1.Preliminary Study on Construction Method of Type 1 Diabetes Model in Banna Miniature Pig

Objective To construct a Type 1 diabetes model in miniature pigs and explore postoperative care strategies for effectively prolonging the survival time of the model pigs.Methods Seven Banna miniature pigs were selected for pancreatectomy.Glucose,vitamins,and antibiotics were administered for 3-5 days after surgery to aid recovery.Blood glucose and urine glucose levels were measured twice a day in the morning and evening to adjust insulin supplementation accordingly.The model pigs were observed daily and records were kept,including orexis,psychosis,weakness,skin ulcer,and feces and urine.Body weight was measured weekly until the death of the model animals.Based on the model pigs'condition,glucose injection and Ringer's lactate solution were administered to supplement nutrition and correct electrolyte imbalances.Results All seven Banna miniature pigs showed typical symptoms of diabetes:random blood glucose levels higher than 11.1 mmol/L after pancreatectomy,far exceeding the average blood glucose level of 6.0 mmol/L in normal pigs;positive urine glucose;and progressive weight loss.These features indicated the successful construction of Type 1 diabetes model.Additionally,Type 1 diabetic pigs that survived more than 8 weeks showed progressive hair loss and skin ulceration.Euthanasia was performed on model pigs when they were unable to stand or even eat independently,and pathological examination and HE staining were conducted on tissues collected from affected organs such as the liver,kidneys,and skin.Pathological sections revealed liver congestion,massive glycogen accumulation,ballooning degeneration of hepatocytes,and progressive liver fibrosis,along with glomerular congestion,vacuolar degeneration in renal tubular epithelial cells,proteinuria,dermal congestion,thinning of vascular walls,and varying degrees of parakeratosis and dyskeratosis in the liver,kidneys,and skin tissues due to prolonged hyperglycemia.The average survival time of the constructed Banna miniature pig diabetes model was 44 d,with a maximum survival time of 121 d.Conclusion Type 1 diabetes model can be constructed successfully in Banna miniature pigs through pancreatectomy.With meticulous postoperative care,a long-term Type 1 diabetes model with significant complications can be achieved,providing a stable large-animal model for Type 1 diabetes treatment strategies.
2.Herbal textual research on Flos Trollii

Flos Trollii is a traditional Chinese medicinal herb in China. The 2020 edition of the Chinese Pharmacopoeia (part 1) did not include the medicinal herb, its source is not clear, and there is a lack of relevantly systematic and comprehensive research. By consulting ancient Chinese herbal medicines, medical books and related literature, the textual research of Flos Trollii was conducted to verify the name, origin and producing area, so as to provide a reference for the clinical application and resource development of Flos Trollii. Through textual research, it could be seen that the name “Jinlianhua” was used as the correct name in the mainstream origin of the past dynasties, and there were still multiple synonyms such as Hanjinlian, Jinmeicao and so on, most of which originated from its growth environment and appearance. According to the distribution of varieties, it could be inferred that the mainstream origin of Flos Trollii in the Qing Dynasty and before was Trollius chinensis Bge. According to historical records, Flos Trollii were mostly produced in northern regions such as Hebei, Inner Mongolia, Shanxi, etc., which was related to the fact that Flos Trollii liked cloudy, humid and cold environments. Based on the textual research results, the author suggested that the mainstream origin of the past dynasties T. chinensis Bge. should be selected for subsequent collection of Flos Trollii.
3.Effects of breast prosthesis implantation on psychological stress and quality of life in patients with breast cancer under double stress

