Atherosclerosis, a chronic inflammatory disease, is markedly influenced by both immune and inflammatory reactions throughout its progression. Dendritic cells, as pivotal antigen-presenting entities, play a crucial role in the initiation of immune responses and the preservation of immunological homeostasis. Accumulating data indicates that dendritic cells are present in healthy arteries and accumulate significantly in atherosclerotic plaques. Novel immunotherapeutic approaches and vaccination protocols have yielded substantial clinical advancements in managing chronic inflammatory diseases, with dendritic cell-centric modalities emerging for atherosclerotic management. In this review, we delineate the essential functions and underlying mechanisms of dendritic cells and their subsets in the modulation of atherosclerotic inflammation and immune responses. We underscore the immense promise of dendritic cell-based immunotherapeutic strategies, including vaccines and innovative combinations with nanotechnological drug delivery platforms for atherosclerosis treatment. We also discuss the challenges associated with dendritic cell immunotherapy and provide perspectives on the future direction of this field.

Polyethylene terephthalate (PET) fibers are characterized by exceptional mechanical strength, and textiles blended with cotton fibers combine both comfort and durability, showcasing widespread use in daily applications. However, improper disposal of discarded polyester-cotton textiles has resulted in severe environmental pollution, necessitating urgent and effective mitigation strategies. Enzymatic recycling of textiles offers superior environmental benefits and holds greater potential for industrial applications than alternative recycling methods. This study aims to explore a large-scale solution for the treatment of waste textiles, particularly addressing the challenge of resource recovery from polyester-cotton blended fabrics. An innovative enzymatic depolymerization process has been developed to achieve the recovery of high-purity terephthalic acid monomers. Experiments were conducted on three different textile blends with polyester-to-cotton ratios of 65/35, 70/30, and 80/20, and the influences of different colors on the process were investigated. Initially, the textiles were pretreated through mechanical grinding, which was followed by depolymerization of cotton fibers with commercial cellulase. The crystallinity of PET in the textiles was reduced through a rapid heating and cooling process. Subsequently, the PET was depolymerized by the engineered PET hydrolase. The results demonstrated that after decolorization and separation of terephthalic acid (TPA) from the reaction system, the monomer recovery rates for the three textile blends (65/35, 70/30, and 80/20) reached 90%, 91%, and 92%, respectively. Characterization analysis by nuclear magnetic resonance (NMR) confirmed that the purity of the recovered TPA was greater than 99%. In conclusion, the fully enzymatic recycling process developed in this study shows considerable promise for large-scale industrial applications and is anticipated to significantly advance the adoption and development of enzymatic recycling technologies for PET in industrial processes.

Objective To establish a dual-mode detection platform for Mycoplasma pneumoniae(MP)based on recombinase aided amplification(RAA),CRISPR/Cas12a,and lateral flow immunochromatographic assay(LFIA).Methods RAA primers and crRNA were designed to target the conserved region of the MP P1 gene.Key parameters including RAA reaction time,core component concentrations of the CRISPR/Cas12a system,and CRISPR reaction time were systematically optimized.The analytical performance of the platform was evaluated via dual-mode of real-time fluorescence monitoring and LFIA naked-eye visual interpretation.Sensitivity was assessed using serially diluted MP plasmid templates(10-1-105 copy/μL),specificity against seven respiratory pathogens,including Mycoplasma hominis and Chlamydia pneumoniae,and clinical concord-ance with 25 throat swab samples validated by real-time fluorescence quantitative PCR(qPCR)as the gold standard.Results The optimal reaction conditions of the established RAA-CRISPR dual-mode platform were determined as follows:RAA reaction time of 15 min,Cas12a concentration of 80 nmol/L,crRNA concentration of 90 nmol/L,and CRISPR reaction time of 40 min.The limit of detection(LOD)for the fluorescence mode was 1 copy/μL with a detection rate of 98.3％(59/60,95％CI:94.7％-99.8％).For the LFIA mode,the LOD was 1 copy/μL with a detection rate of 96.7％(58/60,95％CI:88.4％-99.5％).No cross-reactivity was observed with any tested pathogens.Clinical validation results showed that the platform's detection re-suits were consistent with qPCR,with sensitivity,specificity,and overall agreement all reaching 100.0％.The detection time was relatively short,with the total detection time lasting no more than 55 minutes.Conclusion The established RAA-CRISPR dual-mode platform combines ultrasensitivity,visual interpretation,and operational simplicity,can provide an efficient solution for point-of-care testing(POCT)of MP infections at primary healthcare settings.

