donors and donations inside fixed blood donation sites, registered residence, gender, age, education, occupation, individual donation, group donation, and the annual daily blood collection and delivery data in Tianjin. 【Results】 The number of blood donations and donation units in Feb 2020 decreased by 56.96%(3 226/7 496-1)and 62.40%(5 382/14 314.5-1)compared with the same period in 2019, and decreased by 56.87%(3 226/7 480-1)and 60.70%(5 382/13 695.7-1)compared with the average value in the previous five years. The number of blood donations, donation units and rate of 400 mL donation in 2020 decreased by 13.25%(119 107/137 293-1), 17.7%(205 196.5/249 405-1)and 12.83%(66.0/75.8-1)respectively. Meanwhile the units of group donation increased by 22.59%(44 118.5/35 989.0-1)as compared with those in 2019. The proportion of regular blood donors raised up to 17.31%(20 623/119 107, which was the highest in recent 6 years. During the epidemic period, the number of blood donations of local registered residence, women, over 36 years old, with bachelor and above degree increased by varied extent compared with previous years. Workers, staff members and others were the main groups of voluntary blood donation, and civil servant, medical workers, farmers and teachers also played a certain role during the epidemic. 【Conclusion】 The clinical blood use in Tianjin was basically guaranteed during the COVID-19 epidemic. It is suggested to further optimize the structure of voluntary blood donors, establish a diversified and multi-level group blood donation team, and increase the number of regular blood donors.

Teicoplanin is a glycopeptide antibiotic for treatment of gram-positive bacterial infections in children, especially for methicillin-resistant staphylococcus aureus infection.The body clearance of teicoplanin differs between child and adult.This article explores the suitable testing indicators based on the clinical medication guide and the latest research progress , as well as the pharmacokinetic and pharmacodynamics of teicoplanin , to provide information for individualized dosage regime of teicoplanin in children.