Objective:To investigate the effect of breast prosthesis implantation on psychological stress and quality of life in patients with breast cancer under double stress.Methods:A total of 104 patients with breast cancer who received treatment in the Department of Surgery, The First Affiliated Hospital of Wenzhou Medical University from July 2021 to July 2022 were included in this study. These patients were randomly divided into a control group ( n = 52) and an observation group ( n = 52). All patients underwent radical mastectomy. The patients in the observation group received breast prosthesis implantation in addition to radical mastectomy. Operative time, length of hospital stay, psychological state scores before and 3 months after surgery, aesthetic recovery effect of breast 3 months after surgery, WHO Quality of Life Rating Scale (WHOQOL-100) score, postoperative occurrence of subcutaneous effusion, and flap necrosis were compared between the two groups. Results:There was no significant difference in operative time between the two groups ( P > 0.05). The length of stay in the observation group was (10.89 ± 2.34) days, which was significantly shorter than (21.06 ± 3.82) days in the control group ( t = 16.37, P < 0.001). At 3 months after surgery, the scores of the Self-rating Anxiety Scale (SAS), Hamilton Anxiety Scale (HAMA), Self-rating Depression Scale (SDS), and Hamilton Depression Scale (HAMD) in both groups were significantly lower than those before surgery. At 3 months after surgery, the scores of SAS, HAMA, SDS, and HAMD in the observation group were (39.02 ± 5.21) points, (19.02 ± 2.51) points, (16.03 ± 2.55) points, and (41.89 ± 3.56) points, which were significantly lower than (50.76 ± 4.85) points, (28.97 ± 2.66) points, (53.41 ± 4.56) points, and (24.88 ± 2.32) points in the control group ( t = 11.89, 19.62, 14.77, 18.51, all P < 0.001). At 3 months after surgery, the excellent and good rate of breast recovery in the observation group was [96.15% (50/52)], which was significantly higher than [69.23% (36/52)] in the control group ( Z = 5.40, P < 0.001). At 3 months after surgery, the scores of overall health, physical function, social function, cognitive function, and role function in each group were significantly higher than those before surgery, and the sole of emotional function in the observation group was significantly higher than that in the control group. At 3 months after surgery, the scores of overall health, physical function, social function, cognitive function, and role function in the observation group were (55.87 ± 12.70) points, (55.89 ± 12.34) points, (35.05 ± 11.25) points, (55.87 ± 7.88) points, and (58.77 ± 14.05) points, respectively, which were significantly higher than (43.75 ± 12.58) points, (47.50 ± 12.87) points, (28.04 ± 10.13) points, (39.87 ± 8.06) points, (32.05 ± 13.85) points, (74.52 ± 14.88) points in the control group ( t = 4.89, 3.39, 3.34, 10.24, 9.77, P < 0.001, 0.001, < 0.001, < 0.001, < 0.001, 0.003). The incidence of complications in the observation group was [3.85% (2/52)], which was significantly lower than [17.31% (9/52), χ2 = 4.98, P = 0.026] in the control group. Conclusion:Radical mastectomy combined with breast prosthesis implantation for the treatment of breast cancer patients can shorten the length of hospital stay, increase the excellent and good rate of breast recovery, reduce negative emotions of patients, improve quality of life, and reduce the occurrence of complications.
4.Establishment of PCR Identification Method for Pig Blood Type

ObjectiveXenotransplantation is an effective way to address the shortage of human organ donors, but it faces serious immune rejection reactions, including hyperacute rejection caused by blood type differences. Establishing a stable, convenient, and reliable method for pig blood type identification can quickly screen suitable donor pigs for xenograft research.MethodsBanna miniature inbred pigs, Diannan small eared pigs, and Bama Xiang pigs were selected as the research objects. DNA was extracted from the blood, oral buccal mucosa, and fetal fibroblasts of the three strains of pigs using DNA extraction kits. The target fragment of the ABO homologous gene EAA intron 7 in pigs was amplified using PCR method. Blood agglutination reaction was used to detect hemolysis in pig anterior vena cava whole blood after adding anti A and B antibodies. Immunohistochemical method was used to detect the expression level of A antigen in pig heart, liver, spleen, lung, and kidney tissues. Immunofluorescence method was used to detect the expression level of A antigen in pig oral mucosa. By comparing the results of different methods for determining pig blood types, the stability and reliability of the PCR method were verified, and a convenient PCR based pigblood type identification method was established.Results Firstly, the blood PCR results of 69 inbred strains of Banna miniature pigs, 7 Diannan small eared pigs, and 34 Bama Xiang pigs showed 20 AO blood types, 66 AA blood types, and 24 O blood types. The PCR results of fetal fibroblasts from 47 Diannan small eared pigs showed that all 47 fetuses were O blood type. Among them, the oral mucosal PCR results of 8 gene edited cloned pigs were consistent with those of donor fetal fibroblasts, all of which were O blood type. The oral mucosal PCR results of 8 wild-type pigs (2 inbred lines of Banna miniature pigs, 4 Diannan small eared pigs, and 2 Bama Xiang pigs) were consistent with the blood PCR identification results. Then, 11 inbred lines of Banna miniature pigs, 4 Diannan small eared pigs, and 2 Bama Xiang pigs were randomly selected for blood agglutination reaction validation, and the results were consistent with the PCR identification results of both blood samples and oral mucosa samples. Moreover, immuno-histochemical analysis was performed on the heart, liver, lung, kidney, and spleen tissues of one Banna miniature pig inbred line and two Bama Xiang pigs, and the results were consistent with blood PCR identification and blood agglutination reaction results. Finally, oral mucosal samples were collected from 2 inbred strains of Banna miniature pigs and 1 Bama Xiang pig for immunofluorescence detection, and the results were consistent with the blood PCR identification results.ConclusionBy collecting fetal cells and oral mucosal samples from live pigs for PCR detection, the blood type of pigs can be accurately and efficiently identified, providing a convenient method for blood type screening of xenograft donor pigs.
5.Rape Pollen Promote Androgenic Alopecia Mice by Increasing bFGF and VEGF Levels