Multinodular hepatocellular carcinoma (HCC) poses substantial clinical challenges due to its high tumor burden,pronounced heterogeneity,and poor prognosis. This review summarizes current definitions and staging systems for multinodular HCC and examines the evidence and controversies surrounding surgical management. Particular attention is given to the efficacy and safety of hepatic resection across varying tumor burdens and biological behaviors. The heterogeneity between intrahepatic metastasis and multicentric occurrence subtypes is explored in the context of treatment decision-making. Liver transplantation,while potentially curative and offering notable survival benefits-especially for patients meeting the Milan criteria or within expanded selection frameworks-is limited by donor scarcity and the risk of post-transplant recurrence. Salvage transplantation remains a debated strategy for patients who experience recurrence after resection. In the era of targeted and immune therapies,combining locoregional approaches with systemic treatment has broadened the therapeutic landscape for unresectable multinodular HCC,creating new opportunities for conversion and downstaging. This review also highlights recent advances and persistent challenges in the clinical integration of these strategies. Moving forward,the management of multinodular HCC should prioritize precision medicine,multidisciplinary collaboration,and individualized treatment planning,supported by high-quality clinical research and technological innovation,with the ultimate goal of improving both survival and quality of life for patients.

In early stage after liver transplantation(LT), coagulation function of recipients stays in a fragile balance. Affected by a variety of complex mechanisms, blood is usually hypercoagulable. An imbalance between coagulation factors and physiological anticoagulants, elevated level of vWF, an occurrence of fibrinolysis inhibition and dosing of immunosuppressive agents cause a hypercoagulable state in an early stage after LT. Blood hypercoagulability may lead to such thrombotic complications as hepatic artery, portal vein and deep vein thromboses. Some studies have demonstrated that postoperative prophylactic anticoagulation has some effect in reducing the risks of early postoperative thrombosis. However, there is still a great lack of high-quality evidence. This review summarized the latest researches on early coagulation dysfunction, thrombosis and preventive anticoagulation after LT.

Objective To explore the feasibility of robot-assisted laparoscopic inferior vena cava (IVC) thrombectomy in treating renal tumor with Mayo level Ⅲ-Ⅳ inferior vena cava thrombus.Methods From November 2014 to January 2017,5 cases of renal tumor with Mayo level Ⅲ-Ⅳ inferior vena cava tumor thrombus were treated with robot-assisted surgery.There were 4 males and 1 female with the median age of 59 years (range 54-71 years).Four cases had the renal tumor on the right side and one on the left side.The mean tumor size was 6.8 cm (range 5-9 cm) with 3 cases of T3b and 2 cases of T3c.There were 4 cases of level Ⅲ and 1 case of level Ⅳ inferior vena cava thrombus with the median length of 9 cm (range 7-11 cm).The surgical procedure for Mayo level Ⅲ inferior vena cava thrombus included mobilization of both left and right robes of liver,subsequently controlling the suprahepatic infradiaphramatic IVC and first porta hepatis simultaneously.The surgical procedure for Mayo level Ⅳ inferior vena cava thrombus included cardiopulmonary bypass by multi-disciplinary cooperation among urologists,hepatobiliary and cardiovascular surgeons.The procedures included live mobilization,control of the superior vena cava and first porta hepatis and remove thrombus in the atrium and IVC respectively.Results All operations were completed successfully.The median operative time was 440 min (320-630 min).The blood recovery device was used and the intraoperative estimated blood loss was 2 500 ml (500-6 000 ml) and all cases required intraoperative blood transfusion.The median time of intraoperative occlusion of IVC was 35 min (25-50 min).All patients were transferred to the intensive care unit for median of 4 days (2-8 days) after surgery.The median time to remove the postoperative drainage tube was 9 days (7-12 days).Postoperative pathological diagnosis revealed 5 cases of clear cell carcinoma.Postoperative renal dysfunction occurred in 3 patients and liver dysfunction occurred in 2 patients who improved after medical therapy.During median 19.6 months (12-48 months) of follow-up,1 patient died and 1 patient progressed.Conclusions Despite the high risk of surgery,robot-assisted laparoscopic IVC thrombectomy for renal tumor with Mayo level Ⅲ-Ⅳ thrombus is feasible for experienced surgeons in selected patients.However,the oncological outcomes need further investigation.

The study aims to use CRISPR/Cas9 introducing foreign gene targeted knock-in into chicken EAV-HP genome. First, specific primers were designed for amplification of EAV-HP left, right homologous arms and enhanced green fluorescent protein (eGFP) expression cassette. PCR products of homologous arms were ligated to both sides of eGFP by overlap extension PCR, resulting in full-length donor DNA fragment designated as LER. Then LER fragments were cloned into pMD19-T to obtain donor vector pMDT-LER. Subsequently, the donor vector pMDT-LER was transfected into HEK293T cells to verify the expression of eGFP gene. Furthermore, co-transfection of CRISPR/Cas9 expression vector and pMDT-LER into chicken DF-1 cells was performed to achieve eGFP transgenic cells. Meanwhile, eGFP expression was observed in cells, and the event of eGFP integration into EAV-HP genome was detectable by amplification of target DNA. Finally, the transgenic DF-1 cells were passaged seven times, and the stable integration and expression of eGFP was checked by PCR and Western blotting. These results demonstrated that eGFP gene was knocked into the EAV-HP genome successfully, which provides a new integration site for research of transgenic chicken.

