1.Simultaneous determination of six alkaloid components in Zhachong Shisanwei Pills by QAMS

Objective:To establish a quantitative analysis of multi-components by single marker(QAMS)for the determination of 6 alkaloid components,which is benzoylmesaconine,benzoyl-hypaconine,benzoylaconine,mesaconitine,hypaconitine,and aconitine in Zhachong Shisanwei Pills,and prove the scientificity and feasibility of the method in the quality analysis.Methods:The chromatographic separation was performed on an Agilent Eclipse Plus C18(250 mm×4.6 mm,5 μm)with gradient elution using 0.1 mol·L-1 ammonium acetate(0.5 mL of gla-cial acetic acid per 1 000 mL)(A)-acetonitrile:tetrahydrofuran(25∶15)(B),as the mobile phase(0-50 min,18%B-28%B),the detection wavelength was switched from 235 nm,the column temperature was kept at 40℃and the flow rate was 1.0 mL·min-1.The relative correction factors(fs/i)were established with the other 5 compo-nents to be measured using benzoylaconine as the internal reference,which were used to calculate the mass fraction of each component.At the same time,the mass fractions of the 6 effective constituents in Zhachong Shisanwei Pills were calculated by the external standard method(ESM).By comparing the content results of ESMand QAMS,the accura-cy of QAMS method were evaluated.Results:The relative correction factors(fs/i)of benzoylmesaconine,benzoylhyp-aconine,mesaconitine,hypaconitine,and aconitine in Mongolian medicine Zhachong Shisanwei Pills were reproduci-ble with good reproducibility,which were 0.680 4,0.450 6,0.850 8,0.676 1 and 0.757 0,the result obtained by QAMS approximated those obtained by external standard method(ESM).Conclusion:The method is simple,stable and reproducible,and can be used for the quality control of 6 alkaloid components in Zhachong Shisanwei Pills.
2.SIRT3 protects endometrial receptivity in patients with polycystic ovary syndrome.

BACKGROUND:
The sirtuin family is well recognized for its crucial involvement in various cellular processes. Nevertheless, studies on its role in the human endometrium are limited. This study aimed to explore the expression and localization of the sirtuin family in the human endometrium, focusing on sirtuin 3 (SIRT3) and its potential role in the oxidative imbalance of the endometrium in polycystic ovary syndrome (PCOS).
METHODS:
Endometrial specimens were collected from both patients with PCOS and controls undergoing hysteroscopy at the Center for Reproductive Medicine, Peking University Third Hospital, from July to August 2015 and used for cell culture. The protective effects of SIRT3 were investigated, and the mechanism of SIRT3 in improving endometrial receptivity of patients with PCOS was determined using various techniques, including cellular bioenergetic analysis, small interfering ribonucleic acid (siRNA) silencing, real-time quantitative polymerase chain reaction, Western blot, immunofluorescence, immunohistochemistry, and flow cytometry analysis.
RESULTS:
The sirtuin family was widely expressed in the human endometrium, with SIRT3 showing a significant increase in expression in patients with PCOS compared with controls ( P <0.05), as confirmed by protein and gene assays. Concurrently, endometrial antioxidant levels were elevated, while mitochondrial respiratory capacity was reduced, in patients with PCOS ( P <0.05). An endometrial oxidative stress (OS) model revealed that the downregulation of SIRT3 impaired the growth and proliferation status of endometrial cells and reduced their receptivity to day 4 mouse embryos. The results suggested that SIRT3 might be crucial in maintaining normal cellular state by regulating antioxidants, cell proliferation, and apoptosis, thereby contributing to enhanced endometrial receptivity.
CONCLUSIONS
Our findings proposed a significant role of SIRT3 in improving endometrial receptivity in patients with PCOS by alleviating OS and regulating the balance between cell proliferation and apoptosis. Therefore, SIRT3 could be a promising target for predicting and improving endometrial receptivity in this patient population.

4.Identification of Medicinal Aconitum Species Based on ITS2 Sequences and Analysis of Genetic Relationship

ObjectiveThe internal transcribed spacer (ITS) 2 region of ribosomal gene, a DNA barcode, was employed to identify 12 medicinal Aconitum species and the genetic relationship among the species was analyzed. MethodA total of 30 samples of the 12 species were collected. The DNA was extracted with spin column plant genomic DNA kit and the universal primers of ITS2 sequence were used for polymerase chain reaction (PCR) amplification, followed by electrophoresis detection and bi-directional sequencing. The yielded sequences were aligned and spliced by CodonCode Aligner 17.0 and sequence variation was analyzed by MEGA 7.0. The secondary structure was predicted by ITS2 Database and the neighbor-joining (NJ) method was applied to generate the phylogenetic tree. ResultThe ITS2 sequences of the 12 species were 220-221 bp, with the average guanine and cytosine (GC) content of 64.09%, 140 variable sites, 137 informative sites, and 81 conservative sites. The intraspecific genetic distance (K2P) was smaller than the interspecific genetic distance. According to the secondary structures of ITS2 sequences and NJ cluster analysis, A. scaposum, A. sinomontanum, and A. barbatum had close genetic relationship, while the rest nine showed close kinship, particularly A. soongaricum and A. yinschanicum. ConclusionITS2 sequence is of great value for the molecular identification and genetic relationship determination of Aconitum, which provides a new method for the study of ethnomedicine.