OBJECTIVE To explore the improvement effect of rape pollen on androgenic alopecia mice and its mechanism. METHODS The blank group, model group, positive drug group and administration group were set up, the androgenic alopecia mice model was induced by applying 0.2% testosterone after hair removal. The hair growth rate of mice were observed by using 5% minoxidil as positive drug and 0.4 g·mL-1 rape pollen oil solution as administration group. The hair quality and follicle condition of mice were observed by scanning electron microscope(SEM) and HE staining of skin tissue, respectively. The level of VEGF and bFGF in skin were detected by immunofluorescence staining and Western blotting, while the level of serum sex hormones and reactive oxygen species were detected by ELISA. RESULTS Rape pollen could significantly promote the hair growth in mice and improve the state of mice hair scales compared with model group. Mechanism exploration experiments showed that rape pollen could not promote hair regeneration of mice by regulating hormone levels or anti-oxidative stress. However, rape pollen could increase the expression of bFGF and VEGF related to skin angiogenesis at the modeling site. CONCLUSION Rape pollen can promote hair regeneration in androgenic alopecia mice. Its mechanism may be that it promotes perifollicular vascular regeneration by increasing bFGF and VEGF level.
6.Research progress of liposome drug delivery system in stomatology

Liposomes are spherical vesicles with bilayer membrane structure spontaneously formed by phospholipids dispersed in an aqueous medium. Liposomes are excellent drug carrier with amphiphilic properties. Liposomes have good biocompatibility, biodegradability and no immunogenicity. Liposomes can achieve the delivery of the drug, enhance the solubility, improve the stability, reduce the toxic effect of the drug, and improve the therapeutic effect of the loaded drug. In recent years, liposome drug delivery systems have been widely used in dentistry. This article reviews the application of liposome drug delivery systems in caries, dental pulp diseases, periodontitis, implantation, oral anesthesia and oral candidiasis.

The widespread application of next generation sequencing (NGS) in clinical settings has enabled testing, diagnosis, treatment and prevention of genetic diseases. However, many issues have arisen in the meanwhile. One of the most pressing issues is the lack of standards for reporting genetic test results across different service providers. The First Forum on Standards and Specifications for Clinical Genetic Testing was held to address the issue in Shenzhen, China, on October 28, 2017. Participants, including geneticists, clinicians, and representatives of genetic testing service providers, discussed problems of clinical genetic testing services across in China and shared opinions on principles, challenges, and standards for reporting clinical genetic test results. Here we summarize expert opinions presented at the seminar and report the consensus, which will serve as a basis for the development of standards and guidelines for reporting of clinical genetic testing results, in order to promote the standardization and regulation of genetic testing services in China.
8.Research in effect of entire process nursing intervention during nebulization treatment of acute wheezing episodes in school-age children

Objective To explore the effect of entire process nursing intervention during nebulization treatment of acute wheezing episodes in school-age children.Methods One hundred and forty-one children with acute wheezing episodes were divided into the experimental group (75 patients) and the control group (66 patients) by whether willing to accept nursing intervention or not,respectively.Routine nursing such as cognition of wheezing knowledge and respiration instruction was given to both groups,while specialists accompanied the whole procedure of inhalation treatment and they gave nursing intervention such as respiration,saliva swallow,expiration through nose,grasp of inhalation tank and body position maintenance to the experimental group.The improvement of wheezing and subjective sense by the patients,transcutaneous oxygen saturation (SaO_2),and the compliance of patients were measured and evaluated.Results Improvement of wheezing and comfort feeling were found in 97.3% of the experimental group,while 80.3% of the control group.And SaO_2 value (higher than 95%) was 92.0% in the experimental group,75.8% in the control group.The compliance of the experimental group was also better than the control group.Conclusions Entire process nursing intervention during nebulization treatment can improve clinical effect and the compliance of wheezing children.