Objective To investigate the safety and feasibility of robotic nephrectomy,work bench surgery with robotic kidney autotransplantation in the treatment of complex renal tumors.Methods The clinical data of 5 patients with renal tumors admitted from January 2018 to July 2018 were analyzed retrospectively.There were 4 males and 1 females.The median age was 49 years old,ranging 32-66 years.The median body mass index was 25.6 kg/m2,ranging 21.1-27.8 kg/m2.Serum creatinine level was 87.2 μmol/L,ranging 78.0-88.9μmol/L before bench surgery.5 patients had multiple bilateral renal tumors and had undergone laparoscopic or robotic partial nephrectomy on the contralateral kidney.For bench surgery kidney,4 cases were on the left side and 1 case was on the right side.Each kidney has more than 2 separate tumors,combined with complete endophytic tumors,tumors larger than 7 cm in diameter or hilar tumors.5 patients were all performed robotic nephrectomy,work bench partial nephrectomy with robotic kidney autotransplantation under general anesthesia.The patient was first in a lateral decubitus position for robotic nephrectomy,and the kidney was removed through a median 6 cm periumbilical incision.After kidney removal,kidney tumors were resected and kidney was reconstructed on a hypothermic working table.Then the kidney was packed in a plastic bag,filling with ice slush.The corresponding parts of the plastic bag were cut to expose the renal artery and vein.Finally,the patient was moved to lithotomy position with Trendelenburg tilt of 20°,and the autologous kidney wrapped in the plastic bag was placed through the previous periumbilical incision into the abdominal cavity for robotic kidney autotransplantation.The renal artery and vein were anastomosed end-to-side with the right external iliac artery and vein.The ureter and bladder were anastomosed.Autologous kidneys were placed in abdominal cavity in 4 cases,and placed in right iliac fossa with retroperitonealization in 1 case.Ice slush on the surface of the autologous kidney did not completely melt before the blood supply was restored during the operation,and the autologous kidney immediately urinated after the blood supply was restored.Results All surgeries were performed successfully without conversion to open surgeries.The total operation time was 460 min,ranging (415-645 min),the time of robotic nephrectomy was 120 min,ranging (74-300 min),the time of robotic kidney autotransphntation was 135 min,ranging(103-163 min),the warm ischemia time was 3 min,ranging (1.5-6.0 min),the cold ischemia time was 182 min,ranging(135-210 min),the rewarming time was 50 min,ranging(45-55 min),the estimated blood loss during operation was 100 ml,ranging(50-300 ml),and the hospital stay was 6 d,ranging(5-9 d).The number of resected tumors was 4,ranging(2-6).The pathology reveals clear cell carcinoma in 3 cases and chromophobe cell carcinoma in 2 cases.The surgical margins were all negative.The serum creatinine levels were 111.1 μmol/L (87-217.6 μ mol/L) and 106.1 μmol/L (87.1-172 μmol/L) on the 7th and 30th day after operation,respectively.One month after operation,CT showed that the function and morphology of the autologous kidneys were fine.No recurrence or metastasis was found in 5 patients during a median follow-up of 7 months,ranging (5.4-11.7 mon).Conclusions For patients with complex renal tumors who cannot undergo in situ partial nephrectomy,robotic nephrectomy,work bench surgery with robotic kidney autotransplantation can completely remove the tumors,maximize the preservation of renal function and minimize the trauma of patients,making the ultimate means of nephron-sparing surgery for patients with complex renal tumors more minimally invasive and safe.

Objective To investigate the relationship between maximum blood pressure and the prognosis after discharged from the hospital,among patients with ischemic stroke.Methods A cohort study was conducted which including 471 cases of ischemic stroke patients that were collected from February 2014 to December 2014 at the Second Affiliated Hospital of Harbin Medical University.Values of everyday blood pressure were measured on each patient at the first six days after admitted to the hospital.Maximum blood pressure value of the 6 days was taken as an indicator of the blood pressure levels.The ability on daily living was measured by the modified Rankin score (mRs).Data were analyzed by Chi-square test,t test and multivariate logistic regression analysis.Results Confounding factors would include age,gender,culture,physical activity,income,smoking,alcohol,hypertension,diabetes,coronary heart disease,stroke history,hospitalization mrs,lipid parameters,homocysteine and blood sugar and were adjusted.Results from multivariate logistic regression analysis showed that the maximum SBP was associated with adverse outcomes.Compared with 140-159 mmHg for SBP,OR(95％CI) was 2.51 (1.30-4.85) for 160-179 mmHg,OR (95％ CI) was 2.68 (1.27-5.65) for those pressure levels higher than 180 mmHg,after multiple factors were adjusted.Compared with 90-99 mmHg for DBP,OR(95％CI) was 1.92 (1.00-3.67) for 100-109 mmHg,OR (95％ CI) was 2.78 (1.35-5.69) for the ones higher than 110 mmHg.Conclusion Maximum blood pressure during hospitalization might be associated with adverse outcome of ischemic stroke patients.