Objective:Biobank construction plays an irreplaceable role in the research of accurate prevention and treatment of diseases. Shared biobank network based on a large crowd queue is the way of the future. This subject is one of the key contents of national precision medicine "The Breast Cancer Cohort Study in Chinese Women: (BCCS-CW)" , aiming to solve the bottleneck of insufficient standardization and sharing.Methods:The establishment of "entity library-information library-extension library" , the widely Shared network of biobank of breast cancer specific disease cohort, and the establishment of strict standard setting and quality control standard to construct the standardized biobank.Results:This biobank provides a shared biobank resource for breast cancer risk assessment, prediction and early warning, early screening, classification, individualized treatment, efficacy and safety prediction and monitoring and other accurate prevention and treatment programs and clinical decision-making system research.Conclusion:The data of this biological sample bank is refined and complete, and the sample size of cases is sufficient, which can meet the research needs of medical big data, genomics, metabonomics, epigenetics and other fields.
6.Influences of self-efficacy intervention on compliance of liver transplantation recipients during hospitalization

Objective To explore the influences of self-efficacy intervention on compliance and self efficacy of liver transplantation recipients during hospitalization.Methods A total of 64 liver transplantation recipients in liver transplantation center of 302 Military Hospital of China from October 1 st 201 2 to September 30th 201 4 were recruited and divided into intervention group (targeted measures to enhance self efficacy)and control group (routine nursing and health education)according to the serial number designed by sequence of operation.General information of patients was collected by transplantation registration system.Besides,self-efficacy level before operation,self-efficacy and compliance level at 1 ,6 months after operation was evaluated by questionnaire.And then,self-efficacy and compliance were compared between two groups.Results There were statistically significant differences in the score of self efficacy of patients between two groups at 1 st and 6th month (P <0.01 ).The scores of self-efficacy of patients in two groups before and after intervention were analyzed with repeated measurement analysis of variance.The results showed that it was an obvious difference between two groups.At the same time,there was a statistical difference between two groups with the time changing (P <0.05).The scores of compliance,self-monitoring nursing and life habits of patients in intervention group at 1 st and 6th month were higher than those in control group with significant differences (P <0.01 );however,there was no significant difference in medication compliance and scores of two sub-scales (P >0.05).Besides,there was no significant difference on whether complications occurred (P >0.05 ).Conclusions Self-efficacy intervention can enhance self-efficacy of liver transplantation recipients after operation,improve patients′ability of self-care,promote their health behavior and improve patients′compliance so as to improve the quality of life of patients after operation.
7.Analysis of curative effect for concurrent chemoradiotherapy versus neoadjuvant chemotherapy for stage ⅡB-ⅢB cervical cancer

Objective To observe the effect and side effects of concurrent chemoradiotherapy for stage ⅡB~ⅢB cervical cancer. Methods 126 patients with stage ⅡB~ⅢB cervical cancer were randomly allocated into 2 groups. Concurrent chemoradiotherapy group: radiotherapy carried out same as the neoadjuvant chemotherapy group, i.e.firstly with cisplatin 20 mg iv d1-5, 5-Fu 750 mg iv d1-5, repeated every 28 days, total 4 cycle; the neoadjuvant chemotherapy group i.e.firstly with cisplatin 20 mg iv d1-5, 5-Fu 750 mg iv d1-5, repeated every 28 days, total 2 cycle, after chemotherapy received routine radiotherapy 2 Gy per day, 5 times a week to a total dose of 30 Gy with 192Ir brachytherapy, 7 Gy per week. When total dose reached 46 Gy, the middle field was shielded by plumbum, then continuous radiotherapy, total dose reach A point 65-70 Gy, B point 50-56 Gy. Results All patients were followed-up for more than five years. The follow-up rate was 94.4 %. In concurrent chemoradiotherapy group the 3 year survival rate and the 5 year survival rate were 82.8 %, 65.6 %, In neoadjuvant chemotherapy group the 3 year survival rate and the 5 year survival rate were 67.7 %, 46.8 %. There was a significant difference in two groups (X2=3.86, P<0.05; X2=5.01, P<0.05), no significant difference in toxicity-side effect. Conclusion Concurrent chemoradiotherapy for advanced cervical cancer can significantly improve the 3-year and 5-year survival rate and has little increase in toxicity-side effect.
8.Clinical combination therapy for adrenal metastasis from lung cancer.

BACKGROUNDTo study the results of combination therapy for adrenal metastasis from lung cancer.
METHODSThirty patients with adrenal metastasis were treated in our hospital from Feb. 1995 to Apr. 2001. Forteen patients were small cell lung cancer (SCLC) and 16 patients were non-small cell lung cancer (NSCLC). All patients were treated with chemotherapy and/or radiotherapy. Eighteen patients were treated with combination therapy of chemotherapy and radiotherapy, while twelve patients with chemotherapy alone.
RESULTSThe median survival time was 8 months. The response rate of chemotherapy alone was 25.0% (partial relief 3 cases), while the response rate of combination therapy was 44.4% (complete relief 1 case and partial relief 7 cases). Pain was relieved quickly after radiotherapy for the patients with pain symptom.
CONCLUSIONSCombination of radiotherapy and chemotherapy is better than chemotherapy alone for the treatment of adrenal metastasis from lung cancer.
